Effect of Relative Humidity on Physiology and Antioxidant Metabolism of Grafted Watermelon Seedlings (상대습도가 수박 접목묘의 생리 및 항산화 대사에 미치는 영향)

  • This study investigated the effect of different relative humidity (RH) regimes on graft healing of grafted seedlings of watermelon (Citrullus vulgaris Schrad.). Two watermelon cultivars ('Speed' and 'Sambok Honey') were grafted onto the 'RS-Dongjanggun' bottle gourd rootstock (Lagenaria siceraria Stanld.) and the grafted seedlings were maintained under one of three relative humidity regimes, 95-96% [1.1-0.8 (day) or $0.8-0.6(night)\;g{\cdot}m^{-3}$ vapor pressure deficit (VPD)], 97-98% [ 0.7-0.4 (day) or $0.5-0.3(night)\;g{\cdot}m^{-3}$ (VPD)], or 99-100% [0.3-0.0 (day) or $0.2-0.0(night)\;g{\cdot}m^{-3}$ (VPD)] according to the Mollier diagram based on the air temperature of $25^{\circ}C\;day/18^{\circ}C\;night$ with 16 h photoperiod per day. Among the RH treatments, 97-98% significantly increased plant height and fresh weight of the rootstock and scion of the 'Speed' and it also enhanced the graft union connection of both cultivars after two days of grafting. However, plant height and thickness of the scion of 'Sambok Honey' was increased by the 99-100% RH treatment. Furthermore, both cultivars grown in the 95-96 and 97-98% RH treatments consisted of lower levels of endogenous $H_2O_2$ and less activities of antioxidant enzymes which illustrated the occurrence of less oxidative stress. Hence, the results of this study identified the optimal RH level for the graft healing of watermelon seedlings.

Applicability of Vegetation Index and SPAD Reading to Nondestructive Diagnosis of Rice Growth and Nitrogen Nutrition Status (식생지수와 SPAD를 이용한 벼 생육 및 질소영양상태의 비파괴적 진단 가능성 검토)

  • Precise application of topdressing nitrogen (N) fertilizer is indispensible for securing high yield and good quality of rice and minimizing N losses to the environment as well. For precise N management, growth and nitrogen nutrition status (NNS) should be diagnosed rapidly and accurately. The objective of the study was to evaluate the applicability of vegetation index (VI) calculated from hyperspectral canopy reflectance measurement and SPAD reading to nondestructive in situ diagnosis of growth and NNS of rice. Canopy reflectance, SPAD read­ing, growth parameters, and NNS characteristics were measured from various N treatments to evaluate the relationships among them for two cropping seasons from 2001 to 2002. The correlation coefficient of VIs with variables of growth and NNS increased positively as rice canopy became more closed. Regardless of growth stages, VIs had significantly high correlations with LAI, shoot dry weight (DW), shoot N content and nitrogen nutrition index (NNI). Those correlation coefficients increased steadily before heading stage as rice grew up. However, tiller number and leaf N concentration showed significantly high correlations with VIs only at and after panicle initiation stage (PIS). Among the VIs, RVIgreen had significantly higher correlation with the measured parameters than the other VIs: it showed correlation coefficients greater than 0.8 with leaf and shoot N concentration and DW, and much higher coefficients greater than 0.9 with LAI, shoot N content, and NNI. At LAI of below 2.5, VIs had non-significant or low correlations with the growth and NNS indicators due to the background effects. SPAD reading had significantly high correlation with leaf N concentration and NNI at each growth stage. In addition, it had significant correlations with variables of growth and NNS at PIS and booting stage, particularly, at booting stage. Though SPAD reading had a significantly high correlation value at a given growth stage in each year, it showed very weak relationship with variables of growth and NNS when pooled across growth stages and years. In conclusion, RVIgreen was found to be the most reliable VI to estimate the growth and NNS of rice around at PIS, but SPAD reading had much limitations.

  • In order to evaluate the utility of the anthocyanins of Amaranthys tricolor L. as an edible pigment, the present study was undertaken to investigate the effects of pH. temperature, ascorbic acid, sugars and their degradation products, quercetin, thiourea, sodium pyrophosphate and metal ions on the stability of the anthocyanins in the model systems. The results obtained from this study were as follows. 1. The degradation of total anthocyanins was retarded as the pH levels decreased from 8.0 to 1.0. At pH 1.0, however. the initial degradation reaction proceeded faster than at pH 2.0 to 3.0 2. On heating in buffered aqueous solution at $80^{\circ}C$, the total anthocyanin content was higher at pH 2.0 than other pH levels. Increasing the storage temperature accelerated greatly the pigment degradation. In darkness at $40^{\circ}C$, after 10 days, only 19% of the original amount was left, while at $2^{\circ}C$, under the same conditions of storage, approximately 90% of the pigment was retained. The half-life of the pigment, 63.0 days at $2^{\circ}C$, shortened to 1. 7 days at $40^{\circ}C$. 3. An increase in ascorbic arid concentration from 0. 15 to 0.50 mg/ml lowered the anthocyanin retention. 4. There was no significant difference between glucose and fructose in anthocyanin degradation effect. Furfural was more effective than other sugar degradation products, formic acid or levulinic acid in accelerating anthocyanin breakdown. 5. Neither quercetin nor sodium pyrophosphate had a protective effect on the anthocyanins in the presence of ascorbic acid, while, in the systems 0.5 or 1 mg/ml of thiourea with $150{\;}{\mu}g/ml$ of ascorbic acid, the loss of anthocyanins was significantly reduced. 6. Both mercuric and cupric ions in 30 ppm greatly accelerated anthocyanin degradation.

Studies on Ripening Physiology of Rice plant. -I Difference in Ripening Structure between Jinheung and IR667 (수도(水稻)의 등숙생리(登熟生理)에 관(關)한 연구(硏究) -I 진흥(振興)과 IR667의 등숙구조비교(登熟構造比較))

  • A local rice variety, Jinheung and newly bred IR667-Suwon 214 were grown in $5m^2$ concret pot with two spacings and two nitrogen levels and their ripening structure and its function were comparatively investigated to elucidate the causes of unusually low ripened grain ratio of IR667 lines. The following differences between two varieties were found. 1. Though IR667 had much lower ripened grain ratio (64%) than Jinheung (85%) grain yield(790 kg/10a) of IR667 was higher than that (760 kg/10a) of Jinheung. 2. Number of ripined grain per net assimiration rate (NAR) at 10 days after heading was a little higher in IR667 (6,490) than in Jinheung (6,360) consiting to lower grain weight ($29.9{\times}10^{-3}g$) in IR667 than $31.2{\times}10^{-3}g$ of Jinheung. But number of total grain per NAR was much higher (10,530) in IR667 than 7,290 of Jinheung indicating that it was the probable cause of low ripened grain ratio of IR667. 3. Extinction coeificient (K) was 0.115 in IR667 and 0.200 in Jinheung, thus IR667 could construct greater ripening structure per unit area. 4. Number of grain per LAI was decreased with increasing LAI at heading and the decreasing rate was similar for both IR667 and Jinheung. 5. Critical leaf area index at which crop growth rata (CGR) is maximum was 6.5 for IR667 and 5.2 for Jinheung. Below 5.2 of LAI net assimilation rate was always higher an Jinheung throughout the growing season. 6. The estimated optimum leaf area index having maximum grain yield was 7.4 for IR667 and 6.2 for Jinheung at 10 days after heading. However, actual leaf area index was 6.2 for IR-667 and 4.7 for Jinheung and these were even below critical leaf area index. 7. The decrease of LAI during ripening period was great in IR667 but photosynthesis per $m^2$ was decreased more rapidly in Jinheung. 8. Net assimilation rate (NAR) decreased with the increase of LAI at any time of ripening period. The decreasing rate of NAR with the increase of LAI was greater in IR667 with ripening. The greater decreasing rate of NAR in IR667 seemed to be attributed to low photosynthetic activity and high respiratory loss due to the requirement of higher optimum temperature of ripening. 9. Grain yield-ripened grain ratio curve showed less contribution of dry matter yield after heading to grain yield in IR667 than in Jinheung due to unfavorable ripening environment(specialy air temperature) indicating that yield of IR667 could most effectively increased through the improvement of ripening environment.

Laboratory-scale fruiting body formation of Pleurotus ostreatus using the petri dish culture (느타리의 기내 자실체 형성 및 그 유도조건에 관한 연구)

  • Pleurotus ostreatus, the oyster mushroom, is one of the most widely cultivated and important edible mushrooms in the world. In order to study the developmental process of P. ostreatus and its regulatory mechanism, a new culturing method needs to be established for inducing the fruiting body and sporulation in the laboratory. In this study, we have examined whether the fruiting body of P. ostreatus can be formed on the plastic petri dish which are commonly used for cell culture in the laboratory. The strain was cultured on $60{\times}15mm$ plastic petri dish with potato dextrose agar media at $28^{\circ}C$ for mycelial growth and then at $18^{\circ}C$ for the formation of primordia and fruiting bodies within plant growth chamber. The development of primordia into fruiting bodies was achieved on cultured dishes under air ventilation. At the primordia stage, the normal formation of fruiting body was blocked by sealing the plastic dish with parafilm. The periods requiring for the formation of primordia and fruiting bodies were examined on the dish culture. About 96% and 76% of cultured samples formed primordia and fruiting bodies under the optimal conditions during ten weeks of culture, respectively. These culturing periods, however, were changed by the mechanical injury treatment to mycelia. As other factors affecting the fruiting body formation, the effects of light and cold shock have been tested. No fruiting formation was observed on the cultured dishes under the dark. The cold shock treatment by storing cultured dishes for one day at $4^{\circ}C$ did not have any significant effects in the fruiting body formation. Spores of fruiting bodies acquired from the petri dishes could be germinated on culture media at $28^{\circ}C$. These results suggest that the fruiting bodies of P. ostreatus can be formed on the experimental petri dish and this dish-culturing method is useful for understanding of the developmental process of P. ostreatus in the laboratory. Furthermore, the dish-culturing method is able to shorten the life cycle of P. ostreatus without requiring large area and expensive device.

Strategy for Bio-Diversity and Genetic Conservation of Forest Resources in Korea (생물종(生物種) 다양성(多樣性) 및 삼림유전자원(森林遺傳資源) 보존(保存) 전략(戰略))

  • Due to its topographic complexities and various climatical condition, Korea exhibits diverse forest types. Dominant tree species in this zone are Quercus spp., Betula spp., Zelkova spp., Fraxinus spp., Pinus densiflora, Pinus koraiensis, and Pinus thunbergii ete. Genetic conservation in forest species in Korea there are three ways ; one is in situ, other is ex situ and third is in-facility conservation. In situ conservation include that are the present status of conservation of rare and endangered flora and ecosystem, the reserved forest, the national and provincial park, and the gene pool of natural forests. Ex situ conservation means to be established the new forest from in situ forest stands, progeny and provenance test populations, seed orchard and clone banks, and gene conservation in-facility. As a tool for low temperature storage, several aspects on in vitro system were studied ; (1) establishment of in vitro cultures from juvenile and/or rejuvenated tissues, (2) induction of multiple shoots from the individual micropropagules, (3) elongation of the proliferated shoots. Studies on cold storage for short-and long-term maintenance of in vitro cultures under $4^{\circ}C$ in the refrigerator were conducted. For the cryopreservation at $-196^{\circ}C$, various factors affecting survivability of the plant materials are being examined. The necessity of gene conservation of forest trees is enlarged not only to increase the adaptability for various environments but also to gain the breeding materials in the future. For effective gene conservation of forest trees, I would like to suggest followings ; 1. Forest stands reserved for other than the gene conservation purposes such as national parks should be investigated by botanical and gene-ecological studies for selecting bio-diversity and gene conservation stands. 2. Reserved forest for gene pool should be extented both economically important tree spp. and non-economical species. 3. Reserved forest for progeny test and clone bank should be systematically investigated for the use of Ex situ forest gene conservation. 4. We have to find out a new methodology of genetic analysis determining the proper and effective size of subpopulation for in situ gene conservation. 5. We should develop a new tree breeding systems for successful gene conservation and utilization of the genetic resources. 6. New method of in-facility gene conservation using advanced genetic engineering should be developed to save time and economic resources. 7. For the conservation of species with short-life span of seed or shortage of knowledge of seed physiology, tissue culture techniques will be played a great role for gene conservation of those species. 8. It is are very useful conservation not only of genes but of genotypes which were selected already by breeding program. 9. Institutional and administrative arrangements including legistlation must be necessarily taken for gene conservation of forest trees. 10. It is national problems for conservation of forest resources which have been rapidly destroyed because of degenerating environmental condition and of inexperienced management system of bio-diversity and gene conservation. 11. In order to international cooperation for exchanging data of bio-diversity and gene conservation, we should connect to international net works as soon as possible.

Physiological Interactions Between the Herbicide Pretilachlor and the Safener Fenclorim on Rice (제초제(除草劑) Pretilachlor와 해독제(害毒劑) Fenclorim의 수도(水稻)에 대한 생리적(生理的) 상호작용(相互作用))

  • The incividual and combined effects of the chloroacetanilide herbicide pretilachlor and of the safener fenclorim on the growth and selected physiological processes of rice (Oryza sativa L., var 'Lemont')were evaluated under greenhouse and laboratory conditions. Fenclorim applied at rates ranging from 50 to 300 g a.i./ha antagonized the injurious effects caused by 150 to 900 g a.i./ha of pretilachlor on 15-day old wet-sown rice grown under greenhouse conditions. When used rates of 150 g/ha or higher, fenclorim reversed completely the effects of all doses of pretilachlor on rice. When the two compounds were given simultaneously, fenclorim enhanced the uptake of $^{14}C$pretilachlor into rice leaf mesophyll protoplasts measured for 1 hr, indicating that competition for uptake at the protoplast level is not involved in the protective action of this safener. The safener-induced stimulation of pretilachlor uptake was particularly evident when fenclorim was used at concentrations of 10, 20 and $40{\mu}M$. Following 4 hr of incubation, individual treatments with pretilachlor inhibited the in vitro incorporation of radiolabeled precursors into proteins, DNA, and lipids of rice leaf protoplasts only when used at the high concentration of $100{\mu}M$M. Individual treatments with high concentrations (10 or $100{\mu}M$) of the safener fenclorim inhibited the incorporation of radiolabeled precursors into proteins and lipids of rice protoplasts, but had no DNA synthesis. The combined effects of pretilachlor and fenclorim on the incorporation of radiolabeled precursors into these macromolecules of isolated rice mesophyll protoplasts appeared to be additive or slightly synergistic rather than antagonistic. Fenclorim at $1{\mu}M$ antagonized the effects of pretilachlor on total lipids of rice leaf protoplasts. In addition, individual and combined treat-menu with pretilachlor and fenclorim influenced the incoroporation of$^{14}C$acetate into polar lipids, triglycerides and steryl esters of rice leaf protoplas causing a redistribution of carbon in these lipid fractions. However, these effects were not large enough to explain the herbicidal activity of pretilachlor or to account for the protective action of the safener fenclorim. Overall, the uesults of the present study idnicate that the safener fenclorim does not seem to protect rice against pretilachlor injury by antagonizing its effects on protein, DNA, or lipid syntheses.

A Survey of Disease Occurrence in Korean Black Goats (흑염소 사육농가의 질병발생 실태조사)

  • The goat industry has been developing for many years in Korea to meet demands for food and medicine. However, many complicated patterns of disease have arisen in goat farms as this industry has developed. In this study, disease occurrence patterns in Korean black goats were surveyed in six professional farming households in Imsil and Soonchang in the Jeonbuk province and in Hamyang and Sachon in the Gyeongnam province to understand and extend the goat disease database. We observed morbidity rates between 2.0% and 9.8% for adult goats and between 2.9% and 68.3% for kids. Kids showed a markedly higher incidence of disease when compared to adults. The rate of disease occurrence was 40.0% for floppy kid syndrome (FKS), 37.7% for diarrhea, 16.0% for respiratory disease, and 1.9% for skin disease. The observed mortality rates were 0.7% ~ 10.0%, and 2.2% ~ 24.9% for adult goats and kids, respectively. In addition, FKS, diarrhea, and respiratory disease were observed in 38.3%, 28.9%, and 10.0%, respectively, of dead goats. In conclusion, the majority of diseases in goats occur during the neonatal period, and FKS is the highest single cause of mortality in Korean black goats. Thus careful attention must be paid to kids to reduce the goat mortality rate.

New Technologies for the Removal of Bacteriophages Contaminating Whey and Whey Products as Cheese by-Products: A Review (치즈 부산물인 유청과 유청 제품에 감염된 박테리오파지 제거를 위해 새롭게 개발된 기술: 총설)

  • In general, whey obtained from various cheese batches is being reused, so as to improve the texture and to increase the yield and the nutrient value of the various final milk-based products. In fact, re-usage of whey proteins, including whey cream, is a common and routine procedure. Unfortunately, most bacteriophages can survive heat treatments such as pasteurization. Hence, there is a high risk of an increase in the bacteriophage population during the cheese-making process. Whey samples contaminated with bacteriophages can cause serious problems in the cheese industry. In particular, the process of whey separation frequently leads to aerosol-borne bacteriophages and thus to a contaminated environment in the dairy production plant. In addition, whey proteins and whey cream reused in a cheese matrix can be infected by bacteriophages with thermal resistance. Therefore, to completely abolish the various risks of fermentation failure during re-usage of whey, a whey treatment that effectively decreases the bacteriophage population is urgently needed and indispensable. Hence, the purpose of this review is to introduce various newly developed methods and state-of-the-art technologies for removing bacteriophages from contaminated whey and whey products.

A New Sweetpotato Variety for Table Use, 'Pungwonmi' (식용 고구마 신품종 '풍원미')

  • 'Pungwonmi', a new sweetpotato variety, was developed for table use by Bioenergy Crop Research Institute, National Institute of Crop Science (NICS), RDA in 2014. This variety was derived from the cross between 'Benisatsuma' and 'Luby3074' in 2006. The seedling and line selections were performed from 2007 to 2009, and preliminary and advanced yield trials were carried out from 2010 to 2011. The regional yield trials were conducted at five locations from 2012 to 2014, and it was named as 'Pungwonmi'. This variety has cordate leaf shape, and its leaves, stems, nodes, and petioles are green. Storage root of 'Pungwonmi' has an elliptical shape, red skin, and light orange flesh. 'Pungwonmi' was moderately resistant to fusarium wilt, and resistant to root-knot nematode. Dry matter content was 31.2%, and texture of steamed storage root was intermediate. Total sugar content of raw and steamed storage roots of 'Pungwonmi' was higher than that of 'Yulmi'. ${\beta}$-carotene content of 'Pungwonmi' was 9.1 mg/100g DW. Yield of marketable storage root over 50 g of 'Pungwonmi' was 24.3 MT/ha under the early season culture, which was 46% higher than that of 'Yulmi'. The number of marketable storage roots per plant was 2.8 and the average weight of marketable storage root was 156 g under the optimal and late season culture. Marketable storage root yield of 'Pungwonmi' was 24.1 MT/ha under the optimum and late season culture, which was 26% higher than that of 'Yulmi'. (Registration No. 6428).
