Physicochemical Characteristics of Different Parts of Burdock (Arctium sp.)

  • Burdock (Arctium sp.) is known as a nutraceutical vegetable, especially in Japanese and Korean cuisine. While burdock plants are generally harvested for their tap roots, different parts of the plant are consumed as food or used as traditional medicines. This study investigated the physicochemical properties of the leaves, stems, roots, and peeled roots of the burdock plant based on their pH, soluble solid content, titratable acidity, color values, and mineral content. The pH differed significantly among the different plant parts, with the highest value in the leaves and the lowest in the stems. However, for the soluble solid content, the leaves had the lowest, while the peeled roots had the highest. The titratable acidity of the stems was significantly lower than that of the leaves, roots, and peeled roots. As regards the color values, the lightness value was highest for the stems, while the roots showed the highest redness value, followed by the peeled roots, and the leaves had the highest yellowness value. The leaves and stems contained almost three times more potassium than the roots and peeled roots. Thus, the higher content of different minerals in the leaves and stems of the burdock plant shows that these plant parts could be used as potential sources of dietary minerals.

Antimicrobial Activity of Various Parts of Tomato Plants Varied with Different Solvent Extracts

  • The antimicrobial activity of acetone, hexane, dichloromethane, and methanol extracts from leaves, stems, immature green fruits, and red fruits of tomato plants was examined against six phytopathogens. The minimum inhibitory concentration (MIC) of the acetonic extracts from these four plant parts was lower than that of the other solvents. Among the acetonic extracts, tomato leaves had a lower MIC than the other tomato parts. The acetonic extract from tomato leaves was therefore selected as a source of antimicrobial substances. The acetonic extract from tomato leaves inhibited mycelial growth of Fusarium oxysporum f. sp. lycopersici, Glomerella cingulata, and Rhizoctonia solani. Mycelial growth of R. solani treated with acetone extract from leaves showed more susceptibility than the other phytopathogens. Using 0.31 mg/ml of the acetonic extract from leaves, mycelial growth of R. solani on days 1, 2, and 3 decreased by 50.0, 52.1, and 64.0%, respectively, compared with acetone solvent treatment. The antimicrobial compounds effective against R. solani were identified as linolenic acid and caffeic acid by bioautography and GC-MS. These two compounds were used to treat six phytopathogens to confirm their antimicrobial activities. Linolenic acid inhibited mycelial growth of R. solani, while caffeic acid showed only slight antimicrobial activity. Results indicated that we propose extracts from tomato leaves which included antimicrobial compounds may provide a new lead in the pursuit of new biological sources of agrochemical candidates.

Light- and Relative Humidity-Regulated Hypersensitive Cell Death and Plant Immunity in Chinese Cabbage Leaves by a Non-adapted Bacteria Xanthomonas campestris pv. vesicatoria

  • Inoculation of Chinese cabbage leaves with high titer (107 cfu/ml) of the non-adapted bacteria Xanthomonas campestris pv. vesicatoria (Xcv) strain Bv5-4a.1 triggered rapid leaf tissue collapses and hypersensitive cell death (HCD) at 24 h. Electrolyte leakage and lipid peroxidation markedly increased in the Xcv-inoculated leaves. Defence-related gene expressions (BrPR1, BrPR4, BrChi1, BrGST1 and BrAPX1) were preferentially activated in the Xcv-inoculated leaves. The Xcv-triggered HCD was attenuated by continuous light but accelerated by a dark environment, and the prolonged high relative humidity also alleviated the HCD. Constant dark and increased relative humidity provided favorable conditions for the Xcv bacterial growth in the leaves. Pretreated fluridone (biosynthetic inhibitor of endogenous abscisic acid [ABA]) increased the HCD in the Xcv-inoculated leaves, but exogenous ABA attenuated the HCD. The pretreated ABA also reduced the Xcv bacterial growth in the leaves. These results highlight that the onset of HCD in Chinese cabbage leaves initiated by non-adapted pathogen Xcv Bv5-4a.1 and in planta bacterial growth was differently modulated by internal and external conditional changes.

TMV 감염에 의한 고추의 역병 저항성 유도 (Induction of Resistance by TMV Infection in Capsicum annuum Against Phytophthora Blight)

  • Induction of systemic acquired resistance (SAR) against phytophthora blight and pathogenesis-related (PR) protein accumulation by TMV infection in pepper plant (Capsicum annuum cv. Nockwang) were examined to understand the mechanism of the systemic acquired resistance in pepper plant. The zoospore suspension of Phytophthora capsici was inoculated on stem of pepper plant in which TMV-pepper strain had been inoculated on fully expanded upper leaves, and thephytopha blight incidence was examined. Both disease severity and lesion length of phytophthora blight were much smaller in TMV pre-inoculated pepper plant than in uninoculated control plants. The phytophthora blight incidence was decreased about 50% in the TMV pre-inoculated pepper, compared to the uninoculated control plant at 10 days after P. capsici inoculation. Accumulation of PR1 and PR5 proteins in intercellular fluid of TMV-inoculated and uninoculated upper leaves were monitored by immuno-blot with tobacco P1b and PR5a, antibody during induction of SAR. PR1 and PR5 were detected from 24 hours after TMV inoculation in both TMV-inoculated and uninouclated upper leaves, and increased rapidly in TMV-inoculation in uninoculated upper leaves were defoliated. PR5 could be detected upto 20 days after TMV inoculation in uninoculated upper leaves. These results suggest that TMV infection induces SAR against phytophthora blight in pepper plant, and that PR proteins are accumulated very rapidly during induction of SAR and maintained for quite long time in pepper plant.

Observations of Infection Structures on the Leaves of Cucumber Plants Pre-treated with Arbuscular Mycorrhiza Glomus intraradices after Challenge Inoculation with Colletotrichum orbiculare

  • Resistance inductions on the leaves of cucumber plant by an arbuscular mycorrhiza Glomus intraradices were investigated. In addition, the infection structures were observed at the penetration sites on the leaves of plant inoculated with Colletotrichum orbiculare using a fluorescence microscope. The severity of anthracnose disease caused by Colletotrichum orbiculare was significantly decreased on the leaves of cucumber plant colonized with G intraradices compared with those of non-treated control plants. As a positive control, pre-treatment with DL-3-aminobutyric acid (BABA) caused a remarkable reduction of the disease severity on the pathogen-inoculated leaves. There were no significant differences in the frequency of either germination or appressorium formation of the plant pathogen between mycorrhiza colonized and non-treated plants. It was also the same on the BABA pre-treated plants. However, the frequency of callose formation was significantly high on the leaves of G intraradices colonized plants compared to those of non-treated control plants at 5 days after challenge inoculation. On the leaves of BABA treated plants callose formation was not significantly high than those of non-treated, although the disease severity was more strongly suppressed. It was suggested that the resistance induced by colonization with G. intraradices might be related to the enhancement of callose formation at the penetrate sites on the leaves invaded by the pathogen, whereas resistance by BABA did not.

Photosynthetic characteristics and chlorophyll of Vitex rotundifolia in coastal sand dune

  • Background: This study analyzed the physiological adaptations of a woody plant, Vitex rotundifolia, in Goraebul coastal sand dunes from May to September 2022. Environmental factors and physiological of plants growing under field and controlled (pot) conditions were compared. Results: Photosynthesis in plants growing in the coastal sand dunes and pots was the highest in June 2022 and July 2022, respectively. Chlorophyll fluorescence indicated the presence of stress in the coastal sand dune environment. The net photosynthesis rate (PN) and Y(II) were highest in June in the coastal sand dune environment and July in the pot environment. In August and September, Y(NPQ) increased in the plants in the coastal sand dune environment, showing their photoprotective mechanism. Chlorophyll a and b contents in the pot plant leaves were higher than those in the coastal sand dune plant leaves; however, chlorophyll-a/b ratio was higher in the coastal sand dune plant leaves than in the pot plant leaves, suggesting a relatively high photosynthetic efficiency. Carotenoid content in the coastal sand dune plant leaves was higher in August and September 2022 than that in the pot plant leaves. Leaf water and soluble carbohydrate contents of the coastal sand dune plant leaves decreased in September 2022, leading to rapid leaf abscission. Diurnal variations in photosynthesis and chlorophyll fluorescence in both environments showed peak activity at 12:00 hour; however, the coastal sand dune plants had lower growth rates and Y(II) than the pot plants. Plants in the coastal sand dunes had higher leaf water and ion contents, indicating that they adapted to water stress through osmotic adjustments. However, plants growing in the coastal sand dunes exhibited reduced photosynthetic activity and accelerated decline due to seasonal temperature decreases. These findings demonstrate the adaptation mechanisms of V. rotundifolia to water stress, poor soils, and high temperature conditions in coastal sand dunes. Conclusions: The observed variations indicate the responses of the V. rotundifolia to environmental stress, and may reveal its survival strategies and adaptation mechanisms to stress. The results provide insights into the ecophysiological characteristics of V. rotundifolia and a basis for the conservation and restoration of damaged coastal sand dunes.

Zonate Leaf Spot of Sorghum Caused by Gloeocercospora sorghi in Korea

  • Gloeocercospora sorghi caused the zonate leaf spot on sorghum (Sorghum bicolor Moench) in fields in Korea. The zonate spots were conspicuous on sorghum leaves as circular, reddish purple bands alternating with tan or straw-colored areas, which formed a coarsely zonate pattern. The lesions often occurred in semicircular patterns along the margin of leaves. The fungus produced sporodochia on the surface of infected leaves in a moist chamber. The conidia, born in a pinkish to salmon-colored slimy matrix, were hyaline, elongate to filiform, straight or slightly curved, 3- to 17-septate, and of variable lenght (28-197 x 1.6-$3.4\mu\textrm{m}$). G. sorghi isolated from diseased leaves of sorghum caused characteristic reddish-brown water-soaked leaf spots when inoculated to seedlings.

  • This study was conducted to investigate the effects of plant density and urea supplement on the yield, quality, value and total nitrogen, alkaloid contents of burley tobacco leaves. The results were as fellows; As urea supplement were increased from 0kg/10a to 50kg/10a, yield, total alkaloid, total nitrogen of cured leaf were significantly increased. The lower plant density levels also increased dry weight and leaf area per plant. Price of cured leaves were not influenced by plant density and amounts of urea supplemented. The effects of plant density and urea supplement on the number of leaves were not detected.

NMMP1, a Matrix Metalloprotease in Nicotiana benthamiana Has a Role in Protection against Bacterial Infection

  • Plant matrix metalloproteases (MMPs) are a family of apoplastic metalloproteases closely related to human matrilysins. Up-regulation of Nicotiana benthamiana matrix metalloprotease 1 (NMMP1) expression by treatment with pathogens, ethephon and aging indicates that the gene is related to plant defense and the aging process through ethylene signaling. NMMP1 expression was higher than in normal growth leaves following infection with an incompatible pathogen Pseudomonas syringae pv. tomato T1 or a compatible pathogen P. syringae pv. tabaci and in aged leaves. Transient overexpression of NMMP1 in N. benthamiana leaves lowered the growth of P. syringae pv. tabaci. However, NMMP1-silenced leaves showed increased growth of P. syringae pv. tabaci. These data strongly suggest that NMMP1 in N. benthamiana is a defense related gene, which is positively regulated by ethylene.

수분 부족 스트레스 처리시 Monodehydroascorbate Reductase (MDHAR)의 반응 (Response of Monodehydroascorbate Reductase (MDHAR) in Lettuce (Lactuca sativa L.) Leaves Subjected to Water Deficit Stress)

  • 수분 공급을 제한하여 수분 부족 스트레스를 처리한 상추식물에서 산화적 스트레스와 관련된 monodehydroascorbate reductase (MDHAR)의 활성도, 엽록소 함량, 과산화수소의 함량 등과의 상관관계를 조사한 결과, 생육배지의 수분의 함량이 감소함에 따라 식물체내 과산화수소의 생성량이 증가($R^2$=0.8851)하였으며, 수용성단백질 함량은 점차 감소($R^2$=0.9826)하는 경향을 나타내었다. 총 엽록소함량은 수분 부족 스트레스를 처리한 공시작물에서의 함량이 정상 생육시 보다 그 함량이 대체적으로 낮은 경향을 보였으며, 엽록소 a와 엽록소 b함량 변화도 총 엽록소의 함량변화와 비슷한 경향을 보였다. 그러나 총 엽록소에 대한 카로티노이드의 비율은 수분 부족 스트레스를 처리한 식물에서 정상생육 시 보다 더 높은 경향을 보였다. 수분 부족 스트레스가 진행됨에 따라 아스코브산의 함량은 정상 생육 시 보다 더 높은 경향을 보였으나 환원형인 디하이드로아스코브산의 함량은 수분 부족 스트레스를 처리한 초기에 정상생육 시 보다 더 낮은 경향을 보였다. MDHAR의 활성도는 사이토졸(cytosolic) 분획과 엽록체(chloroplastic) 분획에서 공히 크게 증가하였으며 MDHAR의 mRNA 전사 정도도 수분 부족 스트레스가 진행됨에 따라 크게 증가하였다. 수분함량이 감소함에 따라 MDHAR의 활성도가 크게 증가하였으며, 또한 과산화수소의 함량이 증가함에 따라서도 MDHAR의 활성도가 크게 증가($R^2$=0.9937과 0.8645)되어 수분 부족 스트레스로 나타나는 요인들과 MDHAR 사이에 밀접한 관련이 있음을 확인할 수 있었다.
