Callus Induction and Plant Regeneration from Mature Embryos in Oat

  • Mature embryos of five oat genotypes were cultured to develop an efficient method of callus induction and plant regeneration. Murashige and Skoog(MS) and N6 media supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin were used for callus induction. Percentage of callus induction showed significant among the combinations of plant growth regulators. Callus induction showed high efficiency in medium containing 3 mg/$\ell$ of 2,4-D. The high frequency of callus induction was obtained in Gwiri37. For plant regeneration, calli induced from mature embryos were transferred onto MS and N6 media supplemented with combinations of 6-benzyladenine (BA) and naphthaleneacetic acid (NAA) for 5 weeks. Percentage of plant regeneration showed high in MS medium containing 0.2 mg/$\ell$ of NAA and 1 mg/$\ell$ of BA. The callus initiation medium affected the subsequent plant regeneration. Treatment with 3 mg/$\ell$ of 2,4-D, and 3 mg/$\ell$ of 2,4-D and 3 mg/$\ell$ of kinetin in callus induction media showed high frequency for plant regeneration. Plant regeneration frequency among the genotypes showed significant. Especially, Gwiri37 showed high regeneration frequency. Regenerated shoots were treated with 200, 350 and 500 mg/$\ell$ of indole-3-butyric acid (IBA) transferred onto half-strength MS medium without plant growth regulators. Treatment of shoots with IBA induced root formation rapidly.

Interaction Between time of Nodal Explant Collection and Growth Regulators Determines the Efficiency of Morus alba Micropropagation

  • The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.

Effect of Medium Composition and Growth Regulators on Mass Propagation of Virus-Free Plant from the Meristem Cultures of 'Jarang' Grape ('자랑' 포도 생장점 배양으로부터 무병주 식물 대량번식에 미치는 배지 구성물질과 생장 조절제의 영향)

  • This study was performed to clarify the effect of medium compositions and plant growth regulators on the shoot, root formation and growth of 'Jarang' grape for mass propagation of virus-free plant. The formation and growth of shoot were considerably favorable in half-concentration of MS medium. However, the formation of adventitious root per explants (avg. 2.1) was effective in higher concentration (two times) of MS medium. For sucrose concentration, 1% for the shoot formation, 3% for the adventitious root formation and 1% for the growth were observed as yield significant results. With the addition of 0.05% of activated carbon, the shoot growth was improved, and it was effective for the adventitious root formation and growth as well. A pH of 6.8 in the medium was the most suitable for mass propagation; the results showed significant enhancement in the number of nodes and the length of the shoot, 3.9 and 1.3 cm, respectively. The shoot growth was the most vigorous in BA 1.0 mg/L due to the impact of the growth regulator on the mass propagation in it. Consequently, 16.9 shoots per explant were formed in NAA 1.0 mg/L so good results were obtained.

  • Ethylene was released from leaf and fruit but root of Panax ginseng. Root callus showed higher ethylene release (ER) than fruit ER increased with leaf senesence. Fruit during ripening showed decreasing ER in the order of green stage, early stage of reddening and fully ripened stage. between leaves from the plant with fruits in different stages of ripening showed similar trend of fruit in ER but it was about 10 times higher in leaves than in fruits. Leaves of P. quinquefolius showed about 200 times higher ER than that of P ginseng on 22 July Fruits from the plant treated with ethephon showed higher ER after 109 days. Forty-five day-old seedlings grown with various growth regulators showed a significant decrease of stem length and significant increase of ER only in Uniconazole (0.1 ppm) and H-9 (0.0, 5 ppm) solution.

In vitro Propagation of Junos Orange (Citrus junos Sieb.) through Nucellar Polyembroid Cultures

  • An in vtro nucellar polyembryo propagation method was established with mature seed of the Citrus junos Sieb. 7-8 nucellar polyembryos per seed were induced on MS basal medium without plant growth regulators. The polyembryos developed to complete plantlets on teatment with IBA. These shoots grew further in MS medium without plant growth regulators. Rooting of shoots occurred on MS medium supplemented with IBA. These plantlets were successfully transplanted to small plastic pot containing soil mixture. Somatic embryos were induced from nucellar polyembryo and maturation occurred spontaneously from proliferating cultures on MS medium without growth regulators. Random Amplified Polymorphic DNA (RAPD) marker analysis of in vitro and in vivo grown junos orange showed identical polymorphism indicative of their genetic stability. The RAPD polymorphism produced revealed same banding pattern in each regenerant. Hence, propagaton of junos orange by nucellalr polyembryos was efficient and produced in genetically stable plants under in vitro conditions.

Micropropagation of Plants and Mass Production of Adventitious Roots from Culture of Seedling Explants of Polygonatum odoratum

  • When the leaves, roots and stem segments of seedling of Polygonatum odoratum were cultured on Murashige and Skoog medium with 2.0mg/l BAP, stem segments were the most efficient explants for adventitious shoot inductino. To observe the efficient combination of growth regulators on the adventitious shoot formation , stem segments were cultured on MS medium with various kinds of cytokinins (BAP, kinetin, zeatin). From this experiment, cytokinin treatement was prerequisite for theadventitious shoot formatino,especially BAP was the most effective. Auxin (NAA or IBA) in combination with cyotokinin highly enhanced the adventitious shoot formation. Twenty five percents of explants produced the adventitious shoots on medium with 2.0mg/l BAP solely, while 83% of explants produced the adventitious shoots on medium with 2.0mg/l BAP and 0.1mg/l IBA. Root formationform adventitious shoot was promoted after transfer to 1/2 MS medium supplemented with 0.1mg/l IBA and 0.5mg/l zeatin, thereafter the plantlets with shoots and roots were cultured on 1/2MS medium lacking growth regulators. When the stem segments were cultured to MS medium with 1.0mg/l 2,4 NAA and IBA , yellow and nodulous cali were formed from the stem segments which were developed into adventitious roots. These roots were actively grew after transferred to MS liquid medium lacking growth regulators.

Propagation of a New Anticancer Plant, Cephalotaxus koreana, by Cutting (신규 항암소재의 개비자나무의 삽목증식)

  • This study was conducted to investigate the propagation characteristics of Cephalotaxus koreana by cutting, whose plant has been known to new anticancer treatments. The treatments of plant growth regulators, cutting media temperature and growing media were investigated to know the rooting ability of cuttings. For rooting formation affected by plant growth regulators and its concentration, The application of IBA 100 mg/L was most effective in all of the characteristics including rooting rate of 88.5%. The optimum media temperature by cutting was $24^{\circ}C$ (treatment IBA 100 mg/L, rooting rate 84.6%), predicting that greenwood cutting from June to July is most effective. VPPL (vermiculite+peatmoss+perlite 1:1:1(v/v/v)) showed the highest value in all of the characteristics including rooting rate of 68.5% for the effect of growing media on rooting formation.

  • The research concerned of the regeneration of plants from embryos obtained from anther cultures of ginseng (Panax ginseng C. A. Meyer). The aim was to determine the influence of the regeneration medium on the efficiency of the regeneration process. We conducted to determine the optimum conditions such as cold pretreatment, plant growth regulators and carbon sources on anther culture of P. ginseng. Highest callus formation rate was obtained when flower buds pretreated at $4^{\circ}C$ for 1 day. Among the treated growth regulators with various degrees of concentration in Murashige and Skoog's (MS) medium, 4.53 ${\mu}M$ of 2.4-dichlorophenoxyacetic acid and 4.44 ${\mu}M$ of 6-benzylaminopurine gives the most responsive callus with the frequency of 73.89% and 129.53 g of fresh weight. When we used 3-9% of sucrose and maltose among the different kinds and various concentrations of carbohydrates, callus was formed highest 67.29% in the medium with 3% of sucrose. Shoots induced from callus supplemented with 28.9 ${\mu}M$ of gibberellic acid and rooted in Gamborg's B5 medium supplemented with 14.7 ${\mu}M$ of indole-3-butyric acid.

Influence of Plant Growth Regulators on Shoot Multiplication of Thymus quinquecostatus Celak. (백리향 (Thymus quinquecostatus Celak.)의 기내증식에 미치는 식물생장조절물질의 영향)

  • In vitro culture system was established to induce multiple shoots of Thymus quinquecostatus Celak. by investigating the effects of cytokinins. Stem explants were cultures on MS medium supplemented with either five drfferent plant growth regulators or their combinations under light or dark condition. The most effective cytokininsource was the combination of BA 1.0mg/L and TDZ 0.1mg/L for producing shoots (6.05$\pm$1.51), zeatin 2.0mg/L and TEZ 0.1mg/L for elongating shoots (3.27$\pm$0.66cm) under the light condition. In addition the most effective cytokinin was 2-ip 2.0mg/L for producing shoots(5.20$\pm$1.81), zeatin 2.0mg/L for elongating shoots(5.64$\pm$1.24cm)under the dark condition. Overall, the average percent for in vitro shooting was greater than 89.58%.

In Vitro Shoot Multiplication of Albizzia julibrissin Duraz. (자귀나무(Albizzia julibrissin Duraz.)의 기내 대량증식)

  • In vitro culture system was established to induce multiple shoots of Albizzia julibrissin Duraz. by investigating the effects of cytokinins. Cotyledon, hypocotyl and root explants were cultured on MS media supplemented with either three different plant growth regulators or their combinations. The most effective cytokinin sources were zeatin 2.0 + TDZ 0.5 mg/L in cotyledon, zeatin 1.0 mg/L in hypocotyl, and BA 0.2 + TDZ 0.01 mg/L in root explant for producing shoots ($5.67\;{\pm}\;1.20$, $19.50\;{\pm}\;3.50$, and $20.50\;{\pm}\;2.47$, respectively). Also, zeatin treatment was tended to induce more shoots rather than the combinations of other cytokinins. In addition, the root induced in 1/2 MS medium without any plant growth regulators was longer and thicker than treatments of IBA, NAA, IAA and 2.4-D as auxins. Overall, the highest average percent of in vitro shoot formation was 73% from three different types of explants with treatment of zeatin (1.0mg/L).
