Inspired by the multi-scale nature of hippocampal place cells, a biologically inspired model based on a multi-scale spatial representation for goal-directed navigation is proposed in order to achieve robotic spatial cognition and autonomous navigation. First, a map of the place cells is constructed in different scales, which is used for encoding the spatial environment. Then, the firing rate of the place cells in each layer is calculated by the Gaussian function as the input of the Q-learning process. The robot decides on its next direction for movement through several candidate actions according to the rules of action selection. After several training trials, the robot can accumulate experiential knowledge and thus learn an appropriate navigation policy to find its goal. The results in simulation show that, in contrast to the other two methods(G-Q, S-Q), the multi-scale model presented in this paper is not only in line with the multi-scale nature of place cells, but also has a faster learning potential to find the optimized path to the goal. Additionally, this method also has a good ability to complete the goal-directed navigation task in large space and in the environments with obstacles.
Germinal centers (GCs) are formed in peripheral lymphoid tissues in response to protein antigens. In order to see if immunoglobulin isotype switching takes place in GC B-cells, we isolated GC B-cells (PNA positive cells) from mouse popliteal lymph nodes by a flow cytometer after the staining of lymph node cells with PNA-FITC and anti-B220-PE, and determined the expression of ${\gamma}1$ germline transcript and ${\gamma}1$ mRNA by RT-PCR. ${\gamma}1$ germline transcript and ${\gamma}1$ mRNA were amplified specifically in cDNAs from hybridoma expressing IgG1 or splenocytes stimulated LPS plus IL-4. Germinal center B-cells formed in popliteal lymph nodes of mice immunized with chicken ovalbumin were isolated 7 days after immunization. We sorted GC B-cells five times. Immunoglobulin ${\gamma}1$ germline transcripts were expressed in germinal center B-cells in three out of five sorts whereas two out of five sorts did not express ${\gamma}1$ germline transcripts in GC B-cells. The contents of GC B-cells ranged from 5 to 7% of total lymph node cells in most flow cytometric analyses but those of two sorted cells which did not express ${\gamma}1$ germline transcripts were out of normal range. These results imply that isotype switching of immunoglobulins may take place in GCs.
CD4 and CD8 T cells are key players in the immune response against both pathogenic infections and cancer. CD4 T cells provide help to CD8 T cells via multiple mechanisms, including licensing dendritic cells (DCs), co-stimulation, and cytokine production. During acute infection and vaccination, CD4 T cell help is important for the development of CD8 T cell memory. However, during chronic viral infection and cancer, CD4 helper T cells are critical for the sustained effector CD8 T cell response, through a variety of mechanisms. In this review, we focus on T cell responses in conditions of chronic Ag stimulation, such as chronic viral infection and cancer. In particular, we address the significant role of CD4 T cell help in promoting effector CD8 T cell responses, emerging techniques that can be utilized to further our understanding of how these interactions may take place in the context of tertiary lymphoid structures, and how this key information can be harnessed for therapeutic utility against cancer.
The pattern of elongation in the developing hypocotyl of Glycine max shows that the elongation generally proceeds from base to the cotyledonary node in acropetal diredtion, although earlier elongation takes place through the entire hypocotyl. Because the differentiation of the vascular cambium in the hypocotyl advances also acropetally, it can be seen that the acropetal wave of hypocotyl elongation is associated with the acropetal differentiation of the cambium in the hypocotyl elongation is associated with the acropetal differentiation of the cambium in the hypocotyl elongation is associated with the acropetal differentiation of the cambium in the hypocotyl. In order to examine the mechanism of asbestos clastogenicity, CHO cells were treated with chrysotile and crocidolite. Crocidolite and chrysotile were able to induce lipid peroxidation in a dose dependent manner. Ultrafiltrate of culture media from CHO cells treated with chrysotile/crocidolite induced sister chromatid exchange in CHO cells. Ultrafiltrate of culture media from CHO cells treated with chrysotile induced chromosome aberration but it was not statistically significant. Simultaneous treatment of 3-Aminobenzamide (3-AB) or cytosine arabinoside (Ara C) with crocidolite had no effect on the frequency of chromosome aberration by crocidolite whetease posttreatment of caffeine significantly increased the chromosomel aberration by crocidolite. This indicated that DNA damage by asbestos took place at late stage of cell cycle. The results suggested that the ultrafiltrate of media contained clastogenic factor (CF) and lipid peroxidation might be involved in the formation of CF.
Ultrastructural characteristics of hemocytic differentiation pathway in Sericinus montetla Grey were observed with transmission electron microscope. Hemocytic differentiation took place in loose islets of hemopoietic organs as indicated by the presence of differentiating hemocytes; prohemocytes, plasmatocytes, granular cells and spherule cells. They are differentiatied from the stem cells through individual cell lineages. However, differentiating aspects of the oenocytoids were not observed. According to Brehelin and Zachary (1986), the premature hemocytes around the hemopoietic organs were classified into five types; prohemocytes, plasmatocytes, granular cells, spherule cells and oenocytoids.
Nitric oxide (NO) is now recognized as a mediator of several biological and immunological functions, but unlike classical neurotransmitters. NO simply diffuse of the postsynaptic cells and around affecting cells. Taraxacum officinale (Compositae) has been used for maintenance of vitality, and they still occupy an important place in the traditional Korean medicine. We have examined that the effect of Taraxacum officinale water extract on NO synthesis in microglial cells of murine's brain, using the Griess method. And this study was evident that Taraxacum officinale did not induce NO production without recombinant interferon gamma ($rIFN-{\gamma}$), whereas Taraxacum officinale (10-1000 g/ml) with $rIFN-{\gamma}$ effectively produced NO in microglial cells of brain. As result. NO production in microglial cells increased most significantly in dose of 100 g/ml of the Taraxacum officinale and the production of NO was dependent on the dose of Taraxacum officinale, NG-monomethyl-L-arginine, competitive inhibitor of NO synthase, reduced the NO production by Taraxacum officinale stimulation with $rIFN-{\gamma}$ in microglial cells of murine. The effect of Taraxacum officinale was mainly dependent on Taraxacum officinale-induced tumor necrosis factor- secretion. Conclusively, this study suggested that Taraxacum officinale stimulate NO production at microglial cells in brain, which may be an important factor for mediating immune and neuroendocrinologic regulation in nervous system.

The incorporation of ³H-proline by epithelial and connective tissue elements of rat palatal mucosae was studied in order to investigate the relative levels of protein synthesis by the epithelium and underlying connective tissue cells. Following a sixty minutes incorporation of the radioactive tracer in vitro, it was found that the suprabasal cells had most grains per unit area. Furthermore, the grains were more concentrated over the cytoplasm than the nucleus. This was in contrast with the labeling of basal cells which had twice as many grains over the nucleoplasm than that over the cytoplasm. In intermediate cells; i.e., the spinous layer, the number of silver grains per unit area was decreased from that of the suprabasal cells. In areas where desmosomes were more prominent, many grains were in touch with such desmosomes. However, the labeling appeared to be reduced as soon as the cells became flattened. Moreover, the epidermal keratohyalin granules were relatively free of grains. Except for certain intercellular surfaces the keratinized cells were generally free of grains. On the connective tissue side, silver grains were primarily localized over the fibroblasts with occasional grains being found over palatal muscle cells, neural elements and so on. Most grains over collagenous fibers were found in relation to mature collagen fibrils. Thus, protein synthesis in isolated mucosae of the rat palate appeared to take place both in epithelial and connective elements. There were no apparent tissue alterations caused by the in vitro incorporation procedure utilized under conditions of this study.
Antibody-toxin conjugates, termed immunotoxins, are currently being evaluated as potential new anticancer agents and one of the most extensively studied toxins for construction of immunotoxin is ricin which exists in the seeds of castor bean, Ricinus communis. Another toxic lectin from castor bean is RCA (Ricinus communis agglutinin). Both toxins are very homologous. We reported the puriffcation procedure and biological properties of ricin from the Korean castor bean in another place and here we report those of RCA. The purified RCA shows three bands on denatured SDS PAGE while ricin shows two bands. On cultured $K_{562}$ cells ricin and RCA both inhibit the multiplication of cells extensively. $30{\mu}g/ml$ of ricin shows 73% of inhibition rate at day 4 compared to 68% in same condition of RCA. The inhibition of multiplication of cells are directly proportional to the concentration of toxins and the incubation period. In every case ricin was more toxic than RCA. The $LD_{50}$ dose of ricin on ICR mice was 60 ng at day 3 but that of RCA was $10{\mu}g$.
Attachment and growth of mouse flbroblast cells on block copolypeptides were studied in the pres once or absence of serum proteins. Cells are rapidly attached to she polymer surface within 30 min regardless of substrate in the presence of serum. The number of flbroblast cells attached on the poly mer surface coated by collagen was larger than thats on the bare surface. Attachment of cells Is as a whole suppressed to a low level by the addltion of sodium azide in the absence of serum. Thls suE gests that the active attachment of cells requires the biological metabolism taking place on polymer substrates. In the presence of serum protein, flbroblast cells are more rapidly grown on the bolck co polymer consisting of poly(T-benzyl L-glutamate) (PBLG) and polyoxypropylene(POP) than on other block copolymers. These results were in agreement wish the data obatlned by an Inverted ml croscope.
