Studies on the Artificial Cultivation of Pleurotus cystidiosus O.K.Miller, the Abalone Mushroom (전복느타리버섯(Pleurotus cystidiosus O.K.Miller)의 인공재배에 관한 연구)

  • The study was carried out on the artificial cultivation of the abalone mushroom, Pleurotus cystidiosus O.K.Miller. The pine sawdust substrates with 20% rise bran were good for mycelial growth and high quantity of P. cystidiosus in the bottle cultivation. Moreover, the proper volume for bottle cultivation was 850 ml and the removal of spawn and surface layer of the medium before pin-heading was more efficient. The yields of P. cystidiosus were higher in sawdust substrates added calcium carbonate than those not added calcium carbonate. The volume of 3 kg polypropylene bag is good for yield and biological efficiency in bag cultivation of P. cystidiosus. Cotton wastes were proper substrates for bag cultivation. In the effect of different cultivation temperature, $28{\pm}2^{\circ}C$ cultivation temperature was good for for primordial formation after inoculation.

  • This study investigated the culture characteristics of Cryptoporus volvatus, whichis grow naturally in Korea, to determine the suitable environmental conditions for its cultivation. The physiological characteristics of the mycelia were assessed according to the cultivation conditions, to determine the optimal conditions for artificial cultivation. The visual characteristics of the hyphae of Cryptoporus volvatus KACC52303 included an irregular and uneven surface and a fuzzy or cotton-like texture. Under the microscope, its microstructure showed pre-chlamydospore formation, but no clamps were seen. The appropriate culture temperature was found to be a medium/high temperature of approximately 25-30℃, and the optimal pH was found to have a wide range from weakly acidic (pH 4) to neutral (pH 7). In the optimal nutrient source experiment, hyphal growth was shown to be fair in a mixed medium with 2.5% dextrin as the carbon source and 0.1% yeast extract as the organic nitrogen source. Among the various amino acids, organic acids, and inorganic salts tested, the fastest hyphal growth was observed in the presence of leucine, acetic acid or gluconic acid, and KCl or KH2PO4, respectively. The column test showed that the best mycelial growth occurred in a mixed medium of 80% pine sawdust, 10% rice bran, and 10% corncob sawdust.

Comparison in Cultural Characteristics According to Pot Diameter and Substrate Weight in Pot Cultivation of Pleurotus ostreatus (느타리버섯 봉지재배시 봉지직경 및 배지량에 따른 생육 특성 비교)

  • This study was carried to investigate efficient pot diameter and substrate weight in pot cultivation of Pleurotus ostreatus. Cultivating substrate was pine sawdust+beet pulp+cotten seed flour (50 : 30 : 20, v/v). Ranges of pot diameter and substrate weight were $10{\sim}20\;cm\;and\;600{\sim}2,500\;g/pot$, respectively. Smaller pot diameter resulted in longer pot length according to increasing substrate weight, so mycelial growth period and total cultivation period was retarded. Wider pot diameter resulted in a little lower biological efficiency in the same substrate weight. Heavier substrate weight was brought higher yield but lower biological efficiency. Average yield according to pot diameter was decreased at wider than 14 cm during second flush. And average biological efficiency according to substrate weight was lower than 100% at heavier than 1,200 g/pot during second flush. So suitable pot diameter and substrate weight in pot cultivation of Pleurotus ostreatus were estimated $10{\sim}14\;cm\;and\;800{\sim}1,200\;g$, respectively.

Selection of substitute medium of cotton seed pomace on the oyster mushroom for bottle cultivation (느타리버섯 병재배 면실박 대체배지 선발)

  • Main materials used as media for oyster mushroom cultivation are pine sawdust, beet pulp, cotton seed pomace. Increases in the price and the unbalance of demand and supply of cotton seed pomace was often damage to oyster mushroom cultivation farm, so we investigated agricultural by-product to replace the cotton seed pomace for bottle cultivation of oyster mushroom. Treatment of coconut oil meal or coconut pomace delayed incubation period about 3 days compared with cotton seed waste treatment(control), but yield and income index showed similar to each other in three treatment. Consequently coconut oil meal and coconut pomace could select for cotton seed pomace substitute.

  • This study was carried out investigate optimal additive content of spent medium from oyster mushroom in Volvariella volvacea cultivation. The formation of spent mushroom medium from oyster mushroom consisted of 50% pine sawdust, 30% beet pulp and 20% cotton seed hull pellet. The higher amount of spent medium from oyster mushroom resulted in higher total nitrogen content and C/N ratio of medium decreased according to increasing mixing ratio of spent medium from oyster mushroom. The fruit body yield and biological efficiency of supplemented with 50% spent medium from oyster mushroom was same level of non additive medium.

  • A new commercial strain of Agrocybe chaxingu, "Dasan", was developed with crossing between monokaryotic strain derived from Jinhyang and GWM20502 strain. The optimum temperature, carbon source and nitrogen source for mycelial growth were $25{\sim}30^{\circ}C$, glucose and ammonium chloride, respectively. The period of primordia formation and fruiting body growth were about 12~13days and 6~8days in bottle cultivation using Georgia pine sawdust which supplemented with 30% wheat bran. The yields was 118.2g/850mL bottle, and the color of pileus was brown.

  • This experiment was carried out to find the method for mass production by artifical cultivition and to analyze the components of Paecilomyces japonica according to several media. Time of inoculation of the Paecilomyces japonica using silkworm was on first day of five molting and infection rate was 72.0%. Optium medium for mass production of the Paecilomyces japonica was known effective for increasing dry weight and fruitbody at brown rice 80g plus pupa powder 20g. Dry weight of Paecilomyces japonica using fungus of silkworm was 1.2g including pupa and length of fruitbody was appeared 3.0cm to 3.5cm. Content of $\beta$ - glucan was very high as 40.5% at inoculation on the first day of the five molting while 16.4% at brown rice, 20.7% at pupa, 23.1% at brown rice plus pupa powder, and 28.7% at pine sawdust plus wheat bran. Mycelium was poor and pinkly conidiospore was formed on media of centipede, maggot and powder of silkworm.

  • To study the cultural characteristics and wood rotting ability of the secondary mycellia of Armillaria mellea, it was cultivated on the various media. The optimal mycelial growth condition was 20~27 and pH 5.0~6.5 on PDB. A. mellea grew well on MEA, PDA and GP. Lactose and mannitol as carbon sources and glutamic acid as nitrogen sources were found to be effective as additives. A. mellea employed in this study have the characteristics of white rot types. Pine and oak wood were selected as candidates for sawdust substrate.

Effect of Peatmoss-Based Organic Material Mixtures on Soil pH, Growth and Fruit Quality of Highbush Blueberry(Vaccinium corymbosum L.) Plants (하이부시 블루베리 정식 시 유기물 조성이 토양 pH, 생육 및 과실 품질에 미치는 영향)

  • This experiment was conducted to test the suitability of various organic materials in order to reduce the use dependence of peatmoss as a soil pH regulator and to examine the effect of soil organic matter supply. A 2-year old northern-highbush blueberry (Vaccinium corymbosum L.) 'Duke' plants were planted in the field at $2.0m{\times}2.5m$ spacing in spring. Before planting, organic materials were incorporated into the soil including the planting hole at 20 liter per plant, as the following mixtures: peatmoss only (20 L), peatmoss (10 L) + pine needle (10 L, PN), peatmoss (10 L) + rice hull (10 L, RH), and peatmoss (10 L) + sawdust (10 L, SD). The pH of organic materials was lowest in peatmoss (pH 4.3), followed by PN (pH 4.8), SD (pH 5.7) and RH (pH 7.8). Soil pH measured right after planting ranged from 5.3 to 5.9 and was lower in PM only and PM + PN than PM + RH and SD treatments. In the third year, the pH lowered to the range of 4.2 to 4.5, with PM and PM + PN still maintaining lower values. The early growth was good in the mixed treatment of PM and PN, and the plant height and width and the number of new shoots were good in the PM treatment. Soil water content was maintained highest in PM + PN, followed by PM, PM + SD and PM + RH. Vegetative growth was maintained better in PM and PM + PN, and the number of flower cluster and yield were also slightly higher in those treatments while mean fruit weight was similar among all treatments. Fruit quality indices such as total soluble solids, titratable acidity and firmness were not affected.

Cultivation technique using plastic container and selection the superior strain of nameko mushroom (Pholiota nameko) (맛버섯(Pholiota nameko) 우량균주 선발 및 병 재배법)

  • These experiment was conducted to find the superior strain selection, cultivation technique and optimum environmental condition of nameko mushroom culture using plastic container. The results was following as Mycelium of Pholiota nameko grown well at MCM and Hamada media, and its media acidity was pH 6~7. The optimum temperature condition for growing mycelium was $25^{\circ}C$. Under $15^{\circ}C$ and above $30^{\circ}C$ of temperature condition, mycelium growing speed was delayed remarkably. Among the 29 strains of nameko mushroom, the most productive strains was JNM19007, JNM19026, JNM19027 and JNM19028. The optimum media composition rate for produce fruitbody was pine sawdust 80% + wheat bran 20%. In this condition, the average fruitbody amount was 188g per 1,100cc container. The optimum post-culturing period was 50 days and mushroom sprout appeared 7 days after old mycelia removed. The suitable temperature was $12^{\circ}C$ for induce sprout, growing period was $16^{\circ}C$ and the optimum relative humidity was 95% in all culturing periods.
