Optimal additive ratio of barley flour for substitution of rice bran at cultivation of winter mushrooms (팽이버섯 재배에서 미강 대체용 보리가루의 적정 첨가 비율)

  • This study was carried out to investigated optimum additive ratio of barley flour for substitution of rice bran at cultivation of winter mushrooms. Mycerial growth was faster according to increase of barley flour ratio than those of controls, but only some slow at the addition of 10% barley flour. Yields of fruiting body show the highest to 165.4 g/850 ml of medium which are the addition 30% of barley flour and not significant difference of yields up to the addition of 70% barley flour. Diameter of pileus was the highest at the addition of 30% barley flour. Hardness of pileus and stipes were the highest at the addition of 10% barley flour. The L value of stipes were the hight at the addition of barley flour, but the L value of pileus were decreased at the addition of barley flour, but there was no significant difference in the a-value and the b-value.

Growth characteristics and productivity of oyster mushroom according to adding of Salvia miltiorrhiza (단삼 첨가에 따른 느타리버섯의 생육 특성 및 수량성)

  • This study was carried out to investigated optimum mixing ratio of Korean cultivation Salvia miltiorrhiza for production of functional oyster mushroom. Mycerial growth was slow at addition of Salvia miltiorrhiza, and significant difference by increase of Salvia miltiorrhiza. Yields of fruiting body show the highest to 139.5 g/850 ml of medium which are addition 5 g/bottle of Salvia miltiorrhiza but rapid decrease by increase of Salvia miltiorrhiza. Diameter of pileus and thick of stipes were higher at addition Salvia miltiorrhiza than those of the controls. Thick of pileus and length of stipes were the highest at addition 30g, and 20 g and 30 g of Salvia miltiorrhiza. The L value of stipes were the highest at addition 20 g and 50 g of Salvia miltiorrhiza and the L value of pileus were the highest at addition 30 g of Salvia miltiorrhiza, but there was no significant difference in the a-value and the b-value.

Characteristics of a newly bred Pleurotus eryngii cultivar, Gat_Aeryni (큰느타리(새송이)버섯 신품종 '갓애린이'의 생육특성)

  • In order to breed a new Pleurotus eryngii cultivar with a large pileus and convex shape, which are favorite traits of customers from Europe and North America, single crosses between monokaryotic mycelia derived from basidiospores of KNR2555 were performed to yield the new cultivar $2{\times}12$ chosen by cap shape (convex), diameter of pileus (60.7 mm), and quality (4.9) in a preliminary cultivation. The strain was named Gat_aeryni and was cultivated on a large scale for comparison with Kenneutari 2 ho at the GNARES and at mushroom farms. The yield of Gat_aeryni (71.7 g) was not significantly different from that of Kenneutari 2 ho (71.4 g), and the quality of the new cultivar was 6.8, which was not significantly different from that of a reference cultivar (6.5). Days for harvest, length, and diameter of the pileus in the two cultivars were statistically different by an independent t-test (P < 0.001, P < 0.05, P < 0.001 and P < 0.05 respectively). The new and reference cultivars were discriminated by PCR reactions with a primer set (URP1 and URP10) and simultaneous cultivation.

Physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn according to harvest cycle (액체종균에 의한 표고의 수확 주기에 따른 이화학적 특성 및 항산화 활성)

  • The physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn were compared according to the harvest cycle (1-3 cycles). There was no significant difference in moisture content. The crude lipid and crude protein contents tended to decrease according to harvest cycle. The content of β-glucan was the highest in mushrooms after one cycle. The β-glucan content in cycles 2 and 3 was 68.5% and 62.3% that of the content after cycle 1, respectively. Mineral content in the pileus and stipe was similar. The mineral content was highest in the pileus after two cycles. Composition and contents of free amino acids tended to decrease according to the harvest cycle. Contents of essential amino acids were higher in mushroom after one cycle. Total phenol content was highest in the pileus (31.96 mg/100 g) and the stipe (21.10 mg/100 g) after one cycle and tended to decrease with subsequent cycles. The flavonoid content was 11.13 mg/100 g after one cycle and significantly decreased according to the harvest cycle. Antioxidant activities were also highest after one cycle and significantly decreased according to the harvest cycle. When compared to L. edodes cultured for one cycle, L. edodes cultivated with liquid spawn for three cycles was considered to have similar marketability in terms of quality characteristics.

Characteristics and breeding of 'Geumhyang2ho', a new brown and labor-saving variety of Flammulina velutipes (노동력 절감형 갈색 팽이버섯 신품종 '금향2호'의 육성 및 특성)

  • 'Geumhyang2ho', a new brown and labor-saving variety of Flammulina velutipes, was bred by spontaneous crossing between 'Geumhyang' and 'Garlmoe' during demonstration cultivation in bag culture farm of oyster mushroom in 2013. During bottle cultivation, the temperature was maintained at $16^{\circ}C$; the variety displayed good quality compared to the control ('Geumhyang'), without necessitating vinyl cone treatment. The time periods necessary for mycelial growth, fruit body growth, and primordia formation were 24 days, 11 days, and 8 days, respectively. The total cultivation period was 43 days, which was 2 days longer than that of the control. The pileus diameter was smaller, but the pileus thickness and stipe diameter were greater than those of the control. The pileus color was yellowish ivory, which was very similar to that of the control. The yield of 'Geumhyang2ho' was 118 g per 850 ml bottle, which was markedly lower than that for bottle cultivation with vinyl cone treatment. However, its good quality suggests the potential of the labor-saving bottle-based cultivation of 'Geumhyang2ho' without vinyl cone treatment in oyster mushroom farms.

Growth characteristics of Lentinula edodes treated with bean sprout waste (콩나물 부산물 첨가량에 따른 표고의 생장특성)

  • In this study, the growth characteristics of Lentinula edodes were confirmed by bean sprout waste(BW) as an alternative raw material for rice bran. The mycelium growth of Sanjo701, a major cultivation variety of L. edodes, was compared between a medium mixed with 8:2(v/v) of oak sawdust and a medium mixed with BW 50% and BW 100%. The mycelium growth in BW 50% was 13.5 cm. Compared to the control, BW 50% increased the diameter of the pileus by 1.6 cm. Additionally, the length of the pileus decreased by 0.4 cm when comparing the growth of the fruit body. In contrast, at BW 50%, the diameter of the pileus decreased by 9.6 cm and the length of the stipe decreased by 1.4 cm. According to analysis of the constituent amino acids, BW 50% showed a lower overall nutritional content than the control, whereas BW 100% had a lower amino acid content than the control. However, glutamic acid and aspartic acid, which are flavor-enhancing ingredients, were observed at levels of 3.954 mg/g and 1.436 mg/g, respectively, in BW 100%. Therefore, if bean sprout by-products are efficiently processed and utilized, it is believed that they will be beneficial to farmers as a substitute for rice bran and reduce the cost of manufacturing substrate

Breeding and characterization of a new white cultivar of Pleurotus ostreatus, 'Sena' (갓이 백색인 느타리 신품종 '세나'의 육성 및 특성)

  • The development of automated bottle cultivation systems has facilitated the large-scale production of Pleurotus ostreatus, a commonly cultivated oyster mushroom species in South Korea. However, as the consumption of this product is decreasing and production quantities are exceeding demand, farmers are seeking various other mushroom types and cultivars. In response to this, we have developed a new oyster mushroom cultivar named 'Sena'. This high-yielding cultivar has a white pileus and excellent quality. The white oyster mushroom cultivars 'Goni' and 'Miso' were selected as parental strains from the genetic resources of the National Institute of Horticultural and Herbal Science's Mushroom Division. By crossing their monokaryons, hybrids were developed and subjected to cultivation trials and characteristic evaluations to select the superior cultivar. The optimal temperature for 'Sena' mycelial growth is 25-30℃, with inhibition occurring at temperatures above 30℃, whereas the temperature for mushroom growth is 14-18℃. The mushrooms grow in clusters, with the white pileus having a shallow funnel shape. Optimal mycelial growth occurs in malt extract agar medium. When cultivated in 1,100 cc bottles, the 'Sena' cultivar had 35 available individuals, surpassing the number 16 available from the control cultivar 'Goni'. The yield per bottle also increased by approximately 157 g, a 24% increase over the control cultivar amount. When 300 g samples of harvested mushrooms were packed and stored at 4℃ in a cold storage facility for 28 days, the weight loss rate of 'Sena' was approximately 4.22%, lower than that of 'Goni'. Moreover, the changes in pileus and stipe whiteness (measuring 6.99 and 8.33, respectively) were also lower than those of the control cultivar. Since the appearance of a white cap is crucial for quality assessment, the 'Sena' cultivar is superior to the 'Goni' cultivar in terms of both weight and quality after undergoing low-temperature storage.

Physiological and Genetic Characteristics of Cultivated Mushroom, Hypsizygus marmoreus

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated in Northeast Asia. Japan's annual production is 110,000ton or more. Since 2002, cultivation is expanded in Korea. To investigate the morphological, cultural and microscopic characteristics of Hypsizygus marmoreus, 109 isolates were collected from Korea and other countries. Clamp connection, chlamydospore and arthrospore were present in all tested isolates of H. marmoreus except HYM-002 and HYM-004. Also pilealtrama, gilltrama, basidia, basidiospore and cystidia of fruiting body were no difference among the isolates in the present investigation. Morphological characteristics of fruiting body was that color of pileus was brown and white, irregular as marble, the average size 12~22mm and stipes was $46{\sim}91{\times}6{\sim}10mm$. Isolates HYM-031, HYM-047 and HYM-109 formed grayish-brown pileus with a faint pattern. Molecular analysis with RAPD and ITS rDNA sequence analysis were also performed to check the genetic relationships among H. marmoreus isolates. Based on the RAPD analysis using the URP-PCR, all isolates of H. marmoreus were clustered into large 3 groups but more than 90% showed high similarity. In addition, morphological and geographical differences have been classified as an independent cluster. The brown and white strains enclosed in same cluster. So genetically no significance difference was observed between these two strains. ITS gene sequences of 16 selected isolates which were 640 bp long, were aligned and compared. The similarity in ITS sequence was 94.8 to 99.1% among tested isolates and the H. marmoreus isolates in GeneBank. In conclusion the tested isolates were H. marmoreus. Morphological and molecular observations proved that all tested isolates were belonging to H. marmoreus. For the stable artificial cultivation, composition of optimum media, mature period and light condition were established. Optimal formula of artificial cultivation medium was Douglas sawdust: corn cob: soybean meal: wheat bran = 40:30:15:15. In addition, 7% rice bran and 3% yellow sucrose was the most effective composition for spawn's liquid medium. For the maturation of the isolates was favorable for growing for 20 to 30 days at $25^{\circ}C$ and the LED lights in mixture of white and blue was good for growth period. For effective growth, the temperature, humidity and aeration control in every step was important.

A new oyster mushroom variety 『Hukbaek』 for the bottle cultivation (느타리버섯 신품종 육성 연구(III) -병재배용 신품종 『흑백』 느타리버섯의 특성-)

  • Hukbaek oyster mushroom were bred and cultivated one after mating spores from Aenutari 1-ho irradiated to X-ray and Byungnutari 1-ho. The major charateristics of the mushroom are showing that pileus is black-brown and stems are pure-white and straight. The optimum temperature for the mycelial growth was around $25{\sim}30^{\circ}C$ and that for the pinheading and growth of fruitbody was around $14{\sim}16^{\circ}C$. Around 20 days at $20^{\circ}C$ were required for incubation time and the yield was shown high by 142.8g/bottle. When the cultivating temperature is over $17^{\circ}C$ the color of pileus becomes gray-brown.

Characteristics and breeding of a new brown variety 'Heukhyang' with good taste in Flammulina velutipes (고식미 갈색 팽이버섯 신품종 '흑향'의 육성 및 특성)

  • 'Heukhyang', a new variety with dark brown color, short cultivaltion period, and good taste in Flammulina velutipes, was bred by crossing between monokaryons isolated from 'Garlmoe' and wild strain 'CBMFV-10'. In bottle cultivation, the period of mycelial growth, primordia1 formation, and growth of fruit body was required 23 days, 6 days, and 17 days, respectively. Total culvivation period was 46 days, showing 8 days shorter than that of 'Garlmoe'. Pileus diameter and thickness were greater than those of 'Garlmoe', and pileus color showed dark brown. Fruit body production per 850 ml bottle was 135 g, which was 10% lower than that of 'Garlmoe'. However, the overall acceptance was most excellent in sensory evaluation, and 'Heukhyang' has an advantage to target a niche market of white ones due to differentiated color and taste.
