The present study compared the developmental potential of cloned porcine embryos with mesenchymal stem cells (MSCs), fetal fibroblasts (FFs) and cumulus cells (CCs) by assessing the cleavage and blastocyst rate, total cell number, inner cell mass (ICM) ratio and apoptosis. MSCs were isolated by ficoll gradients from femur of -6 month old female pig, and maintained for primary cultures. FFs from a female fetus at ${\sim}30$ day of gestation were established, and CCs were obtained from cumulus oocyte complexes (COCs) aspirated from $3{\sim}6$ mm follicles in diameter. Donor cells at $3{\sim}4$ passage were employed for nuclear transfer (NT). COCs were matured and fertilized in vitro(IVF) as control. Cleavage rate was significantly (P<0.05) higher in IVF than in NT embryos with MSCs, FFs and CCs ($82.7{\pm}8.9%\;vs\;70.6{\pm}5.4,\;68.7{\pm}5.1\;and\;63.4{\pm}5.6%$, respectively). However, blastocyst rates in IVF and NT embryos derived from MSCs ($24.5{\pm}2.8\;and\;20.4{\pm}8.3%$) did not differ, but were significantly (P<0.05) higher than NT derived from FFs and CCs ($10.6{\pm}2.7\;and\;9.8{\pm}2.1%$). Total cell number and the ratio of ICM to total cells among blastocysts cloned from MSCs ($35.4{\pm}5.2\;and\;0.40{\pm}0.09%$, respectively) were significantly (P<0.05) higher than those from FFs and CCs ($24.9{\pm}6.2%\;vs\;0.19{\pm}0.16,\;23.6{\pm}5.5\;and\;0.17{\pm}0.16%$, respectively). Proportions of TUNEL positive cells in NT embryos from FFs and CCs ($6.9{\pm}1.5\;and\;7.4{\pm}1.7%$, respectively) were significantly (P<0.05) higher than in MSCs ($4.8{\pm}1.4%$) and IVF ($2.3{\pm}0.9%$). The results demonstrate that MSCs have a greater potential as donor cells than FFs and CCs in achieving enhanced production of cloned porcine embryos.

The selection of morphologically normal spermatozoa is critical to obtain high breeding performances in boar breeding farms and artificial insemination (AI) centers. Parameters for the selection of semen mainly include total sperm motility, concentration, and morphology. However, these primary parameters are often not reliable for discriminating between normal and abnormal, non-fertilizable spermatozoa. The present study was designed to compare the motion characteristics, fertilization ability using in vitro fertilization (IVF), and acrosome formation of the semen from boars having low (boar number 2012) and normal (boar number 2004 and 2023) breeding performances. The ultimate goal was to identify additional simple and easy criteria for the selection of normal sperm. There was no significant difference between boar 2004 and boar 2023 sperm total motility in computer assisted sperm analysis. However, boar number 2012 semen presented a significantly reduced population of rapid moving spermatozoa and an increased population of slow moving spermatozoa compared to boar numbers 2004 and 2023. Analysis of detailed motion characteristics revealed that sperm from boar number 2012 had significantly reduced motility in progressiveness, average path velocity, straight-line velocity (VSL), curvilinear velocity (VCL), straightness, and linearity. The assessment of the fertilizing ability by IVF also showed that sperm from boar number 2012 showed a fertility rate of 3.4%, whereas sperm from boar number 2023 had a fertility rate of 75.45%. Interestingly, most of the sperm nuclei were found on the peripheral area of the oocytes, suggesting that the sperm from boar number 2012 lacked penetration ability into the oocyte zonapellucida. The acrosome formation analysis using Pisum sativum agglutinin staining demonstrated that the sperm from boar number 2012 had a defect in acrosome formation. Consequently, primary parameters for selecting semen before AI such as motility are not sufficient to select normal and fertilizable spermatozoa. In conclusion, the present study suggests that the acrosome staining and detailed motion characteristics such as progressiveness, VCL, and VSL should be included in determining semen quality together with primary parameters for successful AI and high breeding performance in the swine industry.
Blastocyst formation, consisting of the inner cell mass (ICM) and trophectoderm (TE), is the first differentiation process during embryonic development in mammals. It has been hypothesized that the proportion of ICM to TE in the blastocyst may be crucial for subsequent developmental competence of early embryos, which it may be expressed as a sensitive indicator for evaluating in vitro systems. In this study ICM/total cell ratio of nuclear transfer (NT) embryos was compared with IVF-derived and in vivo embryos. Somatic cell nuclei obtained from a fetus at Day 40 of gestation were transferred into the enucleated oocyte and then cultured in NCSU 23 medium for 6 days as previously described (Koo et al., Biol. Reprod. 2000; 63:986-992). ICM and TE cells of blastocysts were determined by using a differential staining method (Han et al., Biol. Reprod. 1999; 60:1110-1113). Development rate (9.8$\pm$2.5%, 23/225) to the blastocyst stage of NT embryos was lower than IVF embryos (23.8$\pm$2.7%, 53/223). Thus, a difference was detected in the in vitro developmental rate to blastocyst stage between NT and IVF-derived embryos (P<0.05). In the next experiment, we investigated ICM and TE nuclei to assess the quality of blastocysts that produced by NT, IVF and in vivo, respectively. NT blastocysts (27.6$\pm$8.3) showed a smaller total cell number than IVF-derived (42.6$\pm$17.4) and in vivo embryos (283.9$\pm$103.5) (P<0.05). Ratios of ICM/total cells in NT, IVF and in vivo blastocysts were 15.1$\pm$ 18.6% (n=56), 12.3$\pm$9.2% (n=57) and 30.4$\pm$6.8% (n=40), respectively. Individual blastocysts for the ratio of ICM/total cells were assigned to 3 groups (I; <20%, II; 20 to 40% and III;>40%). As the results, most in vivo blastocysts (97.5%, 39/40) were distributed into group II while most NT (78.6%, 44/56) and IVF-derived blastocysts (82.5%, 47/57) were allocated to group I. Thus, our data show that NT or IVF-derived embryos have aberrant morphology during early development in vitro systems, suggesting that these anomalies may result in developmental failures of the NT embryos to term.
도축된 돼지의 난소에서 난포란을 채취하여 체외 성숙시킨 후 인위적으로 활성화시켜 이배체 배발달을 유기하기 위해 cytochalasin B를 2.5, 5.0, $7.5\;{\mu}g/mL$ 농도로 3, 5, 7 시간 처리한 후 NCSU23 배양액으로 7일간 배양하여 배발달율에 미치는 영향을 검사하였다. 체외 성숙된 돼지 난모세포를 활성화 처리하여 2일째 분할율을 관찰한 결과 각 처리의 분할율은 $39.0{\sim}48.9%$로 나타났으며, 처리별로 유의차가 없었다. 7일간 배양하여 상실배기 이상으로 발달한 난자들의 처리별 발달율 차이를 관찰한 결과는 cytochalasin B를 $5.0\;{\mu}g/mL$ 농도로 3시간 처리구(19.7%)에서 $2.5\;{\mu}g/mL$ 농도로 3, 5시간 처리한 구(9.4%)의 배발달율에 비해 유의적으로 높은 결과를 얻었다. 시간별과 농도별로 분석한 결과 $2.5\;{\mu}g/mL$ 처리구가 5.0, $7.5\;{\mu}g/mL$ 처리구들보다 유의적으로 낮은 배발달율을 보였다. 배반포기까지 발달한 난자들의 처리별 발달율 차이를 관찰한 결과는 cytochalasin B를 $5.0\;{\mu}g/mL$ 농도로 3시간과 5시간 처리구들의 배반포기 발달율은 9.4%와 9.0%로 $2.5\;{\mu}g/mL$ 농도로 3시간 처리구의 배발달율인 0%보다 유의적으로 높은 배발달율을 보였다. 시간별과 농도별로 분석한 결과 농도에 따라 $5.0\;{\mu}g/mL$ 처리구가 2.5와 $7.5\;{\mu}g/mL$ 처리구들보다 유의적으로 높은 배반포기 배발달율을 보였으며, 3, 5, 7시간 처리에 따른 유의적인 차이는 없었다. 이상의 결과로 돼지 난모세포를 65시간 체외 성숙 후 활성화 처리할 때 cytochalasin B $5.0\;{\mu}g/mL$로 $3{\sim}5$ 시간 처리하는 것이 가장 높은 배반포 발달율을 얻을 수 있었으며, 처리 농도가 배발달에 유의적인 영향을 미치는 것을 확인하였다.
본 연구는 체외 성숙된 난자와 동결 융해 정자를 이용한 돼지의 체외 수정 과정에서 난구 세포의 존재가 정자 침투율, 웅성전핵 형성률 그리고 후기배로의 체외 발육에 미치는 영향을 알아보기 위하여 수행되었다. 돼지 난소로부터 난자-난구세포 복합체를 채취하여 eCG/hCG, 10% 돼지 난포액, epidermal growth factor 등이 첨가된 TCM 199 배양액에서 44시간 배양하여 체외 성숙을 유도하였다. 성숙 배양 후 난구 세포를 제거한 난자와 난구 세포가 부착되어 있는 난자를 돼지 동결 융해정액을 이용하여 5mM caffeine과 10mM calcium chloride를 함유한 mTBM배양액에서 8시간 체외 수정하였다. 체외 수정 후 난자를 고정, 염색하여 정자 침투율과 웅성전핵 형성률을 조사하였고(실험 $1{\sim}3$) 일부 수정란을 North Carolina State University-23 배양액에서 체외 수정 후 156시간 배양하여 후기배로의 발육능을 검토하였다(실험 3). 실험 1에서는 정자 농도를 $7.5{\times}10^5/ml$로 조정하여 나화 난자와 난구 세포 부착난자에서 정자 침투율 및 웅성전핵 형성률을 조사하였다. 실험 2에서는 난구 세포 부착 난자의 체외 수정에 적합한 정자 농도를 구하기 위해 2, 3, 4, 및 $5{\times}10^6/ml$의 농도로 난자를 수정한 후 정자 침투율 및 웅성전핵 형성률을 조사하였다. 실험 3에서는 나화 난자 및 난구 세포 부착 난자를 각각 $7.5{\times}10^5/ml$의 정자 농도로 체외 수정한 후 후기배로의 발육률을 조사하였다. 실험 1의 결과 정자 침투율은 나화 난자에 비해 난구 세포 부착 난자에서 유의적으로 감소되었다(35.2% vs. 77.4%; p<0.01). 실험 2에서 다양한 정자 농도에 의한 정자 침투율과 정상 수정률을 바탕으로 판단했을 때 $4.6{\times}10^6/ml$의 정자 농도가 다른 정자 농도에 비해 난구 세포부착 난자의 체외 수정에 적합한 것으로 나타났다. 체외 수정과정에서 난구 세포 부착된 상태로 수정된 난자는 나화 난자에 비해 유의적으로(p<0.05) 높은 분할률(48.8% vs. 58.9%), 배반포 형성률(11.0% vs. 22.8%)과 배반포 세포수$(22{\pm}2\;vs.\;29{\pm}2)$를 나타내었다. 본 연구의 결과로부터 돼지의 체외 수정과정에서 난구 세포의 존재는 정자 침투를 저해하지만 분할률, 배반포 형성률 및 배반포의 세포수를 증가시키는 것으로 사료된다.

본 연구는 lipid peroxides를 생산하는 것으로 알려진 ascorbic acid (Asc; 0.5 mM)와 ferrous sulfate (Fe$^{2+}$; 1 mM)가 돼지 동결-융해 정자의 체외수정능력에 미치는 영향을 검토하였다. 그 결과, 동결정액의 융해 처리시 배양액내 Asc/Fe$^{2+}$ (38%)의 동시첨가시 대조구 (27%)에 비해 정자의 첨체반응이 유의적으로 높게 유기되었다 (P<0.05). 정자침입율 또한 Asc/Fe$^{2+}$ (76%)를 동시에 첨가하므로서 대조구 (55%)에 비하여 유의적으로 높게 나타났다 (P<0.05). 한편 정자의 peroxidation은 상기의 처리구에서 maiondialdehyde (MDA)의 생성에 기초를 두고 평가하였는데, 정자의 처리시 Asc/Fe$^{2+}$의 동시첨가에 의해 MDA의 생성이 증가하였으나 처리구 사이에서 유의적인 차이는 인정되지 않았다. 또한 sulfhydry1(-SH) group의 용량을 측정한 결과 Asc/Fe$^{2+}$를 동시에 첨가한 실험구에서 가장 높게 나타났으나 타처리구와의 사이에서 유의적인 차이는 인정되지 않았다. 또 다른 연구에서 동결-응해정자가 난자의 투명대에 접착하는 정도를 평가한 결과, 대조구와 Asc처리시 Fe$^{2+}$ 처리에 비하여 유의적으로 (P < 0.05) 높은 정자의 접착을 나타냈으나 Asc/Fe$^{2+}$의 동시 처리에 의한 정자접착의 증가는 인정되지 않았다. 결과적으로 lipid peroxidation의 증가를 야기시키는 Asc/Fe$^{2+}$의 동시첨가는 투명대에서 정자의 접착능력을 증가시키지 않았음에도 불구하고 첨체반응과 침입능력을 향상시키는 것으로 나타났다.

본 연구에서는 TCM-199, mSOF, NCSU-23 세 종류의 배지의 자연 단위 발생 유도로 형성된 돼지 배아의 초기 발육에 대한 효과를 조사하였다. 실험 1에서는 세 종류의 배지에서 체외 성숙된 도축장 유래 돼지 난자의 성숙율을 성숙 48 시간째 핵상 관찰로 조사하였다. 각 배지에서의 핵 성숙율은 mSOF군에서 83.1$\pm$2%, NCSU-23 군에서 78.0$\pm$3%, TCM-199군에서 83.5$\pm$2%로 세 배지의 난자 성숙율에 대한 유의적 차이는 없었다(P(0.05). 실험 2에서는 돼지 난자의 전핵 형성률, 단위 발생으로 형성된 배아의 분할율, 6-8세포기까지의 발육율 등이 각각 체외 성숙 55-58 시간, 96시간 그리고 168 시간 후에 관찰되었다. 전핵 형성율 (5.4$\pm$2 % in mSOF and 3.7$\pm$1% in NCSU-23 vs 1.7$\pm$3% in TCM-199) 그리고 6-8cell까지의 발육율은 (3.2$\pm$3% in mSOF and 4.0$\pm$1% in NCSU-23 vs 1.4$\pm$3% in TCM-199) mSOF군과 NCSU-23군에서 유의적으로 높았다(P<0.05). 이상의 결과를 볼 때 mSOF 배지와 NCSU-23 배지에서 장시간 배양에 의한 난자의 단위발생 유도율이 유의적으로 높았다.
