In vitro and in vivo activities of a biocontrol agent, Serratia plymuthica strain A2l-4, was evaluated for the control of Phytophthora blight of pepper, Strain A2l-4 inhibited mycelial growth, germination of zoosporangia and cystospores, and formation of zoospore and zoosporangia of Phytophthora capsici in vitro. In the pot experiment, incidence of Phytophthora blight of pepper in non-treated control was 100% at 14 days after inoculation, while no disease was observed in the plot treated with S. plymuthica A2l-4. In the greenhouse test, infection rate of pepper in the non-treated plots was 74.5%, while it was only 12.6% in the plots treated with A2l-4. Results indicate that S. plymuthica A2l-4 is a potential biocontrol agent for Phytophthora blight of pepper.
Since 1995, boxthorn bight caused by Phytophthora spp. has occurred at Chongyang areas in Chungnam province. Infected plants showed yellows and wilt at early stage, but the symptom rapidly progressed into blight due to the decay of roots and basal stem tissues. The disease was relatively severe in poorly drained lowlands and incidence reached ca. 20% in some fields. Two species of Phytophthora were constantly isolated from freshly infected roots and asal stems. Among 39 isolates collected, 26 were identified as P. nicotianae and 13 as P. drechsleri based on their mycological characteristics. Both fungi showed strong pathogenicity to boxthorn cv. Chongyang No. 1. However, the former expressed stronger pathogenicity than the latter. Phytophthora blight of boxthorn caused by the fungi has not been reported n Korea previously.
Phytophthora crown rot of cymbidium was observed in Cheju island since June of 1996. The disease initiated at the basal portion of infected plant progressed upward to lower leaves. Soon after distinct water-soaking lesions appeared on lower leaves, the plant was wilted, blighted and died. Four orchid farms at Sogwipo out of 16 surveyed in the island were infected by the disease estimating 5~20% infection rates. The causal fungus was identified as P. palmivora based on following distinguishing characteristics. All isolates were heterothallic as A1 types and readily produced chlamydospores with cultural age. Sporangia were conspicuous papillate, ellipsoidal to ovoid, highly deciduous with short pedicels ca. 3~4 ${\mu}{\textrm}{m}$. Koch's rules were satisfied by a pathogenicity test and re-isolation of the fungus from inoculated plants. The pathogen has never been reported in Cheju island previously and its firstly recorded as the cause of Phytophthora crown rot of cymbidium in Korea.
A severe Phytophthora rot of strawberry caused by a species of Phytophthora has been widely occurred at major cultivation areas of Kimhae on August in 1997. Incidence of the disease was obtained in the range of 69.2~83.6% in surveyed 4 fields and showed an average of 75.2%. A species of Phytophthora was mostly isolated from the crown of infected strawberry plants and all the isolates were identified as P. nicotianae var. nicotianae (=P. parasitica). The fungus showed strong pathogenicity on strawberry by inoculation test. As a result of the leaf inoculation using mycelial disks of the fungus, both leaves and petioles were darkly browned, and were finally blighted. As a result of the root inoculation of zoospore suspension, both roots and crowns were rotten with dark brown. Although the fungus produced sporangia either on V-8 juice agar medium or liquid medium, the sporangia observed on the liquid medium appeared to be broadly turbinate and noncaducous. Moreover the fungus cultured on the liquid medium often produced sporangia having two papilla. The number of zoospores in sporangia was found to be ranged from 3 or 4 to as many as 20 or 25. In addition, the released zoospore from the sporangium became the cystospore during the prolonged culture of the fungus. The sporangia were measured as av. 49$\times$35 ${\mu}{\textrm}{m}$ with l/b ratio of 1.43. All isolates from crowns were heterothallic and A1 mating type since oospores were abundantly formed on clarified V-8 juice agar by dual culture with P. capsici A2 mating type. Aplerotic oospores were sized 24-26 ${\mu}{\textrm}{m}$. Antheridia were always amphigynous and recoreded an average of 12$\times$10 ${\mu}{\textrm}{m}$. Hyphal swlling were easily observed, and terminal or intercalary chlamydospores were abundantly formed on V-8 juice agar as well as in C/Z solution and sized av. 28.2 ${\mu}{\textrm}{m}$. This is the first report of Phytophthora rot of strawberry in Korea.

Yuzu (Citrus junos) gummosis disease, caused by Phytophthora nicotianae, was first reported in 1997. As known in citrus, Phytophthora is the most fastidious soil-borne pathogen to control. In order to minimize its damage to Citrus spp., integrated pest management (IPM) approach, including fungicide chemicals and resistant cultivars, is necessary. Therefore, in this study we tried to evaluate tolerance of yuzu cultivars and its related species against yuzu Phytophthora. Trifoliate orange was evaluated as a susceptible host to yuzu Phytophthora by both mycelial growth onto extract media and immature fruit inoculation. However, in zoospores spray-inoculation on 2-year-old cuttings tree, trifoliate orange appeared to have a resistant property as showing less than 6% diseased leaf rate. Among yuzu cultivars only 'Namhae No. 1' appeared resistant property against both P. nicotianae and P. citrophthora. The 'Namhae No. 1' showed 5.7% and 10.6% diseased leaf ratio by P. nicotianae and P. citrophthora, respectively. Clearly, in order to reduce damages caused by two yuzu Phytophthora, we suggest that growers may utilize a trifoliate orange as a rootstock and 'Namhae No. 1' as a scion for fruit production.
Induction of systemic acquired resistance (SAR) against phytophthora blight and pathogenesis-related (PR) protein accumulation by TMV infection in pepper plant (Capsicum annuum cv. Nockwang) were examined to understand the mechanism of the systemic acquired resistance in pepper plant. The zoospore suspension of Phytophthora capsici was inoculated on stem of pepper plant in which TMV-pepper strain had been inoculated on fully expanded upper leaves, and thephytopha blight incidence was examined. Both disease severity and lesion length of phytophthora blight were much smaller in TMV pre-inoculated pepper plant than in uninoculated control plants. The phytophthora blight incidence was decreased about 50% in the TMV pre-inoculated pepper, compared to the uninoculated control plant at 10 days after P. capsici inoculation. Accumulation of PR1 and PR5 proteins in intercellular fluid of TMV-inoculated and uninoculated upper leaves were monitored by immuno-blot with tobacco P1b and PR5a, antibody during induction of SAR. PR1 and PR5 were detected from 24 hours after TMV inoculation in both TMV-inoculated and uninouclated upper leaves, and increased rapidly in TMV-inoculation in uninoculated upper leaves were defoliated. PR5 could be detected upto 20 days after TMV inoculation in uninoculated upper leaves. These results suggest that TMV infection induces SAR against phytophthora blight in pepper plant, and that PR proteins are accumulated very rapidly during induction of SAR and maintained for quite long time in pepper plant.
Phytophthora rot on sword bean, Canavalia gladiata, which has not been reported yet in Korea, occurred in some fields of Jinju in 2003. The disease develops on the basal stem of the plant, but is also often observed on leaves and pods. Rot lesions begin with small dark brown spots and as these are water-soaked, they enlarge rapidly. The magnitude of at the field reached 40%. Abundant sporangia of Phytophthora were formed on the surface of diseased pods and were mummied later. The causal fungus was identified as P. nicotianae with the following mycological characteristics: Sporangium-readily formed in water, papillate, noncaducous, ovoid to spherical, 24-58 (L) ${\times}$ 22-35 (W) in size; Oogonium-spherical, smooth walled, and 22-30; Oospore- aplerotic, spherical, and 18-24; Antheridium- amphigynous, unicellula, and spherical; Chlamydospore- abundant, spherical, and 25-35; Sexuality- heterothallic, and A1 or A2; Optimum growth temperature- about 28$^{\circ}C.$ The fungus showed strong pathogenicity to sword bean. Symptoms similar to those observed in the fields appeared 2 days and 4 days after inoculation with and without wound on pods. This is the first report of Phytophthora rot of sword bean in Korea.
A severe brown rot on peach fruit caused by a Phytophthora sp. has occurred at peach orchards in Taegu of Korea from late June to early August in 1997. Infected fruits showed irregularly round or circular water soaking brown regions. In the severe case, fruits were entirely rotten and surface of the fruits were wrinkled. Occasionally, white mycelia and abundant sporangia were developed on the surface of fruit. Inner tissues of the fruits were also discolored to brown. The causal fungus was identified as Phytophthora cactorum based on following characteristics. Sporangia were ovoid, conspicuously papillate, caducous and measured as 28.4~48.1$\times$21.9~37.2 (av. 39.9$\times$30.4) ${\mu}{\textrm}{m}$. Sexuality of the fungus was homothallic. Oogonia were 25.0~34.0 (av. 29.9) ${\mu}{\textrm}{m}$ in size. Most antheridia were paragynous and measured av. 10.5$\times$13.0 ${\mu}{\textrm}{m}$. Optimum temperature for mycelia growth was around 25~3$0^{\circ}C$. However none of the isolates grew under 7$^{\circ}C$ and over 35$^{\circ}C$. The fungus revealed high pathogenicity to fruits, shoots and leaves of peach, apple and pear with different degrees. Phytophthora fruit rot of peach caused by Phytophthora cactorum has not been reported in Korea previously.
Phytophthora rot on broad bean(Vicia faba) occurred in the experimental field at Gyeongsangnam-do Agricultural Research and Extension Services from 2004 to 2006. The fungus isolated from the diseased plants grew well on potato dextrose agar and showed an arachnoid or rosaceous colony pattern. Sporangia were conspicuously papillated, noncaducous, ovoid to globose, and $25-64{\times}18-44{\mu}m$ in size. Oogonia and oospores were spherical and measured as 20-32 ${\mu}m$ and 16-28 ${\mu}m$ in size, respectively. Oospores were relatively small and aplerotic. Antheridia were amphigynous, spherical, and unicellula. Chlamydospores were globose and 18-40 ${\mu}m$ in size. Optimum temperature for growth was about $28^{\circ}C$ on potato dextrose agar. The disease occurred in all parts of the plant including roots, stems, leaves and pods in the field. The symptoms similar to those of naturally infected plants were induced by artificial inoculation and the pathogen was re-isolated from the plant. On the basis of mycological and pathological characteristics, the causal pathogen of broad bean rot was identified as Phytophthora nicotianae. This is the first report of Phytophthora rot of broad bean caused by P. nicotianae in Korea.
Treatment with antagonistic rhizobactera Burkholderia cepacia strain N9523 or an inducer of resistance DL-$\beta$-amino-n-butyric acid (BABA) effectively inhibited Phytophthora capsici infection on pepper plants in artificially infested pots. Treatment with BABA alone at $1,000\mu\textrm{g}$/ml or together with B. cepacia in combination induced a strong protection from the Phytophthora disease in the greenhouse. In artificially infested field, protection of pepper plants against the Phytophthora epidemic by BABA treatment was maintained at a considerable level. In contrast, soil drench with the antagonist B. cepacia alone, or in combination with BABA did not suppress the Phytophthora epidemic in the field. Mortality of pepper plants caused by P. capsici infection was significantly reduced by treatment with the antagonist Pseudomonas aeruginosa strain 950923-29 and BABA (12-29% plants diseased) relative to the untreated control (41-91% plants diseased) in the naturally infested field. Treatment with the antagonist Ps. aeruginosa strain 950923-29 and BABA also resulted in high levels of protection against Phytophthora blight in pepper plants. In the plastic filmhouse test, the average percentage of plants diseased was significantly low relative to the naturally infested field. Treatment with the antagonist Ps. aeruginosa strain 950923-29 and BABA in combination was most effective in suppressing the Phytophthora disease in field and plastic filmhouse.
