Estimation of the Endogenous Pancreatic/Biliary Zinc Pool and the Effect of Phytate and Calcium on Zinc Homeostasis

  • The pancreas is an important organ in the maintenance of zinc homeostasis. Endogenous zinc is con-tinuously secreted via pancreatic exocrine fluid or to a lesser extent in bile. Much of the endogenous secretion must be reabsorbed to sustain zinc homeostasis. The objective of this study was to estimate the relative size of the pancreatic/biliary zinc pool in comparision to the dietary zinc intake, and to study the effect of the phytate and calcium on the zinc homeostasis using a rat model. At the termination of the experiment, pan-creatic/biliary fluid was collected from the rats. Both radioactivity and total zinc were measured and the relative size of the pancreatic/biliary zinc pool was estimated. To determine the effect of phytate and calcium on zinc homeostsis, dietary zinc intake, the amount of zinc in pancreatic.biliary fluid and fecal zinc excretion were measured. The flow rate of pancreatic/biliary fluid, as corrected for tubing constriction, gives the corrected zinc concentration in the pancreatic/biliary fluid was 2.2 times higher than dietary zinc intake. To maintain zinc homeostasis, zinc absorption/reabsorption was very efficient in the current model; 76%, 88% of absorption/reabsorption for low calcium group and high calcium group 81% for phytate group and non-phytate group, respectively.

Acid Production and Phytate Degradation using a Leuconostoc mesenteroides KC5l Strain in Saccharified-Rice Suspension (현미 당화액에서 Leuconostoc mesenteroides KC51 균주에 의한 산의 생성과 Phytate의 분해)

  • A saccharified-rice was fermented using Leuconostoc(Ln.) mesenteroides KC51 strain in various dry matter (DM) contents (4%, 8%, and 12%) at $30^{\circ}C$ for 18 h. The changes of viable cell number, acid production and phytate degradation in saccharified-rice during fermentation were investigated. The viable cell population of Ln. mesenteroides KC51 was increased rapidly in proportion to DM contents during the 9 h of cultivation. The changes of pH and titratable acidity in saccharified-rice were dependent on DM contents. At high DM content (12%), the viable cell number of Ln. mesenteroides KC51 increased to 9.56 log CFU/g after 6 h of fermentation. The pH and titratable acidity reached to pH 3.38 and 0.93% after 18 h of fermentation, respectively. The phytate, known as an antinutrient factor, in saccharified-rice was degraded by Ln. mesenteroides KC51 cultivation. The decrease of phytate during fermentation approximately coincided with the increase of Ln. mesenteroides KC51 population observed in fermented saccharified-rice. Regardless of DM contents, the levels of phytate were reduced to around 50% of initial concentration.

Degradation of Phytate Pentamagnesium Salt by Bacillus sp. T4 Phytase as a Potential Eco-friendly Feed Additive

  • A bacterial isolate derived from soil samples near a cattle farm was found to display extracellular phytase activity. Based on 16S rRNA sequence analysis, the strain was named Bacillus sp. T4. The optimum temperature for the phytase activity toward magnesium phytate (Mg-$InsP_6$) was $40^{\circ}C$ without 5 mM $Ca^{2+}$ and $50^{\circ}C$ with 5 mM $Ca^{2+}$. T4 phytase had a characteristic bi-hump two pH optima of 6.0 to 6.5 and 7.4 for Mg-$InsP_6$. The enzyme showed higher specificity for Mg-$InsP_6$ than sodium phytate (Na-$InsP_6$). Its activity was fairly inhibited by EDTA, $Cu^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ba^{2+}$ and $Zn^{2+}$. T4 phytase may have great potential for use as an eco-friendly feed additive to enhance the nutritive quality of phytate and reduce phosphorus pollution.

Prognostic Value of $^{99m}Tc-Phytate$ Liver Scintigraphy in Patients with Liver Cirrhosis ($^{99m}Tc-phytate$ 간스캔을 이용한 간 경변증의 예후 평가)

  • To evaluate the prognostic significance of $^{99m}Tc-phytate$ liver scintigraphy in patients with cirrhosis, we measured the grade of extrahepatic uptake (EHU) of $^{99m}Tc-phytate$, from 0 (absent EHU) to 5 (important EHU), according to the relative distribution of the radiotracer among liver, spleen and bone marrow. The results were as follows: 1) EHU score was correlated to the Pugh score (r=0.64) and to survival. 2) The ROC curve on an observed status of death was superior for EHU score. In conclusion, $^{99m}Tc-phytate$ liver scintigraphy nay be a useful prognostic method in patients with liver cirrhosis.

Polyphenol and Phytate Contents and Their Relationship to Antioxidative Activity in Soybeans (콩의 폴리페놀, 피테이트 함량과 항산화능과의 관계)

  • In this study we evaluated polyphenol contents, phytate contents, and antioxidant activities (DPPH radical and hydroxyl radical scavenging activity) in several soybean cultivars. The polyphenol contents of the cultivars ranged from 0.0446 mg/g to 0.0652 mg/g, with the highest level found in the WS82 cultivar, whereas and the lowest in Duyukong. The phytate content of the WS2906 cultivar was the highest at 22.28 mg/g whereas Duyukong showed the lowest level. $IC_50$ values (concentration of soybean extract exerting 50% scavenging of radicals) for DPPH radical and hydroxyl radical scavenging activity showed the highest in WS82. A positive correlation between DPPH radical scavenging activity and polyphenol contents was observed in the soybean cultivars, whereas not significant relationship between antioxidant activity and phytate content was found.

Evaluation on the Usefulness of Alternative Radiopharmaceutical by Particle size in Sentinel Lymphoscintigraphy (감시림프절 검사 시 입자크기에 따른 대체 방사성의약품의 유용성평가)

  • Purpose Sentinel lymphoscintigraphy (SLS) was using only $^{99m}Tc-phytate$. If the supply is interrupted temporarily, there is no alternative radiopharmaceuticals. The aim of this study measure the particle size of radiopharmaceuticals and look for radiopharmaceuticals which can be substituted for $^{99m}Tc-phytate$. Materials and Methods The particle size of radiopharmaceuticals were analyzed by a nano-particle analyzer. This study were selected known radiopharmaceuticals to be useful particle size for SLS. We were divided into control and experimental groups using $^{99m}Tc-DPD$, $^{99m}Tc-MAG3$, $^{99m}Tc-DMSA$ with $^{99m}Tc-phytate$. For in-vivo experiment, radiopharmaceuticals were injected intradermally at both foot to perform lymphoscintigraphy. Imaging was acquired to dynamic and delayed static image and observe the inguinal lymph nodes with the naked eye. Results Particle size was measured respectively Phytate 105~255 nm (81.9%), MAG3 91~255 nm (98.7%), DPD 105~342 nm (77.3%), DMSA 164~ 342 nm (99.2%), MAA 1281~2305 nm (90.6%), DTPA 342~1106 nm (79.4%), and HDP 295~955 nm (94%). In-vivo delayed static image, inguinal lymph nodes of all experiment groups and two control groups are visible to naked eye. however, $^{99m}Tc-MAG3$ of control groups is not visible to naked eye. Conclusion We were analyzed to the particle size of the radiopharmaceuticals that are used in in-vivo. Consequently, $^{99m}Tc-DPD$, $^{99m}Tc-DMSA $are possible in an alternative radiopharmaceuticals of emergency.
