Sensuous Characteristics and Physiological Activity of Cheongtaejeon Tea Produced with the Inoculation of Microbial Strains (균주를 접종하여 제조한 청태전 차의 관능적 특성과 생리활성 효과)

  • This study was conducted to restore our traditional Cheongtaejeon tea and to develop the special products. We inoculated Cheongtaejeon tea with lactobacilli (Lactobacillus plantarum CHO 25) and the mixed microbial strains (L. plantarum CHO 25 + Saccharomyces cerevisiae + Bacillus amyloliquefaciens CHO 104). We also examined the sensuous characteristics and physiological activity of Cheongtaejeon tea which was produced by the inoculation of microbial strains. The external appearance of Cheongtaejeon teas were not significant among the teas which were produced with or without the inoculation of L. plantarum CHO 25 and the mixed microbial strains. The taste of the tea increased most in Cheongtaejeon tea which was produced without the inoculation of microbial strains. The taste and liking of Cheongtaejeon tea which was inoculated with Aspergillus oryzae and Aspergillus niger decreased significantly, and it was not suitable to drink. Total phenolics compound contents, total flavonoid contents and DPPH ($\alpha,\alpha$-diphenyl-$\beta$-picryl-hydrazyl) radical scavenging activity of Cheongtaejeon tea extracts increased much more in the order of that produced with the inoculation of L. plantarum CHO 25, control and that with the mixed microbial strains. However, nitrite radical scavenging activity in 1,000 mg/L Cheongtaejeon tea hot water extracts were in the order of the control (94.4%), the inoculation of L. plantarum CHO 25 (93.6%) and the mixed microbial strains (91.1%). Overall results indicated that the sensuous characteristics increased most in Cheongtaejeon tea which was produced without the inoculation of microbial strains, and those physiological activities in tea with the inoculation of L. plantarum CHO 25.

Physiological Characteristics of Lactobacillus casei Strains and Their Alleviation Effects against Inflammatory Bowel Disease

  • Lactobacillus casei, one of the most widely used probiotics, has been reported to alleviate multiple diseases. However, the effects of this species on intestinal diseases are strain-specific. Here, we aimed to screen L. casei strains with inflammatory bowel disease (IBD)-alleviating effects based on in vitro physiological characteristics. Therefore, the physiological characteristics of 29 L. casei strains were determined, including gastrointestinal transit tolerance, oligosaccharide fermentation, HT-29 cell adhesion, generation time, exopolysaccharide production, acetic acid production, and conjugated linoleic acid synthesis. The effects of five candidate strains on mice with induced colitis were also evaluated. The results showed that among all tested L. casei strains, only Lactobacillus casei M2S01 effectively relieved colitis. This strain recovered body weight, restored disease activity index score, and promoted anti-inflammatory cytokine expression. Gut microbiota sequencing showed that L. casei M2S01 restored a healthy gut microbiome composition. The western blotting showed that the alleviating effects of L. casei M2S01 on IBD were related to the inhibition of the NF-κB pathway. A good gastrointestinal tolerance ability may be one of the prerequisites for the IBD-alleviating effects of L. casei. Our results verified the efficacy of L. casei in alleviating IBD and lay the foundation for the rapid screening of L. casei strain with IBD-alleviating effects.

Sexual Differences of Cocoon weight, Cocoon Shell Weight, and Cocoon Shell Percentage in the Sex- limited Silkworm Strains, Bombyx mori L., (한성계통의 전견중, 견층중, 견층비율에 대한 성차)

  • Using sexual differences between female and male as the indication of the degree of the physiological distrubance in the sex-limited larval marking strains or sex-limited egg colour strains, it was investigated whether the physiological distrubance caused y the translocated autosome fragment on the W-sex determination chromosome, have an effect on expression of the three quantitative characters; cocoon weight, cocoon shell weight, and cocoon shell percentage in the female silkworm. The mean values of the ratios of female to male the above two experimental groups were 127%, 107%, 85%, in cocoon shell weight, and cocoon shell percentage, respectively. On the other hand, those from the three normal stains, namely the basic stock strains, white egg strains, and hybrids were 129% for cocoon weight, 110% for cocoon shell weight, and 85% for cocoon shell percentage, respectively. From the results, it comes to the conclusion that the translocated autosome fragment on the W-chromosome has no influences on the expression of the quantitative characters of the female silkworm because sexual differences of the sex-limited strains were very similar to those of the three normal strains.

  • Intensive taxonomical studies of the Aspergilli have long been made. Altogether 132 species and 18 varieties are recognized in the book "The Genus Aspergillus" written by Raper and Fennell (1965), in contrast to 77 species, 8 varieties and 4 mutations in " A Manual of Aspergilli" written 20 years earlier by Thom and Raper (1945). Classification of the Asperilli by Thom and Raper (1945) and by Raper and Fenell (1965). Classification of the Aspergilli by Thom and Raper (1945) and by Rapher and Fenell (1965) have been based mainly upon morphological and cultural detail both physiological and biochemical activities. In Korean there are many kinds of foods fermented natrually without the employment of selected microorganisms, and there are, of course, many different microorganisms serving in the fermentation fermented foods than other countries, the distribution and biological properties of the Asperigilli, in Korea are more variable. Taxonominical studies with 36 strains of Asperilli were based upon the examinations of morphological, cultural, and physiological characteristics. nineteen strains indigenous to Korea were selected from a lot of strains which had been isolated from meju and kokja and one strain from soil. They were identified according to the group key of Raper and Fennell. Ten strains were donated by Dr.Hesseltine of the Northern Utilization Research and Development Division in the U.S.A. From the Asp. japonicus supplied by Dr.Hesseltine, a white mutant was isolated and also studied. Two strains were donated by Dr. Murakami of the Research Institute of Brewing in Japan, and four strains came from Korean industrial companies.ndustrial companies.

Isolation and Identification of Lactobacilli from Fermented Sausages (발효소시지로부터 유산생성균의 분리 및 동정)

  • Lactobacilli proliferating in fermented sausages of the specific ripening conditions were isolated from fermented sausages, manufactured in the absence of an added starter, during ripening under controlled temperature-humidity conditions. Based on morphological, physiological and bio- chemical characteristics and carbohydrate fermentation of isolated strains, three strains of isolates were identified as Lactobacillus curvatus, two strains as Lactobacillus sake. Optimal temperature and pH for growth of isolated strains were 30$\circ$C and pH 6.0~7.0, respectively. These strains were salt tolerant, multiplying in the presense of 6~8% NaCl.

  • In the previous paper, two strains of cellulase producing microorganism were isolated from night soil samples using shaking culture. This report deals with the physiological and morphological tests carried out according to the methods of Toyama and Chung. 1. Two strains, Ku-3371 and Ku-4383 which should the best growth in Czapek.s liquid medium, were identified as Trichoderma viride. 2. These strains grew the best at about $30^{\circ}C$ and the optimum pH values of growth in Czapek's liquid medium was 4.0. 3. These mould strains utilized monosaccharide as its carbon source and can utilized sodium glutamate, peptone and nitrate form nitrogen but other inorganic nitrogen compoumd are unable to use as its nitrogen source.

김치에서 분리한 유산균의 인공위액과 담즙에서의 생존특성과 항균성

  • This studies were carried out to investigate the physiological characteristics of lactic acid bacteria related to kimchi fermentation. 157 strains of lactic acid bacteria were isolated from home-made kimchi of Taegu district. 144 strains of isolated lactic bacteria showed resistance to artifical gastric juice and bile. Among isolated strains, fifteen strains were selected according to the growth characteristics. The selected strains survived for 24 hours in artifical bile after incubation for 3 hours in artificial gastric jucie (pH 2.5). Especially, three strains such as B-10, K-2, and F-3, showed little viable cell change for 3 hrs incubation in artifical gastric juice. The selected strains showed low viability in artificial gastric juice but high in artifical bile. The selected strains also showed significantly antimicrobial activities to pathogens such as Listeria monocytogenes, Escherichia coli, Bacillus subtili, Staphylococcus aureus and Vibrio parahaemolyticus. The fifteen selected strains were identified as Lactobacillus plantarum, Lactobacillus sake and Leuconostoc mesenteroides.

Flowers in Islands and Mountains of Korea

  • Yeasts were isolated from wild flowers of some islands and mountains such as Jeju-do, Ulleungdo, Yokjido, Seonyudo and Gyejoksan, Oseosan, Beakamsan and Deogyusan in Korea and were identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA or internal transcribed pacer(ITS) 1 and 2 including 5.8S rDNA using BLAST. Seventy two yeast strains of two hundred eighty nine species were isolated from wild flowers in islands and mountains, Korea. Among them, Cryptococcus species were isolated the most dominantly, and Metschnikowia reukaufii were also isolated thirty species, 10.3% of total strains. Twenty-three species including Cryptococcus aureus were overlapped between yeast strains of the islands and mountains. Some physiological functionality of the culture broth and cell-free extracts from two hundred eighty nine yeast strains were determined. The supernatant of Candida sp. 78-J-2 showed antioxidant activity of 22.5%, and supernatant of Metschnilowia reukaufii SY44-6 showed anti-gout xanthine oxidase inhibitory activity of 49.6% and whitening tyrosinase inhibitory activity of 38.4%, respectively.

Isolation and Identification of Staphylococcus sp. from Korean Fermented Fish Products

  • In order to find out if staphylococci occur in significant numbers in Korean fermented fish products, a total of 40 different fermented fish products were collected from different markets in Korea and analyzed for their physico-chemical and microbiological states. The pH, salt concentration and water activity of the products were measured and the total viable cell count and the number of Staphylococcus grown on mannitol salt agar were determined. The identification of the strains of Staphylococcus were made by API Staph Strip and MIS identification kits, and the physiological properties of the identified strains were further characterized by different conventional methods. The pH, salt content and water activity of fermented fish samples varied widely from 4.8 to 7.1, 7.4-28.7$%$ and 0.77-0.84, repectively, depending on the type of product. The total viable cell count varied from $10^4-10^9$ cfu/ml, and most of the samples had $10^5-10^6$ cfu/ml No correlation was found between the viable cell count and the pH, NaCl concentration and water activity of the samples. Among the 35 colonies identified as Staphylococcus strains by the identification kits, S. xylosus was the most frequently occurring strain marking 17, and S. warneri was 8, S. epidermidis 4 and S. cohnii 2. S. hominis, S. saprophyticus, S. haemolyticus and S. aureus were also identified once each. In some samples (K-3, P-6, K-8, G-5 and G-10), 2-3 different species of Staphylococcus were found. Considering the region of sampling, among the 10 samples from Kunsan 5 were identified as S. warneri, while in the other regions S. xylosus was predominant. Although the physiological characteristics of the identified strains were generally consistent with those in Bergey's Manual, some discrepances were also observed. All the strains were highly salt tolerant, growing in the media containing over 18$%$ NaCl. All the strains except S. aureus (G-11) showed negative in hemolysis activity, plasma coagulation and DNase tests. All the strains including S. aureus (G-11) showed negative in enterotoxin test.

Isolation and characterization in the exhausted mine and Jeju Gotjawal (국내 폐광산 및 제주 곶자왈 지역내의 미생물 분리 및 특징 분석)

  • Most of acidophiles are found in the various low pH environments and affect to metal cycle through oxidation and reduction reactions. The present study was carried out above 50 strains as acidophiles isolated from acidic soils of exhausted mine and Jeju Gotjawal. Finally, total 19 strains obtained and were tentatively identified based on comparative similarity analysis for 16S rRNA gene sequence and physiological characterizations. These isolates belonged to Gammaproteobacteria (6 strains), Actinobacteria (5 strains), Betaproteobacteria (4 strains), Alphaproteobacteria (2 strains), and Bacilli (2 strains). We observed that these isolates can grow under low pH culture condition. This case study for analysis physiological characterizations of indigenous microorganisms in acidic soil might provide basic information on useful application.
