Effect of Soyangin-Hyeongbangpaedok-san on Anti-CD3 Stimulated Mouse T Cells In Vivo and In Vitro (소양인 형방패독산(荊防敗毒散)의 마우스 T 세포 활성에 대한 실험적 연구)

  • Soyangin-Hyeongbangpaedok-san(SHBPDS) is a herbal formula used for the common cold or upper respiratory illness. In order to investigate the effect of SHBPDS, mice were orally administered with SHBPDS alcohol extract for 7 days followed by intravenous anti-CD3 injection. In addition, splenocytes and CD4 T cells were cultured with SHBPDS in response to anti-CD3 in vitro and cytokines and transcription factors were evaluated. In vivo treatment with SHBPDS significantly augmented the expressions of the percentage of CD4 T cells and CD 69, an indicator of early T cell activation. Serum levels of IL-4 were significantly increased but those of IFN-${\gamma}$ and IL-2 did not reach statistical significance. The expressions of IFN-${\gamma}$ and T-bet mRNA were significantly downregulated in SHBPDS treated mice while those of IL-4 and C-Maf were significantly upregulated. In vitro stimulation of splenocytes and CD4 T cells by SHBPDS resulted in a reduction in IFN-${\gamma}$ secretion and STAT4 activity. The IL-4 releases from both cells were slightly reduced, but STAT6 activity was rather increased. In conclusion, SHBPDS exerted an inhibition in the expression of IFN-${\gamma}$, T-bet and STAT4 while IL-4, C-Maf and STAT6 were increased. Further studies are required to examine its pharmacological effects using more appropriate animal experiments.

Relationship between the Lipid Accumulation Product Index and Alanine Aminotransferase in Korean Adult Men (한국 성인남성에서 Lipid Accumulation Product 지수와 Alanine Aminotransferase와의 관련성)

  • The LAP (lipid accumulation product) index is an indicator of excessive fat accumulation in combination with the fasting triglyceride concentration and waist circumference. This study examined the relationship between the LAP index and ALT (alanine aminotransferase) level and the effect of the LAP index on the ALT level. Cross-sectional studies were conducted on healthy adult males without a history of liver disease. From January 2015 to June 2017, 13,854 adults between 20 and 70 years of age, who underwent health screening at a general hospital in Gyeonggi-do, were enrolled in this study. The LAP index was calculated as [waist circumference (cm)-65]${\times}$[triglyceride (mmol/L)] for males. Serum ALT abnormalities were set at 40 IU/L or more in males. All subjects underwent measurements of the anthropometric indicators and physiological examinations. A significant increase in ALT with increasing LAP quintiles was observed (p<0.001). The LAP index correlated with ALT (r=0.238, p<0.001). Insulin (r=0.449, p<0.001) and HDL-cholesterol (r=-0.369, p<0.001) were strongly correlated with LAP. In addition, the LAP index is a factor affecting the ALT level (p<0.001). As the LAP index increased, the risk of an elevated ALT was higher (p<0.001). In conclusion, the LAP index was a predictor of an ALT elevation in Korean men.

  • The purpose of this study is to investigate clinical frequency and priority of five pattern identification settled by Korean Standard Differentiation of the Symptoms and Sign for Stroke. The present study was done over 177 hospitalized patient with stroke in the Daejeon University Oriental Medical Hospital in the period of November 2006 to July 2007. Stroke patients had been interviewed by residents and specialists who studied standard operation procedures in Fundamental study for Standardization and Objectification of Differentiation and Pattern Identification of Syndrome of Oriental Medicine for Stroke. In Heat-transformation pattern group, men showed significantly high frequency and in Deficiency of Gi pattern group, women showed significantly high frequency. In Heat-transformation pattern group, the indicators such as 'aversion to heat during fever', 'flushed face', 'full and rapid pulse', 'dryness of the eyes, 'yellow coating of the tongue', 'feverishness of the limbs', 'dryness of the eyes' showed significantly high frequency. In Dampphlegm syndrome group, the indicators such as 'white coating of the tongue', 'yellowish face', 'thick coating of the tongue', 'wheezing in the throat with sputum', 'swollen tongue', 'slippery pulse' showed significantly high frequency. In Deficiency of Gi pattern group, the indicators such as 'pale tongue', 'lassitude', 'pale face', 'weakness pulse' showed significantly high frequency. In Deficiency of Eum group, the indicators such as 'short and rapid pulse', 'mirror-like tongue' showed significantly high frequency. For more sensitive Korean Standard Differentiation of the Symptoms and Signs for Stroke, Large scale study is to be done, giving weight on the important indicators.

Anti-obesity effects of cultivated ginseng, -wild simulated ginseng and -red ginseng extracts (인삼, 산양산삼 및 홍삼 추출물의 항비만 효과)

  • This study was to investigate whether cultivated ginseng (CG), cultivated wild simulated ginseng (CWG) and cultivated red ginseng (CRG) extracts influences on the obesity. The saponin contents of 3 kinds of ginsengs were analysed by HPLC-ESI-TOF-MS. Total saponin contents were determined in CG on the most contents and since red ginseng has the highest PD (protopanaxadiol type) / PT (protopanaxatriol type) ratio, there may be differences between ginseng, wild ginseng, and red ginseng with respect to their pharmacological effects. Male C57BL/6J mice were fed a normal diet(N), HFD (60% Kcal fat, C), HFD with CG, CWG and CRG extracts (800 mg/kg) for 5 weeks. We observed change of total body weight, degree of hepatic lipid accumulation and immunohistochemical change of GLP-1 and insulin-secreting cells. Also this study attempts to use the physiological analysis method to analyze the changes of blood lipids, insulin and leptin concentration. The change of body weight and size of accumulated lipid droplets in liver lobules decreased in all of the experimental groups than the control(C) group. In the pancreas, the immunohistochemical density of insulin-secreting cells were significantly stronger in the CWG and CRG than C group. The levels of serum insulin and leptin significantly decreased 55.6%, 54.3% respectively in CWG and CRG. The changes of triglyceride, total cholesterol in serum decreased in CRG than the C group. Obesity related CG, CWG and CRG extracts might have contribute to improvement of obesity by regulating the levels of blood lipids and biochemical indicator of fat accumulation.

The Effects of Milking Time on Melatonin and Cortisol Concentrations in Raw Milk and Milk Powder during the Summer and Winter Solstice (계절 및 착유시기에 따른 원유와 분유 내 멜라토닌, 코티솔 농도 변화)

  • Melatonin is a hormone produced by the pineal gland in dark conditions. It plays a major role in the regulation of the sleep-wake cycle. Melatonin synthesis is known to be suppressed by environmental light. Cortisol is a steroid hormone that is a major indicator of physiological alterations due to stressful stimuli. It also displays a circadian rhythm, like melatonin. The highest levels are encountered during early morning and the lowest levels are observed at around midnight. In the present study, the effects of milking time on the melatonin and cortisol concentrations of raw milk and milk powder at the summer and winter solstices were examined. The melatonin concentration in milk increased significantly if cows were milked in the dark at night (p<0.05). The melatonin concentration in milk powder showed the same pattern with respect to the milking time (p<0.05). However, no significant difference in the cortisol concentration was observed between day- and night-time milk. Although the time of day did not affect the level of milk cortisol, seasonal factors affected the release of cortisol in milk (p<0.05). In conclusion, night-time milk is rich in endogenous melatonin. In this respect, it has potential applications for the development of melatonin rich-dairy products, which serve as natural sources of melatonin.

Larger Testicular Volume Is Independently Associated with Favorable Indices of Lung Function

  • Background: Men with chronic obstructive pulmonary disease, have reduced endogenous testosterone levels, but the relationship between pulmonary function and endogenous testosterone levels, is inconsistent. Testicular volume is a known indicator of endogenous testosterone levels, male fertility, and male potency. In the present study, the authors investigated the relationship, between testicular volume and lung function. Methods: One hundred and eighty-one South Korean men age 40-70, hospitalized for urological surgery, were retrospectively enrolled, irrespective of the presence of respiratory disease. Study subjects underwent pulmonary function testing, prior to procedures, and testicular volumes were measured by orchidometry. Testosterone levels of patients in blood samples collected between $7{\small{AM}}$ and $11{\small{AM}}$, were measured by a direct chemiluminescent immunoassay. Results: The 181 study subjects were divided into two groups, by testicular volume (${\geq}35mL$ vs. <35 mL), the larger testes group, had better lung functions (forced vital capacity [FVC]: $3.87{\pm}0.65L$ vs. $3.66{\pm}0.65L$, p=0.037; forced expiratory volume in 1 second [$FEV_1$]: $2.92{\pm}0.57L$ vs. $2.65{\pm}0.61L$, p=0.002; FVC % predicted: $98.2{\pm}15.2%$ vs. $93.8{\pm}13.1%$, p=0.040; $FEV_1$ % predicted: $105.4{\pm}19.5%$ vs. $95.9{\pm}21.2%$, p=0.002). In addition, the proportion of patients with a $FEV_1/FVC$ of <70%, was lower in the larger testes group. Univariate analysis conducted using linear regression models, revealed that testicular volume was correlated with FVC (r=0.162, p=0.029), $FEV_1$ (r=0.218, p=0.003), $FEV_1/FVC$ (r=0.149, p=0.046), and $FEV_1$ % predicted (r=0.178, p=0.017), and multivariate analysis using linear regression models, revealed that testicular volume was a significant predictive factor for $FEV_1$ % predicted (${\beta}=0.159$, p=0.041). Conclusion: Larger testicular volume was independently associated, with favorable indices of lung function. These results suggest that androgens, may contribute to better lung function.

Study of 4 Cases with Changes of Unified Parkinson's Disease Rating Scale, Heart Rate Variability and Quality of Life in Parkinson's Disease Patients through Whole Body Gi-Hyeol Therapy (전신기혈요법 치료를 통해 Unified Parkinson's Disease Rating Scale, Heart Rate Variability 및 삶의 질이 변화된 파킨슨 환자 4례에 대한 증례보고)

  • Parkinson's disease is one of the typical neurodegenerative disease and it is caused by the destruction of substantia nigra in brain leading to lack of dopamine secretion, and it presents 4 major motor symptoms such as tremor, bradykinesia, stiffness, postural instability. Furthermore, it causes many non-motor symptoms such as anosmia, REM sleep conduct disorder, orthostatic hypotension, dementia and autonomic ataxia such as lack of adjusting blood pressure, hyperhydrosis, constipation. Dopaminergic therapy is the most commonly used strategy, but long term treatment of levodopa induce various adverse effects. Thus, many people are focusing on new therapies other than established therapies, and there are many tries and approaches with paradigm shift. Our medical team was able to get 4 cases of PD patients who are hospitalized in our hospital, treated by Whole Body Gi-Hyeol Therapy consisting of acupuncture therapy, herbal therapy, and mental therapy, and their conditions improved in perspective of Unified Parkinson's Disease Rating Scale(UPDRS), Heart Rate Variability(HRV), and Quality of life. Among all 4 cases, UPDRS score and quality of life score is gotton better, and among 2 cases SDNN, RMS-SD, TP, LF, HF scores are finely increased. And PDQ-39 score which shows quality of life is also improved. However, in spite of these improvements and positive results, there were no meaningful improvement in a hurt from a fall which is important to the aged, muscular atrophy which causes bone fracture and SMI(Skeletal Muscle Mass Index) which is indicator of osteoporosis. Thus, supplementary treatment about Whole Body Gi-Hyeol Therapy such as more active nutrition intervention, safe and effective kinesitherapy is needed, and from now on continuous case reports and systematic clinical research which has control group must be carried out.

Ecological Characteristic of Clithon retropictus inhabitating in Yeoncho River in Southern Coastal Area (남해안 연초천에 서식하는 기수갈고둥의 생태적 특성 연구)

  • Clithon retropictus has been designated as an endangered wildlife Class II due to its high value as a biological indicator species capable of judging environmental quality such as salinity, water flow, and ground conditions. However, basic research on its physiological and ecological characteristics is still lacking. As such, this study intended to examine the impact of environmental conditions such as salinity and soil particle size on the size and density of Clithon retropictus at the Yeoncho river estuary. The investigation of the salinity, which is a key variable that affects the distribution of organisms in the estuary, showed that Clithon retropictus could grow at a salinity ranging from 0#x2030; (freshwater) to 25‰ (brackish water). The coarse gravel (19-75mm) tended to increase nearer the upper stream (under the Yeoncho weir), while the proportion of particles smaller than sand (less than 19mm) increased toward the downstream. The population and the size of the individuals decreased rapidly in the downstream where water stagnated near the Yeoncho weir, and the salt water joined. The results indicated that Clithon retropictus had a high tolerance to salinity, but the adaptability was weaker toward the extremes since the population, and the size tended to decrease as the salinity increased. The correlation analysis revealed that both salinity and soil particle size affected the population and individual size. The correlation between the individual size and salinity was -0.242 (P <0.01), indicating that the size decreased with increasing salinity. The correlation between individual size and coarse gravel having a particle size of 19mm or more was 0.420 (P <0.01), indicating that the size increased with increasing the particle size.

Photosynthesis and Chlorophyll Fluorescence of Evergreen Hardwoods by Drying Stress (건조 스트레스가 난대 상록활엽수의 광합성 반응 및 엽록소 형광반응에 미치는 영향)

  • This study was carried out to investigate the effects of C. japonica, D. morbifera, D. macropodum, I. anisatum, Q. glauca and R. indica To investigate the photosynthetic ability, chlorophyll content, chlorophyll fluorescence analysis, and physiological environmental. The photosynthetic rate, cancer respiration rate, stomatal conductance, and rate of evaporation tended to decrease as a result of drying stress in the no-water condition for 28 days. I. anisatum, Q. glauca and R. indica showed a low rate of less than 40% until 28 days of no-treatment. The total chlorophyll contents were decreased in the order of D. macropodum> D. morbifera> C. japonica> Q. glauca> M. thunbergii> R. indica> I. anisatum. Chlorophyll fluorescence analysis showed that there was no change in the qP, but after 28 days no $Fv/F_m$, $F_o$, $R_{fd}$, $NPQ_{_-LSS}$ can be a useful indicator for quantitative estimation within a short period of time with a marked reduction rate of PSII quantum yield ${\Phi}PSII$ in the rectified state by continuous light during the nominal adaptation period. In the case of I. anisatum, Q. glauca and R. indica If water management can be carried out at intervals, it may be possible to plant trees in trees and landscape trees.

  • Human intestinal microbiota play an important role in the regulation of the host's metabolism. There is a close pathological and physiological interaction between dysbiosis of the intestinal microflora and obesity and metabolic syndrome. Akkermansia muciniphila, which was recently isolated from human feces, accounts for about 1-4% of the intestinal microbiota population. The use of A. muciniphila- derived external membrane protein Amuc_1100 and extracellular vesicles (EVs) could be a new strategy for the treatment of obesity. A. muciniphila is considered a next-generation probiotic (NGP) for the treatment of metabolic disorders, such as obesity. Faecalibacterium prausnitzii accounts for about 5% of the intestinal microbiota population in healthy adults and is an indicator of gut health. F. prausnitzii is a butyrate-producing bacterium, with anti-inflammatory effects, and is considered an NGP for the treatment of immune diseases and diabetes. Postbiotics are complex mixtures of metabolites contained in the cell supernatant secreted by probiotics. Parabiotics are microbial cells in which probiotics are inactivated. Paraprobiotics and postbiotics have many advantages over probiotics, such as clear chemical structures, safe dose parameters, and a long shelf life. Thus, they have the potential to replace probiotics. The most natural strategy to restore the imbalance of the intestinal ecosystem normally is to use NGPs among commensal bacteria in the gut. Therefore, it is necessary to develop new foods or drugs such as parabiotics and postbiotics using NGPs.
