금강 하구 퇴적토에서의 종속영양 세균의 분포에 미치는 환경요인과 계절적 우점속에 관하여 (Environmental factors affecting seasonal destribution of heterotrophic bacteria in Kum river estuarine sediments)

  • Dominant genera of heterotrophic bacteria were investigated seasonally in Kum River estuarine sediments. Numerical taxonomy on the isolated heterotrophic bacteria were carried out according to their morphological, physiological, nutritional and biochemical characters. Heterotrophic bacteria in the depths of 0 to 10 cm of sampled sediments were almost facultative anaerobes, and 77.9% to 86.9% of them were Gram negative rods. Kominant genera among the isolates were Pseudomonas, Flavobacterium, Micrococcus, Cnterobacteria and Vibrio during sampling periods. According to their character analyses by similarity index(S), the isolates were clustered in fourteen groups at clay type sediments (Site 1) and four groups at sand type sediments (Site 2).

  • 금강호에서 이화학적 수질과 미생물학적 수질을 월별로 측정하고, 조시기간 중 분리된 60균주에 대하여 16S rDNA를 증폭하고 부분석인 염기 서열 분석을 통하여 계통분류학적 분석을 하였다. 측정 결과 조사기간 중 수온은 $2\sim30.8^{\circ}C$, pH $3.9\sim9.14$, DO는 $5.04\sim14.95\;mg\;l^{-1}$의 범주에서 변화하였다. BOD와 무기영양염류($NH_4$-N, $NO_2$-N, $NO_3$-N, $PO_4$-P)의 농도는 금강 중류지역에 비해 비교적 낮은 값을 나타냈다. 종속영양세균과 총대장균군의 균체수 변화는 각각 $4.1{\pm}1.0{\times}10^2\sim6.7{\pm}1.1\times10^3\;cfu\;ml^{-1}$$0{\sim}2.3{\pm}0.6{\times}10^2\;cfu\;ml^{-1}$의 범주에서 변화하였다. 생리적 특성균으로 단백질 분해 세균, 전분 분해 세균, 지방분해 세균 및 셀룰로스 분해 세균의 분포를 측정하였다. 조사기간 중 셀룰로스 분해 세균이 다른 생리적 특성균에 비해 비교적 높은 값을 나타냈다. 분리된 60 균주는 16S rDNA 분석 결과 우점속은 Pseudomonas 속 20 균주로 나타났다.

  • 낙동강 하구 생태계의 종속영양세균의 특성과 이에 미치는 환경요인의 영향을 조사하였다. 해수역에 위치한 정점에서 분리한 세균군집이 담수역에서 분리한 군집보다 광염성 세균으로 이루어져 있으며, 하계에 분리한 군집이 동계군집에 비해 광온성세균으로 구성되어 있었다. 또한 수계의 세균군집은 육상에서 유입되는 비토착성세균에 의해 다양성이 커지며, 환경이 악화되면 세균군집의 다양성이 감소하는 현상이 관찰되었다.

천수만 해역 세균 군집의 수리학적 분석 (Numerical Analysis of Bacterial Community in Cheonsu bay)

  • 서해안의 대표적 내만인 천수만 해역에서 4계절간 분리한 세균들을 대상으로 수리학적 분류를 설시하였다. 총 48가지의 형태학적, 생리적, 생화학적 실험 결과 분리된 계절에 따라 온도에 대한 서로 다른 적응 양상을 보여 주었으며, 전체적으로 다양한 환경변화에 높은 생존 능력을 보여주였다. 각 세균을 동정한 결과 Enterobacteriaceae, Aeromonas Pseudomonas와 Vibrio 등이 우점종으로 나타났으며, 계절별로는 봄 가을에 Aeromonas가, 여름 및 겨울에는 각각 Pseudom monas 와 Enterobacteriaceae가 제l우점종임이 발견되었다 Cluster analysis 결과 젠체 세규 군집은 75%의 유사도 수1 에서 총 29개의 phenetic group으로 나뉘어졌으며 각 group별로 계절적 구분이 뚜렷하였다. 전체적으로 계절에 놔른 서로 다른 생리적 특성과 종 구성이 전반적인 생태계에의 다양성과 안정성을 높여주는 것으로 나타났다.

한국 연안 해역에서 분리한 3-chlorobenzoate 분해 세균에 대한 수리학적 분류 (Numerical taxonomy of 3-chlorobenzoate degrading bacteria isolated from Korean coastal waters)

  • The bacteria utilizing 3-chlorobenzoate as a sole carbon and energy source were isolated by Most-Probable-Number technique and identified in Korean coastal waters. Pseudomonas, Moraxella and Flavobacterium were dominant genera and comprised 70.2% of total isolates. Forty-four biochemical, cultural, morphological and physiological testa were performed and average linding cluster analysis was conducted from the test results. Total 92.7% of isolates were clustered into 17 groups under the 80% similarity level. The degradation of 3-chlorobenzoate was performed by many heterogeneous bacteria and the species diversity of these bacterial group offers informations of the stability of bacterial communities in relation to carbon compound cycling in coastal enviromnents.

고지방식이에 의한 비만 동물모델에서 유산균 마분말의 항비만 효과 (Anti-Obesity Effects of Lactic Acid Bacteria-Fermented Dioscoreae Rhizoma Powder on High Fat Diet-Fed Animal Model)

  • The purpose of this study was to examine the antiobesity effects of lactic acid bacteria-fermented Dioscoreae Rhizoma powder (LDR) in Sprague-Dawley rats with high-fat diet (HFD)-induced obesity. Rats were divided into 5 groups: normal diet group, control (high fat diet-vehicle) group, 0.4 g/kg LDR (high fat diet + 0.4 g/kg lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group, 1 g/kg LDR (high fat diet + lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group, 2.5g/kg LDR (high fat diet + lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group. Our results indicate that LDR administration has effects on decreasing of body weight, fat weight, blood parameters and adipocyte size in the obesity animals. In addition, verified that fat degeneration and ballooning degeneration were alleviated in the experimental group fed on a high fat diet combined with LDR groups. These results indicate that lactic acid bacteria-fermented Dioscoreae Rhizoma powder may reduce elevating body weight and lipid accumulation in rat fed a high fat diet, suggesting its usefulness as a functional food for reducing body fat and obesity.

Genetic and Physiological Characterization of Oxytetracycline-Resistant Bacteria from Giant Prawn Farms

  • Four hundred and thirteen oxytetracycline-resistant bacteria were recovered from six freshwater giant prawn farms with a history of oxytetracycline use. Most oxytetracycline-resistant isolates were Gram-negative bacteria. Six groups of oxytetracycline-resistant bacteria were classified using cluster analysis based on a comparison of levels of oxytetracycline resistance. Complex fingerprint patterns were obtained for 71 isolates studied. In general, the band patterns of isolates from different ponds were very similar, and the data indicated that the isolates were closely related. The exploration for cross-resistance found that most of the 71 oxytetracycline-resistant isolates were also resistant to tetracycline and chlortetracycline, but had a relatively low resistance to doxycycline. Many isolates showed higher chlortetracycline resistance than oxytetracycline resistance. Additionally, the oxytetracycline-resistant isolates were examined for the presence of tetracycline resistance (tet) genes. Fifty percent of the isolates carried one of the 14 known tet genes examined. The most common determinants were TetA and TetD. However, TetB, TetC, TetE, TetK, TetL, and TetM were also found with various frequencies.

삼림토양내(森林土壤內) 저영양세균(低營養細菌)의 형태(形態) 및 생리적특징(生理的特徵)에 관(關)한 연구(硏究) (Studies on Morphological, Physiological Characteristics for Oligotrophnic Bacteria from Forest Soil)

  • 너도밤나무 삼림토양(森林土壤)의 각 층에는 저영양세균(低營養細菌)이 다수(多數) 존재(存在)해 있었다. 각 토양층으로 부터 분리(分離)한 저영양세균(低榮養細菌) 203주를 갖고 형태적(形態的), 생리적성질(生理的性質)을 검토한 결과 세포형태에 의해 크게 4개의 Group으로 대별되었다. (1) regular rods (73주), (2) curved/spiral rods (29주), (3) irregular rods (56주), (4) appendaged rods (45주)였다. 이와같이 저영양세균(低營養細菌)은 세포형태적(細胞形態的)으로 다양(多樣)한 세균의 집합이며, 그중에는 새로운 형태의 세균도 포함되어 있었다. 이들 각 Group에 속하는 균주의 생리적특질(生理的特質)에도 각각 특징(特徵)이 보여졌다. 그중에는 리그닌의 단위구조물질(單位構造物質)인 feruric acid, p-coumaric acid를 이용하는 균, methanol을 이용하는 균 및 질소고정능(窒素固定能)을 갖는 균 등 흥미(興味)있는 성질을 갖는 세균이 포함되어져 있음을 밝혔다.

마황윤폐탕(麻黃潤肺湯)과 Ciprofloxacin의 병용(倂用)이 호기성 Gram(-) 세균주(細菌株)에 대한 시험관내(試驗管內) 항균력(抗菌力)에 미치는 영향 (Synergic Effect of Mahwangyounpae-tang and Ciprofloxacin on 5 Strains of Aerobic Gram-negative Bacteria)

  • In order to evaluate the in vitro synergic effect of Mahwangyounpae-tang which was a traditional poly-herbal formula has been used in the treatment of respiratory diseases in oriental medicine, and quinolone antibiotics, ciprofloxacin (CPFX), the minimal inhibitory concentration (MIC), $MIC_{50}$ and MIC90 of single use of quinolones and concomitant treatment with Mahwangyounpae-tang against 5 strains of aerobic gram negative bacteria, Escherichia coli, Klebsiella peumoniae, Hemophilus influenzae, Citrobacter freundii and Pseudomonas aeruginosae. The obtained results were as follows : In the case of aerobic gram negative bacteria, the MIC, $MIC_{50}$ and $MIC_{90}$ against Klebsiella peumoniae and Pseudomonas aeruginosae was significantly decreased in concomitant treated groups with Mahwangyounpae-tang compared to those of single treated groups of CPFX, respectively. However, no significant changes were demonstrated against Echerichia coli, Hemophilus influenzae and Citrobacter freundii. According to these results, it is considered as the in vitro antibacterial activity of CPFX was dramatically increased by concomitant use of Mahwangyounpae-tang against some strains of aerobic gram negative bacteria and the increase and selectivity of antibacterial activities against strains were chosen by the selectivity of Mahwangyounpae-tang not CPFX activity.

  • Seasonal fluctuations of physico-chemical and biological aspects of the environment were studied in Vladivostok harbour (Golden Horn Bay, the East Sea/Sea of Japan). The benthic community structure was described with a focus on size-spectra (bacteria, meio- and macrofauna) related with the chemical environment and chemical fluxes in sediment and to reveal their possible ecological role in the process of bioremediation of the environment. Samples from two sites with different concentrations of heavy metals (Fe, Zn, Cu, Pb, Mn, Cr, Ni Cd, Co) and petroleum hydrocarbon were assessed by a number of methods. These included plate counts of culturable bacteria, observation through a scanning electron (SEM) and transmission electron microscope (TEM). These approaches were complemented with microscopic assessments of the diversity of the benthic community. The specific communities had a limited number of species, tolerant to abnormally high levels of toxic compounds. The dominant species were presented by several sho.1-lived small polychaetes (Capitella capitata) and nematodes (Oncholaimium ramosum). The highest population density was recorded in microbenthos, in various diatoms, various physiological groups of bacteria which participate in biomineralization: marine heterotrophic bacteria, which oxidized oil, black oil in addition to groups resistant to heavy metals. They have the entire set of mechanisms for neutralizing the negative effect of those compounds, forming the detrital food web and biogeochemical circulation of material in sediments, which results in the biological self-recycling of sea basins. Macro- and meiobenthic organisms were more sensitive to a greater extent of $H_2S$ and petroleum hydrocarbons than to metal content, but the within-site rankings were the same as those achieved for microbiological analyses.
