대한민국 제주도 연안 해수로부터 agarase를 생산하는 균주 S4를 분리하였다. 균주 S4는 그람-음성의 막대형 세포로 부드러운 베이지색 원형 콜로니를 형성하며, 한 개의 극성 편모를 갖는다. S4 균주는 $15-42^{\circ}C$, 0.5-5%(w/v) NaCl, pH 6.0-9.0, 0.5-5%(w/v) NaCl 농도에서 안정된 성장을 보인다. S4 균주의 G+C content는 49.93 mol%, 세포내 주요 지방산 (>15%)은 $C_{18:1}{\omega}7c$, $C_{16:0}$, Summed feature 3(comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH)이다. 16S rRNA 염기서열, 생화학적 및 분류학정 특징에 기초하여 S4 균주를 Vibrio sp. S4로 명명하였다. 0.1% agar를 첨가한 액체배지에서 S4 균주는 72시간에 세포농도와 agarase 활성이 최대치를 보였다. 반면, agar 를 첨가하지 않은 배양액에서의 agarase 활성은 무시할만한 수준이었으며, 이는 균주의 agarase 유전자 발현이 agar에 의해 유도됨을 시사하고 있다. 균주 S4가 세포외부로 분비하는 총 agarase는 $45^{\circ}C$와 pH 7.0에서 최상의 효소 활성을 보였으며, agarose를 분해하여 (neo)agarotetraose와 (neo)agarohexaose를 생산하였다.
Molecular and cultivation techniques were used to characterize the bacterial communities of biobead reactor biofilms in a sewage treatment plant to which an Aerated Up-Flow Biobead process was applied. With this biobead process, the monthly average values of various chemical parameters in the effluent were generally kept under the regulation limits of the effluent quality of the sewage treatment plant during the operation period. Most probable number (MPN) analysis revealed that the population of denitrifying bacteria was abundant in the biobead #1 reactor, denitrifying and nitrifying bacteria coexisted in the biobead #2 reactor, and nitrifying bacteria prevailed over denitrifying bacteria in the biobead #3 reactor. The results of the MPN test suggested that the biobead #2 reactor was a transition zone leading to acclimated nitrifying biofilms in the biobead #3 reactor. Phylogenetic analysis of 16S rDNA sequences cloned from biofilms showed that the biobead #1 reactor, which received a high organic loading rate, had much diverse microorganisms, whereas the biobead #2 and #3 reactors were dominated by the members of Proteobacteria. DGGE analysis with the ammonia monooxygenase (amoA) gene supported the observation from the MPN test that the biofilms of September were fully developed and specialized for nitrification in the biobead reactor #3. All of the DNA sequences of the amoA DGGE bands were very similar to the sequence of the amoA gene of Nitrosomonas species, the presence of which is typical in the biological aerated filters. The results of this study showed that organic and inorganic nutrients were efficiently removed by both denitrifying microbial populations in the anaerobic tank and heterotrophic and nitrifying bacterial biofilms well-formed in the three functional biobead reactors in the Aerated Up-Flow Biobead process.

Arbuscular mycorrhizal fungi (AMF) have great potential for assisting heavy metal hyperaccumulators in the remediation of contaminated soils. However, little information is available about the symbiosis of AMF associated with an antimony (Sb) accumulator plant under natural conditions. Therefore, the objective of this study was to investigate the colonization and molecular diversity of AMF associated with the Sb accumulator ramie (Boehmeria nivea) growing in Sb-contaminated soils. Four Sb mine spoils and one adjacent reference area were selected from Xikuangshan in southern China. PCR-DGGE was used to analyze the AMF community composition in ramie roots. Morphological identification was also used to analyze the species in the rhizosphere soil of ramie. Results obtained showed that mycorrhizal symbiosis was established successfully even in the most heavily polluted sites. From the unpolluted site Ref to the highest polluted site T4, the spore numbers and AMF diversity increased at first and then decreased. Colonization increased consistently with the increasing Sb concentrations in the soil. A total of 14 species were identified by morphological analysis. From the total number of species, 4 (29%) belonged to Glomus, 2 (14%) belonged to Acaulospora, 2 (14%) belonged to Funneliformis, 1 (7%) belonged to Claroideoglomus, 1 (7%) belonged to Gigaspora, 1 (7%) belonged to Paraglomus, 1 (7%) belonging to Rhizophagus, 1 (7%) belonging to Sclervocystis, and 1 (7%) belonged to Scutellospora. Some AMF sequences were present even in the most polluted site. Morphological identification and phylogenetic analysis both revealed that most species were affiliated with Glomus, suggesting that Glomus was the dominant genus in this AMF community. This study demonstrated that ramie associated with AMF may have great potential for remediation of Sb-contaminated soils.
L-Asparaginase (E.C. 3.5.1.1) is an enzyme involved in asparagine hydrolysis and has the potential to effect leukemic cells and various other cancer cells. We identified the L-asparaginase gene (L-ASPG86) in the genus Mesoflavibacter, which consists of a 1,035 bp open reading frame encoding 344 amino acids. Following phylogenetic analysis, the deduced amino acid sequence of L-ASPG86 (L-ASPG86) was grouped as a type I asparaginase with respective homologs in Escherichia coli and Yersinia pseudotuberculosis. The L-ASPG86 gene was cloned into the pET-16b vector to express the respective protein in E. coli BL21 (DE3) cells. Recombinant L-asparaginase (r-L-ASPG86) showed optimum conditions at 37-40℃, pH 9. Moreover, r-L-ASPG86 did not exhibit glutaminase activity. In the metal ions test, its enzymatic activity was highly improved upon addition of 5 mM manganese (3.97-fold) and magnesium (3.35-fold) compared with the untreated control. The specific activity of r-L-ASPG86 was 687.1 units/mg under optimum conditions (37℃, pH 9, and 5 mM MnSO4).
자일란(xylan)을 가수 분해할 수 있는 해양 미생물 PX-1을 제주도 연안의 해수 로부터 순수하게 분리하였다. 16S rRNA 유전자 서열 및 생화학적 분류 결과에 기초하여, PX-1은 Aestuariibacter 속의 한 종으로 확인되어 Aestuariibacter sp. PX-1로 명명하였다. PX-1을 액체 배양한 배양액으로부터 암모늄 설페이트 침전법과 불용성 자일란을 이용한 흡착 크로마토그래피 방법을 이용하여, 세포 외로 분비된 자일라네이즈 후보 단백질을 순수하게 정제하였다. 정제한 후보 자일라네이즈 단백질(XylA)은 sodium dodecyl sulfate-polyacrylamide gel electrophoresis 및 겔여과 크로마토그래피 분석결과, 분자량이 대략 64 kDa인 것으로 추정되었다. XylA는 실제로 beechwood xylan 을 가수분해하는 자일라네이즈 활성을 보였으며, pH 6.0과 45℃에서 최대의 효소 활성을 나타냈다. XylA에 의한 자일란 가수 분해산물을 TLC로 분석한 결과, XylA 는 자일란을 자일로스와 자일로올리고당으로 분해하는 endo-type의 자일라네이즈임을 확인하였다. Beechwood xylan 에 대한 XylA의 Km 및 Vmax 값은 각각 27.78 mM (4.17 mg/ml), 78.13 μM/min이었다.
본 연구는 도시 하수처리장의 미생물 군집 내 항생제 내성 세균의 특징과 하천으로의 항생제 내성 세균 노출의 계절적 변화의 특성을 평가하였다. 일반 종속영양세균 배양을 위한 R2A agar (R2A)와 대장균군을 선택하여 배양하는 MacConkey sorbitol agar (MSA)에 항생제를 첨가 하여 제작한 배지에 하수처리장 시료를 도말하여 배양한 결과 모든 시료에서 다제 항생제 내성 세균을 검출 해 낼 수 있었다. Ampicillin과 sulfathiazole의 내성률이 다른 나라에 비해 높게 측정 되었으며 시료내 유기물의 정도, 사용된 배지에 따라 내성률이 다름을 볼 수 있었다. R2A 배지에서 분리된 다제 항생제 내성 세균은 모두 기존에 알려진 병원성 세균과 그 염기서열이 유사한 것으로 볼 때 병원성 세균의 항생제 내성 정도가 일반 세균보다 높음을 본 연구 결과로 보일 수 있었다. 또한 본 연구에서는 하수처리장이 하천에 미치는 유해성을 계절별로 관찰하여 전체 미생물중 항생제 내성 세균의 비율은 수온과 비례한다는 결과를 얻었다. 이 결과는 지구 온난화가 미생물 유해성을 증가시킬 가능성을 시사한다.
환경친화적이며 원유 분해능이 강력한 균주를 얻기 위하여, 태백산의 토양 일부를 채취하였다. 채취한 토양 sample 로부터 crude oil을 탄소원으로 이용하는 수십 종의 균주를 분리하였으며, 분리된 균주 중 원유분해능 및 biosurfactant 생성능이 가장 우수한 균주를 선별하였다. 또한 분리된 균주의 형태학적 특성을 관찰하고 165 rDNA sequence를 통하여 Bacillus sp. TBM40-3로 동정하였다. 또한 phylogenetic tree를 이용하여 이미 동정된 다른 균주들과의 유연관계를 파악하였다. Bacillus sp. TBM40-3의 배양액의 표면장력은 최저 29 mN/m까지 감소되었고, 생산된 biosurfactant의 유화활성은 soybean oil에서 최대였으며, crude oil에서도 높은 편이였다. 유화안정성은 합성계면활성제와 비슷하거나 우수하였다. 따라서 TBM40-3에서 추출한 biosurfactant는 합성계면활성제를 대체할 수 있는 환경친화적인 생물 계면활성제로 사용될 수 있는 가능성을 보여준다.
수호초로부터 분리된 DUCC5000 균주를 형태적, 분자생물학적 분류를 통해 Paraconiothyrium brasiliense로 동정하였다. 본 균은 기존에 알려진 P. brasiliense 균주의 ITS 염기서열과 100%의 상동성을 보였다. 계통유전학적 분석 결과 P. brasiliense 균주와 같은 clade를 형성하였다. 서로 다른 영양 배지와 pH 조건 하에서 균사생육 특성을 조사한 결과 oatmeal agar 배지와 pH 9에서 최적의 생장을 보였다. 배지 종류에 따라 그리고 pH 조건에 따라 균총 형태가 달라지는 특성을 지니고 있었다. Chroma 반응배지를 이용하여 7가지 세포외 효소의 분비 능력을 조사한 결과 ${\beta}$-glucosidase 활성이 가장 활발하였고 CM-cellulase와 xylerase에서는 활성이 미약하였다. 인공 접종하였으나 수호초에 대한 병원성은 나타내지 않았다. 본 균은 수호초에서는 국제적으로 처음 보고되는 균이다.
1991년에 우리나라에서 복숭아나무(Prunus persica var. persica) 과실에 흰가루 증상을 일으키는 병원균은 형태적 특징에 의거하여 Podosphaera pannosa로 동정되었다. 복숭아 과실이 P. pannosa에 감염된다는 사실은 일본을 비롯한 여러 나라에서 알려져 왔다. 2011년에 Jankovics 등[11]은 세르비아 및 프랑스에서 채집한 10점의 시료를 연구하여 복숭아 과실의 '녹얼룩점(rusty spot)' 증상은 P. leucotricha에 의한 것이며, 천도(Prunus persica var. nucipersica) 과실의 '흰가루(powdery mildew)' 증상은 P. pannosa에 의한 것이라고 보고하였다. 이에 따라, 한국에서 복숭아 과실에 발생한 흰가루병을 4점 채집하여 형태적 검경 및 분자적 분석을 통해 병원균의 정체를 밝혔다. 3점의 시료는 흰가루 증상, 1점의 시료는 녹얼룩점 증상을 나타냈다. 형태적으로는 4점 모두 P. pannosa로 동정되었다. 분자적으로는 2점의 시료에서 ITS 염기서열을 분석하여 Rosa spp. 유래의 P. pannosa와 99% 이상 상동성을 확인하였다. 그리고 maximum likelihood 방법으로 계통수를 작성한 결과 Rosa spp. 유래의 P. pannosa와 분자계통학적으로 동일한 클러스터에 소속됨을 확인하였다. 이상의 결과를 바탕으로 한국에서 복숭아 과실의 흰가루 증상과 녹얼룩점 증상은 모두 P. pannosa에 의해 발생됨을 처음으로 보고한다.
Saprolegniasis is one of the most devastating oomycete diseases in freshwater fish which is caused by species in the genus Saprolegnia including Saprolegnia parasitica. In this study, we isolated the strain of S. parasitica from diseased rainbow trout in Korea. Morphological and molecular based identification confirmed that isolated oomycete belongs to the member of S. parasitica, supported by its typical features including cotton-like mycelium, zoospores and phylogenetic analysis with internal transcribed spacer region. Pathogenicity of isolated S. parasitica was developed in embryo, juvenile, and adult zebrafish as a disease model. Host-pathogen interaction in adult zebrafish was investigated at transcriptional level. Upon infection with S. parasitica, pathogen/antigen recognition and signaling (TLR2, TLR4b, TLR5b, NOD1, and major histocompatibility complex class I), pro/anti-inflammatory cytokines (interleukin $[IL]-1{\beta}$, tumor necrosis factor ${\alpha}$, IL-6, IL-8, interferon ${\gamma}$, IL-12, and IL-10), matrix metalloproteinase (MMP9 and MMP13), cell surface molecules ($CD8^+$ and $CD4^+$) and antioxidant enzymes (superoxide dismutase, catalase) related genes were differentially modulated at 3- and 12-hr post infection. As an anti-Saprolegnia agent, plant based lawsone was applied to investigate on the susceptibility of S. parasitica showing the minimum inhibitory concentration and percentage inhibition of radial growth as $200{\mu}g/mL$ and 31.8%, respectively. Moreover, natural lawsone changed the membrane permeability of S. parasitica mycelium and caused irreversible damage and disintegration to the cellular membranes of S. parasitica. Transcriptional responses of the genes of S. parasitica mycelium exposed to lawsone were altered, indicating that lawsone could be a potential anti-S. parasitica agent for controlling S. parasitica infection.
