Octyl Gallate Inhibits ATP-induced Intracellular Calcium Increase in PC12 Cells by Inhibiting Multiple Pathways

  • Phenolic compounds affect intracellular free $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) signaling. The study examined whether the simple phenolic compound octyl gallate affects ATP-induced $Ca^{2+}$ signaling in PC12 cells using fura-2-based digital $Ca^{2+}$ imaging and whole-cell patch clamping. Treatment with ATP ($100\;{\mu}M$) for 90 s induced increases in $[Ca^{2+}]_i$ in PC12 cells. Pretreatment with octyl gallate (100 nM to $20\;{\mu}M$) for 10 min inhibited the ATP-induced $[Ca^{2+}]_i$ response in a concentration-dependent manner ($IC_{50}=2.84\;{\mu}M$). Treatment with octyl gallate ($3\;{\mu}M$) for 10 min significantly inhibited the ATP-induced response following the removal of extracellular $Ca^{2+}$ with nominally $Ca^{2+}$-free HEPES HBSS or depletion of intracellular $Ca^{2+}$ stores with thapsigargin ($1\;{\mu}M$). Treatment for 10 min with the L-type $Ca^{2+}$ channel antagonist nimodipine ($1\;{\mu}M$) significantly inhibited the ATP-induced $[Ca^{2+}]_i$ increase, and treatment with octyl gallate further inhibited the ATP-induced response. Treatment with octyl gallate significantly inhibited the $[Ca^{2+}]_i$ increase induced by 50 mM KCI. Pretreatment with protein kinase C inhibitors staurosporin (100 nM) and GF109203X (300 nM), or the tyrosine kinase inhibitor genistein ($50\;{\mu}M$) did not significantly affect the inhibitory effects of octyl gallate on the ATP-induced response. Treatment with octyl gallate markedly inhibited the ATP-induced currents. Therefore, we conclude that octyl gallate inhibits ATP-induced $[Ca^{2+}]_i$ increase in PC12 cells by inhibiting both non-selective P2X receptor-mediated influx of $Ca^{2+}$ from extracellular space and P2Y receptor-induced release of $Ca^{2+}$ from intracellular stores in protein kinase-independent manner. In addition, octyl gallate inhibits the ATP-induced $Ca^{2+}$ responses by inhibiting the secondary activation of voltage-gated $Ca^{2+}$ channels.

Antimicrobial Activities and Phenolic Compounds of Pyroligneous Liquor (목초액의 항균활성과 페놀화합물의 함량)

  • Antimicrobial activities of pyroligneous liquor were investigated by determining Minimal Inhibitory Concentration (MIC). The solvent extracts of pyroligneous liquor, which were extracted by using solvents with different polarities such as hexane, ethylacetate, or butanol. The activities were examined by disc diffusion method using MIC against 7 food poisoning microbe type strains. Antimicrobial activities were shown in hexane, ethylacetate, butanol, and aqueous fractions of pyroligneous liquor. Among the four fractions, ethylacetate fraction showed the highest inhibitory effect on the microorganism such as Shigella sonnei, and Yersinia enterocolitica at the concentration of 2.0 mg/disc. The purified P-1 and P-2 fractions isolated by silica gel column chromatography from ethylacetate fraction of pyroligneous liquor had the highest antimicrobial activity. The total phenolic compounds content in ethylacetate, hexane, butanol, and aqueous fraction was 488.3 mg/g, 403.8 mg/g, 83.6mg/g, and 74.5 mg/g, respectively. Taken together, these results suggest that the ethyl acetate fraction could be suitable for the development of isolation and identification of antimicrobial compound from pyroligneous liquor, resulting from the above antimicrobial activity.

Quality of Soybean Paste (Doenjang) Prepared with Lotus Root Powder (연근 분말을 첨가한 된장의 품질 특성)

  • Soybean paste (Doenjang) was prepared by adding Lotus root powder (LRP) at $5\~15\%$ (w/w) to improve quality of the Doenjang and to give some functional properties. Moisture content was ranged about $50.05\~54.04\%$ and amino nitrogen content was $635\~648\;mg\%$ following the LRP contents. Crude protein amounts were $11.55\~12.56\%$ that was no difference between test samples. Carbohydrates contents increased $1.5\~2$ times in the test samples than the control depending on the LRP contents. However, contents of crude lipid $(6.99\~8.55\%)$ and ash $(13.99\~15.17\%)$ were decreased as increased the amounts of LRP. The pH of the product was decreased until 45 days during aging period and then slightly increased without significantly differences among test samples. The values of acidity were $1.85\~2.18\%$ at the early stage of aging but slightly increased with further aging. The total phenolic content in the Doenjang adding $15\%$ LRP was $461.8\;mg\%$ which is higher than $368.6\;mg\%$ in the control. Doenjang prepared with LRP showed an anti-browning effect of the product itself.

Characteristics of Pinus thunbergii×P. massoniana F1 Hybrid Seedlings (Pinus thunbergii×P. massoniana F1 교잡종(交雜種) 유묘(幼苗)의 특성(特性))

  • Pinus thunbergii ${\times}$ P. massoniana $F_1$ hybrids were made by pollinating 15 years old Japanese black pine (P. thunbergii) with Masson pine (P. massoniana) pollen, air-shipped from Taiwan. The crossability between two species were observed. Seedling height, morphological characteristics, and the composition of phenolic compounds of these hybrids and the open-pollinated progenies of P. thunbergii were investigated. The results of the observation were as follows; 1) Number of the hybrid seeds per cone was 2.3, while open-pollinated Japanese black pine produced 26 seeds per cone. 2) All hybrid seedlings outgrew P. thunbergii, averaging 151% of mean height of Japanese black pine seedlings. The growth of hybrids was much variable, ranging from 120% to 208% of mean height of the open-pollinated progenies of P. thunbergii. 3) With regard to needle characteristics, the hybrid needles were longer and softer than those of P. thunbergii. The color of hybrid needles showed yellowish green. The positions of resin ducts in P. thunbergii observed medial, while those of hybrids showed medial and external. 4) No large difference in phenolic compound composition between hybrid and P. thunbergii was found, but phenolic compound of unknown 4 and 6 were observed only in hybrid. Further intensive work is recommended in this area.

Studies on Antioxidant Activity and Inhibition of Nitric Oxide Synthesis from Codium fragile (청각추출물의 항산화 및 일산화질소 합성 저해 연구)

  • This study was carried out to investigate the biological effects from Codium fragile. Methanol extract of Codium fragile increased two times at 2500 ㎍/ml the growth of Lactobacillus plantarum that associated with probiotic properties of lactic acid bacteria of Kimchi. Ethyl acetate extract of Codium fragile inhibited the cellulase activity up to approximately 60% at $2500\;{\mu}g/ml$. Methanol extract of Codium fragile was fractionated into several subfractions and their antioxidant activities were measured by using DPPH radical scavenging and SOD-like activity. Especially the antioxidative activity of ethyl acetate fraction was shown higher than that of other fractions and its fraction showed higher contents of total phenolic compounds, indicating the positive relationship between DPPH radical scavenging effect and total polyphenol content. Stimulation of the macrophages RAW264.7 cells with lipopolysaccharide (LPS) resulted in increased production of nitric oxide (NO) in the medium. However, the methanol extract of Codium fragile showed marked inhibition of NO synthesis in a dose-dependent manner. This result suggest that Codium fragile plays significant role for activation of immune system in the pathogenesis of inflammatory diseases.

Isolation and Identification of Antimicrobial Compounds against Helicobacter pylori from Rosemary (Rosmarinus officinalis L.) Extracts (로즈마리(Rosmarinus officinalis L.) 추출물로부터 Helicobacter pylori에 대한 항균물질 분리 및 동정)

  • Rosmarinus officinalis extracts had a significant antimicrobial activity against Helicobacter pyori. Total phenolic contents and inhibition zone of rosemary extracts were estimated to be 25.7 mg/g and 14 mm at $200{\mu}g/mL$ of phenolic contents, respectively. The Sephadex LH-20 and MCI-gel CHP-20 column chromatographic separations for the phenolic extracts from R. officinalis leaves led to isolation of five acids, whose structures were determined as protocatechuic acid (A), coumaric acid (B), caffeic acid (C), chlorogenic acid (D), and rosmarinic acid (E), from interpretation of spectroscopic data including nagative fast atom bombardment (FAB)-mass, $^1H$-NMR, $^{13}C$-NMR, and IR. All isolated compounds were tested for antimicrobial activity against H. pyori. The purified single compound showed less antimicrobial activity against H. pylori than the mixed purified compounds, which generate A+B, A+E, C+D, C+E (each $200{\mu}g/disc$) excellent as large clear zone by synergy effect. These results indicate rosemary extracts are preventive agents against H. pyori.

Physiological Activity of Extracts from Radish (Raphanus sativus L.) Leaves (무청 추출물의 생리활성)

  • This study was conducted to investigate yield of extract, total phenolic compounds content, electron donating activity, nitrite scavenging activity, superoxide dismutase like activity of the radish leaves (Raphanus sativus L.) extracts. Radish leaves extracts were prepared using water extraction, 50% ethanol extraction and 50% methanol extraction. The yield of Kwandong radish leaves extracts without blanching treatment (DRLK) was $26.56{\sim}32.56%$ and the extract yield of Minongdanbaek radish leaves without blanching treatment (DRLM) was $34.34{\sim}36.90%$. On the other hand, the yield of samples with blanching treatment was a lower range value of $18.70{\sim}23.56%$ than samples without blanching treatment. In the total contents of phenolic compound, samples with and without blanching treatment were $49.1{\sim}59.4mg/g\;and\;53.41{\sim}62.08mg/g$, respectively. Minongdanbaek radish leaves extracts (DRLM, BDRLM) showed higher contents value than Kwandong radish leaves extracts (DRLK, BDRLK) in the total phenolic compounds. Samples without blanching treatment showed electron donating activity above 80%, nitrite scavenging activity of $36.63{\sim}51.17%$ and superoxide dismutase like activity of $38.53{\sim}45.38%$. Generally, Kwandong samples showed a little higher activity more than Minongdanbaek samples in the electron donating activity, nitrite scavenging activity and superoxide dismutase like activity. Also, radish leaves extracts without blanching treatment showed high physiological activities.

Assessing Carotenoid Levels and Antioxidant Properties in Korean Sweet Corn Inbred Lines to Develop High-Quality Sweet Corn Varieties through Breeding (기능성 단옥수수 품종 육성을 위한 자식계통의 카로티노이드 함량 및 항산화 활성 평가)

  • Sweet corn is widely consumed due to its high nutritional content and diverse phytochemical composition, including carotenoids and phenolic compounds, which have several benefits for human health. This study aims to identify breeding materials for developing high-functional sweet corn varieties by evaluating the phytochemical and antioxidant activities of 37 Korean sweet corn inbred lines. The results revealed genetic variation in various components, such as carotenoid content (range of 120.7~1239.3 mg 100 g-1), polyphenol content (490.5~740.6 mg gallic acid equivalent 100 g-1), and flavonoid content (7.3~68.6 mg catechin equivalent 100 g-1). In addition, the free radical scavenging capacity, measured using 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid), also varied among the inbred lines. Therefore, in this study, we identified Korean sweet corn inbred lines with high phytochemical content and excellent antioxidant activity. The development of sweet corn varieties with improved functionality is expected to further expand the role of sweet corn as a source of antioxidants in the Korean diet.

In vitro Antioxidative Activities and Phenolic Composition of Hot Water Extract from Different Parts of Cudrania tricuspidata

  • We evaluated total phenolics and antioxidative activities of water extracts from different parts of Cudrania tricuspidata (specifically, the leaves, stems, roots, and fruits). The antioxidative activities of these samples were determined using five methods, including 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS) radical scavenging, reducing power, ferric reducing ability of plasma (FRAP), and $\beta$-carotene/linoleic acid system. The water extract of leaves exhibited the higher DPPH, ABTS radical scavenging activities, reducing power, and FRAP than water extract of stem, roots, and fruits. Inhibition values on linoleic oxidation of water extracts from leaves, stems, roots, and fruits were calculated as 45.98%, 33.03%, 39.73%, and 25.48% at 10 mg/mL, respectively. The water extract of C. tricuspidata leaves had the highest amount of toal phenolics (73.60$\pm$0.28 mg/g). High-performance liquid chromatography (HPLC) analysis showed that quercetin is the predominant phenolic compound in water extract of leaves. Thus, our study verified that the water extract of leaves has strong antioxidant activities which are correlated with its high level of phenolic compounds, particularly quercetin. This water extract of C. tricuspidata leaves can be used as an effective and safe source of antioxidants.

Optimal Extraction Conditions of Active Components from the Adventitious Roots of Noni (Morinda citrifolia) (노니 부정근으로부터 유효성분의 최적 추출조건)

  • To determine the optimum condition for the extraction of active components, noni adventitious roots were extracted under various conditions employing various solvents including methanolic water at different ratio (20, 40, 60, 80, and 100% water), extraction times and extraction methods. Anthraquinones, phenolic compounds, and flavonoids extraction using different solvents showed that 1 h of ultrasonic extraction was effective in 60-80% methanol, and 2 h of reflux extraction was effective in 80% methanol. To compare the extraction efficiency of active components according to different extraction methods and extraction times for noni adventitious roots, the active components were extracted by ultrasonic extraction, shaking extraction, reflux extraction, homogenizer extraction, high-pressure extraction, and soaking extraction. The highest phenolic contents were found in the extracted from ultrasonic extraction and anthraquinones and flavonoids contents were highest in the reflux extraction.
