Determination of Protoberberine Alkaloids in Phellodendri Cortex and Preparation by Spectrophotometric Method (흡광도측정법에 의한 황백과 제제 중 프로토베르베린 알칼로이드의 정량)

  • The Phellodendri Cortex of Phellodendron amurense (Rutaceae) is known to contain a number of isoquinoline alkaloid, and berberine, palmatine, jateorrhizine, phellodendrine and magnoflorine are the major constituents of protoberberine alkaloids. For the determination of protoberberine alkaloids from Phellodendri Cortex and berberine chloride from the preparation, the new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex ion. Samples were extracted with 0.1 mM hydrochloric acid, potassium biphthalate reagent, thiocyanatocobaltate reagent and 1.2-dichloroethane for 60 min. The absorbance of protoberberine alkaloid complexes in 1.2-dichloroethane solution was measured at 625 nm. Calibration curve for berberine was linear over the concentration range of 0.05~0.30 mg/ml 1.2-dichloroethane. The method proved to be rapid, simple and reliable for the determination of protoberberine alkaloids from Phellodendri Cortex and berberine chloride from the preparation.

Anti-inflammatory Effect of Imyosan Extract is more potent than that of its Component Herb Extracts in Murine Macrophages (마우스 대식세포인 RAW 264.7에 대한 이묘산(二妙散)의 메탄올 추출물의 항염증 효과)

  • Objectives : Imyosan (IMS), a drug preparation comprised of Phellodendri Cortex (PC) and Atractylodis Rhizoma (AR), is commonly used as a traditional herbal medicine in Korea and China for the treatment of various inflammatory diseases. However, little is known about the effect of IMS and its component herbs on inflammatory mediators in RAW 264.7 cells. Therefore, in this study, methanol extracts of IMS and its component herbs were examined to determine if they inhibited inflammatory effects in RAW 264.7 cells. Methods : Cytotoxic activity of IJHT and its components on RAW 264.7 cells was using 5-(3-carboxymethoxyphenyl)-2H-tetrazolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines were measured by ELISA kit. The levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were detected by western blot. Results : Methanol extract of IMS and its component herbs were significantly reduced iNOS and COX-2 expression as well as NO, PGE2, $IL-1{\beta}$ and IL-6 production in RAW 264.7 cells. Conclusions : The results of this study indicated that the anti-inflammatory effect of Imyosan extract is more potent than that of extracts of its component herbs in macrophages.

  • Background and Objectives : There are four kinds of formulas for invigorating the kidney to cure its asthenic syndrome based on the types of preparation formulas : Boshintang, Boshinhwan, Boshinsan, and Boshingaeng. There are 16 kinds of Boshintang, 28 kinds of Boshinhwan, 3 kinds of Boshinsan, and one kind of Boshingaeng. Combination of herbal medicines, carried out in formulas for invigorating the kidney, consists of various kinds depending on medical scientists' personal experience in medical treatment without any general principles, which makes it difficult to apply it to clinical use. The objectives of this study lie in theoretical establishment of Boshintang for curing the asthenic syndrome of kidney through analyzing the component medicines and combination principles of Hyeongok's Boshintang, and furthermore, maximizing the clinical use of Boshintang. Methods : This study analyzed the component medicines and combination principles of Hyeongok's Boshintang based on the theory for properties and tastes of herbal medicines from the ${\ulcorner}$Yellow Emperor's Canon of Internal Medicine${\lrcorner}$ , the theory for principal herbal medicine, assistant herbal medicine, adjuvant herbal medicine, dispatcher herbal medicine, and the five elements doctrine. Hyeongok's Boshintang is an even prescription, composed of 6 kinds of ingredients : No.1 Fructus Lycii (3don;11.25g), No.2 Radix Polygoni Multiflori (1don:3.75g), No.3 Sarcucarpium Corni (1don), No.4 Fructus Schisandrae (1don), No.5 Rhizoma Anemarrhenae (5poon: 1.875g), and No.6 Cortex Phellodendri (5poon) Results : There are three methods for curing the asthenic syndrome of kidney according to the five elements doctrine : invigorating the kidney, invigorating the lung and purging the spleen. First, if you suffer from the asthenic syndrome of the kidney, you need to invigorate your kidney. There are two available methods, including taste and property invigoration according to the theory for properties and tastes of herbal medicines. They each imply the bitter taste and the cold property invigorate the kidney. In the case of taste invigoration, two herbal medicines with bitter taste, Fructus Lycii and Radix Polygoni Multiflori, are combined into the principal and assistant herbal medicine, respectively. For property invigoration, two herbal medicines with the cold property, Rhizoma Anemarrhenae and Cortex Phellodendri, are combined into adjuvant herbal medicines. Secondly, if you suffer from the asthenic syndrome of the kidney, you need to invigorate your lung which is mother in the mother-child relationship in inter-promotion among the five elements. There are two methods to invigorate the lung, including taste and property invigoration according to the theory for properties and tastes of herbal medicines. They each mean the sour taste and the cool property invigorate the lung. Therefore, it is important to use sour herbal medicines for taste invigoration and cool ones for property invigoration. Both bitter and cool herbal medicines, Rhizoma Anemarrhenae and Cortex Phellodendri, are combined into adjuvant herbal medicines. Lastly, if you suffer from the asthenic syndrome of the kidney, you need to purge your spleen which is an element being surpassed in the relationship between the elements surpassed and ones not surpassed in inter-restraint among the five elements. There are two methods to purge the spleen, which include taste and property purgation according to the theory for properties and tastes of herbal medicines. Taste purgation means to purge the spleen with bitter taste and property purgation to purge the spleen with cool property. Therefore, it is important to use bitter herbal medicines for taste purgation and cool ones for property purgation. Both bitter and cool herbal medicines, Rhizome Anemarrhenae and Cortex Phellodendri. were combined to purge the spleen and invigorate the kidney. In addition, Rhizome Anemarrhenae and Cortex Phellodendri are combined as dispatcher herbal medicine, reinforcing the kidney energy. Conclusions : First, to cure the asthenic syndrome of the kidney, the methods of invigorating the kidney and the lung, and purging the spleen should be used according to the five elements doctrine. Secondly, herbal medicines appropriate for those treatment methods should be chosen according to the theory for properties and tastes of herbal medicine and thirdly, the combination of those herbal medicines should be carried out according to the theory for principal herbal medicine, assistant herbal medicine, adjuvant herbal medicine, dispatcher herbal medicine. As a good example, Hyeongok's Boshintang is combined according to the above theories. In conclusion, this formula was created by applying to the theory for properties and tastes of herbal medicines.

  • Berberine is one of alkaloids extracted from Phellodendri Cortex or Coptidis Rhizoma and has extensively used as an antibacterial and antidiarrheal drug. In order to increase the bioavailability of berberine preparation, berberine pamoate was synthesized and investigated on its usage in vitro and in vivo. Berberine was more rapidly extracted from herbal plants by hot water extraction method than soxhlet extraction method. Berberine pamoate was easily synthesized from berberine hydrochloride and potassium pamoate solution and identified using the infrared spectrum. Quantitative analysis of berberine was possible in methanol solution by fluorometric determination. The dissolution rate of berberine pamoate was more decreased than that of berberine hydrochloride in simulated gastric fluid and simulated intestinal fluid. The remaining proportion of berberine pamoate in the small intestine of rat was maintained at high concentration for a long time as compared with that of berberine hydrochloride.

Preparation and Bioavailability of Oriental Medicine Containing Baicalin (I) : Identification and Physicochemical Properties of Coprecipitated Product of Scutellariae Radix and Coptidis Rhizoma (바이칼린 함유생약의 제제화 및 생체이용률 (제 1보): 황금 및 황련 공침물의 확인 및 물리화학적 성질)

  • Precipitation was formed during the preparation of decoction from a mixture of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine were identified in coprecipitated product and these components were the active ingredients of two herbal medicine. The coprecipitated product was very slightly soluble in water and sparingly soluble in ethanol. The stoichiometric ratio of baicalin and berberine was found to be 1:1. The lipid-water partition coefficients of coprecipitated product were increased more than baicalin and berberine in chloroform, but were decreased in other organic solvents. The content of baicalin and berberine in coprecipitated product, determined by HPLC, were 23.08% and 26.75%, but the content of active ingredients in supernatant were 0.66% and 0.26%, respectively. The dissolution profile of baicalin of coprecipitated product was increased more than extract of Scutellariae Radix in artificial gastric juice, but was decreased in artificial intestinal juice. The dissolution rate of berberine of coprecipitated product was lower than extract of Coptidis Rhizoma in artificial gastric juice and intestinal juice commonly.

  • In order to investigate the effect of 'Whangryonhaedok-Tang' on the anti-inflammatory, ileum of mice, blood pressure, blood glucose, total cholesterol and antibacterial activity, those pharmacological actions were conducted. The results of this studies were summarized as follows; Rat hind paw edema induced by acetic acid was inhibited. In mice, tone of intestinal smooth muscle was suppressed with the treatment of sample and intestinal contractions induced by $BaCl_2$ was inhibited. Hypertensive effect was not observed in rabbits but was observed by the water extract of 'Whangryonhaedok-Tang' excluded Phellodendri Cortex in rabbits. The increased blood glucose levels in alloxandiabetic rabbits were significantly decreased by the oral administration of the sample I after 6 days. The total cholesterol levels in alloxan-induced rabbits were significantly decreased by the oral administration of the sample I after 9 days. Antibacterial activity was observed. In addition, we thought the pharmacological actions were significantly noted by the sample III (Methanol soluble fraction) because of the purification of effective components.

  • Precipitation was formed during the preparation of decoction from a mixure of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine, the active ingredients of the two herbal medicine were identified in coprecipitated product. Pills were prepared using the coprecipitated product and various binders. The dissolution rate of baicalin and berberine from pills was increased in at pH1.2 when acacia or tragacanth was used. The absorption rate of baicalin from the coprecipitated product was faster than that from Scutellaria extract, but the absorption of berberine from CPP was slower in stomach, duodenum and jejunum of rats compared with Coptis extract. The time required for the maximum serum concentration (Cmax) of baicalin and berberine from CPP in mice were 150 and 200 min after oral administration, respectively. The maximum serum concentration of baicalin from CPP in mice was higher than Scutellaria extract, but the concentration of berberine was lower compared with Coptis extract. The minimum inhibitory concentration of CPP was below $50\;{\mu}g/ml$ against gram positive bacteria, and was higher than that against gram negative bacteria. The antibacterial activity of CPP was lower than that of herberine, but was more potent than Scutellaria extract. It was found that the inhibition rates of growth by CPP against S. epidermidis, K. pneumoniae, B. cereus and S.aureus were 60.0, 51.1, 45.4 and 39.9%, respectively.

Anticandidal Activity of the Protein Substance from Coptidis Rhizoma (황련에서 분리된 단백질성분의 항진균효과)

  • Antimicrobial peptides are evolutionary ancient weapons for animal and plant species to depend themselves against infectious microbes. In the present study, we investigated if an antimicrobial peptide was produced from Coptidis Rhizoma. For the determination, protein substance from the medicinal plant was isolated by various preparations. Among the preparations, the protein portion dissolved in phosphate-buffered saline solution (CRP-DS) that contained the most amount of protein $(90\%)$ resulted in maximal inhibition of Candida albicans which causes local and systemic infections. Analyses by gel-electrophoresis and gel-permeation chromatography showed the CRP-DS formed a single band of approximately 11.8 KDa as molecular size. Antifungal activity of the CRP-DS was almost equivalent to antifungal activity by fluconazole, resulting in MIC (minimal inhibitory concentration) of approximately $50{\mu}g/ml$. The antifungal activity was a dose-dependent. The antifungal activity appeared to be inactivated by heat-treatment and ionic strength, respectively. In a murine model, the CRP-DS enhanced resistance of mice against disseminated candidiasis. The HPLC analysis demonstrated maximum $4\%$ of berberine as residual content in the CRP-DS preparation resulted in no influence on the antifungal activity. In addition, protein portion isolated from Phellodendri Cortex producing the alkaloid component like Coptidis Rhizoma had no such anticandidal effect. These results indicate that the protein substance from Coptidis Rhizoma was responsible for the antifungal activity.
