B. subtilis MC 31를 이용한 청국장의 품질특성 (Characteristics of Chungkookjang Produced by Bacillus subtillus MC31)

  • GABA 함량이 높은 청국장을 발효하는 Bacillus subtilis MC 31의 청국장 품질특성을 조사하였다. B. subtilis MC 31에 의해 발효된 청국장으로부터 24개의 아미노산이 검출되었고 그 중에서도 leucine이 가장 높은 함량을 나타냈다. B. subtilis MC31에 의해 발효가 된 청국장의 미생물의 생육 변화를 조사한 결과 총 균수가 정지기에 들어서면서 log $9.52{\pm}0.5$~log $9.049{\pm}0.5$ CFU/g까지 증가하였다. 청국장의 일반성분은 수분 $61.7{\pm}0.01%$, 회분 $1.52{\pm}0.01%$, 조단백 $17.66{\pm}0.04%$, 조지방 $8.96{\pm}0.03%$, 조섬유가 2.61%를 함유하였다. 암모니아태, 아미노태, 환원당은 모두 삶은 콩보다 청국장의 함량이 높게 조사되었지만 산도와 총 당은 청국장이 삶은 콩보다 함량이 낮았다. 청국장의 pH는 시간이 지날수록 알칼리화되고 점질물은 $4.7{\pm}0.05%$, protease activity는 $0.519{\pm}7.36$ g/l로 나타났으며 Fibrin plate와 Robbin실험을 통하여 fibrinolytic activity도 높음을 확인하였다. 이상의 결과로 미루어 보아 B. subtilis MC 31은 우수한 청국장 품질을 띄어 제조용 균주로 사용이 가능한 것으로 사료된다.

Effectiveness of cephalosporins in hydrolysis and inhibition of Staphylococcus aureus and Escherichia coli biofilms

  • Importance: Staphylococcus aureus and Escherichia coli contribute to global health challenges by forming biofilms, a key virulence element implicated in the pathogenesis of several infections. Objective: The study examined the efficacy of various generations of cephalosporins against biofilms developed by pathogenic S. aureus and E. coli. Methods: The development of biofilms by both bacteria was assessed using petri-plate and microplate methods. Biofilm hydrolysis and inhibition were tested using first to fourth generations of cephalosporins, and the effects were analyzed by crystal violet staining and phase contrast microscopy. Results: Both bacterial strains exhibited well-developed biofilms in petri-plate and microplate assays. Cefradine (first generation) showed 76.78% hydrolysis of S. aureus biofilm, while significant hydrolysis (59.86%) of E. coli biofilm was observed by cefipime (fourth generation). Similarly, cefuroxime, cefadroxil, cefepime, and cefradine caused 78.8%, 71.63%, 70.63%, and 70.51% inhibition of the S. aureus biofilms, respectively. In the case of E. coli, maximum biofilm inhibition (66.47%) was again shown by cefepime. All generations of cephalosporins were more effective against S. aureus than E. coli, which was confirmed by phase contrast microscopy. Conclusions and Relevance: Cephalosporins exhibit dual capabilities of hydrolyzing and inhibiting S. aureus and E. coli biofilms. First-generation cephalosporins exhibited the highest inhibitory activity against S. aureus, while the third and fourth generations significantly inhibited E. coli biofilms. This study highlights the importance of tailored antibiotic strategies based on the biofilm characteristics of specific bacterial strains.

성인 계단보행 시 계단 너비에 따른 하지의 운동역학적 분석 (A Kinetic Analysis of the Lower Extremity during Walking on Three Different Stair width in Healthy Adults)

  • 이 연구는 성인 남성을 대상으로 계단 너비에 따른 계단 보행의 지지국면 시 하지분절의 역학적 변인을 정량적으로 분석하여, 보다 효과적인 계단의 너비를 제시하고자 실시하였으며, 이를 위해 20대의 남자 대학생 10명이 참여하였다. 계단의 높이는 18cm, 최소폭은 90cm로 동일하나 디딤면의 너비가 각각 26cm, 31cm, 36cm인 세 개의 계단을 사용 하였다. 이때 사용된 계단의 세 번째 디딤면에 지면반력기를 설치하여 지지구간의 하지관절 모멘트를 분석한바 다음과 같은 결론을 얻었다. 상향계단보행에서 발목관절의 굴곡/신전 모멘트변화를 분석한 결과 계단의 너비가 증가 할수록 pull-up 단계의 peak 모멘트가 감소하였으며(p<.05), 무릎관절에서는 굴곡/신전 모멘트를 분석한 결과 계단의 너비가 증가할수록 무릎에 작용하는 신전모멘트가 중지지기에 증가하는 것으로 나타났다(p<.05). 그러나 고관절에서 차이가 없는 것으로 나타났다. 그리고 하향계단보행에서 발목관절의 굴곡/신전 모멘트를 분석한 결과 계단의 너비가 증가할수록 지지기 동안 저측굴곡과 배측굴곡의 차이가 확연히 나타나는 것으로 나타났으며(p<.05), 무릎관절과 고관절에서는 굴곡/신전 모멘트를 분석한 결과 계단의 너비에 따라 신전 모멘트의 차이가 크지 않은 것으로 나타났다.

배양된 사람치은각화상피세포의 미세구조 (CULTURED HUMAN ORAL KERATINOCYTES; ULTRASTRUCTURAL STUDY)

  • In oral and maxillofacial surgery, there are many cases requiring the graft of epidermal tissues such as maxillectomy, and vestibuloplasty. There have been so many challenges for the culture of the epidermal tissue. Observing the ultrastructure of the cultured human oral kertinocytes, we could compare this findings with that of in vivo ones. With that, we could find the differencies and similarities between cultured cells and in vivo ones, and evaluate the clinical applications of cultured tissue. Human gingiva was obtained and the specimen was explanted on 24-well plate. Two types of culture media were used in this culture system. One was for the growth of the keratinocytes (Media I), and the other was for the stratification (Media II). Media I had special ingredients for the epidermal growth. Those were 0.5% dimethyl sulfoxide (DMSO), 30ng/ml of epidermal growth factor (EGF), 30ng/ml of cholera toxin, and $5{\mu}g/ml$ of transferrin. We cultured the oral keratinocytes for 3 weeks, and at that time the cultured keratinocytes were processed to prepare the specimen for the TEM study. The results were as follows.; 1. In the phase contrast micrograph, epidermal outgrowth firstly appeared on the 3rd day after explantation, and the growing keratinocytes were activley mitotic, and had polygonal shape and increased N/C ratio. 2. In the phase contrast micrograph, the outer most cells exhibited areas where broad cytoplasmic processes extended out onto the culture subtratum(fan-like appaearance). 3. In the TEM micrographs, the cultured keratinocytes showed stratification. The cells were in elongated form, and there were no morphologic differencies among the layers usually found in the in vivo gingiva. 4. Most of cellular organelles underwent lysis, and keratohyaline granules were seen. Tonofibrils were dispersed in the cytoplasm. 5. The cells were interconnected by desmosomes, and their frequency of distribution was considered to be lower than that of in vivo keratinocytes. 6. We could conclude the cultured oral keratinocytes exhibited signs of terminal differentiation.

Time series analysis of patients seeking orthodontic treatment at Seoul National University Dental Hospital over the past decade

  • Objective: This paper describes changes in the characteristics of patients seeking orthodontic treatment over the past decade and the treatment they received, to identify any seasonal variations or trends. Methods: This single-center retrospective cohort study included all patients who presented to Seoul National University Dental Hospital for orthodontic diagnosis and treatment between January 1, 2005 and December 31, 2015. The study analyzed a set of heterogeneous variables grouped into the following categories: demographic (age, gender, and address), clinical (Angle Classification, anomaly, mode of orthodontic treatment, removable appliances for Phase 1 treatment, fixed appliances for Phase 2 treatment, orthognathic surgery, extraction, mini-plate, mini-implant, and patient transfer) and time-related variables (date of first visit and orthodontic treatment time). Time series analysis was applied to each variable. Results: The sample included 14,510 patients with a median age of 19.5 years. The number of patients and their ages demonstrated a clear seasonal variation, which peaked in the summer and winter. Increasing trends were observed for the proportion of male patients, use of non-extraction treatment modality, use of ceramic brackets, patients from provinces outside the Seoul region at large, patients transferred from private practitioners, and patients who underwent orthognathic surgery performed by university surgeons. Decreasing trends included the use of metal brackets and orthodontic treatment time. Conclusions: Time series analysis revealed a seasonal variation in some characteristics, and several variables showed changing trends over the past decade.

태권도 품새 우수·비 우수선수 간 앞차고 몸돌아 옆차기의 성·패에 따른 균형성 비교 (Comparison between the Balance of Skilled and Less-Skilled Players during Successful and Failed Front Kick and Turning Side Kick Motions)

  • The aim of this study is to identify the appropriate movement for maintaining postural balance during Front Kick and Turning Side Kick motions. To do so, ten Taekwondo athletes: five skilled players(S, body mass: $65.0{\pm}5.8kg$, height: $172.3{\pm}3.7cm$, age: $20.0{\pm}1.2yrs$, career: $9.0{\pm}1.9yrs$) and five less-skilled players(LS, body mass: $67.1{\pm}5.5kg$, height: $173.2{\pm}5.1cm$, age: $19.4{\pm}1.7yrs$, career: $9.6{\pm}1.7yrs$) participated in this study. A three-dimensional motion analysis was performed on the participants using eight infrared cameras and two force plate(sampling frequency of 200 Hz and 2000 Hz for S and LS players, respectively). The participants' motions were divided into: a front-kick phase(P1) and a turning-side-kick phase(P2). For P2(p<.05), the range and root mean square(RMS) of the ground reaction torque and the M-L mean velocity of COP were greater for LS than for S; similarly, for P2(p<.05), the M-L range, A-P range, and velocity of the COP were greater for LS than for S. Further, the M-L range and maximum velocity of the COP was greater for failure than for success(p<.05). The femoral biceps muscle for bending the knee joint was significantly stronger in S than in LS(p<.05). It is expected that these results will be useful in developing a training program for improving the balance and stability of Taekwondo poomsae athletes and improve their front-kick and turning-side-kick motions.

공정 온도에 따른 사면체 비정질 카본 (ta-C) 코팅의 트라이볼로지적 특성연구 (Effects of Process Temperature on the Tribological Properties of Tetrahedral Amorphous Carbon (ta-C) Coating)

  • In this study, mechanical and tribological properties were investigated by varying the process temperature (50, 100, 125 and 150℃) to reduce internal stress. The internal stress reduction by thermal dissociation ta-C coating film with increasing temperature is confirmed through the curvature radius of the ta-C coating according to the temperature of the SUS plate. As the coating temperature increased, the mechanical properties (hardness, modulus, toughness) deteriorated, which is in agreement with the Raman analysis results. As the temperature increased, the sp2 phase ratio increased owing to the dissociation of the sp3 phase. The friction and wear properties are related to the process temperature during ta-C coating. Low friction and wear properties are observed in high hardness samples manufactured at 50℃, and wear resistance properties decreased with increasing temperature. The contact area is expected to increase owing to the decrease of hardness(72 GPa to 39 GPa) and fracture toughness with increasing temperature which accelerated wear because of the debris generated. It was confirmed that at process temperature of over than 100℃, the bond structure of the carbon film changed, and the effect of excellent internal stress was reduced. However, the wear resistance simultaneously decreased owing to the reduction in fracture toughness. Therefore, in order to increase industrial utilization, optimum temperature conditions that reduce internal stress and retain mechanical properties.

적층내 탄성 유도초음파의 모드 특성에 관한 연구 (A Study on Elastic Guided Wave Modal Characteristics in Multi-Layered Structures)

  • 이 연구에서 다층구조물에서 각 모드에 대한 위상속도, 군속도, 감쇠 그리고 파형구조를 구하는 프로그램을 개발하였다. 판의 수와 재료 물성치를 변화시키면서 각 모드의 파형구조를 얻었다. 유체가 닿아 있는 구조물에서 유도파를 이용한 비파괴 검사의 성공여부는 에너지 손실을 최소화하는 모드선정의 최적화이다. 이 연구에서는 자유표면판재의 표면에서 정규화된 두께방향의 변위가 감쇠의 변화를 예측하기 위해서 사용되었으며 감쇠와 파형구조의 관계를 밝혔다. 이것은 유도파의 감쇠를 물이 닿아 있는 경우 복소수근을 찾는 수학적 어려움을 경감하면서 자유표면에서 두께방향 변위의 변화로부터 손쉽게 얻을 수 있다. 이 연구를 통하여 다층구조물에서 보다 민감하고 효율적인 비파괴 검사를 위한 유도파의 모드선정의 최적화 개념을 완성했다.

여름철 냉감성 의류소재 개발을 위한 비스코스 레이온 중심의 직물 제조 및 PCM 가공 (Preparation of Rayon Filament based Woven Fabric and PCM Treatment for Developing Cool Touch Summer Clothing Material)

  • To develop cool touch feeling fabrics for summer clothing material, it was manufactured several compositions of woven fabrics, having rayon multi-filament yarn (non-twisted) as warp and various kinds of yarn, such as viscose rayon multi-filament yarn (twisted), tencel$^{(R)}$ spun yarn, PET high absorbance quick dry filament yarn, and PET based rayon-like yarn, as weft. After preparing the fabrics, basic properties of the fabrics were investigated, such as air-permeability, tensile strength, absorption rate, drying rate, etc. Also, surface warm / cool sensations of the woven fabrics were assessed by Qmax Warm / Cool Touch Tester. It was observed that the fabrics composed of viscose rayon multi-filament yarn (warp) and PET high absorbance quick dry filament yarn (weft) showed excellent surface cool touch sensation-the highest Qmax value. This is because the fabric having flat shaped PET high absorbance quick dry filament shows the largest contact area with Qmax measuring plate. And, the fabric also showed superior high absorbance and quick dry property as expected. In addition, we treated phase change material (PCM) on the surface of the fabric composed of viscose rayon multi-filament yarn (warp) and PET high absorbance quick dry filament yarn(weft) to improve the cool touch feeling. However, the surface cool touch feeling was impaired by resin treated with PCM during the finishing process.

KTP 단결정 성장 및 광학 특성에 관한 연구 (Growth and Optical Property Characterization of KTP Crystal)

  • K$_{6}$P$_4$ $O_{13}$ flux로부터 30$\times$40$\times$70㎣ 크기의 inclusion이 없는 KTP 단결성을 성장시켰다 성장된 KTP 결정의 SHG 출력 특성 측정 결과 위상정합각은 $\theta$=90$^{\circ}$, $\Phi$=23.3$^{\circ}$이었고, $\Phi$방향의 angular acceptance는 약 2$^{\circ}$이었다. KTP 결정내 굴절율 변화에 따른 위상정합각 편차는 최대 0.17$^{\circ}$까지 변하였다. 또한 Nd : YAG와 brewster plate를 채용한 내부공진형의 diode pumped SHG module에 KTP 결정을 삽입하여 20mW의 녹색 laser를 얻었다.
