High-temperature Oxidation of CrZrN Films in Air (CrZrN 박막의 대기 중 고온산화)

  • Films of CrN, $Cr_{40}Zr_9N$, and $Cr_{31}Zr_{16}N$ were deposited on a steel substrate by closed field unbalanced magnetron sputtering, and their oxidation behaviors at $700^{\circ}C$ and $800^{\circ}C$ for up to 60h in air were investigated. All the deposited films were composed of the CrN phase. Zirconium atoms in $Cr_{40}Zr_9N$ and $Cr_{31}Zr_{16}N$ films partially dissolved in the CrN phase. They advantageously refined the columnar structure, reduced the surface roughness, and increased the microhardness. The CrN film displayed relatively good oxidation resistance, owing to the formation of the highly protective $Cr_2O_3$ on its surface. The $Cr_{40}Zr_9N$ and $Cr_{31}Zr_{16}N$ films oxidized to $Cr_2O_3$ as the major phase and ${\alpha}-ZrO_2$ as the minor one. They oxidized primarily by the inward transport of oxygen. The addition of Zr could not increase the oxidation resistance of the CrN film, because the formed $ZrO_2$ that was intermixed in the $Cr_2O_3$-rich oxide layer was oxygen permeable, and developed the compressive stress in the oxide scale owing to the volume expansion during its formation.

Numerical Discussion on Natural Convection in Soils (지반내 자연대류에 대한 수치해석적 논의)

  • Thermal behavior of soils is mainly focused on thermal conduction, and the study of natural convection is very limited. Increase of soil temperature causes natural convection due to buoyancy from density change of pore water. The limitations of the analysis using fluid dynamics for natural convection in the porous media is discussed and a new numerical analysis is presented for natural convection in porous media using THM governing equations fully coupled in the macroscopic view. Numerical experiments for thermal probe show increase in the uncertainty of thermal conductivity estimated without considering natural convection, and suggest appropriate experimental procedures to minimize errors between analytical model and numerical results. Burial of submarine power cable should not exceed the temperature changes of $2^{\circ}C$ at the depth of 0.2 m under the seabed, but numerical analysis for high permeable ground exceeds this criterion. Temperature and THM properties of the seafloor are important design factors for the burial of power cable, and in this case effects of natural convection should be considered. Especially, in the presence of heat sources in soils with high permeability, natural convection due to the variation of density of pore water should be considered as an important heat transfer mechanism.

Effects of Heme Oxygenase System on the Cyclooxygenase in the Primary Cultured Hypothalamic Cells

  • Endogenous carbon monoxide (CO) shares with nitric oxide (NO) a role as a putative neural messenger in the brain. Both gases are believed to modulate CNS function via an increase in cytoplasmic cGMP concentrations secondary to the activation of soluble guanylate cyclase (sGC). Recently CO and NO were proposed as a possible mediator of febrile response in hypothalamus. NO has been reported to activate both the constitutive and inducible isoform of the cyclooxygenase (COX). Thus, we investigated whether CO arising from heme catabolism by heme oxygenate (HO) is involved in the febrile response via the activation of COX in the hypothalamus. $PGE_2$ which is a final mediator of febrile response released from primary cultured hypothalamic cells was taken as a marker of COX activity. $PGE_2$ concentration was measured with EIA kits. Exogenous CO (CO-saturated medium) and hemin (a substrate and potent inducer of HO) evoked an increase in $PGE_2$ release from hypothalamic cells, and these effects were blocked by methylene blue (an inhibitor of sGC). And membrane permeable cGMP analogue, dibutyryl-cGMP elicited significant increases in $PGE_2$release. These results suggest that there may be a functional link between HO and COX enzymatic activities. The gaseous product of hemin through the HO pathway, CO, might play a role through the modulation of the COX activity in the hypothalamus.

  • 저영향개발(Low Impact Development, LID) 시설들은 미국 일본 등 선진국을 중심으로 관련법에 따라 개발 및 적용 되어왔으나 국내에서는 최근에서야 LID 시설을 적용할 수 있는 제도적인 틀을 구축하고 있는 실정이며, 국내 여건에 맞는 LID 시설을 개발하여 그 효용성을 검증하기 까지는 상당한 시간이 소요될 것으로 판단된다. 서울시는 2014년 '서울특별시 빗물관리에 관한 조례 전부개정조례'를 발표함으로써 한국형 LID 시설을 대단위로 적용 하고 검증 할 수 있는 발판을 마련하였다. 서울시의 2014년 전부개정조례에 따르면 시장 및 구청장은 저영향개발 계획 수립의 실효성 확보를 위하여 저영항개발 사전협의 제도를 마련하여 시행하여야 하며, 시장은 저영향개발 지구단위계획을 수립하여야 한다. 이에 따라, 본 연구에서는 실제로 적용 가능한 투수포장 주차장을 설계 할 수 있도록 투수 주차장형 LID 시설을 검증할 수 있는 투수효율성 검증실험단지를 설계 및 구축 하였다. 과거 도심의 우수배제는 중앙 집중형 시스템으로 단기간에 우수를 차집하여 배제하는 방식이었으나, 근래에는 집중형 우수배제 시스템의 위험성, 경제성 그리고 용량한계 등 여러 가지 문제점이 부각되면서 분산형 시스템으로의 전환이 이루어지고 있다. 물순환도시 및 지속가능한 도시 등이 분산형 우수배제 시스템의 예이며, 주차장, 도로, 건물 등 불투수 표면으로부터의 우수를 지면으로 침투 및 침루시키는 방법 등을 활용하여 건전한 물순환을 꾀하고 있다. 침투 및 침루 능력은 각각 포장체 및 포장면 하부구조의 재료와 밀접한 관련이 있으며 재료의 선정은 하부구조의 안정성 확보를 고려하여 선택되어야 한다. 또한 우수 배제를 위한 유공관은 접합점에서 강도를 유지하면서 효율적으로 유수를 배제할 수 있어야 하며, 저류조 설치는 강수의 활용목적에 맞게 선정되어야 한다. 이러한 투수 주차장형 LID 시설은 하나의 시스템으로서 포장체의 재료에 따른 공학적 성질, 하부구조 구축방법 및 재료 선정 그리고 유공관 배열 등에 따라 그 시스템의 거동이 변화하므로 기존에 행해왔던 단순 재료실험으로는 투수성 주차장의 우수배제 시스템을 평가할 수 없다. 따라서 본 연구에서는 이를 검증할 수 있는 투수효율성 검증실험단지 설계 및 구축하였다.

Solubilization of Quercetin , and Permeability Study of Quercetin and Rutin to Rabbit Duodenal Mucosa (퀘르세틴의 가용화 , 퀘르세틴 및 루틴의 토끼 십이지장 점막 투과성)

  • To increase the solubility of quercetin, which is a practically insoluble flavonoid of Ginkgo biloba leaf, the effects of nonaqueous vehicles. Their cosolvents, water-sol uble polymers and modified cyclodextrins (CDs) were observed. Polyethylene glycols, diethyleneglycol monoethyl ether, and their cosolvents with water showed a good solvency toward quercetin. Also the aqueous solutions of povidone, copolyvidone and Cremophor RH 40 was effective in solubilizing quercetin. Complex formation of quercetin with ${\beta}$-cyclodextrin (${\beta}$-CD), dimethyl-${\beta}$-cyclodextiin (DMCD), 2-hydroxypropyl-${\beta}$-cyclodextrin (HPCD) and ${\beta}$-cyclodextrin sulfobutyl ether (SBCD) in water was investigated by solubility method at $37^{\circ}C$. The addition of CDs in water markedly increased the solubility of quercetin with increasing the concentration. AL type phase solubility diagrams were obtained with CDs studied. Solubilizaton efficiency by CDs was in the order of SBCD >> DMCD > HPCD > ${\beta}$-CD. The dissolution rates of quercetin from solid dispersions with copolyvidone, povidone and HPCD were much faster than those of drug alone and corresponding physical mixtures, and exceeded the equilibrium solubility (3.03${\pm}1.72{\mu}$g/ml). The permeation of quercetin through duodenal mucosa did not occur even in the presence of enhancers such as bile salts, but the permeation was observed when the mucus layer was scraped off. This was due to the fact that quercetin had a strong binding to mucin ($58.5{\mu}$g/mg mucin). However rutin was permeable to the duodenal mucosa. The addition of enhancer significantly increased the permeation of rutin in the order of sodium glycocholate.

Expression of vesicular glutamate transporter in transient receptor potential vanilloid 1-positive neurons in the rat trigeminal ganglion

  • Activation of transient receptor potential vanilloid 1 (TRPV1), a calcium permeable channel expressed in primary sensory neurons, induces the release of glutamate from their central and peripheral afferents during normal acute and pathological pain. However, little information is available regarding the glutamate release mechanism associated with TRPV1 activation in primary sensory neurons. To address this issue, we investigated the expression of vesicular glutamate transporter (VGLUT) in TRPV1-immunopositive (+) neurons in the rat trigeminal ganglion (TG) under normal and complete Freund's adjuvant (CFA)-induced inflammatory pain conditions using behavioral testing as well as double immunofluorescence staining with antisera against TRPV1 and VGLUT1 or VGLUT2. TRPV1 was primarily expressed in small and medium-sized TG neurons. TRPV1+ neurons constituted approximately 27% of all TG neurons. Among all TRPV1+ neurons, the proportion of TRPV1+ neurons coexpressing VGLUT1 (VGLUT1+/TRPV1+ neurons) and VGLUT2 (VGLUT2+/TRPV1+ neurons) was 0.4% ± 0.2% and 22.4% ± 2.8%, respectively. The proportion of TRPV1+ and VGLUT2+ neurons was higher in the CFA group than in the control group (TRPV1+ neurons: 31.5% ± 2.5% vs. 26.5% ± 1.2%, VGLUT2+ neurons: 31.8% ± 1.1% vs. 24.6% ± 1.5%, p < 0.05), whereas the proportion of VGLUT1+, VGLUT1+/TRPV1+, and VGLUT2+/TRPV1+ neurons did not differ significantly between the CFA and control groups. These findings together suggest that VGLUT2, a major isoform of VGLUTs, is involved in TRPV1 activation-associated glutamate release during normal acute and inflammatory pain.

Tat-CIAPIN1 protein prevents against cytokine-induced cytotoxicity in pancreatic RINm5F β-cells

  • Cytokines activate inflammatory signals and are major mediators in progressive β-cell damage, which leads to type 1 diabetes mellitus. We recently showed that the cell-permeable Tat-CIAPIN1 fusion protein inhibits neuronal cell death induced by oxidative stress. However, how the Tat-CIAPIN1 protein affects cytokine-induced β-cell damage has not been investigated yet. Thus, we assessed whether the Tat-CIAPIN1 protein can protect RINm5F β-cells against cytokine-induced cytotoxicity. In cytokine-exposed RINm5F β-cells, the transduced Tat-CIAPIN1 protein elevated cell survivals and reduced reactive oxygen species (ROS) and DNA fragmentation levels. The Tat-CIAPIN1 protein reduced mitogen-activated protein kinases (MAPKs) and NF-κB activation levels and elevated Bcl-2 protein, whereas Bax and cleaved Caspase-3 proteins were decreased by this fusion protein. Thus, the protection of RINm5F β-cells by the Tat-CIAPIN1 protein against cytokine-induced cytotoxicity can suggest that the Tat-CIAPIN1 protein might be used as a therapeutic inhibitor against RINm5F β-cell damage.

Consensus channelome of dinoflagellates revealed by transcriptomic analysis sheds light on their physiology

  • Ion channels are membrane protein complexes mediating passive ion flux across the cell membranes. Every organism has a certain set of ion channels that define its physiology. Dinoflagellates are ecologically important microorganisms characterized by effective physiological adaptability, which backs up their massive proliferations that often result in harmful blooms (red tides). In this study, we used a bioinformatics approach to identify homologs of known ion channels that belong to 36 ion channel families. We demonstrated that the versatility of the dinoflagellate physiology is underpinned by a high diversity of ion channels including homologs of animal and plant proteins, as well as channels unique to protists. The analysis of 27 transcriptomes allowed reconstructing a consensus ion channel repertoire (channelome) of dinoflagellates including the members of 31 ion channel families: inwardly-rectifying potassium channels, two-pore domain potassium channels, voltage-gated potassium channels (Kv), tandem Kv, cyclic nucleotide-binding domain-containing channels (CNBD), tandem CNBD, eukaryotic ionotropic glutamate receptors, large-conductance calcium-activated potassium channels, intermediate/small-conductance calcium-activated potassium channels, eukaryotic single-domain voltage-gated cation channels, transient receptor potential channels, two-pore domain calcium channels, four-domain voltage-gated cation channels, cation and anion Cys-loop receptors, small-conductivity mechanosensitive channels, large-conductivity mechanosensitive channels, voltage-gated proton channels, inositole-1,4,5-trisphosphate receptors, slow anion channels, aluminum-activated malate transporters and quick anion channels, mitochondrial calcium uniporters, voltage-dependent anion channels, vesicular chloride channels, ionotropic purinergic receptors, animal volage-insensitive cation channels, channelrhodopsins, bestrophins, voltage-gated chloride channels H+/Cl- exchangers, plant calcium-permeable mechanosensitive channels, and trimeric intracellular cation channels. Overall, dinoflagellates represent cells able to respond to physical and chemical stimuli utilizing a wide range of G-protein coupled receptors- and Ca2+-dependent signaling pathways. The applied approach not only shed light on the ion channel set in dinoflagellates, but also provided the information on possible molecular mechanisms underlying vital cellular processes dependent on the ion transport.

Characteristics of methane and propane leaking gas images (methane과 propane의 누출 Optical Gas Image의 특성연구)

  • In this paper is image characteristics of main gas can be a basic data for the identification of the type of leaking gas and the estimation of the emission quantity in OGI(Optical Gas Image) technology. The purpose of this research is to observe the differences of leaking gas images of the two important hydrocarbons of methane and propane in the industry. We fabricated a wind shield of quartz-based with infrared-permeable properties was prepared and methane and propane were simultaneous emission and then photographed with an infrared OGI camera and we are analyzed it. We have a stable image with windbreak of quartz-based minimizes the effect of wind. As a result of analyzing the image of two hydrocarbons with a leakage gas reference value of 1 L/min, an easily recognizable distances by OGI camera were 6 m for methane and 9 m for propane. In the distances range of 1 to 10 m between the infrared camera and the leaking gas point, the gas plume size of the propane gas was larger and clear than that of the methane gas plume. Compared with the number of points in the image, propane was 3.8 times more than methane.

Development of Improving Water Quality in Eutrophic Lake Using Microalgal Cultivation (미세조류 배양을 이용한 부영양호 내 수질 개선 기술 개발)

  • There are many eutrophic lakes by point and non-point pollution sources such as in dustrial waste water, domestic raw sewage, and mucks. The eutrophic lakes not only cause algal blooms but also destroy the ecosystem in the lakes due to high nutrient concentrations. The purpose of this study was to improve water quality in eutrophic lakes by cultivating microalgae using photobioreactors (PBRs) with selectively permeable mesh (SPM), supplying nutrients in the lake and inhibiting cell leakage by diffusion and water permeability. Chlorella vulgaris, was cultivated using PBRs with SPM installed in Inkyung Lake located in Inha university, Incheon, Korea. When cultivating C. vulgaris, $8.3g/m^2/day$ of average biomass productivity was obtained at 3 days. Furthermore, concentrations of total nitrogen and phosphorus were reduced by 35.7% and 84.2%, respectively, compared to initial condition and water quality in eutrophic lake was improved to oligotrophic environment. These results suggest that microalgal cultivation using PBRs with SPM in the lake could produce microalgal biomass as well as improve water quality by decreasing nutrient concentrations.
