  • 토양으로부터 penicillinase를 강력히 분비하는 방사균속 일주를 분리한 후 본균의 형태학적 특징, 배양상 및 생리화학학적 특성을 조사하여 동정한 결과 Streptomyces bobili 내지 그 류연균으로 동정되었다.

  • The in vitro activity of penicillins(penicillin G, ampicillin, carbenicillin, methicillin and cloxacillin) and gentamicin to Staphylococcus aureus and coagulase-negative staphylococci isolated from bovine mastitic milk samples was determined. The growth of all isolates of staphylococci tested was inhibited by cloxacillin and methicillin at a concentration of $0.78{\mu}g/ml$ and by gentamicin at a minimum inhibitory concentration (MIC) of $1.56{\mu}g/ml$. Of the 140 strains of Staphylococcus aureus isolated 99(70.7%) gave positive reactions for penicillinase on the starch-iodine test. Of 121 isolates of coagulase-negative staphylococci, 58(47.9%) showed penicillinase production, but all the Staph. xylosus lacked the ability to produce penicillinase. MIC of penicillin G of the penicillinase-positive strains was $0.2{\mu}g/ml$ or more. The combination of cloxacillin with gentamicin showed a synergistic effect by inhibiting regrowth of the tested organism.

  • Streptonyces sp. (YS-40) 가 생산하는 penicillinase의 효소학적 성질을 조사한 결과는 다음과 같다. 본 효소의 작용 적용온도는 45$^{\circ}C$이며 최적pH는 5.0부근이었다. 열처리에 대해서는37$^{\circ}C$에서도 불안정하였으며 6$0^{\circ}C$와 8$0^{\circ}C$에서 60분간 처리했을 때 각각 60%, 80%가 실활되었으나 안정 pH범위는 4.5~8.5로서 비교적 넓었다. 금속 ion의 농도가 $10^{-3}$M로 반응액에 첨가 되었을 때 Cu$^{++}$의 경우 25%정도 실활시켰으며 2 $\times$$10^{-3}$M에서는 50% 정도 실활시켰다. SDS에 의해서 강하게 저해되었으며 EDTA, sodium fluoride 에 의해서 강하게 저해되었으나 MIA, L-cystine, urea에 의해서는 저해를 받지 않았다.

  • 토양에서 분리, 선별한 Streptomyces 속의 한 균주가 생산하는 Penicillinase의 최적생산조건을 조사한 결과를 요약하면 다음과 같다. 탄소원으로서는 glucose, 실소원으로는 L-as-paragine이 가장 우수했으며 금속감중에서 Mn C $l_2$를 첨가했을때 효소생성을 37% 증가시켰다. Amino 산중에서 L-leucine이 본 효소생산을 약간 증가시켰으며 L-histidine L-methionine은 크게 저하시켰다. Riboflavine, i-inositol, hesperidine, niacinamide, biotin, DL-$\alpha$- lipoic acid, folic acid가 효소생산을 증가시켰으며 항생물질중에서 cephradine, cephalexin, ampicillin, cloxacillin의 첨가가 효소생성을 크게 증가시켰다. 본 효소생산의 최고 pH는 7.0이며 최적온도는 28$^{\circ}C$였다. 또 500$m\ell$ Erlenmeyer flask에서 175$m\ell$의 배지를 가하여 3 일간 배양했을 때 효소생성이 최고에 도달했다.

  • Penicillin sensor was prepared by immobilizing penicillinase (Pcase) on $H^{+}$-selective carboxylated poly (vinyl chloride) (PVC-COOH) membrane or cellulose filter membrane. The immobilization techniques are as follows. Pcase was immobilized with GTH on $H^{+}$-selective PVC-COOH membrane or some amount of BSA was dropped on that membrane. Another method to make immobilization is to mix type I Pcase with GTH and drop on a cellulose filter membrane. According to immobilization techniques, there were some differences in response properties of enzyme electrodes, however, all electrodes responded to Pcase-resistant penicillin derivatives. Pcase immobilized on cellulose filter membrane with $H^{+}$-selective PVC membrane eletrode was more stable and more sensitive to penicillinase-resistant penicillin derivatives than any other immobilization techniques.

$\beta$-Lactamase 저해능이 있는 방선균의 선별 (Screening of Microorganisms Having Inhibitory Activity against $\beta$-lactamase)

  • Microorganisms having beta-latamase inhibitory activity were selected from soil samples collected from 63 spots throughout the country. Screening procedures consist of two steps. Those are growth inhibition test of penicillinase-producing Staphylococcus aureus by double-layered agar plate containing penicillin G as a substrate, and that of penicillin sensitive Staphylococcus aureus ATCC 6538 in the similiar condition including penicillinase. Finally, a strain was selected from a soil sample of Pa-ju, Kyeong-gi Do. This strain was classified as a Streptomyces sp. by ISP(International Streptomycete Project) and Bergey's manual.

방선균이 분비하는 불용성 Penicillinase (Prodution and Properties of the Insoluble Penicillinase from Streptomyces)

  • 특이하게도 일단 어떤 원인으로든지 변성이 일어나서 침전이 형성되고 난 후에는 그 변성으로 인하여 다시 용해되지 않는 penicillinase를 생성분비하는 Streptomyces 속 균주 AS-727을 토양으로부터 얻었으며, 이 균의 효소생산성과 이 효소의 일반적성질을 검토한 결과는 다음과 같다. 1) 본 효소의 생산을 위해서는 질소원으로서 sodium nitrate, 탄소원으로서 glucose 나 maltose를 사용하여 4 일간 배양하는 것이 바람직했으며 2) 정제는ammonium sulfate로 완전포화시켜 생성된 침전을 원심분리로 모아 그것을 투석해서 조정제효소를 얻는데, 이때 투석할 때 침전은 용해 되지 않는다. 3) 본 효소의 최적 pH는 중성부근 (6.0~8.0)이며 최적온도는 4$0^{\circ}C$ 근처이고, 4) 안정 pH 범위는 4.0~9.0으로서 비교적 넓었으며 열에 대해서는 4$0^{\circ}C$에서는 안정했으며 6$0^{\circ}C$와 8$0^{\circ}C$에서 60 분간 처리하면 각각 약 30%와 40%가 실활되었다. 5) 본 효소의 활성에 있어서 zinc chloride가 $10^{-3}$ M 농도에서 약 20%, $10^{-2}$ M 이상의 고농도에서는 95% 이상을 조해하였다. 6) 그리고 동결이나 명종 고류 또는 유기용매로 일단 형성된 효소단백침전은 다시 용해되지 않는 특이한 성질을 가졌다.

  • MRSI is the staphylococcal infection having resistance to the methicillin which is semisynthetic penicillinase-resistant agents against penicillinase. These infections are very difficult to treat because they have resistance to almost every antibiotics except for vancomycin. We experienced MRSE(methicilline-resistant staphylococcal epidermis) infected 56 years old man who developed 2 months after arthroplasty for TMJ ankylosis and MRSA(methicilline-resistant staphylococcal aureus) infected 59 years old man who was performed arthroplasty far traumatic TMJ disc displacement.

Characterization and Food Application of a Potentiometric Biosensor Measuring $\beta$-Lactam Antibiotics

  • $\beta$-Lactam antibiotics such as penicillin G, amoxicillin, and ampicillin were determined by a potentiometric biosensor system which exploited penicillinase immobilized on Immobilon cellulose nitrate membrane and a flat-bottomed pH electrode-as the biological component and transducer. The optimum reaction buffer for maximum sensitivity was found as 2 mM of sodium phosphate buffer (pH 7.2). The detection limit of the biosensor could be extended to 1 $\mu{M}$ of the analytes by increasing the enzyme loading for immobilization to 100 units/$m\ell$. The model samples spiked with each of the standard penicillins were measured for their biosensor responses and HPLC peak area, resulting in the relative responses of 82.1-103.5% and 79.5-106.1% for the biosensor method along with HPLC analysis, respectively. This result showed a good precision of the current biosensor method for screening the penicillin compounds.

  • Penicillin sensor was manufactured by immobillizing penicillinase with glutaraldehyde on the $H^+$-selective membrane based on PVC-COOH-TDDA. This membrane was not inter fered by $K^+$ ion in Pc-G potassium salt. When enzyme was immobilized with glutaraldehyde, the PVC-COOH matrix was more effective than PVC matrix. Calibration curve calculated from Nernst equation was not linear. But potential was relative to concentration of Pc-G. And maximal potentiometric velocity was also relative to concentration of Pc-G. Therefore, it may be applied to Michaelis-Menten equation. The penicillin sensor was useful for determination of Pc-G at concentration of 0.1~10mM level.
