Scanning Electron Microscopic Observation of Human Skin Replica

  • The skin is the largest organ of the integument system whose surface is closely related with many physiological and pathological conditions. Various methods are used to understand the structural and functional status of human skin. We would like to present usefulness of scanning electron microscopic (SEM) observation of skin replica and its significance of training module for a novice. The silicon replicas from several regions of the body (hand, finger, forearm, lip, and face) were casted by applying Exafine$^{(R)}$ mixture. The positive replicas were prepared by applying EPON 812 mixture on negative silicon replicas. Some of the negative silicon replicas were cut with a razor blade and surface profiles were observed. The negative and positive replicas were coated with platinum and were observed under the scanning electron microscope. We could investigate the detailed structures of the human skin surface without any physical damage to the subject. The positive replicas depicted real surface structure of the human skin vividly. The cross sectional view of the negative silicon replicas provided surface profile clearly. The scanning electron microscopic observation of the human skin replicas would be useful to study skin surface structures and to evaluate medical and esthetical applications.

Influence of Squid Liver Powder on Accumulation of Cadmium in Serum, Kidney and Liver of Mice

  • In this study, the effect of squid liver powder intake on accumulation of cadmium in mice was investigated. Subjects were divided into 4 groups including the control group (CON), squid liver powder group with lipids not removed (SLP100), and squid liver powder groups with lipids removed (LFSLP50 and LFSLP100). Feed intake and food efficiency ratio of squid liver powder groups was significantly higher than the CON. As a result of investigating cadmium content in hair, serum, liver, and kidney during intake of squid liver powder, all groups showed increase in cadmium accumulation through consistent, long-term intake. Especially, cadmium content in liver and kidney of LFSLP100 was significantly higher than the content of SLP100 and CON. As a result of pathological observation on liver and kidney tissues according to squid liver powder diet, LFSLP100 showed most serious pathological symptoms. In case of kidney tissues, degeneration was significantly more severe in LFSLP100 compared to other groups. Such results suggest that cadmium concentration in human body can be increased by ingestion of whole squid including internal organs and that tissues can be damaged by increased cadmium concentration. More specific and systematic studies are deemed necessary.

  • Enterotoxemia caused by Clostridium perfringens(Cl. perfringens)has occurred sporadically with devastating effect in Korean cattle. The disease is confirmed by detecting bacterial toxin the interstinal contents. however diagnosis has been challenging since the toxin is short lived and it must be demonstrated within one hour following death. The purpose of this paper is to provide additional clinicall and pathological information derived from observation of cattle administered with Cl. perfringens type A and/or its toxins isolated from natural case. Clinically cattle died suddenly in lateral recumbency with dyspnea tachycardia and muscle tremor without increased rectal temperature. hematology showed leukopenia with absolute decrease of all leukocytes. Blood glucose AST and ALT were increased. Grossly lungs were hyperemic haemorrhagic and edematous. Small intestine especially ileum was hyperemic and hemorrhagic with diffuse areas of necrosis. This study demonstrated that death without increased rectal temperature combined with leukopenia increased serum glucose pulmonary haemorrhage and edema plus necrotizing ileitis are important features associated with enterotoxemia due to Cl. perfringens type. An Outbreak of Porcine Pleuropneumonia in Korea (돼지 흉막폐렴(胸膜肺炎)의 발생(發生))

  • Authors observed an outbreak of porcine pleuropneumonia at a piggery in Gyeongbug, Korea, which were diagnosed on the ground of clinical and pathological findings with the following results. 1. One hundred and forty weanlings out of 200 ones died suddenly without noticiable signs during the period of 15 days. All of the dead pigs showed formy, blood-tinged discharge from the nose and mouth. 2. Main clinical findings observed from 60 alive pigs from group consisted of fever, cyanosis severe degree of dyspnea and pleuritic friction rub. 3. On hematological observation for typical 5 cases leukocytes showed marked degenerative left shift with leukopenia (10500-13070/cmm), neutropenia (7-13%) and increase in the number of band neutrophils (6-27%). 4. The most prominent and constant pathological changes observed at necropsy for dead or alive pigs consisted of fibrinous pleurisy with partial adhesion, overlying of the pulmonary surface with fibrinous menbrane and necrotic pneumonic lesions with marked edema and congestion of lung parenchyma. 5. From these results the outbreak was considered to be typical porcine pleuropneumonia, although no bacteriological isolation was attempted.

Pathological study on rabbit haemorrhagic disease in young rabbits

  • We investigated the pathological changes in young rabbits which were experimentally infected with rabbit hemorrhagic disease virus (RHDV). Experimental infection of RHDV was carried out in both thymectomized and non-thymectomized young immature rabbits and adult rabbits. None of young rabbits infected with RHDV died during the experiment. Histologically, single or focal hepatocellular degeneration and necrosis with mild lymphocyte infiltration were observed in the rabbits killed at 30 hours and 5 days PI. Lymphocyte infiltration was more severe at 5 days PI than at 30 hours PI. RHDV antigens were mainly detected in the degenerating hepatocytes adjacent to the infiltrated lymphocytes at 30 hours PI and 5 days PI. In electron microscopical observation, infiltrated lymphocytes in the lesions had large nuclei without cytoplasmic granules and interdigitated with adjacent hepatocytes. It is assumed that infiltrated lymphocytes in hepatic lesions in RHDV infected young rabbits are T-lymphocytes and originate from peripheral lymphoid organs or tissues rather than from thymus.

Pathological findings of paraquat poisoning in mice, rats and rabbits (마우스, 랫트 및 토끼에서 paraquat 중독의 병리학적 관찰)

  • This study was carried out to investigate the pathological changes with paraquat(1.1'-dimethyl-4.4'-dipyrildiylium dichloride) administered by intraperitoneally, orally, skin applied in mice, rats and rabbits. Results were obtained as follows; In 2 days after paraquat administration clinical signs anorexia, depression, tachypnea, and tachycardia, respiratory failure, coma and death were observed in mice, rats and rabbits. Compared toxicity of paraquat with mouse were observed toward to paraquat that resistance was strong than rats and rabbits. Also, mortality of skin application were found the low than intraperitoneal and high than oral administration. In the case of gross observation were appear lips moisture in orally administered rats and rabbits by skin application. Lung of all laboratory animals were observed congestion and haemorrhage, swelling or atrophy. In the case of microscopic findings were severe congestion and haemorrhage, interstitial pneumonia of lung. Congestion and haemorrhage of liver, congestion and haemorrhage, renal tubule epithelium necrosis of kidney were observed in mice, rats and rabbits. Skin application group of mice, rats and rabbits showed infiltration of inflammatory cells and folliculitis of epidermis and dermis. Also, in oral administration group showed congestion and haemorrhage, tachment, necrosis of alimentary tract mucosa.

Clinical Observation and Prognostic Analysis of Pemetrexed plus Platinum as First-line Treatment in Patients with Advanced Non-small Cell Lung Cancer

  • Objective: To determine clinical efficacy, safety and prognostic factors of pemetrexed plus platinum as first-line treatment in patients with advanced non-small cell lung cancer (NSCLC). Materials and Methods: Clinical characteristics, short-term efficacy, survival and adverse reactions of 47 advanced non-squamous NSCLC patients who had received pemetrexed plus platinum as first-line treatment in Shanghai Pulmonary Hospital from January 2009 to June 2011 were retrospectively analyzed. The Chi-squared test was applied to statistically analyze the overall response rate (ORR), disease control rate (DCR) and toxicity reactions in both groups, while survival data wereanalyzed by Kaplan-Meier and logrank methods, and the COX proportional hazards model was adopted for a series of multi-factor analyses. Results: Only two patients were lost to follow-up. The ORR, DCR, medium progression-free survival time (PFS) and medium overall survival (OS) were 31.9%, 74.5%, 5 months and 15.2 months, while 1- and 2-year survival rates were 63.8% (30/47) and 19.2% (9/47), respectively. Single-factor analysis showed that tumor pathological patterns and efficacy were in association with medium PFS (P<0.05), whereas tumor pathological patterns, smoking history and efficacy were closely connected with medium OS (P<0.05). Multi-factor analyses demonstrated that pathological patterns and efficacy were independent factors influencing OS (P<0.05). The rate of toxicity reactions in degree III/IV was low, including hematologic toxicity marked by decline in white blood cell count and decrease in the platelet count (PLT), and non-hematologic toxicity manifested by gastrointestinal reactions, such as nausea and vomiting. Conclusions: Pemetrexed plus platinum as first-line treatment has excellent efficacy and slight adverse reactions with favorable drug-tolerance in patients with advanced non-squamous NSCLC.

Light and electron microscopy of the kidney tissues on viral haemorrhagic disease in rabbits (토끼의 바이러스성(性) 출혈증(出血症) 감염시(感染時) 신장(腎臟)의 광학(光學) 및 전자현미경적(電子顯微鏡的) 관찰(觀察))

  • In order to observe the pathological changes of kidney in rabbits infected with the viral haemorrhagic disease, the kidney tissues from the 91 rabbits infected with the viral haemorrhagic disease were examined by light and electron microscopy. The results observed were as follows: 1. On light microscopic observation, the kidney lesions were identified as haemorrhagic glomerular necrosis(33.0%), membranous glomerulonephritis(20.9%), thrombotic glomerulopathy(19.8%), membranoproliferative glomerulonephritis(8.8%), mesangial proliferative glomerulonephritis(8.8%) ischemic acute tubular necrosis(7.7%), and acute serous glomerulitis(6.6%). 2. On electron microscopic observation, cytoplasmic degeneration of mesangial cells, and irregular thickening of basement membranes with electron dense granular materials were observed. In podocytes swelling of mitochondria, dilatation of endoplasmic reticulum and extensive fusion of foot processes were also observed. Nonenveloped round icosahedral picornaviral particles with a diameter of 28~33nm were detected in the cytoplasm of degenerative endothelial cells, polymorphonuclear leucoytes, and monocytes.

Dose Dependence of the Severity of Radiation-Induced Thymic Lymphoma in Mice

  • The dose dependence of the severity of radiation-induced thymic lymphoma in C57BL/6J mice was studied. Mice were exposed to fractionated irradiation at the total doses of 4.0, 6.0 and 8.0 Gy (four irradiations at 8-day intervals) starting from 33 days after birth. Pathological and histological changes of each mouse were observed after periodical sacrifice at day 75, 100, 125, 150, 175, 200, 250, 300 after the first irradiation. The severity of cancers were classified into 4 stages by clinical signs with respect to the enlargement of the thymus, spleen, liver, the progression of the cancer in the thymus, and the metastasis to the spleen, liver, lung and the lymphatic nodes. Among the 490 mice observed, 146 mice had thymic lymphoma. A clear dose-effect relationship was observed as well as the dose-response relationship. Also, periodical observation showed that thymic lymphoma was first induced in mice sacrificed at day 100 (130days old), and metastasize in the order of spleen, lung, liver and then the lymphatic nodes. The results suggest that radiation may be involved not only as a tumor initiator but also as a tumor promoter, and a tumor progression-enhancing agent.

Single Dose Toxicity Study of Buxus Microphylla var. Koreana Nakai Extract in Mice (마우스에서 회양목 추출물의 단회투여독성시험)

  • The single dose toxicity of Buxus microphylla var koreana Nakai was evaluated in ICR mice. Twenty five mice of each sex were randomly assigned to five groups of 5 mice each and were administered singly by gavage at dose of 0, 222, 667, 2,000 and 5,000 mg/kg body weight. After single administration, signs of toxicity were observed every hour for the first 6 hours and every day for 14 days. At the end of 14-day observation period, all animals were sacrificed for gross postmortem examinations. Neither significant toxic signs nor death was observed during the observation period. In addition, no pathological changes were noticed in macroscopic examination at necropsy. These results indicate that the single oral administration of Buxus microphylla var. Koreana Nakai did not cause any toxic effect at the dose of 5,000 mg/kg body weight for both sexes and $LD_{50}$ of Buxus microphylla var. koreana Nakai is greater than 5,000 mg/kg body weight.
