Detection of Tick-Borne Pathogens in the Korean Water Deer (Hydropotes inermis argyropus) from Jeonbuk Province, Korea

  • The objective of this study was to investigate the prevalence of tick-borne pathogens in the Korean water deer (Hydropotes inermis argyropus). Pathogens were identified using PCR which included Anaplasma, Ehrlichia, Rickettsia, and Theileria. Rickettsia was not detected, whereas Anaplasma, Ehrlichia, and Theileria infections were detected in 4, 2, and 8 animals, respectively. The most prevalent pathogen was Theileria. Of the 8 Theileria-positive animals, 2 were mixed-infected with 3 pathogens (Anaplasma, Ehrlichia, and Theileria) and another 2 animals showed mixed-infection with 2 pathogens (Anaplasma and Theileria). Sequencing analysis was used to verify the PCR results. The pathogens found in this study were identified as Anaplasma phagocytophilum, Ehrlichia canis, and Theileria sp. To the best of our knowledge, this is the first report identifying these 3 pathogens in the Korean water deer. Our results suggest that the Korean water deer may serve as a major reservoir for these tick-borne pathogens, leading to spread of tick-borne diseases to domestic animals, livestock, and humans. Further studies are needed to investigate their roles in this respect.

Infestation and Related Ecology of Chigger Mites on the Asian House Rat (Rattus tanezumi) in Yunnan Province, Southwest China

  • This paper is to illustrate the infestation and related ecological characteristics of chigger mites on the Asian house rat (Rattus tanezumi). A total of 17,221 chigger mites were collected from 2,761 R. tanezumi rats, and then identified as 131 species and 19 genera in 2 families. Leptotrombidium deliense, the most powerful vector of scrub typhus in China, was the first major dominant species on R. tanezumi. All the dominant mite species were of an aggregated distribution among different individuals of R. tanezumi. The species composition and infestations of chiggers on R. tanezumi varied along different geographical regions, habitats and altitudes. The species-abundance distribution of the chigger mite community was successfully fitted and the theoretical curve equation was ${\hat{S}}(R)={37e^{-(0.28R)}}^2$. The total chigger species on R. tanezumi were estimated to be 199 species or 234 species, and this further suggested that R. tanezumi has a great potential to harbor abundant species of chigger mites. The results of the species-plot relationship indicated that the chig-ger mite community on R. tanezumi in Yunnan was an uneven community with very high heterogeneity. Wide geographi-cal regions with large host samples are recommended in the investigations of chigger mites.

Characterization of Bacteriocin produced Lactobacillus bulgaricus acting on bovine mastitis pathogens

  • The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by pretense, which confirmed it as a bacteriocin and referred to 'bulgaricin HJ'. The bulgaricin HJ showed the inhibitory activity against mastitis pathogens, gram-positive and gram-negative bacteria. The optimal conditions for the production of bulgaricin HJ were at the temperature of $30^{\circ}C$ and 10 h after cultivation of L. bulgaricus. Staph. and Strep. agalactiae, common bovine mastitis pathogens, were treated with bulgaricin HJ by the agar well diffusion method and showed antimirobial activities to the bovine mastitis pathogens. The activity of the bulgaricin HJ was maintained at pH 6-7 and $100^{\circ}C$ for 60 min against the mastitis pathogens. The bulgaricin HJ was determined as class IV bacteriocin by various enzyme treatments. Colony forming units analysis with indicator strains by the treatments of bulgaricin HJ indicates that the mode of bacteriocin action was bactericidal rather than bacteriostatic.

  • Aim: The purpose of this study was to identify dominant pathogens and their antimicrobial susceptibilities of endodontic infection in type II diabetes mellitus (DM) patients to determine effective empirical antibiotics. Methodology: Pathogens from endodontic infection in six patients with DM and in six patients without DM were cultured, identified and their antimicrobial susceptibility was tested using Vitek2 systems ($bioM{\acute{e}}rieux$, Marcy l' Etoile, France). The results were analyzed using Chi-square test and Fisher's exact test at P< 0.05 level. Results: Pathogens of opportunistic infection were dominant in DM patients (P=0.015). However, there was no significant difference of antimicrobial susceptibility between DM and non-DM patients. Relatively high percent (27%) of pathogens showed resistance to penicillin. Conclusions: More cautions should be paid to DM patients because they are prone to opportunistic infection. Penicillin is not effective in the control of endodontic infection.

  • In this study, we investigated the antimicrobial activity of herbs related with treatments of intestinal diseases against intestinal pathogens under anaerobic broth system. The water extract of Saussurea lappa Clarke and Myristica fragrans Houtt. showed no growth inhibition against tested pathogens(Eubacterium limonsum ATCC 10825, Escherichia coli ATCC 25922, Bacteroides fragilis KCTC 5013, Clostridium perfringens STCC 3627, Staphylococcus aureus KFCC 11764 및 Salmonella typhimurium ATCC 14028). All tested pathogens were not inhibited in broth containing 100$\mu\textrm{g}$/$m\ell$ of Areca catachu L. Water extract but its extract strongly inhibited the growth of Eubacterium limonsum STCC 10825, Bacteroides fragilis KCTC 5013, Clostridium perfringens ATCC 3627 and Salmonella typhimurium ATCC 14028 at 1,000 to 2,000$\mu\textrm{g}$/$m\ell$ of concentration. Escherichia coli ATCC 25922 and Staphylococcus aureus KFCC 11764 hardly grew in broth containing 2,000$\mu\textrm{g}$/$m\ell$ of Terminalia chebula Retz. water extract.

Relationship between Chemical Structure and Antimicrobial Activities of Isothiocyanates from Cruciferous Vegetables against Oral Pathogens

  • We evaluated the potentials of 10 isothiocyanates (ITCs) from cruciferous vegetables and radish root hydrolysate for inhibiting the growth of oral pathogens, with an emphasis on assessing any structure-function relationship. Structural differences in ITCs impacted their antimicrobial activities against oral pathogens differently. The indolyl ITC (indol-3-carbinol) was the most potent inhibitor of the growth of oral pathogens, followed by aromatic ITCs (benzyl ITC (BITC) and phenylethyl ITC (PEITC)) and aliphatic ITCs (erucin, iberin, and sulforaphene). Sulforaphene, which is similar in structure, but has one double bond, showed higher antimicrobial activity than sulforaphane. Erucin, which has a thiol group, showed higher antimicrobial activity than sulforaphane, which has a sulfinyl group. BITC and iberin with a short chain exhibited higher antimicrobial potential than PEITC and sulforaphane with a longer chain, respectively. ITCs have strong antimicrobial activities and may be useful in the prevention and management of dental caries.

Biosensors and their Applications in Food Safety: A Review

  • Background: Foodborne pathogens are a growing concern with respect to human illnesses and death. There is an increasing demand for improvements in global food safety. However, it is a challenge to detect and identify these harmful organisms in a rapid, responsive, suitable, and effective way. Results: Rapid developments in biosensor designs have contributed to the detection of foodborne pathogens and other microorganisms. Biosensors can automate this process and have the potential to enable fast analyses that are cost and time-effective. Various biosensor techniques are available that can identify foodborne pathogens and other health hazards. Conclusions: In this review, biosensor technology is briefly discussed, followed by a summary of foodborne pathogen detection using various transduction systems that exhibit specificity for particular foodborne pathogens. In addition, the recent application of biosensor technology to detect pesticides and heavy metals is briefly addressed.

Gliotoxin is Antibacterial to Drug-resistant Piscine Pathogens

  • By activity-guided fractionation, gliotoxin was isolated as an antibacterial metabolite of the fungus Penicillium decumbens which was derived from the jellyfish Nemopilema nomurai. Gliotoxin was further evaluated for antibacterial activity against several piscine and human MDR (multidrug resistance) pathogens. Gliotoxin showed significant antibacterial activity against Gram-positive piscine pathogens such as Streptococcus iniae FP5228, Streptococcus iniae FP3187, Streptococcus parauberis FP3287, Streptococcus parauberis SPOF3K, S. parauberis KSP28, and Lactococcus garvieae FP5245. Gliotoxin showed strong activity especially against S. parauberis SPOF3K and S. iniae FP5228, which are resistant to oxytetracycline. It is noteworthy that gliotoxin effectively suppressed streptococci which are the major pathogens for piscine infection and mortality in aquaculture industry. Gliotoxin also showed strong antibacterial activity against multidrug- resistant human pathogens (MDR) including Enterococcus faecium 5270 and MRSA (methicillin-resistant Staphylococcus aureus) 3089.

Diarrheagenic pathogens in calves with diarrhea in Chungcheongbuk-do, Korea, from 2018 to 2021

  • An epidemiological study was conducted to investigate five diarrhea-causing pathogens (coronavirus, rotavirus, E. coli, Cryptosporidium, Giardia) using a rapid diagnostic kit in Hanwoo calves with diarrhea in Chungcheongbuk-do, Korea, from 2018 to 2021. A total of 22,417 fecal samples were collected from calves under 1 year of age; of those, 13,518 (60.3%) were positive for five bovine diarrhea antigens. The antigen positivity rates for rotavirus, coronavirus, E. coli, Giardia, and Cryptosporidium were 34.5%, 11.0%, 8.2%, 4.7%, and 2.0%, respectively. The prevalence of the five pathogens in calves was statistically higher in autumn and winter. The highest prevalence of the pathogens was observed in the under 1 month age group, and the incidence of diarrhea decreased with age. Rotavirus was a major pathogen in calves under 1 month of age, whereas the prevalence of E. coli increased with age. This study provides epidemiological evidence of the prevalence of calf diarrheal pathogens in Chungcheongbuk-do, Korea, which will facilitate early diagnosis and development of measures against calf diarrhea.

Screening of antifungal activities of Bacillus thuringiensis strains for the development of biocontrol agents of plant diseases

  • An attempt was made to screen antifungal activities of Bacillus thuringiensis strains on various plant pathogens, Botryosphaeria dothidea, Diaporthe actinidiae, Botrytis cinerea, Glomerella cingulata, Colletorichum cocodes, Sclerotinia scierotiorum, Alternaria alternata, Helicobuidium mompa, Bipolaris coicis, Fusarium graminearum and Rhizoctosnia solani. Ten and forty-five strains of B. thuringiensis were isolated from animal feces in Korea and Japan, respectively. Inhibitory effects of the strains on the mycelial growth of the pathogens were examined on the mixed media of potato dextrose agar and nutrient agar. Approximately half of the strains inhibited the mycelial growth of one or more pathogens. Most of the pathogens were inhibited by any of the strains but Fusarium graminearum and Rhizoctonia solani were not inhibited at all. This is the first report that B. thuringiensis shows a potent antifungal activity on plant pathogens in Korea.
