건식형 저염 비빔밥에 첨가된 민들레 복합분말의 기호도 및 향균효과 (Acceptance and Antibacterial Effects of Dandelion Compound Powder on Dried Type Sodium Reduced Bibimbap)

  • A reduced salt, dry-type bulgogi bibimbap with AF-343 was prepared, and the acceptance, antioxidant antimicrobiological safety were investigated. The acceptance of the reduced salt and the AF-343 added sample, RW, showed the highest value of 5.37 among the samples, but it did not differ significantly from the other samples of the standard sample group. The acceptances of the appearance, aroma, texture, and overall acceptance were not significantly different in all samples (p>0.05). In the DPPH radical scavenging activity, the values of SW and RW with the AF-343 were 15.05 and 14.77, respectively, which were significantly higher than the 9.57 and 10.42 obtained for SWO and RWO without AF-343, respectively (p<0.05). Pathogenic microorganisms tests that were either negative or not detected in any of the samples representing hygiene safety were met in the dried bibimbap against Escherichia coli, pathogenic microorganisms, and general bacteriological tests. The aerobic plate counts were 30.0 CFU/g, indicating that the dry type salt-reduced bibimbap containing AF-343 was within the safety range that meets the safety standards of microorganisms in the food code for commercialization. Therefore, high-quality dry-type reduced salt bibimbap with AF-343 is helpful for antioxidant action and effective for skin moisturizing and can be produced without affecting the taste and palatability.

2,3,5-Triphenyltetrazolium Chloride를 이용한 병원성 미생물 확인시험에 관한 연구 (Study of 2,3,5-Triphenyltetrazolium Chloride for Detection of Pathogenic Microorganisms)

  • 2,3,5-Triphenyltetrazolium chloride (TTC)는 산화환원지시약으로 미생물의 증식에 의한 산소 소비를 쉽게 확인할 수 있다. 용해 후 무색의 형태를 띠고 있으나 생리활성이 있는 조직에서는 탈수소 효소(dehydrogenases)에 의해 환원되어 빨간색의 불용성 1,3,5,-triphenylformazan (TPF)가 된다. 본 연구에서는 병원성 미생물(Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Enterococcus faecium, Candida albicans)에 TTC 지시약을 활용하여 미생물 성장시험에 대해 확인하였다. 시험 균주에 TTC를 첨가하여 확인한 결과, 모두 탈수소효소 반응으로 인한 TPF 형성으로 붉은색 콜로니를 관찰하였다. 이후 TTC 0.04% 이상의 농도 및 12 h 이상 배양조건으로 최적화 실험 후 균주별 CFU 값을 통해 TPF 발현능을 확인하였다. 결국 TTC가 병원성 세균 및 효모균 성장에 큰 영향을 끼치지 않으며 배양 시 세균의 경우 12 h, 효모균의 경우 48 h 이후부터 확인이 가능하였다. 이러한 결과들로부터 TTC를 활용한 미생물 성장 확인 시험법이 더 신속 정확한 방법으로 화장품 연구에 활용 가능할 것으로 사료된다.

Seasonal Dynamics of Pathogenic Microorganisms (Cryptosporidium, Giardia and Fecal Bacteria) in an Artificial Lake Ecosystem (Sangsa Lake, Korea)

  • This study was performed for the purpose of monitoring monthly levels of two pathogenic microorganisms, Cryptosporidium and Giardia, from November 2005 to August 2007 in Sangsa Lake. Water temperatures, pH and DO fluctuated seasonally at the study site. Annual mean values of BOD, COD and SS were $0.8\;mg\;L^{-1}$, $2.3\;mg\;L^{-1}$ and $1.9\;mg\;L^{-1}$ respectively. Although there was distinct seasonal variation in water chemistry and chlorophyll $\underline{a}$ concentration, the lake generally contains low concentrations of nutrients and chlorophyll $\underline{a}$. The relative abundance of coliform bacteria was always greater than that of fecal coliform. The fecal coliform bacteria comprised $8.5{\sim}22.1%$ of total coliform bacteria. Seasonal analysis of Cryptosporidium and Giardia levels in the study site showed that in winter (November through February), Cryptosporidium oocysts and Giardia cysts were most abundant ($1.1{\sim}1.8\;{\times}\;10\;cells\;L^{-1}$ and $3.8{\sim}5.1\;{\times}\;10\;cells\;L^{-1}$, respectively), while in summer (July through September) the abundance was lowest ($0.0{\sim}0.3\;{\times}\;10\;cells\;L^{-1}$ and $0.9{\sim}2.9\;{\times}\;10\;cells\;L^{-1}$, respectively). Molecular identification revealed two subtypes of Cyrptosporidium parvum in Sangsa Lake.

  • Bacterial infection is a very complex process in which both pathogenic microorganisms and host cells play crucial roles, and it is the outcome of interactions between the two participants. To elucidate the bacterial pathogenesis mechanisms, therefore, it is essential to understand the cellular and systemic responses of the host as well as the virulence factors of the pathogen. 신기술 이용 식중독균 신속검출법 개발 동향 분석 (Trends in Rapid Detection Methods for Food-borne pathogenic Microorganisms by Using New Technologies)

  • 식품으로부터 식중독세균을 검출하는 대부분의 전통적인 방법들은 분리하고자 하는 미생물에 적합한 선택배지에 의존할 수밖에 없다. 이러한 선택배지들은 높은 경제성에도 불구하고 오래 걸려 때를 놓치게 되는 소요 시간이 항상 걸림돌로 작용하고 있으며, 하나의 선택배지로 한의 균만을 선택적으로 분리해 내는 등의 단점이 있다. 이러한 단점을 최소화하기 위하여 IMS 등 면역학적 또는 분자생물학적 기법들을 접목시키고 있고, 증균에서 분리와 확인을 동시에 할 수 있는 package 형태의 제품들이 시판되고 있다. 또한 이러한 제품 중 일부는 국제표준기구로부터 인증을 받았고 대부분 chromogenic 기질을 함유한 선택배지들이 포함되어 있을 만큼 신뢰도가 높다고 할 수 있다. 그러나 선택배지 자체만은 이들 단점을 해결할 수 없어 항체를 활용한 신속, 간편하고 환경친화적인 ELISA법, 검출장비가 불필요하여 가장 신속, 간편하게 현장에서 사용하도록 기술을 향상시킨 paper-strip kit나 ELISA 분석 시 직면하는 시료의 matrix effect를 최소화할 수 있다는 장점을 지닌 형광면역분석법(fluoroimmunoassay)등 새로운 기술이 용도에 맞게 다양하게 개발되고 있다. 또한 동시진단 능력이 우수한 현장 진단형 시스템이 개발될 경우, 향후 신 시장 창출, 바이오칩 기술발전, 식중독균을 포함한 위해 미생물의 동시진단 및 역학조사 등에 지대한 기여를 할 것으로 판단된다. 식품에서 식품위해미생물 검출에 있어서 가장 이상적인 방법은 검출 과정이 간단하고 경제적이며 정확하여야 한다는 것이다. 또한 정량적 결과 산출이 가능하여야 하고, 자동화된 방법으로 전환이 가능하여야 산업에 적용될 수 있다는 것인데, 이러한 이상적인 검출방법은 현재까지 존재하지 않는다. 다만, 여러 방법들이 각각의 장점을 갖고 있을 뿐이다. 즉, 현실적 대안은 현재까지 개발된 각각의 방법을 목적과 대상에 따라 시의적절하게 병용 사용하여야 한다는 것이다.

Antimicrobial Effect of the Wood Vinegar from Cryptomeria japonica Sapwood on Plant Pathogenic Microorganisms

  • The antimicrobial effect of the wood vinegar of C. japanica sapwood and its constituents was evaluated against Ralstonia salanacearum, Phytophthora capsid, Fusarium oxysporum, and Pythium splendens. Phenols and guaiacols had a strong antimicrobial effect against four kinds of microorganisms, but methanol and acetic acid exhibited little or no antimicrobial activity.

신선계육의 유통을 위한 Ozone 처리 효과 (Ozone Treatment for Circulation of Fresh Poultry Meat)

  • The utilization of ozone as a disinfectant for removing poultry meat microorganisms and then cleaning the poultry rinse water was investigated. When microbial suspensions were treated with ozone at 2, 500ppm/min for 40min, microorganisms were not detectable perfectly. The bacteriocidal effect of ozone by temperature was enhanced greater at 7$^{\circ}C$ than $25^{\circ}C$. All poultry meat microorganisms were killed by ozone treatment at 1, 530ppm for 50min. The pathogenic bacteria such as Salmonella sp. were more vulnerable and not detected by ozone treatment for 20min. Ozonation of the suspension for 20min and 50min increased light transmission at 500nm to 58% and 145%, respectively. The order of COD removal was ozone treatment(21%), coagulant((Al)2SO4) treatment(41%), ozone treatment after coagulant treatment(54%).

Enhanced Production, Purification, and Partial Characterization of Lacticin BH5, a Kimchi Bacteriocin Produced by Lactococcus lactis BH5

  • Strain BH5 was isolated from naturally fermented Kimchi and identified as a bacteriocin producer, which has bactericidal activity against Micrococcus flavus ATCC 10240. Strain BH5 was identified tentatively as Lactococcus lactis by the API test and some characteristics. Lactococcus lactis BH5 showed a broad spectrum of activity against most of the non-pathogenic and pathogenic microorganisms tested by the modified deferred method. The activity of lacticin BH5, named tentatively as the bacteriocin produced by Lactococcus lactis BH5, was detected at the mid-log growth phase, reached its maximum during the early stationary phase, and decreased after the late stationary phase. Lacticin BH5 also showed a relatively broad spectrum of activity against non-pathogenic and pathogenic microorganisms as tested by the spot-on-lawn method. Its antimicrobial activity on sensitive indicator cells was completely disappeared by protease XIV or ${\alpha}$-chymotrypsin. The inhibitory activities of lacticin BH5 were detected during treatments up to 100$^{\circ}C$ for 30 min. Lacticin BH5 was very stable over a pH range of 2.0 to 9.0 and was stable with all the organic solvents examined. The cell concentration and bacteriocin production in strain BH5 were maximum when grown at 30$^{\circ}C$ in a modified MRS medium supplemented with 0.5% tryptone, 1.0% yeast extract, and 0.5% beef extract as nitrogen sources. It demonstrated a typical bactericidal mode of inhibition against Micrococcus flavus ATCC 10240. Lacticin BH5 was purified through ammonium sulfate precipitation, ethanol precipitation, and CM-Sepharose column chromatography. The apparent molecular mass of lacticin BH5 was estimated to be in the region of 3.7 kDa, by the direct detection of bactericidal activity after SDS-PAGE. Mutant strain NO141 which was isolated by nitrosoguanidine mutagenesis produced about 4 fold more bacteriocin than the wild type.

활혈거어약(活血祛瘀藥)이 질내(膣內) 미생물(微生物)에 미치는 영향(影響) (Antibiotic Effects of blood-activating stasis-dispelling medicinals on Vaginal Microorganisms)

  • Purpose : The aim of this study is to investigate the antibiotic effects of 14 herbs among blood-activating stasis-dispelling medicinals on vaginal microorganisms. Methods : Staphylococcus aureus, Methicillin-resistant Staphylococcus aureus, Candida albicans, and Gardnerella vaginalis were used for vaginal pathogenic microorganisms. Lactobacillus gasseri, Streptococcus spp. and Escherichia coli HB101 were used for vaginal normal flora. The blood-activating stasis-dispelling medicinals, Mucunae Caulis, Salviae Radix, Persicae Semen, Myrrha, Zedoariae Rhizoma, Achuranthis Radix, Leonuri Herba, Melandrii Herba, Gleditsiae Spina, Lycopi Herba, Scirpi Rhizoma, Caesalpiniae Lignum, Corydlais Tuber and Polygoni Cuspidati Radix were used in this study. In vitro antibiotic activities were observed by optical density and colony test. Results : The optical density and colony test showed that Gleditsiae Spina, Scirpi Rhizoma, Corydlais Tuber, Polygoni Cuspidati Radix and Melandrii Herba of herbs among blood-activating stasis-dispelling medicinals had antimicrobial effect. Gleditsiae Spina had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis and MRSA. Scirpi Rhizoma had antimicrobial susceptibility and selective toxicity in Staphylococcus aureus and MRSA. Corydlais Tuber had antimicrobial susceptibility and selective toxicity in MRSA. Polygoni Cuspidati Radix had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis, Staphylococcus aureus and MRSA. Melandrii Herba had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis. Conclusion : According to the above results, we could suggest that Gleditsiae Spina, Scirpi Rhizoma, Corydlais Tuber, Polygoni Cuspidati Radix and Melandrii Herba of herbs among blood-activating stasis-dispelling medicinals be available to antimicrobial agent of vaginal pathogenic microbial species in vitro.
