Efficient Enantioselective Synthesis of (R)-[3,5-Bis(trifluoromethyl)phenyl] Ethanol by Leifsonia xyli CCTCC M 2010241 Using Isopropanol as Co- Substrate

  • (R)-[3,5-Bis(trifluoromethyl)phenyl] ethanol is a key chiral intermediate for the synthesis of aprepitant. In this paper, an efficient synthetic process for (R)-[3,5- bis(trifluoromethyl)phenyl] ethanol was developed via the asymmetric reduction of 3,5-bis(trifluoromethyl) acetophenone, catalyzed by Leifsonia xyli CCTCC M 2010241 cells using isopropanol as the co-substrate for cofactor recycling. Firstly, the substrate and product solubility and cell membrane permeability of biocatalysts were evaluated with different co-substrate additions into the reaction system, in which isopropanol manifested as the best hydrogen donor of coupled NADH regeneration during the bioreduction of 3,5-bis(trifluoromethyl) acetophenone. Subsequently, the optimization of parameters for the bioreduction were undertaken to improve the effectiveness of the process. The determined efficient reaction system contained 200mM of 3,5-bis(trifluoromethyl) acetophenone, 20% (v/v) of isopropanol, and 300 g/l of wet cells. The bioreduction was executed at $30^{\circ}C$ and 200 rpm for 30 h, and 91.8% of product yield with 99.9% of enantiometric excess (e.e.) was obtained. The established bioreduction reaction system could tolerate higher substrate concentrations of 3,5- bis(trifluoromethyl) acetophenone, and afforded a satisfactory yield and excellent product e.e. for the desired (R)-chiral alcohol, thus providing an alternative to the chemical synthesis of (R)-[3,5-bis(trifluoromethyl)phenyl] ethanol.

High-Throughput In Vitro Screening of Changed Algal Community Structure Using the PhotoBiobox

  • In a previous study, the sequential optimization and regulation of environmental parameters using the PhotoBiobox were demonstrated with high-throughput screening tests. In this study, we estimated changes in the biovolume-based composition of a polyculture built in vitro and composed of three algal strains: Chlorella sp., Scenedesmus sp., and Parachlorella sp. We performed this work using the PhotoBiobox under different temperatures (10-36℃) and light intensities (50-700 μmol m-2 s-1) in air and in 5% CO2. In 5% CO2, Chlorella sp. exhibited better adaptation to high temperatures than in air conditions. Pearson's correlation analysis showed that the composition of Parachlorella sp. was highly related to temperature whereas Chlorella sp. and Scenedesmus sp. showed negative correlations in both air and 5% CO2. Furthermore, light intensity slightly affected the composition of Scenedesmus sp., whereas no significant effect was observed in other species. Based on these results, it is speculated that temperature is an important factor in influencing changes in algal polyculture community structure (PCS). These results further confirm that the PhotoBiobox is a convenient and available tool for performance of lab-scale experiments on PCS changes. The application of the PhotoBiobox in PCS studies will provide new insight into polyculture-based ecology.

  • A design method of fuzzy modeling is presented for the model identification of route choice of traffic problems.The proposed fuzzy modeling implements system structure and parameter identification in the eficient form of""IF..., THEN-.."", using the theories of optimization theory, linguistic fuzzy implication rules. Three kinds ofmethod for fuzzy modeling presented in this paper include simplified inference (type I), linear inference (type 21,and proposed modified-linear inference (type 3). The fuzzy inference method are utilized to develop the routechoice model in terms of accurate estimation and precise description of human travel behavior. In order to identifypremise structure and parameter of fuzzy implication rules, improved complex method is used and the least squaremethod is utilized for the identification of optimum consequence parameters. Data for route choice of trafficproblems are used to evaluate the performance of the proposed fuzzy modeling. The results show that the proposedmethod can produce the fuzzy model with higher accuracy than previous other studies -BL(binary logic) model,B(production system) model, FL(fuzzy logic) model, NN(neura1 network) model, and FNNs (fuzzy-neuralnetworks) model -.fuzzy-neural networks) model -.

Core-loss Reduction on Permanent Magnet for IPMSM with Concentrated Winding (집중권을 시행한 영구자석 매입형 동기전동기의 철손 저감)

  • Interior Permanent Magnet Synchronous motors (IPMSM) with concentrated winding are superior to distributed winding in the power density point of view. But it causes huge amount of eddy current losses on the permanent magnet. This paper presents the optimal permanent magnet V-shape on the rotor of an interior permanent magnet synchronous motor to reduce the core losses and improve the performance. Each eddy current loss on permanent magnet has been investigated in detail by using FEM (Finite Element Method) instead of equivalent magnetic circuit network method in order to consider saturation and non-linear magnetic property. Simulation-based design of experiment is also applied to avoid large number of analyses according to each design parameter and consider expected interactions among parameters. Consequently, the optimal design to reduce the core loss on the permanent magnet while maintaining or improving motor performance is proposed by an optimization algorithm using regression equation derived and lastly, it is verified by FEM.

  • This paper presents an integrated optimum design and a cost effectiveness evaluation method of a viscoelastically damped structural system. The criterion selected for the optimization is the minimization of the life?cycle cost which is a function of structural sizing variables and the amount of the viscoelastic damper. A genetic algorithm is used as a numerical searching technique in order to simultaneously find the optimum parameters of the integrated system. Optimal distributions of design variables according to various seismic characteristics are investigated by applying the proposed design method to a numerical example of a 10?story building structure. The cost effectiveness of viscoelastically damped structural system is also evaluated by comparing the life-cycle cost of the structure without viscoelastic dampers. The results show that the viscoelastic damper is effective in a region of low to moderate seismicity.

  • The optical feedforward is known as a complicated technique in spite of its superior linearization. Here we optimized the feedforward circuit parameters using a systematic approach for optimum efficiency. A systematic approach was verified by transmission experiments of 2.14 GHz two tones. The compared third order IMD was enhanced by 38 dB for two-tone case. The experimental results for transmission of W-CDMA signals were described. The ACPR was enhanced by 20 dB for 1 W-CDMA carrier and by 16 dB for 3 W-CDMA carriers. Also, noise level was reduced by 7 dB.

  • In this study, we measure the radiation dose for every experiments performed during the training education in radiologic science and estimate the radiation dose to each participant in the training education to propose a safe curriculum including operation of the training education. In this paper, we optimized the three parameters and the results show the dramatically reduced radiation dose to each participant. The proposed arrangement of the subjects and operation of the training education will be very helpful to reorganize the curriculum and the subject operation and will protect the students from the radiation dose.

  • Data clustering is one of the most difficult and challenging problems and can be formally considered as a particular kind of NP-hard grouping problems. The K-means algorithm is one of the most popular and widely used clustering method because it is easy to implement and very efficient. However, it has high possibility to trap in local optimum and high variation of solutions with different initials for the large data set. Therefore, we need study efficient computational intelligence method to find the global optimal solution in data clustering problem within limited computational time. The objective of this paper is to propose a combined artificial bee colony (CABC) with K-means for initialization and finalization to find optimal solution that is effective on data clustering optimization problem. The artificial bee colony (ABC) is an algorithm motivated by the intelligent behavior exhibited by honeybees when searching for food. The performance of ABC is better than or similar to other population-based algorithms with the added advantage of employing fewer control parameters. Our proposed CABC method is able to provide near optimal solution within reasonable time to balance the converged and diversified searches. In this paper, the experiment and analysis of clustering problems demonstrate that CABC is a competitive approach comparing to previous partitioning approaches in satisfactory results with respect to solution quality. We validate the performance of CABC using Iris, Wine, Glass, Vowel, and Cloud UCI machine learning repository datasets comparing to previous studies by experiment and analysis. Our proposed KABCK (K-means+ABC+K-means) is better than ABCK (ABC+K-means), KABC (K-means+ABC), ABC, and K-means in our simulations.

Indoor 3D Modeling Approach based on Terrestrial LiDAR (지상라이다기반 실내 3차원 모델 구축 방안)

  • Terrestrial LiDAR emerges as a main mapping technology for indoor 3D cadastre, cultural heritage conservation and, building management in that it provides fast, accurate, and reliable 3D data. In this paper, a new 3D modeling method consisting of segmentation stage and outline extraction stage is proposed to develop indoor 3D model from the terrestrial LiDAR. In the segmentation process, RANSAC and a refinement grid is used to identify points that belong to identical planar planes. In the outline tracing process, a tracing grid and a data conversion method are used to extract outlines of indoor 3D models. However, despite of an improvement of productivity, the proposed approach requires an optimization process to adjust parameters such as a threshold of the RANSAC and sizes of the refinement and outline extraction grids. Furthermore, it is required to model curvilinear and rounded shape of the indoor structures.

Disinfection of E. coli Using Electro-UV Complex Process: Disinfection Characteristics and Optimization by the Design of Experiment Based on the Box-Behnken Technique (전기-UV 복합 공정을 이용한 E. coli 소독 : 실험계획법중 박스-벤켄법을 이용한 소독 특성 및 최적화)

  • The experimental design and response surface methodology (RSM) have been applied to the investigation of the electro-UV complex process for the disinfection of E. coli in the water. The disinfection reactions of electro-UV process were mathematically described as a function of parameters power ($X_1$), NaCl dosage ($X_2$), initial pH ($X_3$) and disinfection time ($X_4$) being modeled by use of the Box-Behnken technique. The application of RSM using the Box-Behnken technique yielded the following regression equation, which is an empirical relationship between the residual E. coli number and test variables in actual variables: Ln (CFU) = 23.57 - 0.87 power - 1.87 NaCl dosage - 2.13 pH - 2.84 time - 0.09 power time - 0.07 NaCl dosage pH + 0.14 pH time + 0.03 $power^2$ + 0.47 NaCl $dosage^2$ + 0.20 $pH^2$+ 0.33 $time^2$. The model predictions agreed well with the experimentally observed result ($R^2$ = 0.9987). Graphical response surface and contour plots were used to locate the optimum point. The estimated ridge of maximum response and optimal conditions for the E. coli disinfection using canonical analysis was Ln 1.06 CFU (power, 15.40 W; NaCl dosage, 1.95 g/L, pH, 5.94 and time, 4.67 min). To confirm this optimum condition, the obtained number of the residual E. coli after three additional experiments were Ln 1.05, 1.10 and Ln 1.12. These values were within range of 0.62 (95% PI low)~1.50 (95% PI high), which indicated that conforming the reproducibility of the model.
