Extractions of Surface-Active Substances from Defatted Rapeseed Meal (Brassica napus L.) by Supercritical Carbon Dioxide (초임계 CO2 유체 추출법을 이용한 탈지 유채박 중 표면활성물질 추출의 최적화)

  • In this study, an attempt is being made to extract surface-active substances from defatted rapeseed cakes by supercritical carbon dioxide fluid. Independent variables for the extraction process, being formulated by D-optimal design, are pressure (150~350 bar), temperature ($33{\sim}65^{\circ}C$ and co-solvent (ethanol, 50~250 g). The dependent variables of the extraction yield, the content of neutral lipids, phospholipids and glycolipids in the extracts were analyzed upon the results through the response surface methodology. As for the extraction yield, it was found to increase with increasing independent variables, among which the co-solvent proved to be a major influencing parameter. Similar trends were found for the content of surface-active substances (i.e, phospholipids and glycolipids) in the extracts, except for the content of neutral lipids. Regression equations were suggested to coincide well with the results from the experiments. Extraction conditions are being optimized to maximize the extraction yields, the content of phospholipids, and glycolipids were 350 bar (pressure), $65^{\circ}C$ (temperature) and 228.55 g (co-solvent), respectively.

Effects of High Dose Gamma Irradiation on Shelf Stability and Lipid Oxidation of Marinated and Precooked Pork Rib Steak (고선량 감마선 조사가 즉석 취식용 양념 돼지 갈비 구이의 저장성 및 지질 산화에 미치는 영향)

  • This study was conducted to evaluate the application of high dose irradiation for ensuring shelf stability of marinated and precooked pork rib steak in the severe environments such as desert or space, etc. Marinated and precooked pork rib steak was manufactured, vacuum-packaged and gamma-irradiated with the absorbed doses of 10, 20, 30, 40 and 50 kGy, and used for the tests of the growth of microorganisms and lipid oxidation during storage at 35t of acceleration condition. Any growth of microorganisms was not observed in irradiated samples after irradiation immediately. However, the growths were observed in 10, 20 and 30 kGy samples at 4, 7 and 14 day storage, respectively. High dose (40 and 50 kGy) gamma irradiation retarded the growth of aerobic microorganisms by the analysis of kinetic parameter. The content of malondialdehyde increased in all samples during storage periods, and gamma irradiation accelerated the increase of lipid oxidation. Therefore, the application of combination of the various food processing technology should be considered for the sterilization of marinated and precooked pork rib steak without any deterioration of the quality occurred by high dose irradiation.

The Topology of Galaxy Clustering in the Sloan Digital Sky Survey Main Galaxy Sample: a Test for Galaxy Formation Models

  • We measure the topology of the galaxy distribution using the Seventh Data Release of the Sloan Digital Sky Survey (SDSS DR7), examining the dependence of galaxy clustering topology on galaxy properties. The observational results are used to test galaxy formation models. A volume-limited sample defined by Mr<-20.19 enables us to measure the genus curve with amplitude of G=378 at 6h-1Mpc smoothing scale, with 4.8% uncertainty including all systematics and cosmic variance. The clustering topology over the smoothing length interval from 6 to 10h-1Mpc reveals a mild scale-dependence for the shift and void abundance (A_V) parameters of the genus curve. We find strong bias in the topology of galaxy clustering with respect to the predicted topology of the matter distribution, which is also scale-dependent. The luminosity dependence of galaxy clustering topology discovered by Park et al. (2005) is confirmed: the distribution of relatively brighter galaxies shows a greater prevalence of isolated clusters and more percolated voids. We find that galaxy clustering topology depends also on morphology and color. Even though early (late)-type galaxies show topology similar to that of red (blue) galaxies, the morphology dependence of topology is not identical to the color dependence. In particular, the void abundance parameter A_V depends on morphology more strongly than on color. We test five galaxy assignment schemes applied to cosmological N-body simulations to generate mock galaxies: the Halo-Galaxy one-to-one Correspondence (HGC) model, the Halo Occupation Distribution (HOD) model, and three implementations of Semi-Analytic Models (SAMs). None of the models reproduces all aspects of the observed clustering topology; the deviations vary from one model to another but include statistically significant discrepancies in the abundance of isolated voids or isolated clusters and the amplitude and overall shift of the genus curve. SAM predictions of the topology color-dependence are usually correct in sign but incorrect in magnitude.

Genetic parameters of milk β-hydroxybutyrate acid, milk acetone, milk yield, and energy-corrected milk for Holstein dairy cattle in Korea (국내 Holstein종에서 milk β-hydroxybutyrate acid, milk acetone, 에너지 보정유량 및 산유량의 유전모수 추정)

  • This study was conducted to estimate the genetic parameters for common ketosis indicators (${\beta}$-hydroxybutyrate acid, BHBA; milk acetone), feed intake efficiency indicator (energy-corrected milk, ECM), and milk yield (MY) in Korean Holstein. A total of 75,072 monthly test-day records from 14,397 first parity cows were collected, between 2012 and 2016, from Korea animal improvement association enrolled farms. Variance components were estimated using a multiple trait random regression model. The heritability of BHBA and acetone levels ranged from 0.06 to 0.15 at different DIMs. The phenotypic and genetic correlations between BHBA and acetone were between 0.73 and 0.90, and between 0.93 and 0.98, respectively. The phenotypic correlation between BHBA and MY, between acetone and MY, between BHBA and ECM, and between acetone and ECM ranged from -0.18 to -0.05, -0.23 to -0.05, 0 to 0.10, and -0.09 to 0.01, respectively. Genetic correlation estimates between BHBA and MY, between acetone and MY, between BHBA and ECM, and between acetone and ECM also ranged from -0.55 to 0.05, -0.62 to -0.04, -0.10 to 0.11, and -0.20 to 0.00, respectively. We hope that these results would greatly assist in the improvement of ketosis disease in the local Holsteins.

Development of Well Placement Optimization Model using Artificial Neural Network and Simulated Annealing (인공신경망과 SA 알고리즘을 이용한 지능형 생산정 위치 최적화 전산 모델 개발)

  • This study presents the development of a well placement optimization model, combining an artificial neural network, which enables high-speed calculation, with a simulated annealing algorithm. The conventional FDM simulator takes excessive time when used to perform a field scale reservoir simulation. In order to solve this problem, an artificial neural network was applied to the model to allow the simulation to be executed within a short time. Also by using the given result, the optimization method, SA algorithm, was implemented to automatically select the optimal location without taking any subjective experiences into consideration. By comparing the result of the developed model with the eclipse simulator, it was found that the prediction performance of the developed model has become favorable, and the speed of calculation performance has also been improved. Especially, the optimum value was estimated by performing a sensitivity analysis for the cooling rate and the initial temperature, which is the control parameter of SA algorithm. From this result, it was verified that the calculation performance has been improved, as well. Lastly, an optimization for the well placement was performed using the model, and it concluded the optimized place for the well by selecting regions with great productivity.

Effect of the Physiologically Active Compounds in Phlomidis Radix on Cell Cycle Regulation in Human Gingival Fibroblasts (속단의 생리활성성분이 치은섬유아세포의 세포주기조절에 미치는 영향)

  • The purpose of present study was to investigate the effects of physiologically active compound (SD62-122) from Phlomidis Radix on the cell cycle progression and its molecular mechanism in human gingival fibroblasts(HGFs). For this purpose, fibroblasts were isolated and cultured from excisioned gingiva during crown lengthening procedure in healthy adult. The following parameter were evaluated that there are cell number counting, MIT assay, cell cycle progression, western blot analysis. The cell number and MIT assay of primary cultured fibroblast was not increased at 2 days but significant increased compare to negative control at 3days(p<0.05). S phase was increased and G1 phase decreased in both $10^{-8}M$ and $10^{-9}M$ of SD62-122 in cell cycle analysis. The cell cycle regulation protein levels of Cyclin $D_1$, Cyclin E, cdk 2, cdk 4 and cdk 6 were increased compare to control in both $10^{-8}M$ and $10^{-9}M$ of SD62-122. The protein levels of p21 and p53 were decreased compare to control, but the level of pRb was not changed compare to control in $10^{-9}M$ of SD2-122. These results suggested that physiologically active compound (SD62-122) isolated from Phlomidis Radix increases the cell proliferation and cell cycle progression in HGFs, which is linked to increased cell cycle regulation protein levels of Cyclin $D_1$, Cyclin E, cdk 2, cdk 4 and cdk 6, and decreased the levels of p21, p53.

A case study for the dispersion parameter modification of the Gaussian plume model using linear programming (Linear Programming을 이용한 가우시안 모형의 확산인자 수정에 관한 사례연구)

  • We developed a grid-based Gaussian plume model to evaluate tracer release data measured at Young Gwang nuclear site in 1996. Downwind distance was divided into every 10m from 0.1km to 20km, and crosswind distance was divided into every 10m centering released point from -5km to 5km. We determined dispersion factors, ${\sigma}_y\;and\;{\sigma}_z$ using Pasquill-Gifford method computed by atmospheric stability. Forecasting ability of the grid-based Gaussian plume model was better at the 3km away from the source than 8km. We confirmed that dispersion band must be modified if receptor is far away from the source, otherwise P-G method is not appropriate to compute diffusion distance and diffusion strength in case of growing distance. So, we developed an empirical equation using linear programming. An objective function was designed to minimize sum of the absolute value between observed and computed values. As a result of application of the modified dispersion equation, prediction ability was improved rather than P-G method.

  • The sensitivity analysis of input parameters was Performed fer an ingestion dose assessment model (U. S. NRC's Regulatory Guide 1.109 model) from routine releases of radionuclides. In this study, three kinds of typical Korean foodstuffs (rice, leaff vegetables, milk) and two kinds of radionuclides $(^{l37}Cs,\;^{131}I)$ were considered. The values of input parameters were sampled using a Latin hypercube sampling technique based on Monte Carlo approach. Sensitivity indices, which represent the influence or the importance of input parameters for predictive results, were quantitatively expressed by the partial rank correlation coefficients. As the results, the ratio of the interception fraction to the yield of agricultural plants and the human consumption rate were sensitive input parameters for the considered foodstuffs and radionuclides. Additionally, in case of milk, the transfer factor of radionuclides from animal intake to milk and the daily intake rate of feedstuffs were sensitive input parameters. The weathering removal half-life and the delay time from food production to human consumption were relatively sensitive for $^{137}Cs$ and $^{131}I$ depositions, respectively.

Forest Thematic Maps and Forest Statistics Using the k-Nearest Neighbor Technique for Pyeongchang-Gun, Gangwon-Do (kNN 기법을 이용한 강원도 평창군의 산림 주제도 작성과 산림통계량 추정)

  • This study was conducted to produce forest thematic maps and estimate forest statistics for Pyeongchang Gun using the kNN technique, which has been applied to produce thematic maps of variables of interest including unobserved plots by combining field plot data, remotely sensed data and other digital map data in forest inventories. The estimation errors for three horizontal reference areas (HRAs), whose radii are 20, 40 and 60 km respectively, were compared. Although the precision for the 40 km radius was lower compared to that for the 60 km radius, the 40 km radius was found to be an efficient HRA because their difference in precision was modest. At a value of k=5 nearest neighbors for the selected HRA, the overall accuracy was high. As a result, using the k=5 neighbors within the HRA of 40 km radius, thematic maps of number of trees, basal area, and growing stock per hectare were generated. As compared to the forest statistics based on field sample plots, the estimated means of each parameter from the produced maps were underestimated.

Prediction of Shelf-life of Sea Tangle Drink (다시마음료의 유통기한 설정)

  • This study was designed to establish the shelf-life of sea tangle drink. Quality changes, including pH, total acidity, color, sensory evaluation, total aerobic bacteria, and coliform, were measured periodically in sea tangle drink kept at 15, 25, and $37^{\circ}C$ for 8 months. The pH level and total acidity did not remarkably change during the storage period regardless of storage temperature. In terms of color, lightness did not change during the storage period, whereas redness decreased. Yellowness did not change at $15^{\circ}C$ during the storage period, although it increased at 25 and $37^{\circ}C$ at 8 months. Color difference (${\Delta}E$) value was the lowest at $15^{\circ}C$ (12.14), followed by $25^{\circ}C$ (12.57) and $37^{\circ}C$ (14.43). During the storage period, total aerobic bacteria and coliform were not detected. There were no changes in appearance, smell, taste, texture, and overall acceptability of sea tangle drink (not exceeding 3 points) throughout the entire storage period. By using correlation coefficients, color value was selected as an indicating parameter for the shelf-life establishment of sea tangle drink. Based on the forecasted distribution day by annual temperature, the shelf-life of sea tangle drink based on color with the highest correlation coefficient was predicted as 27.10 months.
