A DNA Microextractor Using Crossed Field Electrophoresis (교차 전기영동법을 이용한 극소형 DNA 추출기)

  • This paper presents a microextractor for the separation of DNA molecules by their sizes. The DNA microextractor immobilizes the DNA molecules of specific size in the micropillar array by adjusting the period of the crossed electric field, thus providing a starting-point independent target DNA extraction method without separation process monitoring. The DNA microextractor has been fabricated by a three-mask micromachining process. The velocity of three different DNA molecules has been measured at the electric field of E=5V/0.8cm in the fabricated DNA microextractor, resulting in the reorientation times of $4.80{\pm}0.44sec,\;7.12{\pm}0.75sec$, and $9.88{\pm}0.30sec$ for ${\lambda}$ DNA, micrococcus DNA, and T4 DNA, respectively. T4 DNA is trapped in the micropillar array when the crossed electric field of 5V/0.8cm is applied alternately at a 10 second time interval. The present DNA microextractor filters the DNA in a specific size range by adjusting the magnitude and/or the period of the crossed electric field applied in the micropillar array.

  • In this paper, by using the electrophoresis, preparation of YBCO superconducting wire deposited on metal Ag base wire was studied with its Properties. YBCO Powder could be prepared by solid state reactions with calcining and sintering processes. Superconducting wire prepared on metal Ag wire used as cathode of deposition base could be also fabricated in the YBCO/acetone-dispersed solution to obtain several tens of re thick films. And then it could be used as superconducting wire for measurement after calcination, sintering and oxygen absorption processes. In the process of film deposition, a catalyst I$_2$added into the suspension solution was very useful for preparing thick film of YBCO, and BaF$_2$ of additive material was also necessary for preparing crack-free wire of YBCO superconductor. As a result, YBCO superconducting wire added 2~3wt.% of BaF$_2$\ulcorner with catalyst, 12 had better deposition condition for uniform and dense YBCO wires, and critical current density, Jc was calculated at the value of 1,458A/$\textrm{cm}^2$(more than 10$^{3}$A/$\textrm{cm}^2$ ,77K, o[T]) of 30${\mu}{\textrm}{m}$ thick sample by 4 point prove method.

  • To clone ${\beta}$-glucosidase gene from Cellulomonas sp. a gene library was constructed using E. coli JM83 pUC9. Among 2,500 pseudotransformants obtained, 20 clones developed yellow color on the p-nitrophenyl- -D-glucopyranoside filter paper These 20 clones were classified into three groups based on the results of activity staining using nondenaturating polyacrylamide gel electrophoresis and restriction enzyme digestions. Among the three groups, only one group containing pCEl plasmid has specificity for cellobiose.

  • Xylanase(1, 4-beta-D-xylan xylanohydrolase` EC 3.2.1.8) II was purified from Penicillium verruculosum by using the techniques of two anion exchange chromatographies, and gel filtration. The molecular weight of this enzyme was about 22, 000 as determined by SDS-electrophoresis. The enzyme showed hydropytic activity toward xylan but did not catalyze hydrolysis of Rho-nitrophenyl-beta-D-xylopyranoside, Rho-nitrophenyl-beta-D-glucopyranoside, Rho-nitrophenyl-beta-D-cellobiopyranoside, and celluloses such as Avicel, cotton, filter paper, carboxymethylcellulose.

  • We proposed a transflective liquid crystal display (LCD) with high aperture ratio using an electrophoretic particle layer (EPL). The transflective LCD consisted of the stacked LC layer and EPL which was acted as a switchable mirror under in-plane electrode structure. Without separation of reflective part and transmissive part in one pixel, a modeselectable display device can be obtained.

  • Complexation reactions of nitrilotriacetate (NTA) and penicillamine with $Cu^{2+}$ and $Co^{2+}$ have been studied in solution phase using paper electrophoresis technique. The stability constants of the complexes Cu(II)-nitrilotriacetate-penicillamine and Co(II)-nitrilotriacetate-penicillamine have been found to be $6.64{\pm}0.03\;and\;5.86{\pm}0.05$ (logarithm stability constant values), respectively at 35$^{\circ}C$ and ionic strength 0.1 M.

  • Proteomics research includes identification and quantitation of single protein and/or protein complex, profiling of protein expression changes in response to biological perturbations, characterization of protein functions and interactions, and elucidation the linkage between proteins and diseases. In this review paper, recent developments in the basic technologies involved in the proteomics research such as 2-dimensional PAGE and mass spectrometry are discussed. Also, the application areas of proteomics technology such as protein expression mapping and cell map proteomics are introduced with the focus on new drug development.

  • The distribution of $^{51}Cr\;and\;^{128}I$ recoil species following radiative-neutron capture in chromates, dichromates, iodates and periodates has been investigated by using paper-electrophoresis. In view of the effective recoil energy and the effect of the internal conversion, it is unlikely that an atom which has captured a neutron can remain bound in its original molecule. It is also unlikely that the energy of the recoil atom is dissipated in heating a small region of the crystal. However, the results of paper-electrophoresis separation of recoil $^{51}Cr\;and\;^{128}I$ indicated that many more of the recoil atoms were bound in the parent molecule. The disorder model for the reaction was proposed from observations of retention. In considering cations, the greater their radii, the higher is the probability of the recoil atom breaking through the secondary cage. In ammonium salt, the ammonium ion behaved as a reducing agent in the disorder zone and resulted in low retention. Crystal structures with their greater free space have shown low retention.

  • A fungal strain able to produce filter paper activity (FPase) was isolated from soil by testing the ability to hydrolyze using filter paper. The isolated strain was identified as an Aspergilus sp. judging from its morphological and microscopical characteristics. The cellulase-xylanase system of Aspergillus sp. 8-17 was detected in situ after gel electrophoresis in the presence of SDS and showed that each protein pattern had a distinct polypeptide composition. ${\beta}$-1,4-Glucanase, cellobiohydrolase, and xylanase activity profiles differ from protein patterns. The Aspergillus sp. 8-17 hydrolytic enzymes responsible for the hydrolysis of ${\beta}$-glucan, MUC, and xylan have multiple active forms.

The Cell and Genetic Characteristics of Slime Forming Bacteria on Antibiotic Resistance in the Paper-making Process

  • The seven strains, Pseudomonas paucimobilis, Pseudomonas cepacia, Staphylococcus auricularis, Staphylococcus saprophyticus, Acidovorax spp., Acinetobacter calcoaceticus, and Actinobacillus capsulatus were tested with three slimicides. Most of the tested bacteria were inhibited with slimicide K (an isothiazolin based compound), even at its low concentration, except for Actinobacillus capsulatus and Staphylococcus auricularis. Both slimicides B (an organic bromine based compound) and S (aldehydes) also couldn't prevent these two strains even at their highest concentration. Five different sizes of plasmid DNA were isolated from Actinobacillus capsulatus. Staphylococcus auricularis, a gram-positive bacteria, showed the slimy substances around its cell distinctively. The results suggest that two strains, Actinobacillus capsulatus, Staphylococcus auricularis, have presumably developed a resistance to the slimicide, by plasmid DNA or slimy substance. Our findings also suggest that not only gram-negative bacteria, but also gram-positive bacteria should not be neglected
