생약추출물의 Cytochrome P450 약물대사 효소계 저해활성 (Inhibitory Effects of Medicinal Herbs on Cytochrome P450 Drug Metabolizing Enzymes)

  • The MeOH ext., $CH_2Cl_2$ Frac., EtOAc Frac., n-BuOH Frac., and $H_2O$ Frac. of 23 Korean medicinal herbs were prepared and were tested the inhibitory effects on Cytochrome P450 (Cyp) 1A1/2, 2B1/2, 2E1. Among the tested samples, the extracts of Selaginella tamariscina, Euonymus alatus, Salvia miltiorhiza, Angelica acutiloba, Rheum palmatum, Paeonia moutan, Scutellaria barbata, Tribulus terrestris, Hedyotis diffusa, Curcuma zedoaria, Rehmania glutinosa, Trogopterus xanthipes, Melandryum firmum, Achyranthes bidentata, Leonurus sibricus, Panax ginseng, Paeonia lactiflora, Poncirus trifoliata, Cnidium officinale, Cyperus rotundus, Corydalis ternata showed significant inhibitory effects on Cyp 1A1/2, 2B1/2, 2E1. The $IC_{50}$ values of those extracts were found to be below $50\;{\mu}g/ml$.

홍삼과 발효홍삼의 항염증 작용 및 항알러지 효과 비교 (The comparative study of anti-allergic and anti-inflammatory effects by fermented red ginseng and red ginseng)

  • 홍삼은 인삼을 증숙하는 과정에서 생겨나는 화학적으로 변화된 생리활성물질들에 의해 인삼과 차별화된 효과를 보여준다. 또한 홍삼은 장내미생물의 생물학적 전환에 의해 변화되어 생체내 약리활성을 보이는데, 이는 개인의 장내 미생물의 분포 차이에 의해 약리작용의 차이를 보일 수 있다. 최근 이 같은 차이를 극복하기 위해 인위적으로 장내미생물에 의해 사전 발효시킨 발효홍삼은 홍삼과 다른 성분량의 차이를 보임으로써 독특한 약리작용을 보인다고 알려져 있다. 이에 본 연구는 홍삼과 발효홍삼의 추출물이 알러지 및 염증반응에 끼치는 영향에 대해 알아보기 위해 비만세포의 탈과립 과정에서 분비되는 $\beta$-hexosaminidase 분비량 비교, 염증매개 세포부착물질인 ICAM-1 발현비교, 생쥐림프구의 증식능, 각질형성세포 증식능을 상호 비교하였다. 실험결과 홍삼은 1, $10\;{\mu}g/ml$에서 $\beta$-hexosaminidase 분비량 억제 및 ICAM-1 발현 억제 조절 효과를 나타내었으며, 발효 홍삼은 모든 농도에서 유의적인 억제 효과를 나타내었다. 그러므로 홍삼과 발효홍삼에서는 모두 항알러지 및 항염증 효과가 있다고 사료된다. 그러나 LPS에 의한 B세포의 증식능에서 홍삼 $1\;{\mu}g/ml$은 증가효과를 발효홍삼에서는 $100\;{\mu}g/ml$에서 억제효과를 나타내었고, ConA에 의한 T세포의 증식능에서는 홍삼 $100\;{\mu}g/ml$에서 증가효과를 발효홍삼 $100\;{\mu}g/ml$에서는 억제효과를 나타냄으로써 림프구 증식능에 관한 실험에서는 두 추출물간에 상반된 효과를 나타내었다. HaCaT세포를 이용한 각질형성세포 증식능에서는 발효홍삼 1, $10\;{\mu}g/ml$에서만 효과를 나타내었다. 이러한 결과를 종합해 볼 때, 홍삼과 발효홍삼에 존재하는 성분량의 변화에 따라 그 효능에서 차이를 나타내고 있는 것으로 사료되며, 홍삼 및 발효홍삼의 고농도 $100\;{\mu}g/ml$에서는 저농도와는 다른 효과를 보이는 경향을 나타내고 있으므로 홍삼 및 발효홍삼의 사용에 있어 적절한 적응증과 주성분의 검증 및 유효 농도에 대한 연구가 더욱 필요하다고 판단된다.

고려홍삼의 수지상세포 활성화 효과 (Effects of Red-ginseng Extracts on the Activation of Dendritic Cells)

  • 본 연구에서는 정관장 홍삼의 물(water) extract, 식용발효 주정 extract 및 홍삼 추출물로부터 분리 제조한 crude saponin을 이용하여 면역반응을 매개하는 수지상세포의 활성 효과에 대하여 알아보았다. 그 결과 홍삼시료 중, crude saponin $100\;{\mu}g/ml$을 처리하였을 때 수지상세포의 세포표면 분자인 MHC class II, CD40, CD80, CD86의 발현이 증가하였으며, phagocytosis는 감소하였다. 또한 홍삼시료를 처리한 수지상세포와 allogeneic T세포를 함께 배양하였을 때, 홍삼시료의 물 extract, 식용발효주정 extract, crude saponin 모두 allogeneic T세포의 증식반응을 유도하였고, IL-2와 $IFN-{\gamma}$의 생산량을 증가시키는 것을 확인하였다. 또한 $CD4^+$ syngeneic T세포와 $CD8^+$ syngeneic T세포의 반응에서도 T세포의 증식반응을 높게 유도하였으며, $CD4^+$ syngeneic T세포에서 IL-2와 $IFN-{\gamma}$의 생산량을 증가시키고, $CD8^+$ syngeneic T세포에서는 $IFN-{\gamma}$ 생산량을 증가시키는 것을 확인하였다. 이상의 결과로 crude saponin의 경우 수지상세포 의 세포표면 공동자극분자의 발현을 유도하고 성숙을 유도함으로써 T세포의 활성을 증진시키는 것으로 생각되며, 물 extract와 식용발효주정 extract는 crude saponin과는 다른 기작으로 T세포 활성화를 유도하는 것을 알 수 있었다. 따라서 실험에 사용한 홍삼시료, 즉 물 extract, 식용발효주정 extract, crude saponin 모두 수지상세포의 활성을 유도하는 물질로써 암항원 특이적 T세포 활성화를 이용한 항암치료에 이용할 수 있는 가능성이 있다고 사료된다.

Stimulatory Effects of Ginsan on the Proliferation and Viability of Mouse Spleen Cells

  • Ginsan is an acidic polysaccharide purified from Panax ginseng, a famous oriental herb. Although a variety of biological activities of ginsan have been studied, the effects of ginsan on spleen cells are not fully elucidated. We investigated the effect of ginsan on the viability and proliferation of spleen cells. Using Cell Counting $Kit-8^{(R)}$ solution and trypan blue solution, we found that ginsan significantly enhanced viability and proliferation. Multiple clusters, indicating proliferation, were observed in ginsan-treated spleen cells and, carboxyfluorescein succinimidyl ester and surface marker staining assay revealed that ginsan promoted proliferation from $CD19^+$ B cells rather than $CD4^+$ or $CD8^+$ T cells. In addition, ginsan decreased the percentage of late apoptotic cells. Ginsan increased the surface expression of CD25 and CD69 as well as production of interleukin-2 from spleen cells, suggesting increased activation. Taken together, these results demonstrate that ginsan increases the viability and proliferation of spleen cells via multiple mechanisms, valuable information for broadening the use of ginsan in clinical and research settings.

흰쥐의 염산.에탄올 유발 위염 위조직에서 ginsenoside $Rb_1$의 항산화 효과 (Anti-oxidative Effect of Ginsenoside $Rb_1$ on the HCI.Ethanol-Induced Gastric Tissue in Rats)

  • In the previous study, we demonstrated that ginsenoside $Rb_1$ isolated from the butanol fraction of the head of Panax ginseng had significant gastroprotective activity on gastritis and gastric ulcer models in rats. It has been well established that drugs to have capacity of scavenging or inhibiting the generation of reactive oxygen radicals prevent the gastric mucosal injury. Ginsenoside $Rb_1$ was tested on HCl ethanol-induced gastritis in rats, DPPH-induced free radical scavenging effect, MDA assay, GSH activity, and SOD activity in gastric tissue. It showed significant inhibition in HCl ethanol-induced gastritis, and al~o significantly increase of GSH activated SOD. We speculate that the protective effect of ginsenoside $Rb_1$ against HCl ethanol-induced gastric mucosal damage is originated from the increase of GSH and the activation of SOD.

사군자탕(四君子湯) 재료를 첨가한 식빵의 품질특성 (Quality Characteristics of White Pan Bread with Ingredients of Sagoonja-Tang)

  • This study was carried out to determine the functional works and basic ingredients of Sagoonja-Tang on white pan bread. The -experimental groups consisted of 6% mixtures of five(Ed- confirm the number) kinds powders, : B1 for Sagoonja-Tang powder, B2 for Panax ginseng, B3 for Poria cocos, Koidz, B4 for Atractylodes macrocephala, and B5 for Glycyrrhiza uralensis Fisch. The volume of white pan bread somewhat decreased after adding the ingredients, and the volume differences in order from highest to lowest, were white pan bread control>B2>B3>B4>B1>B5. For white pan bread, pH of control, B3, and B5 decreased on the 2nd day during storage, but increased on the 4th and 7th days. In the texture analyses of white pan bread, B4 and B5 showed a higher degree of hardness than that of control, while B2, and B3 were the lowest. In sensory test, the overall acceptability of regular white pan bread in order from highest to lowest was control>B3>B2>B1 ${\cdot}$ B5>B4.

재생불량성빈혈(再生不良性貧血)의 변증론치(辨證論治)에 대(對)한 고찰(考察) (A Study of Bian Zheng Lun Zhi on Aplastic Anemia)

  • Background/Aims: Aplastic anemia is defined as pancytopenia (anaemia, leucopenia, and thrombocytopenia) result from aplasia of the bone marrow. Many studies have shown that survival rate of aplastic anemia is 50-60% with immunomodulation therapy. In Korea, there is a lack of research considering oriental herbal medicine with aplastic anemia. Methods: It was compared and analyzed that recently several experimental or clinical reports of oriental herbal medicine on aplastic anemia. Results and Conclusion: The oriental herb of Panax ginseng radix, Cprdonopsis pilosula radix, Astragalus membranaceus radix, Atractylodes marcrocephala. Cervi Cornu Parvum, Epimedii Herba, Boshniakiae Herba, Morindae Radix, Angelicae gigantis Radix, Cascutae Semen, Lycii Fructus, Polygoni Multiflori Radix potently stimulated hematopoietic stem cell activity, Response rate to oriental herbal medicine of aplastic anemia was 30-60% and effect rate of aplastic anemia was 73-93%, Bian zheng Lun zhi(辨證論治 treatment according to syndrome differentiation) which based on Shen xu(腎虛) is presumed to approach highest degree effect in response rate.

Compound K 측정을 위한 ELISA법 개발 (Development of ELISA Method for the Determination of Compound K)

  • In order to quantify compound K(CK), anticancer component of Panax ginseng C. A. Meyer, high titer rabbit polyclonal antibodies (pAbs) were raised against a conjugate of CK and bovine serum albumin coupled by a periodate oxidation method. Coating antigen (CK-OVA) was also prepared by the same method with OVA. As a result of optimization of antiserum dilution (2,000 fold), coating antigen ($25{\mu}g/ml$) and other condition (incubation time, temperature and washing method), ELISA method for the determination of CK was established. The measuring range extended from 0.5 ng/ml to 25 ng/ml of CK. The antibodies exhibited minor or even no cross reactivities with protopanaxatriol (1.56%) and other tested ginsenosides, $GRb_1$ (0.11%), $GRg_1$ (0.07%) except protopanaxadiol (87.2%) from the structural similarity. And the antibody showed good correlation (r=0.987) between the assay values obtained by this ELISA method and HPLC. Therefore, the ELISA method could be very useful tools for the determination of CK in biological fluids because of their high sensitivity and specificity.

  • The word of stress was wsed in the field of physics as a external force from 17th century, but the meaning of this stress had chafed to the internal and exteral demand fort the human body in medical area. All the stumulants which make stress was called as j stressor. When animals get stress blucose excreted from liver to adapt for the emergent state ant some related hormone secrete convert protein and lipid to glucose for the purpose of energy supply to muscle. As a results heart rate, blood pressure, respiration rate are increased and musclse are strengthed. These physiolgical reactions ate controlled by autonomic nerve system under the control of hyothalamus in brain. Autonomic nerve system and endocrinary system are react harmoiously to stress reaction. According to the stress reaction, adrenomedullary system are stimulated, and epinephrine, morepinephrine are exceted. Author experimented the effect of the immobilizational stress to rat by analyzing the variaition of catecholamine secrction, the Na concentration and the effect of the antistresf effect by Panax Ginseng which is a traditional Korean herb medicine. The concentration of the norepinephrine, epinephrin, and dopamin in normal rat are 1 578 ng/ml, 0.365 ng/ml, and 0.731 ng/ml respectively, but in the immobilyzed stress groIn the concentration were increased to 1.915 ng.ml, 0.854 ng/ml, and 2,361 ng/ml which she the high show the higher concentration of catecholamine to stressor.

인삼 뿌리썩음병균(Cylindrocarpon destructans) 후막포자의 형성 및 발아에 영향을 주는 물리화학적 요인 (Effect of physical and chemical Factors on the Formation and Germination of Chlamydospore of Cylindrocarpon destructans Causing Root Rot of Panax ginseng)

  • 인삼뿌리썩음병균(Cylindrocarpon destructans)후막 포자의 형성은 CD(Czapek`s dox broth)와 SNY(Spezieller Nahrtoffarmer yeast extract broth)배지에서 진탕배양(2$0^{\circ}C$, 120 rpm)하면 양호하였다. 특히 CD배지에서 2$0^{\circ}C$로 30일간 진탕배양하면 5.99 log 후막포자/ml로 형성되었다. 후막포자의 발아율은 12.9~23.3%로 CD배지와 SNY배지에서 좋았으며 발아적온은 5~1$0^{\circ}C$였고, 최적 pH는 6이었다. 그러나 2$0^{\circ}C$이상에서는 전혀 발아하지 않았으며 pH 7.0이상에서는 발아율이 급속히 저하되었다. 인삼추출물 3%, GA 10 ppm, IAA 10 ppm, NOVOZYM\ulcorner234 20 ppm을 처리 할 경우 대조구에 비해 발아율이 증가하였다. 특히 NOVOZYM\ulcorner234 20 ppm 처리구는 발아율이 무처리에 비해 2배 증가하였다. CD배지(pH5)에 NOVOZYM\ulcorner234 20 ppm과 GA 10 ppm을 조합처리하고 5$^{\circ}C$로 정치배양 할 때 후막포자의 발아율은 49.4%로 가장 높았다.
