Panax 속 한약재가 흰쥐 혈관운동성에 미치는 영향에 관한 비교 연구 (The Effects of Water Extract of Genus Panax on Rat Blood Vessels)

  • Objectives : We have examined the relaxational response to the water extract of genus Panax in rat thoracic aorta and mesenteric artery. Methods : Segments of thoracic aorta and mesenteric artery obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. Results : We found that the thoracic aorta segments responded to the water extract of genus Panax with a dose-dependent vasorelaxation. At $10^{-5}m$ 5-hydroxytrptamine (5-HT), the maximal contraction force were 94.9% of the maximum KCl-response. At $10^{-5}m$ 5-HT - induced contraction, The contractile response of thoracic aortic rings were inhibited by 54.7%, 36.3% and 31.3% after addition of the high concentration (100 mg/ml) of water extract of Panax ginseng, Panax japonicus and Panax quinquefolium. The contractile response of mesenteric arteries were inhibited by 88.3%, 87.7%, and 70.3% after addition of the high concentration (100 mg/ml) of water extract of Panax ginseng, Panax japonicus and Panax quinquefolium. Conclusions : In conclusion, water extract of genus Panax - induced relaxation in the isolated rat thoracic aorta and mesenteric artery were composed of endothelium - independent relaxation and dose - dependent relaxation.

고려인삼, 미국삼 및 죽절삼의 생육 및 형태적 특성 비교 (Comparison of Plant Growth and Morphological Characteristics Among the Korean Ginseng, the American Ginseng and the Bamboo Ginseng)

  • An investigation was conducted to ascertain the basic information on characteristics of growth and morphological characters among the Korean (Panax. ginseng), the American (Panax. quinquefolium) and the Bamboo (Panax. japonicus) ginseng. In aerial parts growth of the ginseng species by age, The Korean ginseng and American ginseng's stem and leaf growth was alike in 2-4 years old, but growth cycle changed in 6 years old. The Korean ginseng was more vigorous than the American ginseng. The Korean ginseng roots were highly observed in ratio of red skin roots among three species, whereas The American ginseng roots were highly infected by root rot. It seems to be variable depending on growing stage and species. The Korean ginseng flowered about the middle of May, the American ginseng early June, and the Bamboo ginseng was late of May, The berry color of the ginseng species was observed, The Korean and American ginseng's mature berry color was red, The Bamboo ginseng's berry was three type of color and shape. In root characteristics of the seedling, Korean (p. ginseng), American (p. quinquefolium) ginseng's root shape was similarity in type, the bamboo ginseng showed different type, which root length and root weight was smaller than those of ginseng. In morphological characters of Leaf surface, pollen, and stoma, the Korean ginseng and American ginseng had crystal rosette on epidermis cell, but the Bamboo ginseng didn't has crystal rosette. Pollen shape observed tricolpate pollen and size was media type among the ginseng species, and also guard cell was anomocytic type, which were observed by scanning electronic microscope.

Development of Reproducible EST-derived SSR Markers and Assessment of Genetic Diversity in Panax ginseng Cultivars and Related Species

  • Little is known about the genetics or genomics of Panax ginseng. In this study, we developed 70 expressed sequence tagderived polymorphic simple sequence repeat markers by trials of 140 primer pairs. All of the 70 markers showed reproducible polymorphism among four Panax species and 19 of them were polymorphic in six P. ginseng cultivars. These markers segregated 1:2:1 manner of Mendelian inheritance in an $F_2$ population of a cross between two P. ginseng cultivars, 'Yunpoong' and 'Chunpoong', indicating that these are reproducible and inheritable mappable markers. A phylogenetic analysis using the genotype data showed three distinctive groups: a P. ginseng-P. japonicus clade, P. notoginseng and P. quinquefolius, with similarity coefficients of 0.70. P. japonicus was intermingled with P. ginseng cultivars, indicating that both species have similar genetic backgrounds. P. ginseng cultivars were subdivided into three minor groups: an independent cultivar 'Chunpoong', a subgroup with three accessions including two cultivars, 'Gumpoong' and 'Yunpoong' and one landrace 'Hwangsook' and another subgroup with two accessions including one cultivar, 'Gopoong' and one landrace 'Jakyung'. Each primer pair produced 1 to 4 bands, indicating that the ginseng genome has a highly replicated paleopolyploid genome structure.

Regulatory effects of saponins from Panax japonicus on colonic epithelial tight junctions in aging rats

  • Background: Saponins from Panax japonicus (SPJ) are the most abundant and main active components of P. japonicus, which replaces ginseng roots in treatment for many kinds of diseases in the minority ethnic group in China. Our previous studies have demonstrated that SPJ has the effects of anti-inflammation through the mitogen-activated protein kinase (MAPK) and nuclear factor kappa B (NF-${\kappa}B$) signaling pathways. The present study was designed to investigate whether SPJ can modulate intestinal tight junction barrier in aging rats and further to explore the potential mechanism. Methods: Aging rats had been treated with different doses (10 mg/kg, 30 mg/kg, and 60 mg/kg) of SPJ for 6 mo since they were 18 mo old. After the rats were euthanized, the colonic samples were harvested. Levels of tight junctions (claudin-1 and occludin) were determined by immunohistochemical staining. Levels of proinflammatory cytokines (interleukin-$1{\beta}$ and tumor necrosis factor-${\alpha}$) were examined by Western blot. NF-${\kappa}B$ and phosphorylation of MAPK signaling pathways were also determined by Western blot. Results: We found that SPJ increased the expression of the tight junction proteins claudin-1 and occludin in the colon of aging rats. Treatment with SPJ decreased the levels of interleukin-$1{\beta}$ and tumor necrosis factor-${\alpha}$, reduced the phosphorylation of three MAPK isoforms, and inhibited the expression of NF-${\kappa}B$ in the colon of aging rats. Conclusion: The studies demonstrated that SPJ modulates the damage of intestinal epithelial tight junction in aging rats, inhibits inflammation, and downregulates the phosphorylation of the MAPK and $NF-{\kappa}B$ signaling pathways.

  • 종간, 속간의 잡종에서는 흔히 불임현상이 일어나는데 인삼의 경우도 예외가 아니어서 고려인삼(panax ginseng: P.G.)과 미국인삼(panax quinquefolium: P. Q.) 그리고 죽절인삼(Panax japonicus: P.J) 간의 교잡육종에서 P.G.${\times}$P.J.에서만 아주 희귀하게 종자를 얻을수 있고 다른 종합에서는 전혀 잡종 제이세대를 얻을 수가 없었다고 하므로 본실험에서는 P.G.${\times}$P.Q.와 P.G.${\times}$P.J.의 교잡종의 화기 및 종자형성중의 유리아미노산의 소장을 추구함으로서 불임현상과 물질대사와의 관련성의 일단을 밝히고저 한다. 1. 두 교잡종의 시료에서 검출된 유리아미노산의 Chromatogram은 서로 유사한 pattern을 나타냈으며 그들의 양친이 나타내는 pattern과 유사하나 spot의 종류나 크기 정색도는 여러 가지 점에서 특이한 변화가 있다. P.G.${\times}$P.Q.에서 19종, P.G.${\times}$P.J.에서는 21종의 spot을 검출하였는데 전자에서 Alanine, Valine, Leucine, Phenyl alanine, Proline, Hydroxy proline, Serine, Threonine, Tyrosine, Aspartic acid, Glutamic acid, Lysine, Arginine, ${\beta}$-Alanine, Cysteic acid, Tryptophan, Asparagine, Glutamine, ${\gamma}$-Amino butyric acid를 확인하고 후자의 경우는 상기한 것 외에 Methionine과 한 개의 미지 spot를 얻었다. 2. Alanine, Aspartic acid, Glutamic acid, Cysteic acid와 Asparagine이 전체적으로 보아 주체를 이루고 있는 것은 P.G. P.Q. P.J에서와 같으나 Asparagine이 소포자기와 화분성숙기에 최다량으로 나타나는 것은 이들의 양친의 경우에는 보지 못했던 특이한 일이다. 3. 홍숙기에 Cysteic acid가 감소되는 것은 P.Q. P.J.의 경우와는 유사하나 P.G의 경우와는 반대되는 현상이며 P.G.${\times}$P.J.에서 Methionine이 검출된 것도 특이하다. 4. Proline은 그들의 양친의 경우와는 현저한 차이가 있었는데 소포자기에서 아주 미약한 정색을 나타낼뿐이며 이 때에 Asparagine의 spot가 가장 크고 강한 정색을 나타내었는데 이제까지의 여러 연구 결과를 종합해 보면 Proline의 양과 화분의 임성과는 아주 밀접한 관계가 있으며 이런 관계가 인삼 종자 형성에서도 나타나고 있다. 또한 Proline이 결핍될 때 Asparagine의 축적이 있다는 많은 보고도 있다. 5. 교잡종에서의 결실불능의 원인을 Proline의 과소에 의한 것으로 보며 Proline이 약의 화분퇴화와 중요한 관계를 가지고 있을 것으로 추찰된다. 6. 그밖에 여러 아미노산의 소장이 P.G. P.Q. P.J. 등의 자식계와 그들과의 잡종사이에 있어서 상당한 차이가 생기는 것은 교잡에 의한 Gene-action system의 변혁에 따른 결과라고 추찰되는 것으로 Proline의 대사 및 생리적 영향과 더불어 보다 깊은 조사가 필요한 것으로 생각된다.

Genetic Relationships of Panax Species by RAPD and ISSR Analyses

  • This study was carried out to develop convenient and reproducible methods for identifying the genetic relationship among germplasms of Panax species based on molecular genetics. Using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses, genetic polymorphism of the Panax species was investigated with following cultivars and accessions, such as Chunpoong, Yunpoong, Kopoong, Sunpoong, and Kumpoong in domestic cultivars, Hwangsuk, Jakyung and Suckju in domestic accessions, and Panax quinquefolius L. and Panax japonicus C.A. Meyer in foreign introduced accessions, respectively. Specific DNA fragments ranging from 200 to 3,000 base pairs in size could be obtained with various ISSR and RAPD primers under the optimized PCR conditions. The dissimilarity coefficients among the genetic polymorphisms of ginseng cultivars and accessions were calculated from 0.26 to 0.90 in RAPD and from 0.12 to 0.89 in ISSR analysis, respectively. Eleven plant samples were grouped siblings together with cultivars and parents based on cluster analysis of genetic distance depending on genetic property such as origin of the species. In results, both RAPD and ISSR analyses were useful for identifying the genetic relationship among cultivars and accessions of Panax species at DNA level.

Saponins from Panax japonicus ameliorate age-related renal fibrosis by inhibition of inflammation mediated by NF-κB and TGF-β1/Smad signaling and suppression of oxidative stress via activation of Nrf2-ARE signaling

  • Background: The decreased renal function is known to be associated with biological aging, of which the main pathological features are chronic inflammation and renal interstitial fibrosis. In previous studies, we reported that total saponins from Panax japonicus (SPJs) can availably protect acute myocardial ischemia. We proposed that SPJs might have similar protective effects for aging-associated renal interstitial fibrosis. Thus, in the present study, we evaluated the overall effect of SPJs on renal fibrosis. Methods: Sprague-Dawley (SD) aging rats were given SPJs by gavage beginning from 18 months old, at 10 mg/kg/d and 60 mg/kg/d, up to 24 months old. After the experiment, changes in morphology, function and fibrosis of their kidneys were detected. The levels of serum uric acid (UA), β2-microglobulin (β2-MG) and cystatin C (Cys C) were assayed with ELISA kits. The levels of extracellular matrix (ECM), matrix metalloproteinases (MMPs), tissue inhibitors of metalloproteinases (TIMPs), inflammatory factors and changes of oxidative stress parameters were examined. Results: After SPJs treatment, SD rats showed significantly histopathological changes in kidneys accompanied by decreased renal fibrosis and increased renal function; As compared with those in 3-month group, the levels of serum UA, Cys C and β2-MG in 24-month group were significantly increased (p < 0.05). Compared with those in the 24-month group, the levels of serum UA, Cys C and β2-MG in the SPJ-H group were significantly decreased. While ECM was reduced and the levels of MMP-2 and MMP-9 were increased, the levels of TIMP-1, TIMP-2 and transforming growth factor-β1 (TGF-β1)/Smad signaling were decreased; the expression level of phosphorylated nuclear factor kappa-B (NF-κB) was down-regulated with reduced inflammatory factors; meanwhile, the expression of nuclear factor erythroid 2-related factor 2-antioxidant response element (Nrf2-ARE) signaling was aggrandized. Conclusion: These results suggest that SPJs treatment can improve age-associated renal fibrosis by inhibiting TGF-β1/Smad, NFκB signaling pathways and activating Nrf2-ARE signaling pathways and that SPJs can be a potentially valuable anti-renal fibrosis drug.

Comprehensive comparative analysis of chloroplast genomes from seven Panax species and development of an authentication system based on species-unique single nucleotide polymorphism markers

  • Background: Panax species are important herbal medicinal plants in the Araliaceae family. Recently, we reported the complete chloroplast genomes and 45S nuclear ribosomal DNA sequences from seven Panax species, two (P. quinquefolius and P. trifolius) from North America and five (P. ginseng, P. notoginseng, P. japonicus, P. vietnamensis, and P. stipuleanatus) from Asia. Methods: We conducted phylogenetic analysis of these chloroplast sequences with 12 other Araliaceae species and comprehensive comparative analysis among the seven Panax whole chloroplast genomes. Results: We identified 1,128 single nucleotide polymorphisms (SNP) in coding gene sequences, distributed among 72 of the 79 protein-coding genes in the chloroplast genomes of the seven Panax species. The other seven genes (including psaJ, psbN, rpl23, psbF, psbL, rps18, and rps7) were identical among the Panax species. We also discovered that 12 large chloroplast genome fragments were transferred into the mitochondrial genome based on sharing of more than 90% sequence similarity. The total size of transferred fragments was 60,331 bp, corresponding to approximately 38.6% of chloroplast genome. We developed 18 SNP markers from the chloroplast genic coding sequence regions that were not similar to regions in the mitochondrial genome. These markers included two or three species-specific markers for each species and can be used to authenticate all the seven Panax species from the others. Conclusion: The comparative analysis of chloroplast genomes from seven Panax species elucidated their genetic diversity and evolutionary relationships, and 18 species-specific markers were able to discriminate among these species, thereby furthering efforts to protect the ginseng industry from economically motivated adulteration.

Discovery, semisynthesis, biological activities, and metabolism of ocotillol-type saponins

  • Ocotillol-type saponins are one kind of tetracyclic triterpenoids, sharing a tetrahydrofuran ring. Natural ocotillol-type saponins have been discovered in Panax quinquefolius L., Panax japonicus, Hana mina, and Vietnamese ginseng. In recent years, the semisynthesis of 20(S/R)-ocotillol-type saponins has been reported. The biological activities of ocotillol-type saponins include neuroprotective effect, antimyocardial ischemia, antiinflammatory, antibacterial, and antitumor activities. Owing to their chemical structure, pharmacological actions, and the stereoselective activity on antimyocardial ischemia, ocotillol-type saponins are subjected to extensive consideration. In this review, we sum up the discovery, semisynthesis, biological activities, and metabolism of ocotillol-type saponins.
