Characteristic study on the chemical components of Korean curved ginseng products

  • Dried ginseng (DG) is in fact the representing ginseng product in the worldwide market. Although it is made in various packages depending on the processing method, size and age of DG, basic scientific data reporting the chemical components are limited. In this study, 4-year-old curved ginseng (CG), one of the domestic DG products, was selected for further investigation. Eighty-six samples of 30 and 50 piece-grade CG, which are the most widely distributed in the market, were collected for 5 yr. Their major components, such as moisture, total sugar, acidic polysaccharides, total phenolic compounds, and saponins, were analyzed to figure out the standard quality characteristics. The moisture content of all CG samples was less than 15%. The total water-soluble sugar contents were 22.9% to 47.8% and 23.2% to 49.5% in the 30 and 50 piece-grade CG, respectively. The acidic polysaccharide contents were 3.6% to 6.7% and 2.9% to 6.9% in the 30 and 50 piece-grade CG, respectively. The total phenolic compound content was 0.4% to 0.5% in CG, regardless of the piece-grade. The crude saponin content, which represents the active component of ginseng, was over 2% in all samples. In 30 piece-grade CG samples, the contents of major ginsenosides, Rb1, Rf, and Rg1, were 2.2 to 4.7 mg/g, 0.4 to 1.3 mg/g, and 1.6 to 4.0 mg/g, respectively. The ginsenoside contents in 50 piece-grade CG samples were 2.1 to 3.9 mg/g (Rb1), 0.5 to 1.2 mg/g (Rf), and 1.3 to 3.4 mg/g (Rg1). Overall, since there were relatively high standard deviation and coefficient of variation in all the chemical component contents that were assessed, we found some difficulties in showing the CG standard chemical component characteristics by average, standard deviation, and other statistical analysis factors.

A Comparative Study of High-Performance Liquid Chromatographic Method for Analysis of Ginseng Saponin (인삼(人蔘)사포닌분석(分析)을 위한 고속액체(高速液體)크로마토그래피법(法)의 비교연구(比較硏究))

  • Ginseng saponins separated from Panax ginseng C.A. Meyer were analyzed by high-performance liquid chromatography using a carbohydrate analysis column. The effect on the resolution and retention time of each ginsenosides, as well as the addition effect of n-butanol on a acetonitrile/water system, was examined using various proportions of acetonitrile/water system (80/20-94/6) which have been used with typical solvent of carbohydrate analysis column. The retention time of each ginsenosides was greatly affected by the compositions and mixture proportions of the mobile phase and also markedly increased as the proprotion of acetonitrile in mobile phase increased. It was proved that acetonitrile/water system (80/20) and acetonitrile/water/n-butanol system (86/14/10) were very effective mobile phases for diol and triol sapoin analyses, respectively. According to the result obtained by this method, the PT/PD ratios of white and red ginseng saponins were 0.401 % and 0.561 %, respectively. Red ginseng increased PT/PD ratio, compared with white ginseng. This is attributed to the change of saponin pattern by processing method and the change of PT/PD ratio would greatly influence on the biochemical and pharmacological effects of ginseng and its products.

Characteristics of New Cultivars in Panax ginseng C.A. Meyer (고려인삼 신품종 특성)

  • This paper reports the characteristics of 8 new cultivars for selected from Korean ginseng. The occurance of multi stems were the highest in Yunpoong(45%) and the lowest in Gumpoong(7%), but growth of aerial parts were the highest in Gumpoong and the lowest In Yunpoong among new cultiyars. The ratio of seeds harvest were the highest in Gumpoong(85.4%) and the lowest in Chunpoong(69.1%), but number of seeds per plant were the highest in Yunpoong(108.3ea) and the lowest in Chunpoong(77.5ea) among new cultivars. The ratio of leaf burning were the highest in Chunpoong but the lowest in Yunpoong among new cultivars. In weight distribution of the different parts of the ginseng roots, the ratio of main root were high in Jakyungjong(63.1%) but low in new cultivars(49%-55.9%), but the ratio of lateral root were high in new cultivars(19.3-23.3%), but low in Jakyungjong(13.2%), the ratio of fine root were not different. Root yield declined in the order of Yunpoong, Gumpoong, Gopoong, Chunpoong, Sunpoong, Jakyungjong. The length of main root were the longest in Chunpoong(8,0cm) but the shortest in Yunpoong(6,4cm), The ratio of rusty-root was low in new cultivars(0,2-9,5%), but high in Jakyungiong(16,3%). The grade of red ginseng roots decreased in the order of Chunpoong, Gumpoong, Gopoong, Sunpoong, Yunpoong, Cheongsun, Jakyungjong. The total ginsenoside contents per dry weight in main roots was high in Gumpoong(8.53mg), Yunpoong(8.13mg), Gopoong(7,47mg), but low in Chunpoong(5.73mg), Sunpoong(4.87mg).

Ginsenoside Rg1 suppresses early stage of adipocyte development via activation of C/EBP homologous protein-10 in 3T3-L1 and attenuates fat accumulation in high fat diet-induced obese zebrafish

  • Background: Ginsenoside Rg1 is a class of steroid glycoside and triterpene saponin in Panax ginseng. Many studies suggest that Rg1 suppresses adipocyte differentiation in 3T3-L1. However, the detail molecular mechanism of Rg1 on adipogenesis in 3T3-L1 is still not fully understood. Methods: 3T3-L1 preadipocyte was used to evaluate the effect of Rg1 on adipocyte development in the differentiation in a stage-dependent manner in vitro. Oil Red O staining and Nile red staining were conducted to measure intracellular lipid accumulation and superoxide production, respectively. We analyzed the protein expression using Western blot in vitro. The zebrafish model was used to investigate whether Rg1 suppresses the early stage of fat accumulation in vivo. Results: Rg1 decreased lipid accumulation in early-stage differentiation of 3T3-L1 compared with intermediate and later stages of adipocyte differentiation. Rg1 dramatically increased CAAT/enhancer binding protein (C/EBP) homologous protein-10 (CHOP10) and subsequently reduced the $C/EBP{\beta}$ transcriptional activity that prohibited the initiation of adipogenic marker expression as well as triglyceride synthase. Rg1 decreased the expression of extracellular signal-regulated kinase 1/2 and glycogen synthase kinase $3{\beta}$, which are also essential for stimulating the expression of $CEBP{\beta}$. Rg1 also reduced reactive oxygen species production because of the downregulated protein level of nicotinamide adenine dinucleotide phosphate hydrogen (NADPH) oxidase 4 (NOX4). While Rg1 increased the endogenous antioxidant enzymes, it also dramatically decreased the accumulation of lipid and triglyceride in high fat diet-induced obese zebrafish. Conclusion: We demonstrated that Rg1 suppresses early-stage differentiation via the activation of CHOP10 and attenuates fat accumulation in vivo. These results indicate that Rg1 might have the potential to reduce body fat accumulation in the early stage of obesity.

Ginsenoside Rb2 suppresses cellular senescence of human dermal fibroblasts by inducing autophagy

  • Background: Ginsenoside Rb2, a major active component of Panax ginseng, has various physiological activities, including anticancer and anti-inflammatory effects. However, the mechanisms underlying the rejuvenation effect of Rb2 in human skin cells have not been elucidated. Methods: We performed a senescence-associated β-galactosidase staining assay to confirm cellular senescence in human dermal fibroblasts (HDFs). The regulatory effects of Rb2 on autophagy were evaluated by analyzing the expression of autophagy marker proteins, such as microtubule-associated protein 1A/1B-light chain (LC) 3 and p62, using immunoblotting. Autophagosome and autolysosome formation was monitored using transmission electron microscopy. Autophagic flux was analyzed using tandem-labeled GFP-RFP-LC3, and lysosomal function was assessed with Lysotracker. We performed RNA sequencing to identify potential target genes related to HDF rejuvenation mediated by Rb2. To verify the functions of the target genes, we silenced them using shRNAs. Results: Rb2 decreased β-galactosidase activity and altered the expression of cell cycle regulatory proteins in senescent HDFs. Rb2 markedly induced the conversion of LC3-I to LC3-II and LC3 puncta. Moreover, Rb2 increased lysosomal function and red puncta in tandem-labeled GFP-RFP-LC3, which indicate that Rb2 promoted autophagic flux. RNA sequencing data showed that the expression of DNA damage-regulated autophagy modulator 2 (DRAM2) was induced by Rb2. In autophagy signaling, Rb2 activated the AMPK-ULK1 pathway and inactivated mTOR. DRAM2 knockdown inhibited autophagy and Rb2-restored cellular senescence. Conclusion: Rb2 reverses cellular senescence by activating autophagy via the AMPK-mTOR pathway and induction of DRAM2, suggesting that Rb2 might have potential value as an antiaging agent.

Rapid Determination of Ginsenosides Rb1, Rf, and Rg1 in Korean Ginseng Using HPLC (HPLC를 이용한 고려인삼 중 진세노사이드 Rb1, Rf 및 Rg1의 신속분석 방법 개발)

  • A simple gradient HPLC method for rapid determination of major ginsenosides ($Rg_1$ and $Rb_1$) and unique ginsenoside (Rf) of Korean ginseng (Panax ginseng C.A. Meyer) was developed. Within 50min, three ginsenosides have been separated and identified on $\mu$-Bondapak $C_{18}$ column ($3.9{\times}300\;mm$, $10{\mu}m$) with gradient elution using water and acetonitrile as a mobile phase. The method was validated in terms of linearity, accuracy, and precision. The correlation coefficients ($r^2$) for calibration curves of ginsenosides were over 0.9997. The developed HPLC method was successfully applied to the analysis of ginseng samples and the recoveries of ginsenosides were in the range of $101.1{\sim}115%$ with RSD<3.2%. The developed method could be used for rapid evaluation of the ginsenosides $Rg_1$, $Rb_1$, and Rf.

Extracting Conditions for Promoting Ginsenoside Contents and Taste of Red Ginseng Water Extract (홍삼 물 추출액의 사포닌 함량 및 맛의 증진을 위한 추출 조건)

  • In this study, red ginseng extract solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total red ginseng extracts was 64.6 mg / 100 ml which was extracted at $75^{\circ}C$ for 18 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest total content of $Rb_2$ and Re was 11.8 mg / 100 ml at $75^{\circ}C$ for 12 hours which decreased according to the increased extraction temperature and time. The prosapogenin content of red ginseng extract was increased at $75^{\circ}C$ and $85^{\circ}C$ while the content decreased at $95^{\circ}C$, in which the highest prosapogenin content was 34.9 mg / 100 ml at $85^{\circ}C$ for 24 hours. The total sugar content and cloudness were increased according to the increased extraction time at $95^{\circ}C$, but pH and hue value were decreased according to the increased extracted time. The highest sweetness content was 4.0% which was found at $95^{\circ}C$ for 24 hours extract. Therefore, the most appropriate red ginseng extracting method was lower the temperature for saponin content at first time in combination with raise the temperature for taste at second time.

Antioxidative Effect of Crude Saponin Fraction Prepared from Culture Product of Basidiomycota cultured with Fresh Ginseng as Substrate (수삼을 기질로 한 담자균 배양물로부터 분리한 조사포닌의 항산화 효과)

  • Antioxidative activity of crude saponin fraction (CSF) prepared from Basidiomycota cultured with fresh ginseng (Panax ginseng C.A. Meyer) as substrate was investigated by analyzing CSFs for ginsenoside and phenolic compounds. On TLC chromatogram, ginsenosides such as $Rg_{2},\;Rg_{3}$, and $Rh_{1}$ which were rare in fresh ginseng, were identified. CSF of Phellinus linteus culture product showed the highest total phenolic content and electron donating ability (EDA), suggesting phenolic compounds contribute to EDA. In vitro lipid peroxidation was inhibited most by CSF of Ganoderma lucidum, indicating that the highest EDA does not imply highest inhibition against lipid peroxidation. Tyrosinase was also inhibited mostly by CSF of G. lucidum. These results suggest culture of Basidiomycota with fresh ginseng has more active substances than fresh ginseng alone.

Ginsenosides Protect the High Glucose-induced Stimulation of IGFs in Mesangial Cells (Mesangial 세포에서 고포도당에 의해 유도되는 insulin-like growth factor 분비 촉진작용에 대한 ginsenosides의 차단 효과)

  • Panax ginseng C. A. MEYER is one of the most widely used herbal medicines in the Asian countries and has diverse functions including anti-diabetic action. The dysfunctions of mesangial cells in hyperglycemic conditions are implicated in the development of diabetic nephropathy. Insulin-like growth factors (IGFs) are also associated with the onset of diabetic nephropathy. Thus, we examined the effect of ginsenosides against high glucose-induced dysfunction of primary cultured rat mesangial cells. In the present study, high glucose increased IGF-I and IGF-II secretion in mesangial cells. Ginsenoside total saponin (GTS) prevented high glucose-induced increase of IGF-I and IGF-II secretion in mesangial cells. In addition, GTS prevented high glucose-induced increase of lipid peroxide formation and decrease of GSH contents. GTS also ameliorates high glucose-induced increase of arachidonic acid release and decrease of prostaglandin $E_2$. In conclusion, GTS prevented high glucose-induced dysfunction of mesangial cells via inhibition of oxidative stress and arachidonic acid pathways.

  • In this paper, new methods for the determination of the total and the individual saponin glucosides were proposed. One of them was a colorimetric method following Two-dimensional Thin layer chromatography. And the method employing Thinchrograph TFG-10 or Densitorol DMU-33C followed the separation of the saponins by means of a preparative thin layer chromatography. In accordance with the density of the chromatogram of each saponin, Thinchrogram or Densitogram of the individual saponin fraction was plotted and determined.
