Chemical Composition and Immunostimulating Activity of the Fermented Korean Ginseng (Panax ginseng C.A. Meyer) with Mushroom Mycelium by Solid Culture (균사체를 이용한 수삼 고체발효물의 화학적 조성 및 면역 활성)

  • For the utilization of Korean ginseng (Panax ginseng C.A. Meyer) in the functional drink, we prepared the fermented Korean ginseng with mushroom mycelia (Ganoderma lucidum; WG-GL, Hericium erinaceum; WG-HE and Phellinus linteus; WG-PL) by solid culture. A proximate analysis showed that the fermented Korean ginseng contained significantly more crude fat (4.66$\sim$12.02%) than Korean ginseng (WG, 1.61%) whereas crude protein content of WG (13.64%) was higher value than those of the ferments (7.60$\sim$12.57%). When we also evaluated effects of the fermented Korean ginseng on the mitogenic activity, hot-water extract from WG-PL was significantly higher than those of WG or mycelia only fermentation (GL, HE and PL) as analyzed by IL-2 production (1.64-fold of the saline control) and proliferation of splenocytes (1.47-fold). In addition, the lysosomal phosphatase activity (WG-HE; 1.32-fold) and NO/TNF-$\alpha$ production (WG-HE; 2.27-fold of the saline control at 50 ${\mu}g$/mL, WG-PL; 3.56-fold, respectively) from macrophage in the presence of the fermented Korean ginseng were higher than those of WG or mycelia fermentation. These results indicate that hot-water extracts from the fermented Korean ginseng with mushroom mycelia by solid culture contain chemical ingredients different from the Korean ginseng, and that it might provide beneficial immunostimulating activity.

Antioxidant Activity of Panax ginseng Flower-buds Fermented with Various Microorganisms (발효 미생물에 따른 인삼꽃의 항산화 활성)

  • To improve the use of ginseng flower-buds, antioxidant activities of ginseng flower-buds fermented using a variety of useful microorganisms were analyzed. Non-fermented grape pomace was used as a control, while fermentation was carried out using Bacillus subtilis (BS), Lactobacillus plantarum (LP), Lactobacillus casei (LC), Candida utilis (CU), Saccharomyces cerevisiae strain CHY1011 (Y1), Saccharomyces cerevisiae strain ZP 541 (Y2), and a mixed-strain culture with LP, LC, and CU (M). The total polyphenol content of ginseng flower-buds was highest in the control compared to the other fermented ginseng flower-buds. DPPH radical and ABTS radical scavenging activity were also highest in fermented group by BS. The FRAP value (10 mg/mL) was highest in the control group but did not show a significant difference in the fermented group by BS. The highest reducing power activity was in the fermented group by LC compared to the other group, including the control. Therefore, the fermentation of ginseng flower-buds using various microorganisms, shows that fermentation with the Bacillus subtilis strain increases antioxidant activity. More research of its effects on other physiological activities will be needed.

Ginseng saponin metabolite 20(S)-protopanaxadiol relieves pulmonary fibrosis by multiple-targets signaling pathways

  • Background: Panax ginseng Meyer is a representative Chinese herbal medicine with antioxidant and anti-inflammatory activity. 20(S)-Protopanaxadiol (PPD) has been isolated from ginseng and shown to have promising pharmacological activities. However, effects of PDD on pulmonary fibrosis (PF) have not been reported. We hypothesize that PDD may reverse inflammation-induced PF and be a novel therapeutic strategy. Methods: Adult male C57BL/6 mice were used to establish a model of PF induced by bleomycin (BLM). The pulmonary index was measured, and histological and immunohistochemical examinations were made. Cell cultures of mouse alveolar epithelial cells were analyzed with Western blotting, coimmunoprecipitation, immunofluorescence, immunohistochemistry, siRNA transfection, cellular thermal shift assay and qRT-PCR. Results: The survival rate of PPD-treated mice was higher than that of untreated BLM-challenged mice. Expression of fibrotic hallmarks, including α-SMA, TGF-β1 and collagen I, was reduced by PPD treatment, indicating attenuation of PF. Mice exposed to BLM had higher STING levels in lung tissue, and this was reduced by phosphorylated AMPK after activation by PPD. The role of phosphorylated AMPK in suppressing STING was confirmed in TGF-b1-incubated cells. Both in vivo and in vitro analyses indicated that PPD treatment attenuated BLM-induced PF by modulating the AMPK/STING signaling pathway. Conclusion: PPD ameliorated BLM-induced PF by multi-target regulation. The current study may help develop new therapeutic strategies for preventing PF.

  • The balance between metabolic activation of xenobiotics and detoxification of their active metabolites may playa vital role in controlling mutagenic and carcinogenic processes. To assess the possible role of P. ginseng C.A. Meyer in detoxification of xenobiotics, we studied the effects of ginseng on several parameters of the monooxygenasd system, including benzo(a) pyrene monooxygenase(AHH) and benzo(a) pyrene epoxide hydratase(EH) as well as effects of ginseng on the conjugation system. Test animals receiving ginseng saponin-fraction induced epoxide hydratase activity to over $150\%$ (20mg/kg b.w.) of the control and increased glutathione transferase activity (GSH-T) up to $140\%$ (20mg/kg b.w.) of the control, whereas no significant changes were observed in the benzopyrene monooxygenase activity (AHH). Such a selective induction of the inactivation enzyme epoxide hydratase, combined with a marked elevation of the detoxifying enzyme glutathione transferase, without a concurrent induction of benzopyrene monooxygenase which is responsible for the formation of carcinogenic intermediates, demonstrates that ginseng has the potential to alter the metabolic course of carcinogenic polycyclic aromatic hydrocarbons, and thereby enhance detoxification. Thus, ginseng may play an important role in the prevention of tumors caused by carcinogens.

Study on The Preventive Effect of Ginsenosides Against Hypercholesterolemia and Its Mechasnism (인삼사포닌의 고 cholesterol 혈증 강하작용에 관한 연구)

  • The Preventive effect of the saponin fraction of Panax ginseng C.A. Meyer against hyperchole- sterolemia was demonstrated by assaying the cholesterol and triacylglyceride level of the blood serum and liver of rats fed high-cholesterol diet with and/or without ginsenoside. To understand the mechanism of the preventive action of ginsenoside, ginsenoside effect on LDL receptor binding ability, cholesterol level, and cAMP level of Chinese hamster ovary (CHO) cells cultured under various conditions were examined. When LDL (20 $\mu$g/ml) was added to the culture medium for CHO cell culture, LDL receptor binding activity of the cell was suppressed down to 58% of that of normal group. Ginsenosides at 10--2% and 10-3% level (w/v) were shown to exert an appreciable stimulatory effect on CHO cell LDL receptor activity, which partially overcame the suppression due to the presence of LDL (20 $\mu\textrm{g}$/ml) in the medium. Ginsenosides also reduced the increased cholesterol level of test group almost to that of normal group, and it increased cAMP level, which was usually reduced to 55% of that of the normal group due to the presence of LDL in the medium. Comparison of Kd and Bmax value of CHO cells cultured under different conditions revealed that this stimulation was due not to the receptor's binding affinity but to its number. Addition of ginsenoside (10-2%) decreased the uptake of taurocholic acid as much as 20% at the actively transporting everted ileal sacs, but it failed to form a large mixed micelles with taurocholic acid, which was one of the proposed mechanisms by which ginsenoside inhibits bile acid reabsorption. From the above results, it seemed likely that ginsenoside prevented hypercholestrolemia by decreasing cholesterol level in cells thereby relieving the inhibition of LDL receptor synthesis by cholesterol and also by inhibiting bile acid reabsorption from the small intestine.

  • As Korean ginseng is hybrid, an individual variation is very severe, and it takes long times in new breeding because it is required 4 years to pick the seed. But, transformation technique makes the high-functional breeding in short time. The focus of these ginseng studies is to find and secure the useful gene. And it is urgent to accumulate the fundamental data for the molecular breeding and secure the useful genes. Therefore, transformation and soil acclimatization technique are necessary to molecular breeding in use of the introduction of functional genes. In this study, it add to secure of new regulation gene and useful gene as to accumulate the fundamental data for the place where it will contribute to raise the national competitive power. To analyze the useful genes in large scale, we constructed CDNA libraries with various tissues, species, and treated tissue. EST analysis of ginseng perform in large scale and build the EST database of ginseng. We perform the full length sequencing about the selected lots of clones that include the entire open reading frame of the amino acid residues and construct cDNA chip with the parental EST clones. Establishment of the transformation and a soil acclimatization system throuth the re-introduction of the selected ginseng gene that related with the secondary metabolism and anti-stress into the ginseng.

Effect of Blue and Yellow Polyethylene Shading Net on Growth Characteristics and Ginsenoside Contents in Panax ginseng C. A. Meyer (청색과 황색 해가림이 인삼의 생육 및 진세노사이드 함량에 미치는 영향)

  • Yield and ginsenoside contents of ginseng (Panax ginseng C. A. Meyer) is affected by light intensity and quality, and the color and the thickness of PE shading net when PE net is utilized for shading material. This study was carried out to investigate the effect of light quality on root yield and ginsenoside contents off-year-old ginseng by using polyethylene shading net with each blue and yellow color, Spectral irradiance under blue and yellow shading net showed the peak at 498 nm and 606 nm, respectively, which made distinct difference in light quality. Heat injury ratio of blue shading net was increased distinctly more than that of yellow shading net in summer season because of higher transmitted quantum (23%)and air temperature (0.3 $^{\circ}$C) in blue shading net than those of yellow shading net. Chlorophyll content and leaf area under yellow shading net were higher than those of blue shading net, and its heat injury ratio was lower than those of blue. These effects may led to 48% higher increase of root yield under yellow shading net than that under blue shading net. The content of total ginsenoside in taproot was not significantly differed between blue and yellow shading net, while the content in lateral and fine root was significantly increased in blue shading net compared to yellow shading net. PDM ratio of blue shading net showed more significant increase in lateral root than that of yellow shading net. All of Rb$_1$/Rg$_1$ ratio in three parts of root under blue shading net was higher than that of yellow shading net, but there were no significant increase in the ratio of lateral root.

  • To define the effects of synthetic auxins on rooting from the ginseng stern cutting and the root growth in diameter after the rooting of the cuttings, stern cuttings with palmate leaves obtained from seedlings and 2-year old ginseng plants were planted in rooting media treated with solutions of the synthetic auxins. All the roots induced from the cuttings were adventitious fibrous roots at first, but a few adventitious roots of the cutting were thickened in diameter to over 2 to 3mm at 120 days after cutting and the rest of them disappeared. IBA was the most effective auxin for rooting and root growth in diameter after rooting from the cuttings. The shape of the roots that thickened in diameter could be divided into two types. Both types of thickened roots were fully lignified at 120 days after cutting and those thickened roots did not dry up or die by July of the next year, however no rhyzomes or shoot primodia were induced from them.

Changes in Chemical Composition of Korean Red Ginseng (Panax ginseng C.A. Meyer) Extract With Alcohol Extraction

  • We extracted red ginseng with various alcohol concentrations and evaluated total carbohydrate, uronic acid, polyphenols compounds and ginsenoside contents, and yields of alcohol extract. The water extraction (0% alcohol extraction) showed a high level of total carbohydrate content. 10% and 20% alcohol extraction showed the highest uronic acid contents (7,978.8 and $7,872.7\;{\mu}g/mL$ of extract, respectively). The efficiency order of the red ginseng extract (RGE) preparations in liberating polyphenols was: $0{\sim}50%$ alcohol${\geq}\;60%$ alcohol> $70{\sim}90%$ alcohol. Solid contents in RGE were decreased with increased alcohol concentration; the same tendency as with the results of total carbohydrate content. Total ginsenoside contents in $20{\sim}50%$ alcohol extracts showed similar levels ($442,962.9{\sim}47,930.8\;{\mu}g/mL$ of extract). Water extraction showed the lowest ginsenoside content ($14,509.4\;{\mu}g/mL$ of extract). The ginsenoside contents at above 60% alcohol were decreased with increased alcohol concentration. Generally, ginsenoside (Rg2, Rg1, Rf, Re, Rd, Rb2, Rc and Rb1) contents were increased with increased alcohol concentrations. However, Rg3 content was decreased with increases in alcohol concentration.

Production and Identification of Anthocyanin in Hairy Root Cultures of Ginseng (인삼 모상근 배양에 의한 Anthocyanin의 생산과 동정)

  • In hairy root cultures of ginseng (Panax ginseng C.A. Meyer) transfonned by Agrobacterium rhizogenes, the effects of light, carbon source and various honnone on hairy root growth and anthocyanin production were investigated. Anthocyanin synthesis began to first occur 5 days after exposure to light, and then maximum yield of anthocyanin was 0.36 mg/g(fr wt) in MS medium after 30 days. Of the nutritional factors concentration of 60 mM nitrogen and sucrose as a carbon source showed marked effects on the growth and anthocyanin productiom MS medium supplemented with 0.5 mg/L IAA was most suitable for the hairy root proliferation, and the best accumulation of anthocyanin was obtained at 1 mg/L IAA treatment (0.41 mg/g, fr wt). Whereas 2,4-D tended to restrain the pigment synthesis. From the isolation and identification of anthocyanin pigments, main anthocyanin in ginseng hairy root was identified as pelargonidin-glucoside.coside.
