V. A. Mycorrhizae가 미국삼의 생육에 미치는 영향 (Effects of Vesicular-Arbuscular Mycorrhizae on The Growth of American Ginseng)

  • Newly sprouted American ginseng (Panax quinquefolium L.) seedlings were transplanted to forest pots with mycorrhizae-infested soil and grown in screenhouse for 2 years. Growth patterns, mortality rate and fresh root weight were investigated. Plants in VAM soil had lower mortality rales than control. In soils infested with two species of mycorrhizal fungi (Glomu deseyicola, frappe, Bloss and Merge and G. intraradices, Schenck and Smith), 28-35% of plants produced 3-prongs in the second season and significantly increased fresh root weight by 41 to 43%.

Metabolite Analysis of Panax ginseng C. A. Meyer by HPLC According to Root Age

  • In order to investigate the major metabolite patterns of aged Panax ginseng C.A. Meyer roots, the ginsenoside contents for white ginseng roots of various ages were compared. The 1-year to 6-year old roots were extracted with methanol, and then the methanol-soluble metabolites were analyzed by high performance liquid chromatography (HPLC). The metabolite contents of the 1-year and 2-year roots, including the ginsenosides and minor components, were not different, but the $Rg_1$, Re, and Rc ginsenoside contents between the 2-year and 3-year roots showed significant differences. $Rg_1$ and Rc increased significantly in the 1-year to 2-year roots, and Re increased significantly from the 3-year root age. Rd increased slightly until the 2-year age and decreased from the 3-year age. Based on the ginsenoside distributions and contents at various root ages, we have suggested 2 biogenesis schemes using the ginsenosides that have been isolated from the roots of P. ginseng so far.

고려인삼(Panax RiwenR) Invertase의 화학조성과 안정성 (Chemical composition and Stabilities of Invertase from Korean Ginseng, Panax ginseng)

  • The chemical composition and stabilities of the purified ginseng invertase were investigated. The purified enzyme was found to be a glycoprotein composed of 80.2% protein and 19.7% total sugar. The protein component of the enzyme was composed of acidic amino acid (9.3%), basic amino acid (48.9%), nonpolar amino acid (21.4%), polar amino acid (20.4%) and 6.1% S-containing amino acid. It showed especially high contents of histidine and serine. The enzyme was inactivated almost completely by the treatment with some proteases (papain, pepsin. trypsin, pancreatin and microbial alkaline pretense) and protein denatllrants (8M urea and 6M guanidine-HC1), bolt not with glyrosidase (${\alpha}$-amylase, ${\beta}$-amylase. glcoamylese and cellullase). btonosaccharides sllch as glilrose, fructose, galactose and mannose did not exert any influence on the enzyme activity. The activity of the enzyme was inhibited by Ag+, Mn2+, Hg2+, Zn2+ and Al3+, whereas Ca2+, Mg2+, Ba2+ and Fe3+ gave rather activating effects on the enzyme activity. The enzyme was relatively stable in the VH range of VH 6 and 8, and at the temperatures below 35$^{\circ}C$.

Biological control of Colletotrichum panacicola on Panax ginseng by Bacillus subtilis HK-CSM-1

  • Background: Biological control of plant pathogens using benign or beneficial microorganisms as antagonistic agents is currently considered to be an important component of integrated pest management in agricultural crops. In this study, we evaluated the potential of Bacillus subtilis strain HK-CSM-1 as a biological control agent against Colletotrichum panacicola. Methods: The potential of B. subtilis HK-CSM-1 as a biological control agent for ginseng anthracnose was assessed. C. panacicola was inoculated to ginseng plants and the incidence and severity of disease was assessed to examine the efficacy of the bacterium as a biological control against C. panacicola. Results: Inoculation of Panax ginseng plants with B. subtilis significantly suppressed the number of disease lesions of C. panacicola and was as effective as the chemical fungicide iminoctadine tris(albesilate). The antifungal activity of B. subtilis against C. panacicola was observed on a co-culture medium. Interestingly, treatment with B. subtilis did not significantly affect the diameter of the lesions, suggesting that the mechanism of protection was through the reduction in the incidence of infection related to the initial events of the infection cycle, including penetration and infection via spore germination and appressorium formation rather than by the inhibition of invasive growth after infection. Conclusion: Our results suggest that B. subtilis HK-CSM-1 can be used as an effective and ecologically friendly biological control agent for anthracnose in P. ginseng.

Seed and Root Rots of Ginseng (Panax quinquefolius L) Caused by Cylindrocarpon destructans and Fusarium spp.

  • Ginseng (Panax quinquefolius L.) has become one of the most valuable herb crops grown in North America. However, traditional cropping practices are favourable to disease and significant losses due to root disease are common, despite frequent use of fungicides. Seedlots are often contaminated with pathogens, however, little is known about the causes of seed decay and the role of seed pathogens as incitants of root rots. It was shown that both Fusarium spp. and Cylindrocarpon destructans were able to rot seeds and that C. destructans was more virulent than Fusarium spp. on seedling roots. A modified rose bengal agar MRBA) medium (1 g KH$_2$PO$_4$; 0.5 g MgSO$_4$; 50 mg rose bengal; 10 g dextrose; 5 g Bacto peptone; 15 g Bacto agar; 30 mg streptomycin sulfate; 250 mg ampicillin; 10 mg rifampicin; 500mg pentachloronitrobenzene; 500 mg dicloran; and 1 L distilled water) was superior to potato dextrose agar in detecting C. destuctans in diseased roots. Isolation of C. destructans from diseased seedlings arising from seeds sown in replant soil supported the hypothesis that this pathogen is a cause of ginseng replant failure in North America.

  • This study was undertaken to evaluate the automatic decision-making on the grading of 6-year-old fresh ginseng (Panax ginseng C.A. Meyer) by an image analyzer. The best input method for the 6-year-old fresh ginseng was under condition of a low resolution (128u 128 pixel) and illumination direction from bottom to up (light box). It was possible to identify the main root, lateral root, and rhizome of fresh ginseng by application of OPEN process in a function of an image analyzer. Finally, we developed the grade decision-making programs, GinP-1. The fitness rates for the fresh ginseng standards which were classified by experts were 94.6, 80.6, 81.5, and 100.0% for 1st, 2nd, 3rd, and 4th grade of fresh ginseng, respectively, and the total time of decision-making was about 4.3 seconds per one root. The decision-making time was reduced to 0.8 seconds per one root by enhancemeat of the Image analyzer, which was tested by the technical company of the image analyzer,'Carl Zeiss (Germany). As a result of this study, the automatic decision-making on the grade of fresh gin send by image analyzer seems to have high possibility.

고려인삼(Panax ginseng C.A.Meyer)의 간이법에 의한 홍삼제조 및 사포닌 성분분석 (Saponin Analysis and Red Ginseng Production using the Simplified Method of Korean Ginseng (Panax ginseng C.A.Meyer))

  • 홍삼 제조공정을 단순화하면서 효율적으로 유효성분을 증대시키기 위해서 4년근 수삼을 구입하여 깨끗이 수세한 후 $96-98^{\circ}C$에 3시간 정도 수증기로 증삼한 후 30시간 정도 열풍건조하여 홍삼제조를 위한 간이법을 개발하였다. 제조한 홍삼으로부터 홍삼엑스($60^{\circ}$ brix 이상)를 제조하였으며 홍삼엑스의 추출수율은 60% 이상이었다. 간이법으로 제조한 홍삼을 재료로 하여 농축한 홍삼엑스의 사포닌 함량을 조사하기 위해서 HPLC를 이용하여 총 사포닌(total saponin)을 분석하였다. 그 결과 PD계 사포닌 중 암세포전이억제효과와 평활근이완작용이 탁월한 ginsenoside-Rg3에는 H사에 비하여 2배 이상 검출되었으며, 간상해 억제작용과 혈소판 응집억제작용이 있는 것으로 알려진 ginsenoside-Rh1종의 경우 3배 이상 검출이 되었다.

인삼 및 모상근의 프로테옴 분석을 위한 단백질 추출 방법 (Purification of Crude Protein Mixture from Panax ginseng and Hairy Root for Proteome Analysis)

  • 인삼 (Panax ginseng C.A. Meyer)은 우리나라의 전통약재로 수 세기 동안 사용되어 왔으며, 약효 및 성분에 대한 연구가 매우 활발하게 이루어져 왔으나, 인삼의 분자생물학적, 단백질 화학적 측면에서 생리연구는 매우 미미하였다. 본 연구에서는 프로테옴믹스를 이용한 인삼 단백질 군을 연구하기 위한 첫 단계로 인삼 (천풍)및 모상근의 단백질 추출 방법을 확립하고, 이차원 전기영동을 통하여 추출방법의 유용성을 확인하였다. Homogenizer와 황산암모늄 침전을 통한 단백질 추출과 액체질소 및 TCA를 이용한 단백질 추출 방법을 수행하여 비교해 본 결과 추출 단백질 양에는 큰 차이가 없으나 이차원 전기영동 수행 시 액체질소 및 TCA를 통한 방법이 프로테옴 연구에 적합하여 훨씬 해상도가 높은 gel 이미지를 얻었을 뿐 아니라, gel당 660개 이상의 단백질 sopt을 확인하였다. 또한 인삼과 모상근의 이차원 전기영동을 비교한 결과 상당히 다른 발현 패턴을 보여, 생장조건 등 외부 환경과 생리상태의 차이에 따라, 같은 유전자를 가지고 있는 조직체라 할지라도, 매우 다른 프로테옴 (proteome)을 보일 수 있음을 보여주었다.

Anti-melanogenic property of ginsenoside Rf from Panax ginseng via inhibition of CREB/MITF pathway in melanocytes and ex vivo human skin

  • Background: Ginsenosides of Panax ginseng are used to enhance skin health and beauty. The present study aimed to investigate the potential use of ginsenoside Rf (Rf) from Panax ginseng as a new anti-pigmentation agent. Methods: The anti-melanogenic effects of Rf were explored. The transcriptional activity of the cyclic adenosine monophosphate (cAMP) response element binding protein (CREB) and the expression levels of tyrosinase, microphthalmia-associated transcription factor (MITF), and tyrosinase-related proteins (Tyrps) were evaluated in melanocytes and UV-irradiated ex vivo human skin. Results: Rf significantly inhibited Forskolin (FSK) or UV-stimulated melanogenesis. Consistently, cellular tyrosinase activity and levels of MITF, tyrosinase, and Tyrps were downregulated. Furthermore, Rf suppressed MITF promoter activity, which was stimulated by FSK or CREB-regulated transcription coactivator 3 (CRTC3) overexpression. Increased CREB phosphorylation and protein kinase A (PKA) activity induced by FSK were also mitigated in the presence of Rf. Conclusion: Rf can be used as a reliable anti-pigmentation agent, which has a scientifically confirmed and reproducible action mechanism, via inhibition of CREB/MITF pathway.
