A Mutagenic Study of β-1,4-Galactosyltransferases from Neisseria meningitidis

  • N-terminal His-tagged recombinant $\beta$-1,4-galactosyltransferase from Neisseria meningitidis was expressed and purified to homogeneity by column chromatography using Ni-NTA resin. Mutations were introduced to investigate the roles of, Ser68, His69, Glu88, Asp90, and Tyr156, which are components of a highly conserved region in recombinant $\beta$-1,4 galactosyltransferase. Also, the functions of three other cysteine residues, Cys65, Cys139, and Cys205, were investigated using site-directed mutagenesis to determine the location of the disulfide bond and the role of the sulfhydryl groups. Purified mutant galactosyltransferases, His69Phe, Glu88Gln and Asp90Asn completely shut down wild-type galactosyltransferase activity (1-3%). Also, Ser68Ala showed much lower activity than wild-type galactosyltransferase (19%). However, only the substitution of Tyr156Phe resulted in a slight reduction in galactosyltransferase activity (90%). The enzyme was found to remain active when the cysteine residues at positions 139 and 205 were replaced separately with serine. However, enzyme reactivity was found to be markedly reduced when Cys65 was replaced with serine (27%). These results indicate that conserved amino acids such as Cys65, Ser68, His69, Glu88, and Asp90 may be involved in the binding of substrates or in the catalysis of the galactosyltransferase reaction.

Hydrothermal Synthesis and Structural Characterization of x mol% Calcia-Stabilized ZrO2 Nanopowders (x mol% 칼시아-안정화 지르코니아 나노분말의 수열합성 및 구조적 특성평가)

  • Pure zirconia and $x$ mol% calcia partially stabilized zirconia ($x$ = 1.5, 3, and 8) nanopowders were synthesized by hydrothermal method with various reaction temperatures for 24 hrs. The precipitated precursor of pure zirconia and $x$ mol% calcia doped zirconia was prepared by adding $NH_4OH$ to starting solutions; resulting sample was then put into an autoclave reactor. The optimal experimental conditions, such as reaction temperatures and times and amounts of stabilizer CaO, were carefully studied. The synthesized $ZrO_2$ and $x$ mol% CaO-$ZrO_2$ ($x$ = 1.5, 3, and 8) powders were characterized by XRD, SEM, TG-DTA, and Raman spectroscopy. When the hydrothermal temperature was as low as $160^{\circ}C$, pure $ZrO_2$ and $x$ mol% CaO-$ZrO_2$ ($x$ = 1.5 and 3) powders were identified as a mixture of monoclinic and tetragonal phases. However, a stable tetragonal phase of zirconia was observed in the 8 mol% calcia doped zirconia nanopowder at hydrothermal temperature above $160^{\circ}C$. To observe the phase transition, the 3 mol% CaO-$ZrO_2$ and 8 mol% CaO-$ZrO_2$ nanopowders were heat treated from 600 to $1000^{\circ}C$ for 2h. The 3 mol% CaO-$ZrO_2$ heat treated at above $1000^{\circ}C$ was found to undergo a complete phase transition from mixture phase to monoclinic phase. However, the 8 mol% calcia doped zirconia appeared in the stable tetragonal phase after heat treatment. The result of this study therefore should be considered as the preparation of 8 mol% CaO-$ZrO_2$ nanopowders via the hydrothermal method.

  • Forty seven strains of yeast were isolated from traditional Meju and were identified as Saccharomyces spp. (7 strains), Zygosaccharomyces spp. (7 strains), Kluyveromyces spp. and Hansenula spp. (each 5 strains), Rhodotorular spp. (8 strains), Candida spp. (12 strains), Pichia spp. and Debaryomyces spp. from results of their microbiological characteristics. The optimal medium for growth of all the yeasts was YM media and the optimal initial pH of the medium was 6.0. The optimum temperature for growth was 30$circ$ and among them, Sacch. exiguus OE-5, Sacch. cerevisiae OE-16, Sacch. kluyveri C-1 strains were thermotolerant yeasts which could grow at 40$circ$C.

  • This experiment was conducted to develop ethylene adsorbents for removing the ethylene produced from fruit during storage. The effect of ethylene adsorbent on the removing ethylene was the highest when $ICI_3$and $KBrO_3$ were used as the base of reactant and $H_2SO_4$ auxiliary reagent of reactant. It was decreased under the saturated water vapor condition and was sligthly decreased under the high carbon dioxide condition.

  • Objectives : This study aimed to investigate the relationship between personality characteristics of medical students at their admission and the level of depression, anxiety, and resilience at their third year. Methods : Self-reported questionnaires were conducted to the students at the beginning of the first year and the third year. When the students in their first year, they performed the personality assessment inventory, the Beck depression inventory, and the Beck anxiety inventory. When they were in the third year, the students answered the composite Scale of morningness and Conner-Davidson resilience scale-10 in addition to the BDI and BAI. Multiple linear regression analyses were performed to identify predictors of the level of depression, anxiety, and resilience. Results : Of the subscales of depression in the PAI, the physiological depression (${\beta}=-0.285$, p=0.049) associated with increase in the depression from the first to the third year. Among the anxiety-related disorder subscale in the PAI phobia (${\beta}=0.119$, p=0.022) and traumatic stress (${\beta}=0.375$, p=0.007) associated with the level of depression at third grade. Somatization (${\beta}=0.631$, p<0.001) in the PAI associated with the level of anxiety at third year. The anxiety (${\beta}=-0.531$, p<0.001) in the PAI was positive associated with the resilience at third grade. Conclusions : This study showed that certain personality characteristics at admission had significant relationships with the level of depression, anxiety, and resilience at the third grade medical students.

Design preference of male according to the Consciousness of Body Size. (남성의 신체 크기 인식에 따른 디자인 선호도)

  • The purpose of this study is to analyze the design preference according to the body consciousness for 20~26 years old male. The consciousness of body size and the satisfaction with body proportion were investigated. And the relationship between the body consciousness and the design preference was analyzed. Most subjects thought their body size was proper generally and their body proportion was proper or a little unsatisfied. The subjects preferred 'Round neckline', 'loose sleeve', and 'loose and long tops'. They didn't like the style that tops are under pants on the waist. They also preferred 'loose and long pants'. When the subjects chose clothing design, especially 'V-neckline tops' and 'short pants', they considered their height. They also considered their girth size and it meant they thought much of looseness. There was not any preference in clothing design according to the consciousness of height. The most significant relationships between the size consciousness and the preference in design was found in tops more than pants. The higher satisfaction with their body proportion was connected with the higher preference in some design. From all these result, it was found that the preference of clothing design could vary according to the consciousness of body size. The results from this study could give the important tips to improve the satisfaction with the clothing for each person.

  • The authors studied clothing buying motive and clothing buying behavior. The date were obtained from 352 women college students living in Seoul and Taejon. As the result of factor analysis. four buying motive factors were found: fashion and individuality, economy, status, advertising and impulsive buying. Three buying behaviors(information using, clothing selection standard, and shop selection standard) were factor analyzed. Three factors were founded in information using: human, electric wave communication, advertising and observation factors. Four factors were found in clothing selection standard: utility, design, marketing, fashion and status. Two factors were found in shop selection standard: merchandise and service. Clothing buying motive and clothing buying behavior were related, and they were different according to the levels of spending money and major in college.
