알로에 성분 Ny932의 혈관생성 촉진작용과 그 작용기전

  • Aloe의 추출 시료들 중 혈관생성을 촉진하는 물질이 G1M1D1M1 분획에 있음을 chorioallantoic-membrane(CAM) assay, in vitro angiogenesis assay, u-PA, PAI, u-PAR, 및 matrix metalloprotease(MMP)의 유전자 발현조사, gelatin zymogram assay, modified CAM assay를 하여 확인 하였다. 그런 다음 G1M1D1M1 분획을 다시 분리하여 단일 성분인 NY932와 3종의 분획들을(U>3,000, 3,000>U>1,000, U<1,000) 얻었다. 이들 각각에 대한 혈관 생성 촉진 작용을 CAM assay 방법으로 조사한 결과 순수단일 성분인NY932가 혈관 생성 촉진 활성을 가장 높게 나타났다. 즉 NY932의 농도를 100ng, 2, 10, 35, 60 $\mu$g으로 투여하였을 때, 각각 2/18($11\%$), 4/10($49\%$), 16/22($73\%$), 15/17($88\%$), 9/9($100\%$)의 혈관생성 촉진 효과를 보였다. 따라서 순수 단일 성분인 NY932의 혈관 생성 촉진 작용기전을 규명하기 위하여 혈관 생성에 관련된 효소들[matrix metalloprotease(MMP)s, urokinase-plasminogen activator(uPA) 등]과 그 저해제들(TIMPs, PAI)의 유전자 발현을 조사하였으며, wounding migration assay등을 수행하였다.

Growth Analysis of Red Pine (Pinus densiflora) by Stem Analysis in the Eastern Region of Korea

  • The study was performed to analyze the growth of the red pine (Pinus densiflora) in the Eastern Region of Korea. Stem profile data from a total of 8 dominant trees of which each tree represents its site were collected and used for the analysis. The stem volumes were calculated using Smalian's formula and much higher than the ones by Korea Forest Research Institute in larger than dbh 40 cm. The total bark volumes were more proportionally related to dbh and stem volume than to height and age. The bark thickness and volume decreased as relative height increases but increased as relative dbh increased. The average PAI of dbh and height reached the highest at 0.8 cm/yr (age 15) and at 0.45 m/yr (age 30) respectively, while the average PAI of volume steadily tended to increase up to age 80. The growth percentages of dbh, height and volume tended to decrease with age and volume growth percentage was higher than dbh and height.

Effects of Multicultural Society Recognition and Multicultural Education Experience on Cultural Sensitivity of Middle and High School Students in Daejeon: Focusing on the Mediating Role of Multicultural Education Needs Perception (대전시 중·고등학생의 다문화사회 인식과 다문화교육 경험이 문화적 민감성에 미치는 영향: 다문화교육 필요성 인식의 매개효과를 중심으로)

  • The objective of this study is to underline the important role of needs for multicultural education in the formation of middle and high school students' cultural sensitivity for the sake of coping multicultural society. This study focuses on how adolescents' multicultural society recognition and education experiences shape and influence their cultural sensitivity, with the mediating role of multicultural education needs perception. A survey 303 middle school students and 236 high school students Daejeon was conducted. The results of this study as follows. First, high school students' open minded and social participation were significantly higher than those of middle school students, while the latter multicultural education experience level was higher than the former. Second, it was found that multicultural society recognition and education experiences have an indirect impact on cultural sensitivity mediating multicultural education needs perception as well as have a direct effect. However, adolescents' cultural sensitivity was shaped by mediating role of multicultural education needs perception rather than by direct effect of multicultural society recognition and education experiences. A major contribution of this research is the identification of the role of multicultural education needs perception in fostering adolescents' cultural sensitivity.

Pedestrian path search based on the shortest distance algorithm using Map API (Map API를 활용한 최단 거리 알고리즘 기반 보행자 경로 탐색 연구)

  • There are casualties due to inundation and flooding due to intensive typhoons or heavy rains in summer. Due to such damage, the biggest disaster is flood, and in order to reduce human damage, this paper proposes a shortest distance algorithm-based pedestrian path search study using Map API. This system selects Map API through comparative analysis and provides the shortest route. The route explored is in JSON format and the data of the shelter is stored in the database. The route search system designed and implemented based on this data locates pedestrians and provides evacuation routes in case of flash floods. In addition, if the route cannot be entered while moving to the evacuation route, the current location of the pedestrian is identified, the route is re-searched, and a new route is provided. Therefore, it is believed that the pedestrian route search system proposed in this paper will prevent negligent accidents.

Radiation Induced Changes in the Expression of Fibronectin, Pai-1, MMP in Rat Glomerular Epithelial Cell (백서 사구체 상피세포에서 방사선에 의한 Fibronectin, Pai-1, MMP 발현의 변화)

  • Purpose: Renal irradiation can lead to the development of radiation nephropathy, and this is characterized by the accumulation of extracellular matrix and final fibrosis. To determine the possible role of the glomerular epithelial cell, the radiation-induced changes in the expression of its genes associated with the extracellular matrix were analyzed. Materials and Methods: Rat glomerular epithelial cells (GEpC) were irradiated with a single dose of 0, 2, 5, 10 and 20 Gy with using 6 MV LINAC (Siemens, USA), and the samples were collected 6, 24, 48 and 72 hours post-irradiation, respectively. Northern blotting, western blotting and zymography were used to measure the expression level of fibronectin (Fn), plasminogen activator inhibitor-1 (Pai-1), matrix metalloproteinases-2, 9 (MMP-2, 9), tissue inhibitor of metalloproteinase-2 (TIMP-2), tissue-type plasminogen activator (t-PA) and urokinase-type plasminogen activator (u-PA). Results: Irradiation with a single dose of 10 Gy resulted in a significant increase in Fn mRNA since 24 hours post-irradiation, and a single dose of 5 and 10 Gy significantly increased the Fn immunoreactive protein measured 48 hours post-irradiation. An increase in Pai-1 mRNA and protein was also observed and especially, a single dose of 10 Gy significantly increased the mRNA measured 24 and 48 hours post-irradiation. The active MMP-2 measured 24 hours post-irradiation slightly increased in a dose dependent manner, but this increase did not reach statistical significance. The levels of MMP-9, TIMP-2, t-PA and u-PA appeared unaltered after irradiation. Conclusion: Irradiation of the glomerular epithelial cells altered the expression of genes associated with the extracellular matrix, implying that the glomerular epithelial cell may be involved in the development of radiation nephropathy.

Glutamic Acid Rich Helix II Domain of the HIV-1 Vpu has Transactivation Potential in Yeast

  • The transactivation potential of HIV-1 Vpu was identified from the yeast two-hybrid screening process. The helix II domain of HIV-1 Vpu protein and mutant Vpu protein lacking the transmembrane domain exhibited transactivation of the LacZ and Leu2 reporter genes carrying LexA upstream activating sequences, but full-length HIV-1 Vpu and the helix I domain of HIV-1 Vpu did not. The helix II domain of HIV-1 Vpu consists of a number of acidic amino acids, and is especially rich in glutamic acid, a characteristic of many transcription factors. This result suggests that protein-protein interaction may occur through the acidic helix II domain of HIV-1 Vpu.

  • To study the mechanism of lipocortin-1, the 37 kDa protein, one of the annxin superfamily thought to be a second messenger during the Glucocorticoid dependent anti-inflammatory action, the gene for lipocortin-1 binding protein was isolated using the yeast two hybrid assay, the yeast based genetic assay recognizing the protein-protein interaction. The results showed that this gene has a weak homology to the for the human serine proteinase.

  • To find the interacting protein with the cytoplasmic domain of HIV-1 gp41, the yeast two hybrid system was used for the expression cloning. Among the $1.4 \times 10^6 colonies, 20 colonies were selected as the final candidate for the interacting protein gene. The nucleotide sequencing revealed three kinds of protein, acidic ribosomal protein P0, beta tubulin, alpha catenin. These proteins interacted with the gp41 specifically in yeast system.

  • Background: Pulmonary hypertension in COPD patients is the result of a direct effect of tobacco smoke on the intrapulmonary vessels with the abnormal production of the mediators that control vasoconstriction, vasodilatation, and vascular cell proliferation, which ultimately lead to aberrant vascular remodeling and physiology. COPD patients are prone to the developmint of an acute and chronic thromboembolism with an elevation of the plasma procoagulant and fibrinolytic markers However, the roles of the coagulation and fibrinolysis system on the right ventricular dysfunction in COPD patients are not well defined. We examined the alteration of the coagulation and fibrinolysis system in COPD patients according to the right ventricular function measured using cardiac multidetector computed tomography (MDCT). Methods: The right ventricular ejection fraction (RVEF) was measured using cardiac MDCT in 26 patients who were diagnosed with COPD according to the definition of the GOLD guideline. The plasma level of thrombin antithrombin (TAT) and plasminogen activator inhibitor (PAI)-1 were measured using an enzyme linked immunoassay. Results: The plasma TAT was markedly elevated in COPD patients ($10.5{\pm}19.8{\mu}g/L$) compared with those of the control ($3.4{\pm}2.5{\mu}g/L$) (p<0.01). However, the plasma PAI-1 in COPD patients ($29.6{\pm}20.7ng/mL$) was similar to that in the controls. The plasma TAT showed a significant inverse relationship with the RVEF measured by the cardiac MDCT in COPD patients (r=-0.645, p<0.01). However, the plasma PAI-1 did not show a relationship with the RVEF (r=0.022, p=0.92). Conclusion: These results suggest that the coagulation system in COPD patients is markedly activated, and that the plasma level of TAT might be a marker of a right ventricular dysfunction in COPD patients.

  • Exponential decrease of saturation absorption signal was reported in pump beam spacially keeping off from probe beam. Optimum angle between pump beam and probe beam in the differential-velocity-selective saturation absorption spectroscopy was computed theoretically, $1.33^{\circ}$ in cesium atoms and $1.08^{\circ}$ in Rb 87 atoms, and was good agreement with the experimental results.
