Antioxidative capacity of hydrolyzed rapeseed cake extract and oxidative stability of fish oil-in-water emulsion added with the extract

  • Rapeseed cake was extracted with 80% ethanol and then fractionated with $H_2O$ (fraction I) as well as with 30% (II), 50% (III), 70% (IV), and 100% ethanol (V). Total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric-reducing antioxidant potential, and Trolox equivalent antioxidant capacity were in the order of fractions II > III > I > IV > V. The three fractions with high antioxidant activities and TPC (I, II, and III) were pooled and hydrolyzed by NaOH solution, resulting in 18.97 mg sinapic acid/g hydrolyzed extract and 21- and 2.2-fold increases in TPC and DPPH radical scavenging activity, respectively. Hydrolyzed rapeseed cake extracts (200, 500, and 1,000 ppm) and catechin (200 ppm) as a comparison were added to 10% fish oil-in-water emulsion, and their effects on oxidative stability were investigated by measuring hydroperoxide values (PV) during refrigerated storage. PVs were significantly lower in the emulsions with added hydrolyzed extract as compared to the control (p<0.05) and significantly decreased with increasing extract concentration (p<0.05) over a period of 29 days. The emulsion added with hydrolyzed extract showed higher PV than that added catechin at the same concentration (200 ppm) during 13-22 days (p<0.05), but after then, the PV was not significantly different (p>0.05). This study indicates that hydrolyzed rapeseed cake extract rich in sinapic acid may inhibit oxidation in a fish oil-in-water emulsion in a concentration-dependent manner.

Preculture Condition for Stable Recovery of Strawberry (Fragaria × ananassa Duch.) Shoot Tips after Cryopreservation Using Droplet Vitrification

  • This study describes an efficient and stable droplet vitrification following cryopreservation of strawberry shoot tip (Fragaria × ananassa Duch.) accessions 'Massey' and 'MDUS3816'. The shoot tips were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7M). Precultured explants were osmoprotected with loading solution (LS, C4) containing 17.5% glycerol and 17.5% sucrose for 40 min and exposed to dehydration solution (B1) containing 50% glycerol and 50% sucrose for 40 min at 25oC. Subsequently, the explants were transferred onto droplets containing 2.5 µL PVS3 on sterilized aluminum foils (4 cm× 0.5 cm) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regrowth rate (%) in both the cultivars was obtained when the shoot tips were precultured with 0.3M sucrose for 30 h + 0.5M sucrose for 16 h at 25oC. The cryopreserved shoots tips exhibited 57.8 % recovery rate by culturing in NH4NO3-free MS medium supplemented with 3% sucrose, 1.0 g/L casein, 1.0mg/L GA3, and 0.5 mg/L BA for 5 weeks and in MS medium supplemented with 0.5 mg/L GA3 for 8 weeks. Variation was not observed in both of ploidy analysis and morphological investigation on plantlets of two accessions cryopreserved under variable preculture conditions.

한국에서의 감자 바이러스병 발생과 그 연구에 대한 고찰 (Review on the Occurrence and Studies of Potato Viral Diseases in Korea)

  • The occurrence of potato(Sotanum tuberosum) viral diseases caused by Potato virus X(PVX), Potato virus Y (PVY), Potato leafroll virus(PLRV), Potato vims S(PVS), Potato virus M(PVM), Potato virus A(PVA), Potato virus T(PVT), Alfalfa mosic virus(AIMV), Tobacco mosic virus(TMV), Potato mop top virus(PMTV) Tobacco rattle virus(TRV) and Potato spindle tuber viroid(PSTVd), potato witches' broom phytoplasma, have been identified so far in Korea. Major viral diseases such as PVX, PVY and PLRV had been studied more deeply, however, the others are just identified and only partially characterized since the first study on the relation between PVX nucleic acid and virus protein by Kim in 1961. The most studies on potato viral diseases are mainly focused on the problems of seed potato production. The National Alpine Agricultural Experiment Station(NAAES), since it began its activities in 1961, has given special attention to this problem by doing studies to identify, characterize and control potato virus diseases. This effort resulted in the development of new potato virus detection methods as a basis for elaborating new method of control, such as the production of seed potato free of virus and the selection of new virus-resistant transgenic potatoes. The further studies of potato viral diseases required would be fallowings: the continuous monitoring for the occurrence of identified or not identified potato viruses in Korea, the isolation of resistant viral genes, the development of control method for the non-persistently transmitted viruses like PVY, special vectors such as nematode and fungus transmitted viruses, TRV and PMTV and the development of control methods against potato viral diseases by viral cross protection, therapy, transgenic plant, and the use of the agents or molecules, such as virus inhibitors and antiviral proteins, etc., blocking viral replication.

The shoot tips of strawberry were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7M). Precultured explants were treated with loading solution (LS, C4) containing glycerol 17.5% and sucrose 17.5% for 40 min and exposed to dehydration solution (B1) containing 50% of glycerol and 50% of sucrose for 60 min at $25^{\circ}C$, and then transferred onto droplets containing $2.5{\mu}l$ PVS3 on sterilized aluminum foils ($4cm{\times}0.5cm$) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regeneration rate (%) was obtained when shoot tips were precultured with treatment-2 (exposing of shoot tips to MS + 0.3M Sucrose for 30 h and then treated with MS+0.5 M sucrose for 16 h) at $25^{\circ}C$ in both the cultivars. The viability of cooled samples, followed by culturing on MS medium for 4 weeks was 77.8% and 60.0% for 'Derunoka' and 'Jumbo pure berry', respectively. This result shows droplet-vitrification would be a promising method for cryobanking strawberry germplasm.

A Successful Regeneration from Shoot Tips of Chrysanthemum morifolium (Ramat.) following Cryopreservation by Droplet-vitrification

  • This study describes an efficient and widely applicable droplet-vitrification following cryopreservation for shoot tips of Chrysanthemum morifolium (Ramat.) cvs. 'Borami' and 'Yes morning'. The shoot tips of Chrysanthemum were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7 M). Precultured explants were treated with loading solution (LS, C6) containing glycerol 20% and sucrose 20% for 30 min and exposed to dehydration solution (B5) containing 40% of glycerol and 40% of sucrose for 60 min at $25^{\circ}C$, and then transferred onto droplets containing $2.5{\mu}l$ PVS3 on sterilized aluminum foils ($4cm{\times}0.5cm$) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regeneration rate (%) was obtained when shoot tips were precultured with treatment-2 (exposing of shoot tips to MS + 0.3M sucrose for 30 h and then treated with MS+0.5 M sucrose for 16 h) at $25^{\circ}C$ in both the cultivars. The viability of cooled samples, followed by culturing on $NH_4NO_3$-free MS medium for first 5 days was increased to two-fold (80.7%) regrowth rate over those cultured on normal MS medium or MS medium containing plant growth regulators. This result shows droplet-vitrification would be a promising method for cryobanking chrysanthemum germplasm.

Cryopreservation of in vitro Grown Shoot Tips of Korean Potato Varieties by Droplet-vitrification

  • Potatoes are the world's 4th major food crop after maize, rice, and wheat and also are a staple food for 1.3 billion people. Due to their wide adaptability to various environmental conditions, their yeild capacity, and high commercial value, potatoes have contributed to global food security. Many potato germplasms are commonly preserved as whole plants in fields or in storage to maintain their particular genetic combinations. However, field maintenance is expensive and has the risk of potential losses from diseases, pests, plant ageing and climate change. Over the past four decades, meaningful efforts have been made toward the safe long-term conservation of potatoes through cryopreservation methods such as droplet-vitrification. In this study, we tested 4 Korean potato varieties('Golden Egg', 'Golden Ball', 'Ja-Young' and 'Ha-Ryeong') with the modified potato droplet -vitrification protocol. Potato shoot tips are precultured in a sucrose-enriched medium(0.3 and 0.7M for 7 and 17hrs, respectively) and submitted to a loading step with C4 solution for osmoprotection. The treated explants were dehydrated with Plant Vitrification Solution(PVS)2 which is 80% A3 solution in ice for 30 minutes. Thawing and unloading steps were performed with 0.8M sucrose solution for 30 sec(40℃) followed by 30min(25℃, room temperature). In a potato post-culture medium(MS+0.1 mg·L-1 GA3+0.1 mg·L-1 kinetin), we obtained a survival rates of post-thawed explants ranging 16.1-82.2%. The results suggest that modified and optimized protocols are required dependinig on every cultivar, genetic and ecological types. To achieve higher survival and regeneration rates, each step within the cryoprocedure must be carefully optimized.

Efficient Cryopreservation of in vitro Grown Shoot Tips of Pear (Pyrus spp.) by Droplet-vitrification

  • In this study, cryopreservation by droplet-vitrification was applied to pear (Pyrus spp.) germplasm. We focused on the development and assessment of various strategies for the selection of suitable tissue, osmoprotection, and dehydration. We also evaluated post-thaw recovery of cryopreserved explants by droplet-vitrification. Preferentially, we tested the effects of preculture and loading treatments to determine which tissues were more suitable, either the apical shoot tips or the axillary buds. Apical shoot tips showed the better regrowth rate than in vitro axillary buds. The most effective techniques for cryopreservation were as follows. Shoots from in vitro seedlings which had been cultured for about 5-6 weeks were cold-hardened at 4℃ for one week, excised shoot tips were precultured on liquid MS medium including 0.3 M sucrose for 31 hours and 0.7 M sucrose for 17 hours, osmoprotected in loading solution (LS) for 40 min, and then cryoprotected in dehydration solution (PVS3) for 90 min. In addition, we found that regrowth rates of explants on regrowth medium after exposure to liquid nitrogen (LN) were higher than those on MS medium. Results indicated that the highest regrowth percentage was 95.6% for 'Bartlett' cultivar and 68.9% for 'BaeYun No.3' cultivar. Consequently, apical shoot tips of two pear cultivars, 'Bartlett' (P. communis) and 'BaeYun No.3' (P. pyrifolia), were successfully cryopreserved by droplet-vitrification. Results of this study show that the enhanced droplet-vitrification method described in the present study could be used as an effective means for long-term storage of pear genetic resources.

레트로 바이러스 벡터를 이용한 돼지난자에의 유전자 전이에 있어 Sucrose와 Polybrene의 효과 (Effect of Sucrose and Polybrene on the Gene Transfer into Porcine Oocytes using Retroviral Vector)

  • 본 연구에서는 레트로 바이러스를 이용하여 형질전환 돼지를 생산하기 위한 기초 연구로써 유전자 도입 효율을 증진시키기 위해 sucrose와 poly-brene을 사용하여 그 효과를 조사하였다. 유전자(hGH) 전달체로는 vesicular stomatitis virus gly-coprotein pseudotyped retroviral vector (VSV-G)를 사용하였다. 체외에서 성숙된 돼지 난자는 매우 협소한 위란강내 공간을 가지며 따라서 위란강내 공간의 확장은 필수조건이다. sucrose 처리는 수정율및 배발달에는 영향을 미치지 않았으며 sucrose 처리를 통해 돼지난자는 위란강의 공간이 확장되어 충분한 양의 바이러스 벡터의 주입이 가능하였다. PCR 분석 결과 8.3 %의 유전자 도입율을 나타낸 무처리군에 비해 0.5, 1, 2, 3 % sucrose 처리군에서 39.3, 43.3, 35.7, 40.7 %의 높은 유전자 도입율을 나타내었다. 추가적으로 돼지난자는 sucrose 처리에 의해 다정자 침입의 억제 효과를 나타내었다. 일반적으로 레트로 바이러스를 이용한 세포에의 높은 유전자 도입 효과를 나타내는 것으로 알려진 polybrene을 바이러스와 공동 주입한 결과 0.5, 5, 50$\mu\textrm{g}$/$m\ell$의 농도에서 각각 56.5, 50.0, 57.1 %의 도입율을 나타냄으로써 바이러스 단독 주입군의 34.6%보다 높은 효율을 나타내었다. 그러나 50$\mu\textrm{g}$/$m\ell$의 농도를 주입하였을 경우, 분할율과 배반포발달율이 43.3 및 4.6%를 나타냄으로써 바이러스 단독주입군의 72.0, 17.3%보다 유의적으로 낮았다. 이상의 결과를 종합할 때, sucrose의 처리는 돼지 난자의 발달에 영향을 주지 않으며 유전자 도입효율을 증진시킬 수 있었으며, 추가적으로 다정자 침입억제효과를 보였다. 또한 polybrene의 사용은 낮은 농도에서 유전자 도입효율을 증진시켰다. 이러한 결과들은 형질 전환 돼지의 생산 가능성을 높이는데 이용될 수 있다고 사료된다.

양파 Pellet 종자의 발아 및 포장출아 특성 (Characteristics of Pellet Seed on Germination and Emergence in Onion(Allium cepa L.))

  • 양파의 직파재배를 정착시켜 국제경쟁력을 제고 시키기 위한 기초연구의 일환으로 양파종자에 pellet 처리하는 방법을 개발하고, pellet종자의 제 특성을 조사하였던 바 그 결과를 요약하면 다음과 같다. 1) Pellet 종자의 모양형성은 일차적으로 pellet물질의 종류에 따라 좌우되며 kaolin, clay, ash,석고 등은 모양형성이 잘 되었으며, bentonite와 diatomite는 모양형성은 되지만 pellet 종자의 표면이 매끄럽지 않았으며, clay는 모양형성은 잘되지만 건조하는 과정에서 pellet종자의 표면에 균열이 발생하였다. PG를 polymer로 PVS(8%)를 binder로 하여 pellet한 종자는 모양 형성, 경도 등에서 가장 적합하였다. 2) Pellet종자의 경도는 polymer종류, binder의 종류와 농도의 영향을 받는데 그 정도는 polymer에서 더 컸다. pellet물질 중 경도가 높았던 것은 gysum, coal ash 등이었으며, 생석회는 경도가 가장 낮았다. 3) PG로 pellet한 종자의 기내 발아율과 출아율은 각각 93.6, 91.8%로 pellet 물질 중 가장 높았고, 출아율은 처리온도 20, $25^{\circ}C$에서 비슷하였다. 토양 수분함량별 출아율은 각각 91% 이상으로 토양 수분함량간 큰 차이는 없었다. 종자의 크기별 평균 출아율은 pellet종자 입경 5와 6mm에서 각각 92.0%로 가장 높았다. 파종심도별 출아율은 파종심도 10mm에서 92.7%로 가장 높았으며 파종심도가 점차 깊어질수록 낮아졌다. 토양수분함량별 평균 출아소요시간은 토양수분함량 70%에서 158시간으로 대조구 138시간에 비해 20시간이 지연되었다.

전북지방의 씨감자 생산을 위한 적지선정 및 감자 바이러스병에 관한 연구(II). 씨감자의 바이러스병 검정 (A Survey of Potato Virus Diseases and Insect Vectors at Seed Potato Production Area in Jeon Bug(II). Serological Test on Virus Diseases of Seed Potatoes)

  • 전북지방의 씨감자 생산적지를 선정하기 위한 기초자료로서 고랭지대인 전북 무주군 무풍면 상오정 1대(700-800m)와 평야지인 전주에서 대관령 고랭지 시험장산 원종 남작(Solanum tuberosum Var. Irish Cobbler, Shimabara)과 무주산 재래종 씨감자(품종미상)를 재배하면서 씨감자의 바이러스 이병율 및 재배기간중의 지역별 감염율등을 혈청학적으로 검정하여 다음과 같은 결과를 얻었다. 1. 우리나라에서 현재 유통되고 있는 씨감자의 감자 바이러스 혼합감염율은 $50.88\%$이며 바이러스별로는 Potato virus Y(PVY)$(34.87\%)$$(40.33%)$$(41.00\%)$< Potato Virus(PVX)$(87.10\%)$로서 PVX가 가장 높은 이병율을 나타내고 있었다. 2. 감자바이러스의 1년차 감염율은 무주지방이 $11.7\%$, 전주지방이 $18.95\%$로서 무주지방이 낮고 특히 곤충매개가 심한 PVY의 감염율은 $6.45\%$ 밖에 안되어 이는 씨감자 생산지로서 적합함을 인정할 수 있었다. 3. 씨감자 자체의 바이러스 이병율은 대관령산이 $41.75\%$, 무주 재래종이 $42.17\%$로서 대관령산과 비슷하였고 작황은 대관령 산이 좋았다.
