Investigating the Induced Systemic Resistance Mechanism of 2,4-Diacetylphloroglucinol (DAPG) using DAPG Hydrolase-Transgenic Arabidopsis

  • Plant immune responses can be triggered by chemicals, microbes, pathogens, insects, or abiotic stresses. In particular, induced systemic resistance (ISR) refers to the activation of the immune system due to a plant's interaction with beneficial microorganisms. The phenolic compound, 2,4-diacetylphloroglucinol (DAPG), which is produced by beneficial Pseudomonas spp., acts as an ISR elicitor, yet DAPG's mechanism in ISR remains unclear. In this study, transgenic Arabidopsis thaliana plants overexpressing the DAPG hydrolase gene (phlG) were generated to investigate the functioning of DAPG in ISR. DAPG was applied onto 3-week-old A. thaliana Col-0 and these primed plants showed resistance to the pathogens Botrytis cinerea and Pseudomonas syringae pv. tomato DC3000. However, in the phlG transgenic A. thaliana, the ISR was not triggered against these pathogens. The DAPG-mediated ISR phenotype was impaired in transgenic A. thaliana plants overexpressing phlG, thus showing similar disease severity when compared to untreated control plants. Furthermore, the DAPG-treated A. thaliana Col-0 showed an increase in their gene expression levels of PDF1.2 and WRKY70 but this failed to occur in the phlG transgenic lines. Collectively, these experimental results indicate that jasmonic acid/ethylene signal-based defense system is effectively disabled in phlG transgenic A. thaliana lines.

A Study on the Durability Complement of Lightweight Photovoltaic Module (경량화 태양광 모듈의 내구성 보완에 관한 연구)

  • In this study, we fabricated light-weight solar module for various applications such as building integrated photovoltaics (BIPV), vehicles, trains, etc. Ethylene tetra fluoro ethylene (ETFE) film was applied as a material to replace the cover glass, which occupies more than 65% of the weight of the PV module. Glass fiber reinforced plastic (GRP) was applied to the ones with a low durability by replacing the cover glass to ETFE. Moreover, to achieve a high solar power conversion in this study, we applied a shingled design to weight reduced solar modules. The shingled module with GRP shows 183.7 W of solar-to-power conversion, and the output reduction rate after weight load test was 1.14%.

  • 유무기 하이브리드 금속-할라이드계 페로브스카이트(organic-inorganic metal halide perovskite) 페로브스카이트 반도체 소재는 광전자 소자와 소재 연구에 새로운 연구 흐름을 만들고 있다. 태양전지 성능이 불과 과거 몇 년 사이의 짧은 연구 기간에도 불구하고, 광-전 변환 소자 중에서도 단일 소자와 적층 소자(tandem)에서 높은 광-전 변환 효율을 나타내기 때문이다. 이러한 급격한 연구 성과와 성장에도 불구하고, 페로브스카이트 소재의 다양한 광전자 특성의 평가와 결과에 대한 논의가 필요한 상황이다. 특히 내부 이온 이동이 광전자 원거리 이동 특성 평가와 해석에 영향을 주는 경우, 페로브스카이트 소재를 기반으로 한 다양한 광전자 소자의 성능 향상과 해석에 여전히 모호함을 준다. 달리 얘기하면, 이 소재의 기초 특성을 이해하고자 적용하는 다양한 기존 특성 평가 분석법의 활용과 해석에도 복잡한 영향을 미치고 있다고 할 수 있다. 이러한 페로브스카이트 소재 내에서 광전자 원거리 이동을 측정하는 새로운 방법을 소개하고자 한다. 첫 번째 방법으로, Quasi-steady 상태에서 광전도도를 전기적 특성으로 측정하고, 광조사 하에 투과 및 반사를 광학적으로 측정하여, 전도도와 광전자 밀도를 동시에 평가하는 방법으로, photo-induced transmission and reflection (PITR) 분광분석법이다. 이 분광분석법은 실제 소자의 구동조건을 구현한 상태에서 광전자의 원거리 이동에서 발생하는 광전자 밀도 변화를 반영한 광전자 이동도 특성 평가라는 장점을 가지고 있다. 두 번째 방법으로, 기존의 연속 전압 인가 방법 대신 펄스형 전압 인가 방식을 도입하는 방법으로, pulsed voltage space charge limited current (PV-SCLC) 분석법이다. 이는 펄스형 전압 인가 방법으로 이온의 이동을 최소화하여, 전류-전압 측정에서 히스테리시스가 없고 측정결과의 재현성과 신뢰도가 매우 높은 장점이 있다.

  • Laboratory studies were carried out to measure the sound velocity (V/SUB p/) and quality factor (Q/SUB p/, inverse attenuation) in the horizontal (H) and vertical (V) direction on the core sampled sediment of deep-sea basin (1,850 meter water depth), East Sea of Korea (Sea of Japan). Sampled core was about 250 cm long and 500 kHz ultrasonic p-wave transducer was used for a sound soured. V/SUB p/ varies from 1,480 m/sec to 1,500 m/sec, it is not clear which direction is faster, V/SUB PH/ or V/SUB pv/, within${\pm}$ 1.0% anisotropy (A/SUB p/). It is thought because the core sediment facies is highly (or slightly) bioturbated homogeneous mud with very high porosity (more than 80%). The general trend of Q/SUB p/ is decreasing 10 to 5 with the buried depth, it is strongly affected by the variation of sediment texture (increasing silt, decreasing clay) with increasing of CaCO$_3$ and organic matter content, But Q/SUB PH/ is jumping up to 14.9 near the bottom of core sediment as including volcanic ash richly. The relationship between V/SUB PH/ and Q/SUB PH/ shows the mirror image nearly, it is interpreted that not only the geotechnical properties and texture but also sea-water characteristics (high Q/SUB p/, low V/SUB p/) according to rich water content affect strongly in the upper part of the unconsolidated deep-sea basin sediment.

Comparative Study of the Rheologic Properties of Elastic Impression Materials (수종 고무인상재의 유변학적 특성에 관한 비교)

  • The rheologic properties of elastic impression materials is a very important role as taking high accuracy impression. But, the studies that are focused on the rheologic properties of Korean elastic impression materials are not sufficient. The purpose of this study is to help clinical high accuracy impression taking by testing rheologic properties of elastic impression material that is made by Korea and other countries. Six type III elastic impression materials are tested. Subjects are 2 Korean polyvinylsiloxane(PVS), 2 imported PVS, 1 polyether, and 1 polysulfide. HAAKE RheoStress $1^{(R)}$(Thermo Electron Co. Germany)is used in measuring. HAAKE RheoStress $1^{(R)}$ is plate to plate type rheometer. All subjects is tested 3 times and measuring time is 900 seconds. We measured G′ and loss tangent after mixing. All elastic impression materials had a sigmoid shape on increasing G′ by time and decreasing loss tangent after setting, maximum G' is appeared highest in polyether, and lowest in polysulfide. Initial loss tangent is highest in polyether, and is lowest in Koreans PVS. Significant difference is showed in initial loss tangent between Korean PVS and imported PVS.

Comparative Efficacy of Synthetic and Natural Tenderizers on Quality Characteristics of Restructured Spent Hen Meat Slices (RSHS)

  • In the present study, comparative efficacy of natural as well as synthetic tenderizers on the quality characteristics of restructured spent hen meat slices (RSHS) was studied. Four different batches of RSHS viz. Control (without any tenderizer), T1 (1.25% calcium chloride replacing salt in formulation), T2 and T3 (1.5% each of pineapple rind and fig powder, replacing binder in the formulation) were developed in pre-standardized formulation. Vacuum tumbling was performed for 2.5 h and cooked product (RSHS) was assayed for quality attributes. Samples were packaged in aerobic conditions, stored for 21 days under refrigeration ($4{\pm}1^{\circ}C$) and were evaluated for pH, oxidative and microbial quality parameters at regular interval of 7 days. Water holding capacity of T2 was recorded the highest and significantly higher (p<0.05) than all other samples. The textural attributes of T2 were comparable to T1 but significantly higher (p<0.05) than C and T3. The colour attributes ($L^*$, $a^*$, and $b^*$ value) of T2 and T3 were improved due to use of natural tenderizers. During sensory evaluation, tenderness scores for T2 samples were recorded the highest. Throughout storage period, thiobarbituric acid reactive substances (TBARS), free fatty acids (FFA) and peroxide value (PV) followed an increasing trend for control as well as treated products; however, T2 showed a significantly (p<0.05) lower value than control and other treated samples. It can be concluded that good quality RSHS with better storage stability could be prepared by utilizing 1.5% pineapple rind powder as natural tenderizer.

The Calmodulin-Binding Transcription Factor OsCBT Suppresses Defense Responses to Pathogens in Rice

  • We previously isolated the OsCBT gene, which encodes a calmodulin (CaM)-binding protein, from a rice expression library constructed from fungal elicitor-treated rice suspension cells. In order to understand the function of OsCBT in rice, we isolated and characterized a T-DNA insertion mutant allele named oscbt-1. The oscbt-1 mutant exhibits reduced levels of OsCBT transcripts and no significant morphological changes compared to wild-type plant although the growth of the mutant is stunted. However, oscbt-1 mutants showed significant resistance to two major rice pathogens. The growth of the rice blast fungus Magnaporthe grisea, as well as the bacterial pathogen Xanthomonas oryzae pv. oryzae was significantly suppressed in oscbt-1 plants. Histochemical analysis indicated that the hypersensitive-response was induced in the oscbt-1 mutant in response to compatible strains of fungal pathogens. OsCBT expression was induced upon challenge with fungal elicitor. We also observed significant increase in the level of pathogenesis-related genes in the oscbt-1 mutant even under pathogen-free condition. Taken together, the results support an idea that OsCBT might act as a negative regulator on plant defense.

Channeling of Intermediates Derived from Medium-Chain Fatty Acids and De novo-SYnthesized Fatty Acids to Polyhydroxyalkanoic Acid by 2-Bromooctanoic Acid in Pseudomonas fluorescens BM07

  • 2-Bromooctanoic acid (2-BrOA) is known to block the formation of polyhydroxyalkanoic acid (PHA) in Pseudomonasfluorescens BM07 without any influence on the cell growth when grown on fructose, but it inhibits the cell growth when grown on octanoate (OA) (Lee et al., Appl. Environ. Microbiol. 67: 4963- 4974, 2001). We investigated the role of 2-BrOA in the PHA synthesis of the bacterium grown with mixtures of fructose and fatty acids. OA, 11­phenoxyundecanoic acid (1 1-POU), and 5-phenylvaleric acid (5-PV) were selected as model substrates. When supplemented with 50 mM fructose, all these carboxylic acids suppressed the formation of PHA from fructose, however, the ~-oxidation coenzyme A monomers derived from the carboxylic acids were efficiently polymerized, but the conversion yield [(mol of carboxylate substrate converted into PHA)/(mol of carboxylate substrate in the feed)] was low (e.g., maximally $\~53\%$ for 5 mM 11-POU). Addition of 2-BrOA (up to 5 mM) to the mixed carbon sources raised the conversion yield sensitively and effectively only at low levels of the acid substrates (e.g., 2 mM 1 1-POU or 5 mM OA): For instance, $100\%$ of 2 mM ll-POU were converted into PHA in the presence of 5 mM 2-BrOA, whereas only $\~10\%$ of the 1 1-POU were converted in the absence of 2-BrOA. However, at highly saturated suppressing levels (e.g., 5 mM ll-POU), 2-BrOA inhibitor showed no significant additional effect on the conversion ($60- 70\%$ conversion irrespective of 2-BrOA level). The existence of competitive and compensative relationship between 2­BrOA and all the carboxylic acid substrates used may indicate 'Present address: Section on Brain Physiology and Metabolism, Bldg. 10, Rm. 6N202, National Institute on Agmg, National Institute of Health, Bethesda, MD 20892, U.S.A. that all the acid substrate-derived inhibiting species bind to the same site as the 2-BrOA inhibiting species does. We, therefore, suggest that 2-BrOA can be used for efficiently increasing the yield of conversion of expensive substituted fatty acids into PHA and then substituted 3-hydroxyacids by hydrolyzing it.

Genetic Analysis on the Bacterial Blight Resistance Gene from a Wild Relative, Oryza minuta (야생벼 Oryza minuta에서 유래한 수원506호의 흰잎마름병 저항성유전자에 대한 고찰)

  • Bacterial blight (BB), cuased by the vascular pathogen Xanthomonas oryzae pv. oryzae, is one of the major threats in rice fields worldwide. In Korea, two resistance genes against BB, Xa1 and Xa3 had been intensively used for developing high quality japonica rice cultivars. Those traditional resistance sources have being rapidly ified by the adopting of BB pathogen through mutations of the corresponding avr-genes, such as K3a exhibiting high compatibility to both Xa1 and Xa3. To expanding genetic resource against BB in Korea, the Suweon506, an introgression line between a Korean japonica cultivar, Hwaseong and a wild relative, Oryza minuta, was be subjected for genetic analysis owing to the BB resistance. Through association analyses between the pathotyping and genotyping results for each $F_2$ progenies, derived from a cross between Suweon506 and a Tongil type cultivar, Milyang23, a major resistant dominant gene is localized on the subterminal region of rice chromosome 4, where at least three BB resistancde genes, Xa1, Xa2, and Xa22, were reported previously.

Influence of Dispersed and Anaerobic Bacteria in Aerobic Paper-making Wastewater Treatment (호기적 제지폐수 처리공정중에의 분산 혐기성 미생물의 영향)

  • Tn order to be helpful to control dispersed microorganisms for stabLlization of wastewater treatment in a paper-makillg process, dominant strains were isolated aerobically and anaerobically. and identified and physiological characteristics were also analyzed. Pseudomonas carboxydohydrogena, Cardiobacten'm hominis, lvIicrococcus lylae, XanfomonCls campestris p" juglandis, Micrococcus diversus, and Comamonas terrigencl as aerobic dominants, and Streptococcus bovis and Prevotella buccae as anaerobIc dominants were identified fi'om the supernatent of the primary settling tank. It seemed that microflora in the treatment process would consist of many kinds of microorganisms, whose dominant would change easily according to environmental conditions, They all grew well at $37^{\circ}C$ and at different initial medium pH's. Especially, some of them required sulfate ion for their growth, which came from a chemical coagulant of aluminium sulfate in the primary settling tank. Interestingly. many anaerobes grew well even in the aerobic wastewater treatment process and seemed to have some functions. Population of anaerobes increased three times in the supematant of primary settling tank and ten times in Lhe bottom sludge of primary settling tank than in the prime wastewater. Therefore, these anaerobes contributed to the producH tion of offensive gases, which would make some microorganisms not precLpitate and be buoyant.
