Overexpression of the bacteriophase PRD1 DNA polymerase

  • In order to overexpress bacteriophage PRD1 DNA polymerase in E. coli cells, the 2 kb HaeII fragment was isolated from phage genomic DNA. This fragment was then cloned into pEMBL/sup ex/ 3-expression vector. A specific 57bp deletion was performed by using uracil containing ss DNA and oligonucleotide spanning each region to remove an unwanted non-coding region. After this deletion, the PRD1 DNA polymerase gene is totally under the control of the vector promoter and SD sequence. Upon heat induction, a protein with an apparent size of 68 kdal was overexpressed as an active PRD1 DNA polymerase. The expression of PRD1 DNA polymerase was about 1% of total E. coli protein.

PRD 강환 말뚝의 연직지지력 특성에 관한 사례 연구 (Case Study on the Characteristics of Vertical Bearing Capacity for Steel Pipe Pile Installed by PRD)

  • Construction case of PRD (Percussion Rotary Drill) pipe pile and matters to be attended in construction of PRD pile were reviewed. The compressive and uplifting static pile load tests for PRD piles were performed and, also, analysis by Pile Driving Analyzer was done. Based on these results, bearing components in each resisting part (that is: steel toe, external skin, and internal skin) were measured separately. The measured resisting force was compared to the value calculated by the estimated formula. The pile capacity was mobilized in steel toe area and the external skin friction and the internal friction were not produced. Thus, it could be considered that toe of PRD pile should be supported in hard bearing stratum (for example, the fresh soft rock).

  • The PRD1 DNA polymerase is a small multi-functional enzyme containing conserved amino acid sequences shared by family B DNA polymerases. Thus the PRD1 DNA polymerase provides an useful model system with which to study structure-functional relationships of DNA polymerase molecules. In order to investigate the functional and structural roles of the highly conserved amino acid sequences, we have introduced three mutations into a conserved amino acid of the PRD1 DNA polymerase. Genetic complememtation study indicated that each mutation inactivated DNA polymerase catalytic activity.

CNG 용기용밸브의 압력방출장치 성능에 관한 연구 (A Study on Performance of Pressure Relief Devices of CNG Cylinder Valves)

  • 이 연구는 천연가스버스에 장착된 용기가 국부화염에 집중적으로 노출되었을 때나 외기온도가 높은 여름철 충전과정에서 과 충전되었을 때 용기파열을 방지하기 위한 방안으로 압력방출장치의 성능을 확인하고 확대 적용할 목적으로 의도되었다. 열사이클링 실험결과, $-40^{\circ}C{\sim}82^{\circ}C$ 인증기준에서 가스누출요건을 모두 충족하였지만 $-45^{\circ}C{\sim}135^{\circ}C$의 가속조건에서는 3개사 시험편중 2개사 시험편의 오링이 손상되었다. 또한 온도감응형 PRD의 평균작동시간은 액체봉입식이 1분 39초가 걸렸고, 가용합금식이 2분 31초가 소요되어 액체봉입식이 가용합금식보다 약 1분 정도 빠름을 나타내었다. 또한 압력감응형 PRD의 작동압력은 가속조건에서 약 32.1 MPa을 나타내었고 인증기준에서의 압력감응형 PRD의 파열압력은 $30.7{\sim}32.1\;MPa$를 기록하였다. 이상의 실험을 통해서 압력방출장치의 성능은 온도감응형 PRD의 경우, 액체봉입식이 가용합금식보다 화염에 더 효과적이며, 압력 감응형 PRD의 파열압력은 외부온도 및 반복가압이 파열판의 작동압력에 크게 영향을 미치지 않음을 확인할 수 있었다.

기술현황분석 - 원전 압력방출장치의 용량인증기관 현황

  • 원전발전소에 납품되는 원전용 안전밸브의 성능은 ASME나 KEPIC와 같은 기술기준코드에 따라 설정압력, 블로우다운(blowdown), 풀리프트(Full lift), 누설(leakage), 방출용량(discharge flow rate)등과 같은 특성인자로 평가된다. 그 중에서도 방출용량은 ASME SEC III NB 7000 코드에 따라 ASME 용량인증이 요구되지만, 아직 국내에는 국제적으로 공인되니 압력방출장치 용량시험기관이 부재하여 외화유출 및 국내 PRD 기술개발에 많은 어려움이 있어왔으며, 이에 따라 원전 PRD 인증기관 개발을 통하여 세계적인 인프라 구축의 필요성이 부각되고 있다. PRD 인증기관 구축은 국내 원전 핵심기자재의 고급기술 국산화에 이바지 할 수 있고, PRD 성능 및 용량시험지원을 통하여 국내 밸브기술 경쟁력 확보하는 데 기여할 것으로 기대된다.

PRD Pile Driving공법의 진동저감 평가를 위한 실험적 연구 (Experimental Study on Vibration Reduction Estimation of PRD Pile Driving Method)

  • In this study, ground vibrations and aero space noises that is produced by the PRD(percussion rotary drill) were analyzed by work processes. Ground vibration equations were suggested by $2.798(SD)^{-0.793}$, $3.485(SD)^{-0.793}$, $3.705(SD)^{-0.793}$, according to experiment result, and these equations have reliability of 50%, 90%, 95%, respectively. Ground vibration values by the PRD method correspond to level of $5{\sim}34%$ of values that are assumed by the Attewell & Farmer's equation, and these result compares in reliability 50%. Also, those values were analyzed that correspond to level of $12{\sim}26%$ of ground vibration values by the Prof. Park etc.'s equations. But, the aero space noise was evaluated that is assumed by 88.9 dB(A) at separated distance 50m and is not satisfied even 85dB(A) that is the most negative noise value that present in domestic noise standard. The PRD method was analyzed that noise decrease effect exists hardly comparing with general pile driving method of construction. When is based in these results, the PRD method is judged that it is desirable that classify by the Low-vibration method more than the Non-violation noise method.

NO REDUCTION PROPERTY OF Pt-V2O5-WO3/TiO2 CATALYST SUPPORTED ON PRD-66 CERAMIC FILTER

  • The effect of Pt addition over $V_2O_5-WO_3/TiO_2$ catalyst supported on PRO-66 was investigated for NO reduction in order to develop the catalytic filter working at low temperature. Catalytic filters, $Pt-V_2O_5-WO_3/TiO_2/PRD$, were prepared by co-impregnation of Pt, V, and W precursors on $TiO_2$-coated ceramic filter named PRD (PRD-66). Titania was coated onto the pore surface of the ceramic filter using a vacuum aided-dip coating method. The Pt-loaded catalytic filter shifted the optimum working temperature from $260-320^{\circ}C$(for the catalytic filter without Pt addition) to $190-240^{\circ}C$, reducing 700 ppm NO to achieve the $N_x$ slip concentration($N_x\;=\;NO+N_2O+NO_2+NH_3$) less than 20 ppm at the face velocity of 2 cm/s. $Pt-V_2O_5-WO_3/TiO_2$ supported on PRD showed the similar catalytic activity for NO reduction with that supported on SiC filter as reported in a previous study, which implies the ceramic filter itself has no considerable interaction for the catalytic activity.

  • The bearing capacity of drilled shafts that take excavation by Percussion Rrotary Drilling(PRD) into consideration was evaluated using static and dynamic pile load tests. The emphasis was on quantifying the allowable bearing capacity and point load-transfer at the pile tip on seven instrumented steel piles. Of the seven instrumented piles, five piles are placed to the bottom of the excavation by rotary and pushing into the final depth of the excavation, as opposed to the two driven piles. Based on the results obtained, it is shown that the skin friction mobilized by PRD is much greater than point resistance, whereas in driven piles, the point resistance is greater than skin friction. It is also found that much greater pile capacity was proved in the case of drilled shafts, compared to the driven piles and thus, the excavation by rotary drilling gives reliable pile capacity required to design axially loaded piles.

  • 이암은 건조 조건에서는 일정한 강도와 강성을 가지나, 지하수 침투 등에 의한 습윤, 포화 시 고유의 강도 및 강성이 저하되어 급속히 풍화되는 특성을 가지며, 포항지역에서 자주 발생하는 절취사면의 활동은 주로 이암이 풍화되어 전단강도가 급격히 저하되는 것으로 볼 수 있다. 본 연구는 시추조사시 강성이 충분한 것으로 평가된 이암층에 PRD강관말뚝을 시공한 후 정재하 시험을 실시한 결과 소요지지력이 부족하여, 이를 보강하기 위한 가장 우수한 방법을 찾기 위하여 여러 가지 공법을 검토하였다. 말뚝지지력 증대에 가장 효과적인 방법을 찾기 위해 말뚝주변 및 선단부에 MSG공법, 마이크로파일 보강후 주변그라우팅공법, 선단보강 그라우팅 및 콘크리트 속채움 등의 방법으로 시험시공을 실시하였고, 시험말뚝에 대한 정재하시험 결과 MSG공법으로 보강한 말뚝의 지지력이 가장 우수함을 알 수 있었다.

Neutrophil Functions and Cytokines Expression Profile in Buffaloes with Impending Postpartum Reproductive Disorders

  • The study was conducted to correlate the periparturient immune status in terms of neutrophil functions and cytokine expression in peripheral blood mononuclear cell culture with impending postpartum reproductive disorders in buffaloes. Forty pregnant buffaloes were observed for occurrence of postpartum reproductive disorders (PRD), i.e., metritis, endometritis and delayed uterine involution etc., during one week prepartum to four weeks postpartum period. A representative number (n = 6) of buffaloes that did not develop any PRD were included in group I (healthy, control), while the animals which experienced PRD were assigned into group II (PRD, n = 8). The blood samples were collected at weekly interval from one week prepartum to four weeks postpartum period considering the day of calving as 'd 0'. Differential leucocytes counts, superoxide and hydrogen peroxide production activity in isolated neutrophils and the mRNA expression profile of cytokines i.e., IL-2, IL-4 and IFN-${\gamma}$ in PBMC culture were studied in all the samples. A higher total leucocytes, neutrophil and band cells count along with impaired neutrophil functions i.e., lowered level of production of superoxide and hydrogen peroxide before parturition and during early postpartum period were observed in buffaloes developing PRD. Further, a lower expression of IL-2, IFN-${\gamma}$ and IL-4 mRNA in PBMC culture was observed at calving in buffaloes that subsequently developed PRD at later postpartum. Thus, suppression in neutrophil function and cytokine expression at prepartum to early postpartum period predisposes the buffaloes to develop postpartum reproductive disorders. Hence, monitoring of neutrophils function and cytokine expression profile would be effective to predict certain reproductive disorders at late pregnancy or immediately after parturition in buffaloes. In future, this may be a novel approach for determining suitable management and therapeutic decisions for prevention of commonly occurring reproductive disorders in farm animals.
