Monitoring mRNA Expression Patterns in Macrophages in Response to Two Different Strains of Probiotics

  • As an initial study to elucidate the molecular mechanism of how probiotics modulate macrophage activity, we monitored mRNA expression patterns in peritoneal macrophages (PMs) treated with two different strains of probiotics. After treatment with either Weissella cibaria WIKIM28 or Latilactobacillus sakei WIKIM50, total RNAs from PMs were isolated and subjected into gene chip analyses. As controls, mRNAs from vehicle (phosphate-buffered saline, PBS)-treated PMs were also subjected to gene chip analysis. Compared to vehicle (PBS)-treated PMs, WIKIM28-treated and WIKIM50-treated PMs exhibited a total of 889 and 432 differentially expressed genes with expression differences of at least 4 folds, respectively. Compared to WIKIM28-treated PMs, WIKIM50-treated PMs showed 25 up-regulated genes and 21 down-regulated genes with expression differences of more than 2 folds. Interestingly, mRNA transcripts of M2 macrophage polarization marker such as anxa1, mafb, and sepp1 were increased in WIKIM50-treated PMs comparing to those in WIKIM28-treated PMs. Reversely, mRNA transcripts of M1 macrophage polarization marker such as hdac9, ptgs2, and socs3 were decreased in WIKIM50-treated PMs comparing to those in WIKIM28-treated PMs. In agreement with these observations, mRNA expression levels of tumor necrosis factor-α and interleukin-1α were significantly reduced in WIKIM50-treated macrophages compared to those in WIKIM28-treated macrophages. These results may indicate that probiotics can be classified as two different types depending on their ability to convert macrophages into M1 or M2 polarization.

  • 부신(副腎)의 기능(機能)이 생식기관(生殖器官)에 미치는 영향(影響)을 구명(究明)코자 21일령(日齡)의 암흰쥐(體重(체중): $58.7{\pm}3.53g$) 210마리를 정상(正常)흰쥐실험군(實驗群)(이하(以下) Int.-)과 부신척출실험군(副腎剔出實驗群)(이하(以下) Adx.-)으로 나누고, 각군(各群)을 다시 대조군(對照群)(이하(以下)-Cont.), PMS 투여군(投與群)(이하(以下) - PMS) 및 PMS와 cortisol의 병용투여군(이하(以下)-PMS+corti)의 3개군(個群)으로 나누어 35마리씩 배치(配置)한 다음 부신척출실험군(副腎剔出實驗群)에 대(對)하여 부신척출(副腎剔出)을 실시(實施)한 5일후(日後)(흰쥐 일령(日齡) : 26일(日))에 PMS투여군(投與群)에는 마리당 25IU의 PMS를, PMS와 Cortisol의 병용투여군에는 마리당 25IU의 PMS와 $30.0{\mu}g$의 Cortisol의 처리후(處理後) 48, 54, 60, 66, 72, 78 및 84시간(時間)에 5마리씩 도살(屠殺)하여 생식기능(生殖機能)의 중량(重量)을 측정(測定)하였다. 1. 난소(卵巢)의 중량(重量)은 모든 처리군(處理群)이 처리후(處理後) 기간(期間)이 경과(經過)할수록 증가(增加)하여 전관찰시간(全觀察時間)에서 비교군간(比較群間)에 고도(高度)의 유의성(有意性)(P<0.01)이 인정(認定)되었으나 대조군(對照群)인 Int.-Cont. 군(群)과 Adx.-Cont. 군(群)은 변화(變化)가 인정(認定)되지 않았다. 다중검정결과(多重檢定結果) 부신(副腎)을 척출(剔出)한 Adx.-PMS군(群)과 Adx.-PMS+Corti. 군(群)은 정상(正常)흰쥐 실험군(實驗群)인 Int.-PMS군(群)과 Int.-PMS+Corti. 군(群)에 비하여 유의(有意)(P<0.05)하게 낮은 값을 보였고, Cortisol 투여(投與)에 따른 효과(效果)는 인정(認定)되지 않았다. 2. 자궁(子宮)의 중량(重量)은 모든 관찰시간(觀察時間)에서 비교군간(比較群間)에 고도(高度)의 유의성(有意性)(P<0.01)이 인정(認定)되었는데, 처리후(處理後) 66시간(時間)까지는 정상(正常)흰쥐 실험군(實驗群)인 Int.-PMS군(群)과 Int.-PMS+Corti.군(群)이 높은 값을 보였고, 72시간(時間) 후(後)부터는 반대(反對)로 부신척출실험군(副腎剔出實驗群)인 Adx.-PMS군(群)과 Adx.-PMS+Corti.군(群)이 높은 값을 나타냈으며, 다중검정결과(多重檢定結果) Int.-PMS군(群)과 Int.-PMS-Corti.군(群), 그리고 Adx.-PMS군(群)과 Adx.-PMS군(群)과 Adx.-PMS+Corti.군(群) 상호간(相互間)에는 유의성(有意性)이 인정(認定)되지 않았다. 3. 정상(正常)흰쥐 실험군(實驗群)에서의 부신(副腎)의 중량(重量)은 Int.-PMS 군(群)에서 근소(僅少)하게 높은 값을 나타냈으나 비교군간(比較群間)에는 통계적(統計的)인 유의성(有意性)은 인정(認定)되지 않았고, 조직소견(組織所見)에서도 특기(特記)할 만한 변화(變化)를 인지(認知)할 수 없었다.

과배란 유기된 rat 난소에 퇴축난포와 성장난포에 대한 면역조직화학적 연구 I. 동원된 난포수에 대하여 (Immunohistochemical study on the atretic and the growing follicles after experimental superovulation in rats I. Number of follicles by superovulation)

  • This study was designed to investigate the number of the growing and mature follicles following gonadotrophin treatments for superovulation in mature rats. Eighteen mature rats (Sprague-Duwely, initially 190~230gm) were randomly alloted into 3 groups. One group was control group, another FSH-treated group was injected intramuscularly with 0.5 units of follicular stimulating hormone (FSH) / rat, and third PMS and HCG-treated group was intramuscularly injected with 20~25IU of pregnant mare serum (PMS) / rat and then at the 48 hrs later, with 20~25IU of human chorionic gonadotrophin (HCG) / rat. The uteri and ovaries of rats were collected and then were observed grossly and serial sections of paraffin embedding ovaries were stained with H-E. Number of ovarian follicles by following 3 grades of large, middle and small follicles from secondary and tertiary follicles were investigated by LM photography of preparations. Small follicles were classified as secondary follicles of preantral follicles with more than 2 layers of granulosa cells surrounding the oocyte and middle follicles were classified as secondary follicles with early signs of antral cavity or with more than one small cavity on either side of the oocytes and large follicles were classified as tertiary follicles with a single medium sized antral cavity or large well-formed antral cavity. In gross findings, the uteri were slightly swelling in FSH-treated group and markedly swelling or filled with fluid in the uterine lumen in PMS and HCG-treated group. In histological findings, the shape and size of the follicles were diverse in middle and large follicles of FSH-treated group and PMS and HCG-treated group, and proportion of atretic follicles was increased in FSH-treated group and PMS and HCG-treated group than those in control group. The uteri of FSH-treated group and PMS and HCG-treated group were hypertropied or filled with fluid in the lumens and walls of uteri. The wall tissue layers were flattened and their blood and lymph vessels were dilated. The mean number of follicle per ovary in control group were appeared to be $17.1{\pm}5.6$($14.0%{\pm}4.6%$), $37.8{\pm}9.1$($30.9{\pm}7.4%$) and $67.6{\pm}30.1$($55.2{\pm}24.6%$) respectively at large, middle and small follicles and total number of these 3 grade follicles were appeared to be $122.5{\pm}40.0$. The mean number of follicle per ovary in FSH-treated group were appeared to be $22.8{\pm}7.0$($17.4%{\pm}5.3%$), $43.4{\pm}6.6$($33.2{\pm}5.1%$) and $64.5{\pm}13.0$($49.3{\pm}9.9%$) respectively at large, middle and small follicles and total number of these 3 grade follicles were appeared to be $130.7{\pm}16.6$. The mean number of follicle per ovary in PMS and HCG-treated group were appeared to be $29.7{\pm}11.0$($16.3%{\pm}6.0%$), $61.9{\pm}17.2$($33.9{\pm}9.4%$) and $91.1{\pm}28.2$($49.9{\pm}15.4%$) respectively at large, middle and small follicles and total number of these 3 grade follicles were appeared to be $182.6{\pm}32.7$. The above findings reveal that large follicles were increased 29.8% in FSH-treated group and 73.7% in PMS and HCG-treated group than those in control group and in histologic findings, proportion of atretic follicles were more increased in ovaries with more number of more developing follicles.

PMS 처리한 미성숙 쥐의 腦下垂體 排卵호르몬에 미치는 卵巢스테로이드의 影響 (The Role of Ovarian Steroids on Pituitary Ovulating Hormone in PMS-treated Immature Female Rats)

  • 본인은 생후 24일 된 쥐에 PMS와 더불어 estradiol을 처리하면 排卵이 예정보다 24시간 앞당겨 일어남을 보고한 바 있다. 본 실험은 이의 계속 연구로 腦下垂體前葉에서 분비되는 preovulatory LH surge가 PMS 처리 후 51$\\sim$56시간에 일어남을 보았다. 그러나 estradiol을 PMS와 동시에 처리하면 예정보다 24시간 단축된다. 卵巢를 제거한 쥐에 0, 24, 48시간에 $2.5\\sim 40\\mu g$의 estradiol을 처리하면 51$\\sim$56시간에 腦下垂體의 LH가 증가되지 않는다. 그러나 estradiol을 처리하고 이어 48시간에 1mg의 progesterone을 처리하면 卵巢를 제거한 쥐에서도 腦下垂體의 LH level은 증가된다. 본 실험의 결과로 estradiol처리가 PMS로 유도한 排卵을 24시간 앞당기는 사실은 estrogen이 progesterone과 길항작용으로 腦下垂體에서 LH분비를 24시간 앞당겨 일으키고, 따라서 排卵도 24시간 단축되는 것이라고 생각된다.

  • 2002년도 제주도내 폐결핵 발생자로 등록된 262명에 대한 조사를 실시한 결과 보건소의 관리실태와 비교해 볼 때, 의원급의 진단 및 추구검사 시행이 제대로 되지 않고 있는 것을 확인하였다. 이런 상황에서 의원 및 병원의 치료 성공률은 각각 38.5%, 63.3%로, 이를 합친 민간의료기관에서의 치료 성공률은 59.8%로 조사되어 보건소의 치료 성공률 80%에 비하여 낮은 수준을 보였다. 민간의료기관에서 치료 실패의 주 원인은 임의의 치료중단 이라는 점에서, 현재 국가결핵관리사업의 이원화된 체계 하에서 민간의료기관에 대한 시급한 시스템보완이 필요하다.

  • The present study is an analysis of some alterations occurring in the population of folliclesin the ovary treated with the gonadotrophins in intact and unilaterally ovariectomized mice. the differences between the findings inthe intact and semispayed animals and those in gonadotrophintreated intact and semispayed animals were disucssed. 1) When mice were semispayed at the stage of diestrus, the remaining ovary showed an increase of secondary follicles in number, while a decrease of atretic folicles, compared with those in intact animals. 2) After injection of PMS and HCG following semispaying at the diestrus stage, the remaining ovary contained larger number of secondary follicles and corpora lutea than those in semispayed animals with no treatment , while showed smaller number of atretic follicles. 3) It is sure that follicular development is remarkably accelerated in the intact mice treated with PMS and HCG. 4) The effect of PMS and HCG on the follicular development was gradually decreased with time in the intact and semispayed animals afte rexogenous hormone therapy. 5) It is believed in the experiment that a decrease of follicular atresia in number and an increase of secondary follicles may be responsible for compensatory reaction after semispaying.

닭의 자궁과 질 접합부의 정자선내에 정자 저장 (Sperm storage of the utero-vaginal glands in domestic hens)

  • The present observations were focussed mainly on the morphological findings of the utero-vaginal(U-V) glands in normal laying domestic hens and the storage of cock spermatozoa in the U-V glands at various times after artificial insemination(AI). These domestic hens were assigned to three group of PMS-treated, GnRH-treated before last AI, and control group. The hens were sacrified at intervals of 1,3,7,12 and 19 days after AI. Histological sections of U-V junctions were prepared and the morphological structures of the U-V glands were observed and then were scored about the spermatozoa presence in the U-V gland. 1. The U-V glandular tubules were mostly unbranched with single columnar epithelium. Also these tubules were occassionally observed as one circular-rotated tubules or 2 to 3 branched convoluted tubules in special shapes. The numbers of the convoluted curves per tubule were $4.3{\pm}3.3$ and the ranges of convoluted curve number were straight to 16 curves. 2. The inside and outside diameters of the glandular tubules were $6.5{\pm}3.5{\mu}m$, and $35.2{\pm}4.7{\mu}m$, respectively, and the tubular lengths of the U-V glands were $219.3{\pm}115.7{\mu}m$. 3. Storaged spermatozoa in the U-V glands of all three group hens were intensively stained by hematoxylin, and packed in tight, longitudinally parallel bundles within the tubules. In addition, numbers of completely spermatozoa-filled glands were tend to increase or decrease in proportion to the numbers of partially spermatozoa-filled glands. Also U-V glands containing spermatozoa tend to be present collectively in the any zone of U-V junction. 4. In the control group, the numbers of glands containing spermatozoa in the hens at 1,3,7,12, and 19 days after AI were found to be 22.9, 33.3, 35.8, 8.6, and 0% respectively. 5. In the PMS-treated group, the numbers of glands containing spermatozoa in the hens at 1,3,7,12, and 19 days after AI were found to be 33.6, 29.7, 26.8, 8.2 and 0% respectively. 6. In the GnRH-treated group, the numbers of glands containing spermatozoa in the hens at 1,3,7,12, and 19 days after AI were found to be 19.7, 40.8, 20.4, and 0% respectively.

과배란 유기된 rat 난소에 퇴축난포와 성장난포에 대한 면역조직화학적 연구 2. 동원된 난포의 퇴축과 성장에 대하여 (Immunohistochemical study on the atretic and the growing follicles after experimental superovulation in rats 2. Atresia and growing of follicles)

  • This study was designed to investigate the effects of superovulation on the growing and mature follicles following gonadotrophin treatments in mature rat by immunohistochemical methods. Eighteen mature rats (Sprague-Duwely, 190~230gm) were randomly alloted into 3 groups. One group was control group, another FSH-treated group was injected intramuscularly with 0.5 units of follicular stimulating hormone(FSH) / rat, and third PMS and HCG-treated group was injected intramuscularly with 20~25IU of pregnant mare serum(PMS) / rat and then at the 48 hrs later, with 20~25IU of human chorionic gonadotropin(HCG) / rat. Half the number of rats were administrated intraperitoneally with bromodeoxyuridine(Brdur, 0.2mg/gm BW once) at 2 hours before exanguination and the remainder of rats were sacrified without Brdur administration. The investigation by immunohistochemical methods using paraffin sections of ovaries was performed by using anti-Brdur antibody and PCNA(proliferating cell nuclear antigen) antibody for labeling proliferating cells in follicles. In immunohistochemical findings, follicles squeezed by peripheral corpus luteum or follicles large follicles with loosly and irregularly distributed granulosa cells and although with compacted granulosa cells, middle follicles with dilated round or oval follicular antrum were confirmed as atretic follicles. The proportions of atretic follicles in control group were 29.8%, 21.7% and 14.2% respectivley at large, middle and small follicles and mean proportions of these all 3 grade follicles were 26.7%. The proportions of atretic follicles in FSH-treated group were 35.4%, 24.9% and 10.4% respectively at large, middle and small follicles and mean proportions of these all 3 grade follicles were 28.1%. The proportions of atretic follicles in PMS and HCG-treated group were 44.7%, 24.0% and 12.7% respectively at large, middle and small follicles, and mean proportions of these all 3 grade follicles were 29.7%. The above findings reveal that the group with higher proportion of atretic follicles were ordered as large, middle and small follicles in size, and these proportions were increased in hormone treated two groups with more number of more growing and mature follicles when compared with control group.

PRE-MAIN SEQUENCE EVOLUTIONS OF SOLAR ABUNDANCE LOW MASS STARS

  • We present the Pre-Main Sequence (PMS) evolutionary tracks of stars with $0.065{\sim}5.0M_{\odot}$. The models were evolved from the PMS stellar birthline to the onset of hydrogen burning in the core. The convective turnover timescales which enables an observational test of theoretical model, particulary in the stellar dynamic activity, are also calculated. All models have Sun-like metal abundance, typically considered as the stars in the Galactic disk and the star formation region of Population I star. The convection phenomenon is treated by the usual mixing length approximation. All evolutionary tracks are available upon request.

PMSG 반복투여가 Rat의 과배란에 미치는 영향에 대한 면역조직화학적 연구 1. 투여회수에 따른 난포수의 차이 (Immunohistochemical Study on the Superovulation Effected by Repeat of PMSG Administration in Rats 1. The Frequency of PMSG Administrations and Number of Follicles)

  • The purpose of this study was attempted to investigate the number changes of the growing and mature follicles in ovary following repeats of pregnant mare serum gonadotropin(PMSG) treatments for superovulation in nulliparous rats. Thirty two rats(Sprague-Dawely, about 200~250 gm) were randomized into 4 groups. Control group rats were sacrified at estrus phase confirmed by vaginal smear. PMSG-treated group 1 rats, PMSG-treated group 2 rats and PMSG-treated group 3 rats were sacrified at 48 hrs after injection once with PMSG 25 IU, after 2 repeated injection by a week interval, and 3 repeated injection, respectively. The uteri and ovaries of rats were removed and weighed and then were observed grossly and serial sections of all ovaries and some sections of uteri by paraffin embedding were stained with H-E. Number of ovarian follicles about 3 grades of small, middle and large follicles from seondary and follicles were investigated by LM photographies of ovary preparations. The criteria of the small, middle, and large follicles were based as small follicle with preantral follicles with 2~4 layers of granulosa cells surrounding the oocyte, as secondary follicles with more than 5 layers of granulosa cells and early signs of antral cavity or with small clefts on either side of the oocytes, and as tirtiary follicles with a single medium sized antral cavity or large well-formed antral cavity, respectively. In gross findings, the wall of the uteri in control group were thin, and those in 3 PMS-treated group were markedly thickened and some uterine lumen of those filled with fluid. In histological findings, the walls of the uteri from 3 PMSG-treated groups were hypertrophied and their blood and lymph vessels were dilated than those of control group. The ovaries fo 3 PMSG-treated groups were more increased in size and the cortexes were more developed and increased in width but there are no difference of development and changes in 3 PMSG-treated groups. The weight of the uteri and ovaries per rat in PMSG -treated group 1, 2 and 3 were a, pp.ared to be significantly increased 171.4$\pm$47.6%, 162.3$\pm$43.9%, 206.9$\pm$30.4%, respectively than those of control groups. The mean number of follicle per ovary in control group were a, pp.ared to be 17.1$\pm$3.5, 46.2$\pm$14.5, and 74.3$\pm$22.7 at large, middle and small follicles, respectively and total number of these 3 grade follicles per ovary were a, pp.ared to be 137.7$\pm$31.7. The mean number of follicle per ovary in PMSG-treated group 1 were a, pp.ared to be 25.6$\pm$7.3, 78.1$\pm$29.9, and 83.2$\pm$34.0, at large, middle and small follicles, respectively and total number of these 3 grade follicles were a, pp.ared to be 187.5$\pm$58.8. The mean number of follicle per ovary in PMS-treated group 2 were a, pp.ared to be 21.9$\pm$5.2, 67.8$\pm$16.8, and 68.0$\pm$14.9 at large, middle and small follicles, respectively and total number of these 3 grade follicles were a, pp.ared to be 157.7$\pm$26.2. The mean number of follicle per ovary in PMS-treated group 3 were a, pp.ared to be 21.7$\pm$4.8, 61.5$\pm$17.0, and 59.7$\pm$16.2 at large, middle and small follicles, respectively and total number of these 3 grade follicles were a, pp.ared to be 143.5$\pm$29.6. The number of follicles in PMSG-treated group 1 a, pp.ared to be more number than other 2 PMSG-treated gruops and tended to be decreased by frequency of PMSG-treatment.
