Application and Improvement of Complex Frequency Shifted Perfectly Matched Layers for Elastic Wave Modeling in the Frequency-domain (주파수영역 탄성파모델링에 대한 CFS-PML경계조건의 적용 및 개선)

  • Absorbing boundary conditions are used to mitigate undesired reflections that can arise at the model's truncation boundaries. We apply a complex frequency shifted perfectly matched layer (CFS-PML) to elastic wave modeling in the frequency domain. Modeling results show that the performance of our implementation is superior to other absorbing boundaries. We consider the coefficients of CFS-PML to be optimal when the kinetic energy becomes to the minimum, and propose the modified CFS-PML that has the CFS-PML coefficient ${\alpha}_{max}$ defined as a function of frequency. Results with CFS-PML and modified CFS-PML are significantly improved compared with those of the classical PML technique suffering from large spurious reflections at grazing incidence.

  • Promyelocytic leukemia (PML) gene, through alternative splicing of its C-terminal region, generates several PML isoforms that interact with specific partners and perform distinct functions. The PML protein is a tumor suppressor that plays an important role by interacting with various proteins. Herein, we investigated the effect of the PML isoforms on oncostatin M (OSM)-induced signal transducer and activator of transcription-3 (STAT-3) transcriptional activity. PML influenced OSM-induced STAT-3 activity in a cell type-specific manner, which was dependent on the p53 status of the cells but regardless of PML isoform. Interestingly, overexpression of PML exerted opposite effects on OSM-induced STAT-3 activity in p53 wild-type and mutant cells. Specifically, overexpression of PML in the cell lines bearing wild-type p53 (NIH3T3 and U87-MG cells) decreased OSM-induced STAT-3 transcriptional activity, whereas overexpression of PML increased OSM-induced STAT-3 transcriptional activity in mutant p53-bearing cell lines (HEK293T and U251-MG cells). When wild-type p53 cells were co-transfected with PML-IV and R273H-p53 mutant, OSM-mediated STAT-3 transcriptional activity was significantly enhanced, compared to that of cells which were transfected with PML-IV alone; however, when cells bearing mutant p53 were co-transfected with PML-IV and wild-type p53, OSM-induced STAT-3 transcriptional activity was significantly decreased, compared to that of transfected cells with PML-IV alone. In conclusion, PML acts together with wild-type or mutant p53 and influences OSM-mediated STAT-3 activity in a negative or positive manner, resulting in the aberrant activation of STAT-3 in cancer cells bearing mutant p53 probably might occur through the interaction of mutant p53 with PML.

  • This study is to develope the quantitative risk assessment program for consequence analysis of fire and explosion (PML-Chem), which is applicable to the chemical plants. The advantages of PML-Chem is easy to use and acquire results. Especially, PML-Chem was embedded real weather condition database for major chemical plants in ${\bigcirc}{\bigcirc}$ complex in country. Also, reliability of PML-Chem was verified through comparing PML-Chem with PHAST-Professional which is already commercial.

  • The PML developed for the radio wave propagation is a powerful numerical domain truncation technique. We perform an analytic study on the reflection from the PML inserted in the ocean bottom. In the ocean bottom, we show the PML to have the improved performance but simultaneously the degeneration below the critical angle of the fast ocean bottom. The degeneration of the PML can be simply relaxed by stretching the thickness of the PML or putting the attenuation coefficient to the ocean bottom. As a better solution, we propose the improved truncation technique based on the PML and the non-local boundary condition. Finally, we apply the PML to the acoustic wave propagation using split-step Pade PE solver. For the problems of the ocean waveguide, the numerical efficiency of the PML is examined and the usefulness of the PML is confirmed.

  • The Perfectly Matched Layer(PML) provide good performance in absorption over a wide frequency range and is an appropriate ABC for waveguides with high dispersion. In this paper, a novel algorithm is proposed to improve the computational efficiency of the PML. In the input and output ports, the fields are decomposed into a series of modes, and then an appropriate ABC is applied to each mode. CPU time and memory storage requirements are greatly reduced, since the computational region is analyzed in one dimension. A WG-90 rectangular waveguide with a thick asymmetric iris is analyzed by Finite-Difference Time-Domain(FDTD) simulations with the conventional PML and the proposed one-dimensional (1-D) PML. Numerical results show that the computational efficiency is significantly improved by the proposed method.

  • Various numerical methods in simulation of seismic wave propagation have been developed. Recently an innovative numerical method called as the Spectral Element Method (SEM) has been developed and used in wave propagation in 3-D elastic media. The SEM that easily implements the free surface of topography combines the flexibility of a finite element method with the accuracy of a spectral method. It is generally used a weak formulation of the equation of motion which are solved on a mesh of hexahedral elements based on the Gauss-Lobatto-Legendre integration rule. Variational formulations of velocity-stress motion are newly modified in order to implement ADE-PML (Auxiliary Differential Equation of Perfectly Matched Layer) in wave propagation in 3-D elastic media, because a general weak formulation has a difficulty in adapting CFS (Complex Frequency Shifted) PML (Perfectly Matched Layer). SEM of Velocity-Stress motion having ADE-PML that is very efficient in absorbing waves reflected from finite boundary is verified with simulation of 1-D and 3-D wave propagation.

  • 본 논문에서는 PML-트리라는 공간색인구조를 제안한다. PML-트리는 object distribution heuristics를 사용하여 공간 데이터 객체를 여러 개의 데이터 공간에 균일하게 배치함으로써 질의처리 속도를 향상시킨다. 두 가지의 object distribution heuristics(absolute crowd index와 relative crowd index)가 제안이 된다. PML-트리는 공간 객체를 분배함으로써 R+-트리의 말단 노드 내에 존재하는 데이터의 중복을 제거하면서, R-트리의 단점인 색인 사각형들 사이에 중첩을 허용치 않는다. PML-트리의 성능은 여러 타입의 테스트 데이터를 사용하여 MXR-트리와 비교된다. PML-트리는 MXR-트리에 비해 높은 공간활용도와 빠른 질의 반응시간을 보임으로써 공간 데이터베이스를 위한 효율적인 색인구조로 사용이 될 것으로 기대된다.

Promyelocytic Leukemia (PML) Gene Mutations may not Contribute to Gastric Adenocarcinoma Development

  • Gastric cancer is the second most common cause of cancer death worldwide. Environmental as well as genetic factors have been shown to be involved in its genesis. Among genetic factors, loss of function of a tumor suppressive gene named promyelocytic leukemia (PML) has been demonstrated in gastric cancer. In order to cast light in the mechanism by which PML protein is under-expressed in gastric cancer cells, we analyzed all exons and intron-exon boundaries of PML gene in 50 formalin-fixed paraffin-embedded tissue blocks from gastric carcinoma tumors by means of PCR-SSCP and CSGE, with direct sequencing of abnormally shifted bands. We found a novel sequence variant of unknown significance localized in intron 5 in 3 samples (c.1398+84delA). We did not detect any deleterious mutations of the PML gene. This study shows that PML mutations may not contribute to gastric adenocarcinoma development. Post-translational modifications or protein degradation might be mechanisms by which PML is not expressed in gastric tumors.

  • The purpose of this paper is developing a PML server which is capable of saving and managing effectively the information of vehicles using RFID technology in the logistical and distributive environment. The PML server logs and manages the real-time static and dynamic information about the vehicles in EPC code format which is represented by XML schema and transmitted by the RFID reader to the server. It receives information from SAVANT the RFID middleware and transmits the data to the traffic information management application The main system we developed consists of the EPC data registry, query processor, data converter, vehicle product and instance data storage, system management tool and application API parts.

In Silico Interaction and Docking Studies Indicate a New Mechanism for PML Dysfunction in Gastric Cancer and Suggest Imatinib as a Drug to Restore Function

  • Gastric cancer as one of the most common cancers worldwide has various genetic and environmental risk factors including Helicobacter pylori (H.pylori) infection. Recently, loss of a tumor suppressor gene named promyelocytic leukemia (PML) has been identified in gastric cancer. However, no mutation has been found in this gene in gastric cancer samples. Cag A H.pylori protein has been shown to exert post transcriptional regulation of some tumor suppressor genes. In order to assess such a mechanism for PML degradation, we performed in silico analyses to establish any interaction between PML and Cag A proteins. In silico interaction and docking studies showed that these two proteins may have stable interactions. In addition, we showed that imatinib kinase inhibitor can restore PML function by inhibition of casein kinase 2.
