Tropical plant supplementation effects on the performance and parasite burden of goats

  • Objective: Examine the effects of supplementing bahiagrass hay (BG) with potentially anthelmintic quantities of hays of perennial peanut (PEA) or sericea lespedeza (LES) or seeds of velvet bean (Mucuna pruriens L.; MUC) or papaya (PAP) on the intake and nutritive value (Experiment 1), and the performance and parasite burden (Experiment 2) of goats. Methods: In Experiment 1, 38 male goats ($27.4{\pm}5.7kg$ body weight) were randomly assigned to each of 5 treatments: i) BG alone and BG plus; ii) PEA; iii) LES; iv) MUC; and v) PAP. Goats were fed for ad libitum consumption and adapted to the diets for 14 d followed by 7 d of measurement. The PEA, LES, MUC (50%, 50%, and 10% of the diet dry matter [DM], respectively), and PAP (forced-fed at 10 g/d) were fed at rates that would elicit anthelmintic effects. In Experiment 2, goats remained in the same treatments but were allocated to 15 pens (3 pens per treatment) from d 22 to 63. All goats were infected with parasites by grazing an infected bahiagrass pasture from 0800 to 1500 h daily and then returned to the pens. Results: Dry matter intake tended to be greater in goats fed PEA and LES than those fed BG (757 and 745 vs 612 g/d, respectively). Digestibility of DM (59.5% vs 54.9%) and organic matter (60.8% vs 56.0%) were greater in goats fed MUC vs BG, respectively. In Experiment 2, feeding PAP, LES, and PEA to goats reduced nematode fecal egg counts by 72%, 52%, and 32%, reduced abomasal adult worm counts by 78%, 52%, and 42%, and decreased plasma haptoglobin concentrations by 42%, 40%, and 45% relative to feeding BG alone, respectively. Conclusion: Supplementation with PEA, LES, and PAP decreased the parasite burden of goats but did not increase their performance. PAP was the most effective anthelmintic supplement.

The Effect of Root Exudate and Chemotaxis on Host Recognition in Soybean-Bradyrhizobium Symbiosis (대두(大豆)-근류균(根瘤菌) 공생(共生)에서 뿌리분비물(分泌物)과 화학주성(化學走性)이 숙주인식(宿主認識)에 미치는 영향(影響))

  • This study was carried out to research the effect of the chemotaxis of Bradyrhizobium japonicum KCTC 2422 and its mutant toward soybean root exudate and to elucidate the effect of the lectin of host specificity (Host Recognition) in soybean-Bradyrhizobium symbiosis. The results obtained were as follows: The homogeneities of the purified lectins from soybean and pea seed was ascertained chromatographically and electrophoretically. Gel electrophoresis of soybean lectin in the presence of sodium dodecyl sulfate appeared a single protein band, whereas pea lectin appeared two protein bands. Soybean lectin from 2 cultivars formed immunoprecipitin arcs at same position with anti-soybean lectin rabbit IgG, but pea lectin did not form immunoprecipitin lines with anti-soybean lectin rabbit IgG. Chemotactic responses of KCTC 2422, LPN-100 and LCR-101 toward proline in capillary assays were 3.1, 1.3 and 1.0-fold above background, respectively. The chemotactic responses of KCTC 2422, LPN-100, and LCR-101 toward Paldal crude root exudate in capillary assays were 3.5, 1.4 and 1.4-fold above background, respectively. The present work shows that B. japonicum and its mutants are capable of very different responses toward root exudate fraction. The chemotactic responses of KCTC 2422 was most with neutral fraction, least with anionic fraction and intermediate with cationic fraction. The nitrogenase activity of soybean nodule was shown in 15days after inoculation with LCR-101. However, we couldn't find out the nodules when soybean was inoculated with LPN-100. From these result we can suppose that the chemotaxis of Bradyrhizobium plays inportant the role of forming the nodule (host recognition) in the soybean-B. japonicum symbiosis.

Effect of Heating Temperature on Elution Patterns of Soluble Carbohydrate of Legume Starches and the Properties of Starch Gels (가열온도가 두류전분의 가용성 탄수화물의 용출양상과 전분겔 특성에 미치는 영향)

  • Effect of heating temperature on elution patterns of soluble carbohydrate of legume starches and the texture of starch gels was investigated. The elution profiles of soluble carbohydrate obtained by Sepharose 2B-CL showed that the larger molecules were leached as the heating temperature increased. The elution profiles of soluble carbohydrate of cow pea and mung bean starches were similar, but those of kidney bean starch were different. The gel structures of cow pea and mung bean were stable with showing high values in hardness and cohesiveness. But red bean and kidney bean gels were weak in hardness and cohesiveness. As the heating temperature rised, hardness and cohesiveness were increased in gels prepared by heating for 15 min. However, the gels made by heating for 1 hour showed that less hard and more cohesive gels from cow pea and mung bean, while harder and more cohesive gels from red bean and kidney bean were obtained as the heating temperature increased from $85^{\circ}C\;to\;95^{\circ}C$.

Growth and Optimum Harvesting Time of Pod-edible Peas (Pisum sativum L.)

  • The present study was performed to obtain the basic information about growth and quality related characteristics and optimum harvesting time for podedible pea which is a new crop in Korea but believed to have a great deal of potentials for both domestic and overseas markets. They can be consumed either as a fresh succulent vegetable or as tender green pods. The daily green pod yield of pod-edible peas started to increase from ten days after flowering and the maximum yield was recorded on 26 days after flowering. Ninety percent of pod yields could be harvested from 16 to 36 days after flowering. Mean green pod yield for the tested varieties was approximately 8.0 t/ha. Total vitamin C content of pod-edible peas showed continuously decreasing trends from five days after flowering. The highest sucrose content was obtained at ten days after flowering. The highest panel score based on sweetness, chewiness, and hardness for the processed green pods was shown at 10-15 days after flowering in all varieties tested, indicating that the optimum harvesting time for pod-edible peas was considered to be 10-15 days after flowering.

  • A pot experiment was conducted to find out the effect of zeolite application on the growth and $N^2$ fixation of pea plants. The results are as follows. 1) The dry weight of pea plants grown with N-fertilizer application was smaller than that without N, but In was increased by application of zeolite 2) The N content in pea plants was increased by zeolite application. The N increment was smaller in 2% zeolite plots than in 1% zeolite plots. Larger amount of Ammonium were remained in the former plots. 3) The N amounts accumulated in different parts of plant derived from $^{15}N$-labelled fertilizer were decreased by application of zeolite. 4) The N derived from atmuspheric $N_2$ was increased by zeolite application. However, the application of fertilizer nitrogen depressed the enhancement of fixed-N accumulation.

Effects of Temperature on the Oviposition, Feeding and Emergence of the Azuki Bean Weevil (Callosobruchus chinensis L.) in the stored beans (저장두류(貯藏豆類)에 대(對)한 팥바구미의 산란(産卵).섭식(攝食) 및 우화(羽化)에 미치는 온도(溫度)의 영향(影響))

  • This study was conducted to study the effects of temperature on the oviposition, feeding and emergence of the azuki bean weevil(ABW) (Callesobruchus chinensis L.) in the stored beans. And ovipositional preference, hatching and emergence on the preference and non-preference hosts were also investigated. ABW has four generations a year and overwinters as larva stage in the bean grains. Optimum oviposition temperature of the ABW ranges from $25^{\circ}C\;to\;30^{\circ}C$. Oviposition preference in different hosts was in the order of azuki bean, mung bean, soy bean, kidney bean, black-soybean, pea. At $25^{\circ}C$, optimum development temperature emergence of period the ABW averaged 29 days on azuki bean, 31 days on mung bean, 49 days on soy bean, 46 days on black-soybean, 34 days on Pea. Percent hatch in different hosts was in the order of azuki bean, mung bean, soy bean, pea, black-soybean, kidney bean. But percent emergence was in the order of azuki bean, mung bean, pea, black-soybean, kidney bean. Especially, in azuki bean and mung bean, percentage of hatching and emergence was very high. And in soy bean and black-soybean, percentage of hatching was high but emergence was low. The kidney bean was hatching but not emergence at all. The longevity at the different temperatures appeared to be $4{\sim}5$ days at $35^{\circ}C,\;5{\sim}7$ days at $25^{\circ}C{\sim}30^{\circ}C$, $10{\sim}15$ days at $20^{\circ}C\;and\;19{\sim}24$ days at $15^{\circ}C$. Kidney bean of feeding non-preference host oviposited at Seed coat and Endosperm, but no larva developed. And cotyledon and embryo parts were not oviposited at all. The percentage of germination of egg laid bean grain and damaged bean grains was low. Especially, the percentage of germination of damaged bean grains was 0% on azuki bean, 5% on black-soy bean, 7% on Pea.

  • Interfacial properties and microfailure degradation mechanisms of the bioabsorbable composites fur implant materials were investigated using micromechanical technique and nondestructive acoustic emission (AE). As hydrolysis time increased, the tensile strength, the modulus and the elongation of poly(ester-amide) (PEA) and bioactive glass fibers decreased, whereas these of chitosan fiber almost did not change. Interfacial shear strength (IFSS) between bioactive glass fiber and poly-L-lactide (PLLA) was much higher than PEA or chitosan fiber/PLLA systems using dual matrix composite (DMC) specimen. The decreasing rate of IFSS was the fastest in bioactive glass fiber/PLLA composites whereas that of chitosan fiber/PLLA composites was the slowest. AE amplitude and AE energy of PEA fiber decreased gradually, and their distributions became narrower than those in the initial state with hydrolysis time. In case of bioactive glass fiber, AE amplitude and AE energy in tensile failure were much higher than in compression. In addition, AE parameters at the initial state were much higher than those after degradation under both tensile and compressive tests. In this work, interfacial properties and microfailure degradation mechanisms can be important factors to control bioabsorbable composite performance.

Molecular Structural Properties of Legume Starches (두류 전분의 분자구조적 특성)

  • Molecular structural properties of legume starches were investigated. In intrinsic viscosity and degree of Polymerization of amylose and amylopectins, cow pea and mung bean were high, but kidney bean was low. Low molecular weight fractions for kidney bean starch were much eluted by gel chromatography. In the elution profiles of their amylose by Sepharose 2B-CL, molecular weight of kidney bean amylose was smaller than that of other amylose Molecular weights of cow pea and mung bean amyloses were large, but that of kidney bean amylose was small and red bean amylose was medium. The elution profiles by Sephadex G-50 after debranching amylopectins with pullulanase showed similar patterns.

Physicochemical Properties of Starch from Cow Pea (동부 전분의 이화학적 특성)

  • Starch granules of cow pea were oval with their sizes of $6-22{\mu}$m. X-ray diffraction pattern of starch granules showed some crystallinity at $2{\theta}$ $15^{\circ}$, $17.1^{\circ}$, $18.1^{\circ}$, $22.9^{\circ}$. The blue value of starch was 0.370, amylose contents 28.1% and water binding capacity 70.6%. Swelling power of the starch increased rapidly from $65^{\circ}C$ to $75^{\circ}C$ thereafter increased less rapidly. Solubility showed the same pattern as the swelling power. Optical transmittance of 0.2% starch suspension was increased rapidly from $70^{\circ}C$ and the gelatinization between $70^{\circ}C$ and $90^{\circ}C$ was single stage. Amylogram of 4% starch solution showed no peak viscosity but 6% starch solution showed peak viscosity.

  • The indole-3-acetic acid(IAA) oxidase activity of shoot and root tips of etiolated pea seedlings (Pisum sativum L. var. Sparkle) during and after chilling was determined. The IAA oxidase level of root tips was 4 to 15 times as high as that of shoot tips. During chilling the seedlings apical and subapical 5 mm shoot sections increased in IAA oxidase activity but apical and subapical 5 mm root sections decreased. When chilled plants were returned to $25^{\circ}C$ to recover, the enzyme activity had a tendency to restore to the activity level of controlled plants.
