Vitexin, a natural flavonoid compound identified as apigenin-8-C-${\beta}$-D-glucopyranoside, has been reported to exhibit antioxidative and anti-inflammatory properties. In this study, we investigated its effect on hypoxiainducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) in rat pheochromacytoma (PC12), human osteosarcoma (HOS) and human hepatoma (HepG2) cells. Vitexin inhibited HIF-$1{\alpha}$ in PC12 cells, but not in HOS or HepG2 cells. In addition, it diminished the mRNA levels of hypoxia-inducible genes such as vascular endothelial growth factor (VEGF), smad3, aldolase A, enolase 1, and collagen type III in the PC12 cells. We found that vitexin inhibited the migration of PC12 cells as well as their invasion rates, and it also inhibited tube formation by human umbilical vein endothelium cells (HUVECs). Interestingly, vitexin inhibited the hypoxia-induced activation of c-jun N-terminal kinase (JNK), but not of extracellular-signal regulated protein kinase (ERK), implying that it acts in part via the JNK pathway. Overall, these results suggest the potential use of vitexin as a treatment for diseases such as cancer.
The effects of sesamin on dopamine biosynthesis in PC12 cells were investigated. Sesamin at concentration ranges of 20-75 ${\mu}M$ significantly increased intracellular dopamine levels and tyrosine hydroxylase (TH) activities at 24 h: 50 ${\mu}M$ sesamin increased dopamine levels to 132% and TH activities to 128% of control levels. Sesamin (50 ${\mu}M$) induced the phosphorylation of TH, cyclic AMP-dependent protein kinase (PKA) and cyclic AMP-response element binding protein (CREB) for 0.5-24 h. Sesamin (50 ${\mu}M$) also increased the mRNA levels of TH and CREB for 3-24 h. In addition, sesamin (50 ${\mu}M$) associated with L-DOPA (50 and 100 ${\mu}M$) further increased the intracellular levels of dopamine for 24 h compared to L-DOPA alone. These results suggest that sesamin enhances dopamine biosynthesis and L-DOPA-induced increase in dopamine levels by inducing TH activity and TH gene expression, which is mediated by PKA-CREB systems in PC12 cells. Therefore, sesamin could serve as an adjuvant phytonutrient for neurodegenerative diseases.
본 연구에서는 마른진흙버섯 자실체를 메탄올과 열수를 이용해 추출한 물질의 anti-xanthine oxidase, anti-cholinesterase 및 염증 저해 효과에 대한 연구를 수행하였다. 마른진흙버섯 자실체의 메탄올 추출물과 열수 추출물의 xanthine oxidase에 대한 저해효과는 양성대조군으로 사용한 allopurinol과 대등하게 높은 효과를 나타냈다. Acetylcholinesterase에 대한 메탄올 추출물의 1.0~2.0 mg/mL 농도에서의 저해활성은 양성대조군인 galanthamine과 유사하게 높았지만 butyrylcholinesterase에 대한 메탄올과 열수 추출물의 저해활성은 양성대조군에 비해 실험에 사용한 모든 농도범위에서 유의하게 낮았다. PC-12 세포에 glutamate의 처리에 의해 유도된 독성은 40 mg/mL와 100 mg/mL 농도의 메탄올 추출물과 100 mg/mL 농도의 열수추출물의 처리에 의해 크게 완화되어 PC-12 세포의 생존율이 유의하게 증가하는 것이 관찰되었다. 마른진흙버섯의 메탄올과 열수 추출물의 염증 저해 실험에서 RAW 264.7 대식세포에 메탄올 추출물을 2.0 mg/mL 농도로 처리하고 염증을 매개하는 LPS를 추가로 투여한 후 RAW 264.7 세포에 생성되는 NO를 측정한 결과, 양성대조군에 비해 3.37배 높은 저해효과를 나타냈고, 처리한 자실체 메탄올 추출물의 농도가 증가함에 따라 생성된 NO의 양이 현저하게 감소하는 경향을 나타내었다. 또한 기염제인 carrageenan에 의해 흰쥐 뒷발에 유도된 부종 저해 실험에서는 투여한 버섯 추출물의 농도가 증가함에 따라 흰쥐의 뒷발에 유도된 부종의 용적이 농도 의존적으로 감소하는 경향을 나타냈다. 따라서 마른진흙버섯 자실체에 함유된 물질은 acetylcholinesterase과 butyrylcholinesterase 등의 cholinesterase에 대한 저해작용과 glutamate에 의해 유도된 PC-12세포의 독성을 완화하고 또한 염증을 저해하는 효과를 나타내 기억력이 감퇴되는 초기 알츠하이머병과 염증을 완화하는 천연소염제로의 이용이 가능할 것으로 사료된다.
Although it is known that neuronal cell death during development occurs by apoptosis, the mechanisms underlying excitatory amino acid-induced neuronal cell death remain poorly understood. In this study we have examined the mechanism by which L-glutamate, an excitatory amino acid neurotransmitter, induces cell death in PC12 cell lines. To characterize cell death, we employed sandwich enzyme-linked immunosorbent assay(ELISA) method for cellular DNA fragmentation, DNA agarose gel electrophoresis and chromatin staining by acridine orange and ethidium bromide after treating the PC12 cells with L-glutamate. L-Glutamate caused dose-dependent cell death with a maximum at 24 hrs after the treatment. These cellular fragmentation was blocked by pretreatment of MK-801, a noncompetitive N-methyl-D-aspartic acid(NMDA) receptor antagonist, and nerve growth factor(NGF). Analysis of DNA integrity from L-glutamate-treated cells revealed cleavage of DNA into regular sized fragments, a biochemical hallmark of apoptosis. The PC12 cells that were induced to die by L-glutamate treatment exhibited classical chromatin condensation under the light microscopy after acridine orange and ethidium bromide staining. These results suggest that apoptosis is one of the key features that are involved in L-glutamate-induced excitotoxic cell death in PC12 cells, and these cell death are mediated by NMDA receptor and depend on NGF.
The water extract of Hwansodan has been traditionally used for treatment of ischemic brain damage in oriental medicine. However, little is known about the mechanism by which the water extract of Hwansodan rescues cells from neurodegenerative disease. PC12 pheochromocytoma cells have been used extensively as a model for studying the cellular and molecular mechanisms of neuronal cell damages. Under deprivation of growth factor and ischemic injury, PC12 cells spontaneously undergoes apoptotic cell death. Serum and glucose deprivation markedly decreased the viability of PC12 cells, which was characterized with apparent apoptotic features such as membrane blebbing as well as fragmentation of genomic DNA and nuclei. However, the aqueous extract of Hwansodan significantly reduced serum and glucose deprivation-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Pretreatment of Hwansodan also ingibited the activation of caspase-3, in turn, degradation of ICAD/DFF45 was completely abolished in serum and glucose deprivated cells. Furthermore, pretreatment of Hwansodan obviously increased heme oxygenase 1 (HO-1) expression in PC12 cells. Taken together, the data suggest that the protective effects of Hwansodan against serum and glucose deprivation induced oxidative injuries may be achieved through the scavenging of reactive oxygene species accompanying with HO-1 induction.
PC12 신경세포를 이용하여 구아바 열매와 잎 추출물이 $H_2O_2$로 유도된 신경세포 독성에 대한 보호 효과를 조사하였다. 구아바 열매와 잎 열수 추출물의 총페놀성 화합물 함량은 각각 11.75 및 293.25 mg/g이었고, gallic acid 함량은 각각 22.78 및 117.34 mg/100 g이었다. $H_2O_2$ 처리한 PC12 cell내의 활성산소 생성억제효과를 측정한 결과 구아바 잎열수 추출물에서 높은 활성산소 생성 억제효과를 보였다. MTT방법을 이용하여 $H_2O_2$로 유도된 PC12 신경세포에 대한 보호효과를 측정한 결과 구아바 열매와 잎 열수 추출물은 높은 신경세포 보호효과를 보였고, LDH release 실험결과 모든 농도에서 세포막 보호효과를 나타내었다. 본 연구 결과를 종합해 볼 때, 구아바 열매와 잎 열수 추출물의 항산화활성과 산화적 스트레스로부터의 신경세포 보호효과를 나타내어 퇴행성 뇌신경질환 등의 예방 및 개선을 위한 산업화 소재로서의 활용 가능성이 높다고 판단된다.
The present study investigated neuroprotective effects Rehmanniae Radix on PC12 cells and hippocampal neural cells. PC12 cells were damage by $H_2O_2$ and nitric oxide and organotypic hippocampal slice cultures were damaged by oxygen-glucose deprivation. Then methanol extract of Rehmanniae Radix was treated with 0.5, 5, and $50\;{\mu}g/ml$ in culture media. Effects of Rehmanniae Radix were evaluated with cell viability assay, PI-staining, and TUNEL-labeling. Treatment of Rehmanniae Radix ($with\;5\;and\;50\;{\mu}g/ml$) produced significant increase of cell viability of PC12 cells damaged by $H_2O_2$ and by SNP-induced nitric oxide. Treatment of Rehmanniae Radix produced significant decrease of PI-uptake % in CA1 ($with\;5\;and\;50\;{\mu}g/ml$) and DG ($with\;50\;{\mu}g/ml$) regions of organotypic hippocampal slice cultures damaged by oxygen-glucose deprivation. Moreover, treatment of Rehmanniae Radix produced significant decrease of TUNEL- positive cells in CA1 ($with\;5\;and\;50\;{\mu}g/ml$) and DG ($with\;50\;{\mu}g/ml$) regions of organotypic hippocampal slice cultures damaged by oxygen-glucose deprivation. These results suggest that methanol extract of Rehmanniae Radix has neuroprotective effects on PC12 cells damaged by oxidative stress and on organotypic hippocampal slice cultures damaged by oxygen-glucose deprivation.
Objective : Alzheimer's disease (AD) is a geriatric dementia that is widespread in old age. With an aging populace, AD is a looming problem in public health service. Alzheimer's disease is characterized by specific neuronal degeneration in certain areas of the brain. Mutations and abnormal expression of several genes are associated with ${\beta}-amyloid$ deposits and Alzheimer's disease; among them APP, PS1, and PS2, SOD, free radical, ROS. Methods:We studied herbal medicines that have a relationship to brain degeneration. From pre-modern times, although a variety of oriental prescriptions of Aster tataricus have been traditionally utilized for the treatment of AD, their pharmacological effects and action mechanisms have not yet been fully elucidated. Result : Based on morphological observations by phase-contrast microscope, TUNEL assay and MTT in the culture media, $H_20_2-induced$ cell death was significantly inhibited by Aticus. We examined by ROS formation, catalase activity and GSH activity. We studied the protective effect and inhibitory effects of neurotoxicity in $H_20_2-induced$ PC-12 cells by Aticus. Findings from our experiments show that Aticus inhibits apoptosis, which has neurotoxicities and cell damage in PC-12 cells. In addition, treatment with Aticus ($>25{\mu}g/ml$ for 6hrs) partially prevented $H_20_2-induced$ cytotoxicity in PC-12 cells, and induced a protective effect. Conclusion : As the result of this study, in the Aticus group, the apoptosis in the nervous system was inhibited, protected against the degeneration of PC-12 cells by $H_20_2$. Taken together, Aticus exhibited inhibition of $H_20_2-induced$ apoptotic cell death. Aticus was found to induce protective effect on GSH and catalase in PC-12 cells. Based on these findings, Aticus may be beneficial for the treatment of AD.
We introduced mechanical stimuli and micropatterned substrate with micro fibers to investigate the effects of those on neurite outgrowth along with nerve growth factor (NGF) in vitro. Microfiber substrates were fabricated using an electrospinning process. And PC-12 cells cultured on substrates were simulated with nerver growth factor and laminar flow shear stress in a fluid flow system The results suggest that microfiber substrates and fluid-induced shear stress are promising for simulating neuronal regeneration in a desired direction.
The effects of liriodenine, an aporphine isoquinoline alkaloid, on dopamine content in PCl2 cells were investigated. Treatment of PC12 cells with liriodenine decreased dopamine content in a dose-dependent manner (33.6% inhibition at $10\;{\mu}M$ for 12 h). The $IC_{50}$ in value of liriodenine was $8.4\;{\mu}M$. Dopamine content decreased at 3 h and reached a minimal level at 12 h after the exposure to liriodenine. Under these conditions, the activities of tyrosine hydroxylase and aromatic L-amino acid decarboxylase were also inhibited at $10\;{\mu}M$ of liriodenine by 10.1% and 20.2% relative to control, respectively. In addition, liriodenine inhibited the increase in dopamine content induced by L-DOPA Treatments $(50-100\;{\mu}M)$ in PC12 cells. These results suggest that liriodenine inhibited dopamine biosynthesis and L-DOPA-induced increase in dopamine content by reducing the activities of tyrosine hydroxylase and aromatic L- amino acid decarboxylase in PC12 cells.
