The Treatment Effect of Younggamgangmisinhayin-tang through Control of Th 2 cell Differentiation in Allergic Rhinitis (영감강미신하인탕이 Th 2 세포 분화 조절을 통해 알레르기성 비염에 미치는 효과)

  • Objectives : Younggamgangmisinhayin-tang is clinically used for the treatment of the aller -gic rhinitis. The aim of this study is to investigate the effects of Younggamgangmisinhayin -tang on allergic rhinitis. Methods : The thirty rats were divided into three groups : control group, AR(allergic rhinitis eliciated) group, YT(Younggamgangmisinhayin-tang treated after allergic rhinitis elicitation) group. To induce allergic rhinitis in AR group, YT group rats were sensitized intraperitoneally with 0.1% ovalbumin(OVA) solution 3 times at intervals of 1 week. Then intranasal sensitization was performed by diffusing 0.1% ovalbumin(OVA) solution 3 times at intervals of 2 days. After that time rats in YT group were oral administration treated by Younggamgangmisinhayin-tang for 7days. The change of the amounts of esinophil, PAS, ZO-1, MIP-2, COX-2, IL-4 were observed in each group. Th 2 skewed condition in EL4 cell was induced by using PMA and OPl. Younggamgangmisinhayin-tang was added into EL4 cell by density. And then IL-4 mRNA was observed. Results : In esinophil study YT group was proved significant inhibitary effect. In PAS and ZO-1 YT group were preserved well. In MIP-2, COX-2 study YT group was proved significant inhibitary effect. In IL-4 and IL-4 mRNA study YT group was proved significant inhibitary effect. Conclusion :According to the above results, it is considered that Younggamgangmisinha- yin-tang has inhibitory effects on the allergic rhinitis rat models.

The Cytotoxicity and Antitumor Activity of Palsun Brewing Water (팔성 양조용수의 세포독성 및 항암활성)

  • In the present study, we have evaluated cytotoxic effects, antitumor activities and metastasis inhibitory effects of Palsun brewing water in NIH 3T3 cells, human epitheloid carcinoma cells, and human skin melanoma cells. The light microscopic study showed morphological changes, AG-NOR (argyrophylic nucleolar organizer region) by silver chloride stain, and glycoprotein by PAS (periodic acid stain) reaction of the treated cells. Disruptions in cell organelles were determined by colorimetric methods; MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide) and SRB (sulforhodamine B protein) assays. These results suggest that Palsun Brewing Water retains no cytotoxic effects in NIH 3T3 cells and a growth-inhibitory activity in cancer cell lines.

AN EXPERIMENTAL STUDY ON THE EFFECT OF THE DEMETHYLCHLORTETRACYCLINE ON MANDIBULAR SYMPHYSIS IN RATS (Demethylchlortetracycline이 백서 하악골결합부의 발육에 미치는 영향에 관한 실험적 연구)

  • This study was undertaken to assess the effects of demethylchlortetracycline on bone growth of mandibular symphyseal region in rats. Demethylchlortetracycline at 30mg/kg body weight, respectively, were daily administered by mouth to the sewen female rats from 10th day of pregnancy to 13th day. Thirty six new-born rats from these experimental animals were used for histological examination at 1, 5, 10, 15, 20, 25 and 30 days. All these new-born rats were killed by an overdose of ether. Mandibular bodies were removed and fixed in $10\%$ neutral formalin,. Carney and aceton. Specimens were embedded, sectioned and stained with H-E, Van Gieson, PAS and prepared for alkaline phosphatase. The results were as follows; 1. Until erupting of incisors, hyaline cartilage was located in relatively large symphyseal space, but bone trabeculae of ossifying area at incisal side were arranged irregularly in experimental group. 2. During this period, PAS reaction was moderately positive, but alkaline phosphatase reaction was slightly positive. 3. By erupting or incisors, symphyseal space appeared narrower like control group, but alkaline phosphatase reaction tended to slow down. 4. By erupting of molars, symphyseal space appeared muck narrower, and cartilane plate was reduced and aealed off like control group. Alkaline phosphatase reaction tended to slow down severely.

Phenolic Composition and Antioxidant Activities of Different Solvent Extracts from Pine Needles in Pinus Species

  • The purpose of this study was to investigate the phenolic acid, proanthocyanidin (PAs), and flavonol glycoside contents, as well as the antioxidant activities of pine needle extracts from six species of young pine trees. The extracts were prepared from Section Pinus (Diploxylon): P. densiflora, P. sylvestris, P. pinaster and P. pinea, and Section Strobus (Haploxylon): P. koraiensis and P. strobus. Phenolics were extracted from pine needles with 80% acetone to obtain the soluble free fraction, and insoluble residues were digested with 4 M NaOH to obtain bound ethyl acetate and bound water fractions. Phenolics were analyzed by HPLC, and the hydrophilic antioxidant activity was measured using oxygen radical absorbance capacity (ORAC). Total phenolic and flavonoid contents of the soluble free fraction were higher than those of the bound ethyl acetate and bound water fractions. The main phenolics were monomers and polymers of PAs in the soluble free fraction, and phenolic acids and flavonol glycosides in bound ethyl acetate fraction. Flavonol glycosides found in different species of pine needles were qualitatively similar within fractions, but composition varied among Pinus sections. High levels of kaempferol arabinoside and an unknown compound were present in all Strobus species. The soluble free fraction had the highest antioxidant activity, followed by bound ethyl acetate and bound water fractions.

  • This study investigated Greene and Corwder(1984)'s modified PAS model, according to which, in a short-term memory recall task, the recency and suffix effects existing in auditory and visual conditions are mediated by the same mechanisms. It also investigated whether the auditory information and mouthed information are encoded by the same codes. Though the experimental manipulation of the phonological nature, the presence of differential recall effect of consonant-and vowel-varied stimuli in auditory and mouthing conditions which has been supposed to interact with the recency and suffix effects, was investigated. The result shows that differential recall effect between consonant and vowel exists only in the auditory condition, but not in the mouthing condition. Thus, this result supported Turner.

  • The effects of folic acid on the oral mucosa of albino rats were histochemically studied in the twenty four male rats, weighing abut 100gm. Seven groups of 3 each were injected with 0.2ml folic acid (folic acid 0.5mg. was dissolved in physiological saline 0.2ml) subcutaneously, during 1,2,3,5,7,10 and 14 days respectively. Oral mucosa of rats sere removed from upper molar region and fixed in 10% formalin, cold absolute alcohol, Carnoy's solution and acetone. The serial sections were histochemically stained by McManu's PAS reaction, Mowry's metachromasia, alloxan-Schiff reaction, and azo dye method for alkaline phosphatase. The comparative staining method was hematoxylin-eosin stain. The results were as follows: 1) Alkaline phosphatase reaction of stratum spinosum and stratum granulosum tended to increase after 7 and 10 days of folic acid administration. 2) PAS reactions of basement membrane and lamina propria increased after folic acid administration. 3) Metachromasia of stratum spinosum and stratum granulosum were slightly increased after 3,5 and 7days of folic acid administration and returned to the level of Control after 10 days. 4) In the oral mucosa, alloxan-Schiff reaction increased after 7 days of folic acid administration.

  • The structure of skin was studied in Iksookimia longicorpus based on the micro-anatomical investigation of skin fragments taken from four regions. The epidermis was distinguished by two types of skin glands, a small mucous cell and a large club cell. The mucous cell was acid sulfomucins (some sialomueins) but the club cell did not give any histochemical tests for mucosubstances. The presence of a well defined lymphatic system with small lymphocytes was established in the stratum germinativum layer of the epidermis. A large number of blood capillaries run very close to each other just below the basement membrane, and a definite area giving AB and PAS positive was present between the basement membrane and scale.

  • This study was conducted for the histological observation of the infundibulum of the oviduct of the laying Korean native pheasants. The results are as follows : 1. The infundibular wall is composed of the epithelium, lamina propria, muscle layer(inner circular and outer longitudinal muscle), and serosa. The funnel lip is divided into the inner, and outer lip of the epithelium and muscle layer. 2. The epithelium of the funnel lip and most region of the cranial part of the funnel are composed of ciliated columnar cells. In the surface and lateral part of the folds, ciliated cells and non-ciliated secretory cells tend to alternate in the epithelium of the caudal funnel and the necks, but are also found in groups of the simple cuboidal epithelium at the bases of the grooves between the ridges and tubular glands found in the subepithelium. 3. The secretory material of the non-ciliated secretory cills contains PAS-positive and alcian blue-positive granules, and these materials show purple colour in the basic fuchsin-methylene blue stain. 4. The cells of the glandular groove and tubular gland of the neck portion of the oviduct mostly show weak PAS-positive, and alcian blue stain negative reaction. The tubular gland cells of the infndibulum contain pink of purple colour granules, and without reaction in the anterior neck portion of the infundibulum in basic fuchsin and methylene blue stain.

  • The blood proteins of pine moth, Dendrolimus spectabilis Butler of different developmental stages were investigated by disc electrophoresis in acrylamide gels. Blood protein concentration was also determined during the metamorphosis. Protein concentration increased gradually with the growth of larva, reaching a maximum in themature larva, and the increase of protein bands also was accompanied. As the larva transforms into the prepupa the number of protein bands as well as the protein concentration dropped. A total of 22 bands were identified throughout the stages. Histochemical staining of the acrylamide gels by the PAS method, Toluidine blut O, and Sudan black demonstrated that the carbohydrate, mucopolysaccharide, and lipid were associated with certain blood proteins.

The Study of Breath Group Based on Oral Airflow in Reading by Healthy Speakers (구강기류 분석에 근거한 정상 성인의 문단 읽기 시 호흡그룹의 특징)

  • Breath group generally refers to one of units of speech production. It is an integral component of structural and contextual features of utterances with some responsibility for fluctuations in speech intelligibility. The purpose of this study was to know the characteristics of breath group in reading passages spoken by healthy speakers, specifically in view of aerodynamic aspects. Eighteen female speakers aged from 20 to 30 years old without communication problems and in healthy condition were participated in this study. PAS (Phonatory Aerodynamic System) was used for aerodynamic measurement of breath group. Results showed that the mean value of breath group in reading tasks was 16.03 per minute (SD=3.1), and the spoken syllables per one breath group were 17.95. And the mean time (m) of breath group was 3.06 (SD=0.62), and the ratio of exhalation and inhalation was appeared in the 1:5. The results need to be discussed in values of normality of breath group and clinical viewpoint, especially their potential implication from speech intelligibility caused by brain damage.
