Effect on the Growth of Pllioblastus pygmaed and Soil Characteristics as Affected by Difference of Soil Thickness and Soil Mixture Ratio in the Shallow-Extensive Green Roof Module System (저관리 옥상녹화 모듈에서 토심, 배합비의 차이가 토양의 특성 및 흰줄무늬사사의 생육에 미치는 영향)

  • The objectives of this study were to compare growth of Pllioblastus pygmaed and soil characteristics as affected by difference of soil thickness and mixture ratio in shallow-extensive green roof module system, and to identify the level of soil thickness and mixture as suitable growing condition to achieve the desired plants in green roof. Different soil thickness levels were achieved under 15cm and 25cm of shallow-extensive green roof module system that was made by woody materials for $500{\times}500{\times}300mm$. Soil mixture ratio were three types for perlit: peatmoss: leafmold=6:2:2(v/v/v, $P_6P_2L_2$), perlit: peatmoss: leafmold=5:3:2(v/v/v, $P_5P_3L_2$) and perlit: peatmoss: leafmold=4:4:2(v/v/v, $P_4P_4L_2$). On June 2006, Pllioblastus pygmaed were planted directly in a green roof module system in rows. All treatment were arranged in a randomized complete block design with three replication. The results are summarized below. In term of soil characteristics, Soil acidity and electric conductivity was measured in pH 6.0~6.6 and 0.12dS/m~0.19dS/m, respectively. Organic matter and exchangeable cations desorption fell in the order: $P_4P_4L_2$ > $P_5P_3L_2$ > $P_6P_2L_2$. $P_6P_2L_2$ had higher levels of the total solid phase and liquid phase, and $P_4P_4L_2$ had gas phase for three phases of soil in the 15cm and 25cm soil thickness. Although Pllioblastus pygmaed was possibled soil thickness 15cm, there was a trend towards increased soil thickness with increased leaf length, number of leaves and chlorophyll contents in 25cm. The growth response of Pllioblastus pygmaed had fine and sustain condition in order to $P_6P_2L_2$ = $P_5P_3L_2$ > $P_4P_4L_2$. However, The results of this study suggested that plants grown under $P_4P_4L_2$ appear a higher density ground covering than plants grown under $P_6P_2L_2$. Collectively, our data emphasize that soil thickness for growth of Pllioblastus pygmaed were greater than soil mixture ratio in shallow-extensive green roof module system.

  • Seasonal variations of the contents of some chemical constituents of the estuary water at two definite stations of the laver bed in Nack Dong River have been determined over one tidal cycle in spring tide from Nov. 1962 to Oct. 1963. The ranges of annual variations of the contents at station 1 and station 2 are as follows: water temp. $2.2-30.8^{\circ}C$, $3.3-28.0^{\circ}C$; pH 7.8-8.5, 7.9-8.4; chlorosity 0.025-19.66 g/l, 4.31-19.56 g/l; magnesium 0.00355-1.565 g/l, -1.524 g/l; calcium 0.00557-0.482 g/l, - -0.590 g/l; saturation % of dissolved oxygen 71.8-123.2%, 88.2-113.8%; silicate-Si 8.00-125.5 ${\mu}$g-at./l, 6.70-100.5 ${\mu}$g-at./l; phosphate-P 0.12-1.47 ${\mu}$g-at./l, 0.11-1.09 ${\mu}$g-at./l; ammonia-N 4.88-25.45 ${\mu}$g-at./l, 4.12-17.58 ${\mu}$g-at./l; nitrite-N 0.07-0.75 ${\mu}$g-at./l, 0.08-0.58 ${\mu}$g-at./l; nitrate-N 2.11-6.89 ${\mu}$g-at./l, 1.85-7.43 ${\mu}$g-at./l each. The annual tidal variations of the constituents at station 1 are more remarkable than of station 2. The chlorosity, magnesium and calcium contents are decreased nearing the slack after ebb, and increased abruptly then one hour after the slack. The contents of the other constituents are varied according to the chlorosity variety. The values of pH, chlorosity, magnesium and calcium contents are lower in summer than winter, while the difference of seasonal variations of the % saturation of dissolved oxygen is not remarkable. The phosphate-P and total nitrogen contents have a tendency of increasing within a definite range, while the silicate-Si increase proportionally, to the increasing of mixing percentage of fresh water. The average values of Si/P and N/P are several times greater than of the normal in sea water. The chemical composition considered from the value of Mg/Cl or Ca/Cl of estuarine water varies according to the variety of chlorosity, even at the high chlorosity of 19 g/l.

Studies on Antioxidative Effect and Lactic Acid Bacteria Growth of Persimmon Leaf Extracts (감잎 추출물의 유산균 생육 및 항산화 효과에 관한 연구)

  • This study was investigated the growth effect of persimmon leaf extracts on various Lactic acid bacteria, the effect as freeze-dry cryoprotectan, by the changes of vitamin C content, electron donating ability, total cell count and SOD-like activity. The total cell count of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 in the absence of persimmon leaf extracts(10%) at 37℃ after 48hr were 3.2×10/sup 9/cfu/mL, 2.9×10/sup 9/cfu/mL, 1.1×10/sup 9/cfu/mL, 1.6×10/sup 9/cfu/mL and 3×10/sup 9/cfu/mL, respectively, and pH and total acidity of those were 3.82∼3.88, and 1.697∼1.842. On the other hand, the total cell count of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 in the presence of persimmon leaf extracts at 37℃ after 48hr were 4.4×10/sup 9/cfu/mL, 4.3×10/sup 9/cfu/mL, 2×10/sup 9/cfu/mL, 3.3×10/sup 9/cfu/mL, and 3.4×10/sup 9/cfu/mL, respectively, and pH and total acidity of those were 3.74∼3.82 and 1.528∼1.805. The total cell counter of lactic acid bacteria in the presence of persimmon leaf extracts(10%) at 37℃ after 48hr higher than those in the absence of persimmon leaf extracts. In freeze-dry chryoprotectan, the survival rate of L. acidphilus MG501 decreased about 30%. In case of L. brevis MG19 and L. bulgaricus MG515, the survival rate decreased about 10%. However, the survival rate of L. lactis MG530 increased about 10% and L. casei MG311 showed the almost same effect. The changes of vitamin C content were the highest in L. lactis MG530(190.26mg/mL) and the lowest in L. acidophilus MG501(56.05 mg/mL). The electron donating ability indicated to L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 added by 10% persimmon leaf extracts, respectively. when 10% persimmon leaf extracts were added L. brevis MG19, the electron donating ability is the highest of all lactic acid bacteria. The SOD-like activity of L. acidophilus MG501, L. brevis MG19, L. bulgaricus MG515, L. lactis MG530 and L. casei MG311 showed lower than that of control.

  • We have selected an valuable pmbiotic strain; Lactobacillus sakei BK19 which has wide antagonic spectrum against fish pathogens . Present study investigated cultural characterization of L. sakei BK19 including pH tolerance , susceptibility of antibacterial agents and growth pattern with different environment such as nutritions, temperature and salinity. L. sakei BK19 showed Significantly higher resistance at low pH(around pH 4) environment and relative high antibiotic tolerance . In the study of optimal culture condition, maltose and saccharose provided the optimal nutritional culture condition while lactose and mannitol were unable to supply its carbon source for the fermentation of L. sakei BK19. Moreover. L. sakei BK19 showed good growth at the temperature range of 15 to $45^{o}C$ und the NaCl concentration of 0 to 7%. Hence, this particular probiotic strain may be benificial both in seawater and fresh weter conditions.

  • A dynamic model was applied to compare the environmental parameters and effluent characteristics of refinery wastewater in activated sludge treatment. The measured DO, pH, MLSS, and COD values were compared with the simulated their results in the activated sludge reactor. As results of simulation. good agreements between the measured and simulated results were investigated. The simulated and measured effluent COD were 19.0~19.4mg/L and 23.1mg/L. respectively. The simulated and measured DO were 1.9~2.4mg/L and 2.2mg/L. respectively.

Immunogenicity of the ChAdOx1 nCoV-19 vaccine in patients with hematologic malignancies

  • Purpose: The present study aimed to study the immunogenicity of the ChAdOx1 nCoV-19 vaccine in patients with hematologic malignancies. Materials and Methods: This prospective cohort study of hematology patients aimed to evaluate their antibody levels against the receptor-binding domain of the severe acute respiratory syndrome coronavirus 2 spike protein and seroconversion rates following two doses of the ChAdOx1 nCoV-19 vaccine. Between June and July 2021, we enrolled 61 patients and included 44 patients in our analysis. Antibody levels were assessed 8 and 4 weeks after the first and second injections, respectively, and compared with those of a healthy group. Results: Eight weeks after the first dose, the geometric mean antibody level was 1.02 binding antibody units (BAU)/mL in the patient group and 37.91 BAU/mL in the healthy volunteer group (p<0.01). Four weeks after the second dose, the geometric mean antibody level was 9.44 BAU/mL in patients and 641.6 BAU/mL in healthy volunteers (p<0.01). The seroconversion rates 8 weeks after the first dose were 27.27% and 98.86% in the patient and healthy volunteer groups, respectively (p<0.001). The seroconversion rate 4 weeks after the second dose was 47.73% in patients and 100% in healthy volunteers. Factors leading to lower seroconversion rates were rituximab therapy (p=0.002), steroid therapy (p<0.001), and ongoing chemotherapy (p=0.048). Factors that decreased antibody levels were hematologic cancer (p<0.001), ongoing chemotherapy (p=0.004), rituximab (p<0.001), steroid use (p<0.001), and absolute lymphocyte count <1,000/mm3 (p=0.009). Conclusion: Immune responses were impaired in individuals with hematologic malignancies, particularly patients undergoing ongoing therapy and B-cell-depleting therapy. Additional vaccinations should be considered for these patients, and further investigated.

Heterocyclic Amines Removal by Binding Ability of Lactic Acid Bacteria Isolated from Soybean Paste (된장에서 분리된 유산균의 결합력에 의한 Heterocyclic Amines 제거)

  • The objective of the this study was to investigate the binding capacity and removal ability of lactic acid bacterial strains obtained from Korean soybean paste for mutagenic heterocyclic amines (HCAs) formed during cooking of protein-rich food at high temperature. Among 19 strains identified by carbohydrate fermentation and 16S rRNA sequencing, the live cell or cell-free culture supernatant of Lactobacillus acidophilus D11, Enterococcus faecium D12, Pediococcus acidilactici D19, L. acidophilus D38, Lactobacillus sakei D44, Enterococcus faecalis D66, and Lactobacillus plantarum D70 inhibited the mutagenesis caused by either 3-amino-1,4-dimethyl-5H-pyrido[4,3-b] indole (Trp-P-1) or 3-amino-1-methyl-5H-pyrido[4,3-b] indole (Trp-P-2) in Salmonella typhimurium TA98 and TA100. The bacterial cells of the isolated strains showed greater binding activity than the pure cell wall, exopolysaccharide, and pepetidoglycan. The carbohydrate moieties of the cell wall or protein molecules on the cell surface have a significant role in binding Trp-P-1 and Trp-P-2, since protease, heating, sodium metaperiodate, or acidic pH treatments significantly (P<0.05) reduced the binding efficacy of the tested bacteria. Addition of metal ions or sodium dodecyl sulfate decreased the binding ability of E. faecium D12, L. acidophilus D38, and E. faecalis D66. Therefore, the binding mechanisms of these strains may consist of ion-exchange and hydrophobic bonds. Especially, the high mutagen binding by L. acidophilus D38 and L. plantarum D70 may reduce the accumulation or absorption of Trp-P-1 and Trp-P-2 in the small intestine via increased excretion of a mutagen-bacteria complex.

Increasing Cell Concentration by the Automatic Addition of Glucose, Ammonium and Phosphate in the Cultivation of a Baker′s Yeast in Alcohol Distillery Wastewater (알콜증류폐액을 이용한 빵효모배양에서 포도당, 암모늄 및 인산의 자동첨가에 의한 증균)

  • Automatic addition of glucose, ammonium and phosphate to alcohol distillery wastewater and their control at low concentrations have been carried increase the cell concentration of a baker's yeast cultivated in the wastewater. Glucose was automatically added using dissolved oxygen as the control parameter, and maintained below 300 mg/L. Ammonium was automatically added by a pH-stat method and maintained in the low range of 12.6~17.4 mM. An automated FIA system, which used an ascorbic acid-based method was developed for the automatic analysis nad addition of phosphate. With this system, the phosphate concentration was succesfully analysed and controlled afrer 19.4 hr in the range 23.3~43.4 mg/L. The cell concentration was increased by 33.0-fold by the addition of these three nutrients. The overall specific growth rate of the yeast was 0.19 $hr^{-1}$.

Isolation, Identification and Physiological Characteristics of Biofertilizer Resources, Insoluble Phosphate-Solubilizing Bacteria (미생물비료 생물자원인 불용성인산 가용화 세균의 분리, 동정 및 생리적 특성)

  • To develop environment-friendly biofertilizer solubilizing insoluble phosphates, a bacterium possessing a high ability to solubilize $Ca_{3}(PO_{4})_{2}$) was isolated from the rhizosphere of peas. On the basis of its morphological, cultural, physiological characteristics, and Vitek analysis, this bacterium was identified as Pantoea agglomerans. The optimal medium composition and cultural conditions for the solubilization of insoluble phosphate by P. agglomerans R-38 were 3% of glucose.0.1% of TEX>$NH_{4}NO_{3}$, 0.02% of $MgSO_{4}\cdot\7H_{2}O$, and 0.06% of $CaCl_{2}\cdot\2H_{2}O$ along with initial pH 7.5 at $30^{\circ}C$. The highest soluble phosphate production under optimum condition was 898 mg/L after 5 days of cultivation. The solubilization of insoluble phosphate was associated with a drop in the pH of the culture medium. The strain produced soluble phosphate to the culture broth with the concentrations of 698 mg/L against CaHPO$_4$, 912 mg/L against hydroxyapatite, 28 mg/L against $FePO_{4}\cdot\4H_{2}O$, and 19 mg/L against $AIPO_{4}$, respectively.

Evaluation on Adaptation of Zosia japonica as Effected by Different Green Roof System under Rainfed Conditon (무관수 옥상녹화시스템의 차이에 따른 들잔디 적응성 평가)

  • This study proposes a guideline of a green roof system suitable for the local environment by verifying the growth of Zoysia japonica in a shallow, extensive, green roof system under rainfed condition. The experimental soil substrates into which excellent drought tolerance and creeping Z. japonica was planted were made with different soil thicknesses(15cm, 25cm) and soil mixing ratios(SL, $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$, $P_4P_4L_2$). The plant height, green coverage ratio, fresh weight, dry weight and chlorophyll contents of Z. japonica were investigated. For the soil thickness of 15cm, the plant height of Z. japonica was significantly as affected by the soil mixing ratio and it was shown in the order SL= $P_4P_4L_2$ < $P_7P_1L_2$ = $P_5P_3L_2$ < $P_6P_2L_2$. For the soil thickness of 25cm, the plant height was increased in order to SL < $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$ < $P_4P_4L_2$. The green coverage ratio was not observed by soil the mixing ratio or soil thickness. However, the green coverage ratio was 86~90% with a good coverage rate overall. The chlorophyll contents of Z. japonica were not significantly affected by the soil mixing ratio in the soil thickness of 15cm, but were higher in the natural soil than in the artificial soil at 25cm soil thickness. The fresh weight and dry weight of Zoysia japonica were heavier in the 25cm thickness than in the 15cm thickness and in the artificial soil mixture than in the natural soil. The result indicated that the growth of Zoysia japonica was more effective in the 25cm soil thickness with artificial soil than in the 15cm soil thickness with natural soil in the green roof system under rainfed condition.
