  • 1. Objectives The purpose of this study was to investigate the effects of the enzyme activity of Palmulgunja-tang with administered orally solution on cytochrome P450 isozyme 2. Methods This study was carried on through following methods. We treated the rat with the {3-naphthoflavone (${\beta}NF$) of 80mg/kg for 3 days i.p injection. Firstable, microsomal protein was separated and total intracellular protein test was done. Then GOT and GPT were measured and assay of cytochrome P450 IAI/2 enzyme activity was performed according to the method of EROD and MROD. (Ethoxyresorufin-O-deethylase(EROD) activity was used to measure cytochrome P450 lAI activity and methoxyresorufin O-demethylase(MROD) activity was used to measure cytochrome P450 lA2 activity. ) 3. Results and Conclusions 1) PGT recovered the liver damage on ${\beta}NF$ inducible CYP IAI/2 by pre-post and high-low condition. 2) At concentration of post-treated 50mg!kg of PGT, the inhibiting of $\betaNF$ metabolites to liver of rat cytochrome P450 lAl was inhibited by 53.0% respectively. 3) PGT showed 36.0% inhibition of ${\beta}NF$-induced lA2 activity at the concentration post-treated 50mg/kg.

Influence of Five Herbal Medicines on Cytochrome P450 3A4 Drug-Metabolizing Enzymes in Human Liver Microsomes (수종 지혈 한약물이 Cytochrome P450 3A4 활성에 미치는 영향)

  • Objects : The aim of this study was to investigate the influence of five herbal medicines on cytochrome P450 3A4 drug-metabolizing enzymes in human liver microsomes. Methods : To use human liver microsomes, an extract of five herbal medicines, which are Artemisia princeps Pampan, Sophora jeponica Linne, Panax notoginseng F. H. Chen, Lithospermum Erythrorhizon Sieb., and Cirsium maackii Maxim, which together are called Jihyulyak(止血藥, drugs for arresting bleeding, hemostatics), was co-incubated and measured for relative enzyme activity in incubation condition compared to ketoconazole, a representative inhibitor of CYP 3A4. Results : We showed that all five of the traditional herbal medicines had no inhibition effect of CYP 3A4 at 10, 20, 30, 40, and $50{\mu}g/ml$ doses in human liver microsomes, although Sophora japonica Linne(SJL) showed a little inhibition at about 81% inhibition rate of control. However, this result is not enough to prove that SJL has a CYP 3A4 inhibition effect. Moreover, we can't make sure that those rates had significant induction effect on CYP 3A4. Conclusions : The result of this study could support that those herbal medicines are safer than chemical drugs, even if this is the basic step to prove that result.

Inhibitory Effect of Licorice Ethanol Extracts and Glycyrrhizin on Cytochrome P450 Drug-Metabolizing Enzymes in Human Liver Microsomes (감초 물 추출물 및 Glycyrrhizin이 인체 간 Microsome에서 Cytochrome P450 약물대사효소에 미치는 영향)

  • Objective : The aim of present study is to evaluate the inhibitory potential of licorice extract and glycyrrhizin on cytochrome P450(CYP) in human liver microsomes. Methods : Using human liver microsomes, water extract of licorice and glycyrrhizin as an inhibitor were co-incubated with each probe drug representing selective CYP isoform activity. We measured relative metabolic activity in incubation condition compared to that with no extract of licorice using HPLC system. Results : Both water extracts of licorice and glycyrrhizin showed inhibitory effect on CYP-catalyzed reactions. CYP2C19 $(IC_{50}=126.7{\mu}g/ml)$ is most potently inhibited by water extract than other tested CYP isoforms$(IC_{50}>450{\mu}g/ml)$, but glycyrrhizin exhibited potent inhibition on CYP1A2$(IC_{50}=106.9{\mu}g/ml)$ followed by CYP2C9 and CYP2D6. Conclusion: These results indicate that water extract of licorice and glycyrrhizin have inhibitory potential on CYP-catalyzed reaction in human liver microsomes. But the mechanism of inhibition was slightly different between them Water extract of licorice mainly inhibited CYP2C19, and glycyrrhizin primarily inhibited CYP1A2. The inhibition by water extract of licorice and glycyrrhizin on CYP isoforms may cause drug interaction with co-administered drug leading to toxicity or treatment failure.

Inhibitory effect of honokiol and magnolol on cytochrome P450 enzyme activities in human liver microsomes

  • Honokiol and magnolol, the major bioactive neolignans of magnolia officinalis, are the most important constituents of the crude drug prescriptions that are used in the therapy of neuroses and various nervous disorders. There have been limited reports on the effects of neolignoid compounds on human cytochrome P450 activity. Therefore, the inhibitory effects of honokiol and magnolol on seven human cytochrome P450 s were evaluated in human liver microsomes. Honokiol and magnolol showed the most potent inhibition of CYP1A2-mediated phenacetin O-deethylase activity ($IC_{50}$ values of 3.5 and 5.4 mM, respectively) among the seven P450s tested. These in vitro data indicate that neolignan compounds can inhibit the activity of CYP1A2 and suggest that these compounds should be examined for potential pharmacokinetic drug interactions in vivo.

Acetone Enhancement of Cumene Hydroperoxide-supported Microsomal Cytochrome P450-dependent Benzo(a)pyrene Hydroxylation

  • In vitro effects of acetone on cytochrome P450 (P450)-dependent benzo(a)pyrene (B(a)P) hydroxylation supported by cumene hydroperoxide (CuOOH) or NADPH/$O_2 $ systems were studied using 3-methylcholanthrene-pretreated rat liver microsomes. The maximal rate of B(a)P hydroxylation at constant concentration ($80\;{\mu}M)$ of the substrate was observed in the presence of $30\;{\mu}M$ CuOOH. However, at concentrations higher than $30\;{\mu}M$ CuOOH the hydroxylation rates were rapidly decreased. In contrast to CuOOH, at a concentration of $200\;{\mu}M$ NADPH, B(a)P hydroxylation rate reached a plateau. At concentrations higher than $200\;{\mu}M$ NADPH, the rates of substrate hydroxylation were maintained at the maximal rate with no inhibition. Acetone at 1% (v/v) enhanced both CuOOH- and NADPH/$O_2$-supported B(a)P hydroxylation at the optimal concentrations of the cofactors. At concentrations higher than 1% (v/v) acetone, substrate hydroxylation was sterero specific under the support of these two cofactors; it was strongly enhanced with $30\;{\mu}M$ CuOOH, but rather inhibited in the $200\;{\mu}M$> NADPH/$0_2 $ system. The lipid peroxidation rate induced during CuOOH-supported P450-dependent B(a)P hydroxylation was increased as CuOOH concentrations were increased. Acetone in the concentration range of 2.5~7.5%(v/v) inhibited lipid peroxidation during CuOOH supported B(a)P hydroxylation. The finding that CuOOH-supported B(a)P hydroxylation is greatly enhanced by acetone suggests that acetone may contribute more to the activation of oxygen (for the insertion of oxygen into the substrate) in the presence of CuOOH than with NADPH/$O_2$. Acetone may also contribute to the partial inhibition of destruction of microsomal membranes by lipid peroxidation.

  • Biotransformation of exogenous (-)-(4R)-isopiperitenone in cell suspension cultures of Mentha piperita resulted in oxidized products, with (-)-7-hydroxyisopiperitenone being the major compound. The mass of products obtained $unde^{18}O_2$, atmosphere was two units higher than that under normal atmosphere. The biotransformation was inhibited by several cytochrome P450-specific inhibitors as well as by carbon monoxide. Carbon monooxide inhibition was substantially overcome by irradiation of cells with blue light including light at 450nm wavelength. These results suggested that a cytochrome P450-type monooxygenase was involved in the biotransformation.

Mechanism of Inhibition of Human Cytochrome P450 1A1 and 1B1 by Piceatannol

  • The resveratrol analogue piceatannol (3,5,3',4'-tetrahydroxy-trans-stilbene) is a polyphenol present in grapes and wine and reported to have anti-carcinogenic activities. To investigate the mechanism of anticarcinogenic activities of piceatannol, the effects on CYP 1 enzymes were determined in Escherichia coli membranes coexpressing recombinant human CYP1A1, CYP1A2 or CYP1B1 with human NADPH-P450 reductase. Piceatannol showed a strong inhibition of CYP1A1 and CYP1B1 in a concentration-dependent manner, and $IC_{50}$ of human CYP1A1 and CYP1B1 was 5.8 ${\mu}M$ and 16.6 ${\mu}M$, respectively. However, piceatannol did not inhibit CYP1A2 activity in the concentration of up to 100 ${\mu}M$. Piceatannol exhibited 3-fold selectivity for CYP1B1 over CYP1A1. The mode of inhibition of piceatannol was non-competitive for CYP1A1 and CYP1B1. The result that piceatannol did not inhibit CYP1B1-mediated $\alpha$-naphthoflavone ($\alpha$-NF) metabolism suggests piceatannol may act as a non-competitive inhibitor as well. In human prostate carcinoma PC-3 cells, piceatannol induces apoptosis and prevents Aktmediated signal pathway. Taken together, abilities of piceatannol to induce apoptotic cell death as well as CYP1 enzyme inhibition make this compound a useful tool for cancer chemoprevention.

Methods: By using of human liver microsomes, we extracted Cnidium officinale Makino, Rhus verniciflua Stokes, Prunus persica Batsch, Corydalis remota Fisch, Carthamus tinctorius Linne, which are called Hwalhyulgeoouhyak(活血祛瘀藥). Then they were incubated and measured for relative enzyme activity under incubation conditions compared to ketoconazole, which is known as a representative inhibitor of CYP 3A4. Results: We showed that all of five traditional herbal medicines had no inhibition effect of CYP 3A4 at 10, 20, 30, 40, and 50${\mu}g/m{\ell}$ doses in human liver microsomes, although Rhus verniciflua Stokes (RVS) showed a little inhibition as about 95% enzyme activity of control. However, this result was not enough to prove that RVS has a CYP 3A4 inhibition effect. Moreover, we can't confirm that those rates have significant induction effect on CYP 3A4. Conclusions: The result of this study could support that those herbal medicines are more reliable than chemical drugs, even if this is a basic step to prove that result.
