Dark Fermentative Hydrogen Production using the Wastewater Generated from Food Waste Recycling Facilities (혐기 발효 공정을 통한 음식물류 폐기물 탈리액으로부터 수소 생산)

  • The authors examined the effects of operating parameters on the $H_2$ production by dark fermentation of the wastewater generated from food waste recycling facilities, in short "food waste wastewater (FWW)". Central composite design based response surface methodology was applied to analyze the effect of initial pH (5.5-8.5) and substrate concentration (2-20 g Carbo. COD/L) on $H_2$ production. The experiment was conducted under mesophilic ($35^{\circ}C$) condition and a heat-treated ($90^{\circ}C$ for 20min)anaerobic digester sludge was used as a seeding source. Although there was a little difference in carbohydrate removal, $H_2$ yield was largely affected by the experimental conditions, from 0.38 to 1.77 mol $H_2$/mol $hexose_{added}$. By applying regression analysis, $H_2$ yield was well fitted based on the coded value to a second order polynomial equation (p = 0.0243): Y = $1.78-0.17X_1+0.30X_2+0.37X_1X_2-0.29X_1{^2}-0.35X_2{^2}$, where $X_1$, $X_2$, and Y are pH, substrate concentration (g Carbo. COD/L), and hydrogen yield (mol $H_2$/mol $hexose_{added}$), respectively. The 2-D response surface clearly showed a high inter-dependency between initial pH and substrate concentration, and the role of these two factors was to control the pH during fermentation. According to the statistical optimization, the optimum condition of initial pH and substrate concentration were 7.0 and 13.4 g Carbo. COD/L, respectively, under which predicted $H_2$ yield was 1.84 mol $H_2$/mol $hexose_{added}$. Microbial analysis using 16S rRNA PCR-DGGE showed that $Clostridium$ sp. such as $Clostridium$ $perfringens$, $Clostridium$ $sticklandii$, and $Clostridium$ $bifermentans$ were main $H_2$-producers.

  • Purpose ; 본 연구는 X-염색체와 관련된 장애 중에서 가장 흔하고 심한 Duchenne Muscular Dystrophy(DMD)의 세포유전학 및 분자유전학적 특성을 설명하기 위해서 DMD에 영향을 받고 있는 두 가계의 13명을 대상으로 가계도 분석과 염색체 분석 및 DNA 분석을 하였다. Method ; DNA분석은 DNA probe을 이용한 Southern blotting method로써 RFLPs와 DMD유전자 부위의 exon소실 유무를 조사하여 아래와 같은 결과를 얻었다. Conclusion ; A 염색체 분석 : 말초혈액과 양수를 표본으로 High-Resolution GTG염색에서 A가계와 B가계의 염색체 분석에서 12명의 염색체는 정상 X-염색체였으나 B가계의 I-2(DMD여성)에서 46, x,-x,+t(2:x)(q 21.1 : p21.2)로 나타난다. B. DNA분석3 : 1) RFLPs의 분석 J66,XJ-1.1,754-11로써 B가계의 RELPs(Restriction Fragment Length Polymorphisms)에서 J66/Pst I은 1.7hb(E), 1.6kb(e)을 보여 주었고 XJ-1.1/Taq I은 3.6kb(F), 3.0kb(f), 754-11/EoR I은 4.2kb(G), 2.0kb(g)의 대립인자를 나타내었다. 이상의 결과를 바탕으로 영향을 받고 있는 남자 (II-2)의 haplotype는 보인자인 어머니의 한쪽 인자를 받았으며 어머니와 딸은 보인자이고 임산부의 태아는 남아였고 태아의 인자들은 그의 할아버지로부터 물려받아 DMD에 영향을 받지 않은 것으로 진단되었다. 2) DMD 유전자의 exon 소실에 대한 분석 cDNA probe 8과 cDNA probe 2b-3으로써 소실에 대한 진단은 영향을 받은 남자(II-2)는 cDNA probe 8에서 12, 7.3, 6.6, 4.2kb에 소실이 있고 cDNA 2b-3은 1.7kb에 소실에 나타났다.

The Relationship Between Body Composition and Bone Mineral Density in College Women (여대생의 신체조성과 골밀도의 관계)

  • Purpose: To investigate body compositons and bone mineral density(BMD) in college women and to find the relationship between them. Method: From January to March of 2001, BMD at four parts(forearm, lumbar, femur and whole body), body mass index(BMI), body fat mass(BFM), lean body mass(LBM) and body fat percentage(%Fat) were measured with the Dual Energy X-ray Absorptiometry. Other physical characteristics were measured with a scale, a height measurer, and questionnaires. Result: Grouping by the BMI, 43.2% showed low weight, and 5% over weight. When applying the percent Fat, 43.8 % was diagnosed as obesity group. The fact indicate that a majority of college women have unbalanced body composition with high percent Fat, compared to their body weight. Assessing the BMD with the WHO standards, 91.4~95.7% of the BMD of forearm and whole body was normal. But, 40.3% and 33.1~43.9% showed osteopenia at lumbar and femur, and 104 %, 0.7~7.2% showed osteoporosis. The BMD at all parts showed significant correlation each other(r=.29~.89, p=.001~.000). Body weight and BMI showed correlations to with BMDs at all parts of the body(r=.19~46, p=.025~.000; r=18~.45, p=.039~.000). But the percent Fat had a correlation with only femur neck BMD(r=.19, p=.024). Conclusion: This study showed a majority of healthy college women were exposed to the risk for osteoporosis. Additional study is required to develop nursing interventions to remove the risk factors of osteoporosis. In particular, the acquisition of balanced body composition is necessary, increasing body weight and BMI through the increase of LBM, not through the quantitative increase of BFM.

  • In this parer, by means of adding nonsuperconductive phase sliver into YBCO matrix, the superconductivity of Melted Texture Growth (MTG) YBa$_2$Cu$_3$O$\_$7-x/ was improved remarkably. In order to eliminate the crack inthe YBCO and the weak linkin the grain boundary, Ag contents from 2wt% to 18Wt% were doped in the YBCO It was found that J$\_$c/ of YBCO increase with the increasing Ag content till 14 wt% over 14wt% of Ag content, the Jc tends to stable . The grain size of YBCO became fine when Ag was added in the YBCO and X-ray diffraction showed that the YBCO crystal prepared by the above technique had (001) orientation and growing plane of YBCO was a-b plane. Using Bens medel, the J$\_$c/ was calculated and the best result was J$\_$c/ 76000A$\textrm{cm}^2$(77K, 100Gs).

  • For efficient and economical production of Btrc,illus tlz~rr.ingi~r1~sstirsa in NT0423 as amicrobial-control agents, a new culture medium and culture condition were developed. Prediction of Thermal Diffusivities of Pork Meat Products (축육 가공품의 열확산도 추정에 관한 연구)

  • To obtain a thermal diffusivity predicting equation for meat products, minced pork meat was mixed with some additives such as lard, isolated soybean protein, 1.5% of table salt and 2% of polyphosphate to control the composition and texture of the products and then stuffed in a model can. Heat penetration curves were measured in the temperature range of 80.76~121.03$^{\circ}C$ by using a thermocouple fixed at the cold point of the model can and the thermal diffusivities were calculated from the plotted heat penetration curves. At constant heating temperature, the thermal diffusivities of pork meat with water content of 49.01~77.55% increased linearly with increasing water content. The thermal diffusivities of the products with constant water content also increased linearly with increasing heating temperature and the values could be predicted by following equation: $\alpha$p=(2.1394+0.5X$_{w}$).$\alpha$$_{w}$+0.0035.10$^{-6}$ .X$_{w}$-0.2785.10$^{-6}$ ,(m$^2$.s$^{-1}$ ). The maximal difference of the values predicted with this equation on the basis of the practical measured values were less than 1.7%. 1.7%.

Effects of Compost Application on Soil Loss and Physico-Chemical Properties in Lysimeters (퇴비시용(堆肥施用)이 토양유실량(土壤流失量)과 토양이화학성(土壤理化學性)에 미치는 영향)

  • This study was conducted to investigate the effects of compost application on the soil loss and physico-chemical properties at the slope land from 1990 to 1991. Lysimeters with 15% slope. 5m slope length. 2m width and 1.2m depth were constructed in 1984 and filled with sandy loam, loam, clay loam. and clay soils. Treatments were bare soil, compost(1.5ton/10a) and non-compost with soybean-barley cropping system from 1984. Losses of soil were increased with the order of sandy loam, clay, loam and clay loam. Run-off was increased with the order of sandy loam, loam, clay loam and clay, but leachate was decreased with the same order. Compost treatments decreased 33.6-44.6% of soil loss and 17.0-24.0% of run-off but increased 17.1-33.7.% of leachate as compared with the non-compost treatments. The amount of soil loss was positively correlated with the amount of run-off by Y = 12.125+0.063X (r=$0.970^{**}$)and negatively correlated with the leachate by Y=43.425-0.096X(r=$-0.917^{**}$). The application of compost increased soil pH, OM, CEC and extractable cations. Application of compost decreased bulk density but increased porosity, water stable aggregate and available water. These results provide that the compost application plays an important role in conserving soil and water, and improving soil physico-chemical properties.

Rapid Detection of Salmonella spp. in Fresh-Cut Cabbage by Real-Time PCR (Real-Time PCR을 이용한 신선편이 양배추에서 Salmonella spp.의 신속검출)

  • This study was conducted to find out the minimal time needed for detection of Salmonella spp. which exist at very low concentration in foods by using real-time PCR. The sal-F and sal-R sequences were used as primers and sal-P was used as a probe. The detection limit of Salmonella spp. was $3.77{\times}10^2\;cfu/mL$ in buffered peptone water (BPW). Microbial growth was monitored after artificially inoculated Salmonella spp. into BPW. The obtained growth curve was well fitted with the equation, y=$0.0127x^2$+0.5927x-0.4317 ($R^2$=0.99), if assuming that 1 cell exists in 25 g sample (0.04 cfu/mL). The microbial concentration will be reduced to 10 fold by adding BPW during sample treatment, so actual initial concentration at the starting point of enrichment is 0.004 cfu/mL. At this condition, real-time PCR detection would be possible only when microbial concentration increase occurs to exceed the detection limit (377 cfu/mL). The time needed for microbial increase was calculated from the growth curve equation as 7 hours and 20 minutes. Therefore the total time required for detection was less than 10 hours including the PCR operating time.

Synthesis and Rietveld Structure Refinement of Mn-Tourmalines (Tsilaisite) (Mn-전기석(Tsilaisite)의 합성 및 리트벨트 구조분석)

  • Synthetic Mn-tourmalines (tsilaisite) were obtained by hydrothermal synthesis under the condition of 2 Kbar, $375{\sim}700^{\circ}C$, and 50 day-run-time with complete substitution of Mg in dravite by Mn (Mn%=0, 25, 50, 75, and 100%). They are all 6 samples containing Mn-tourmaline with some amounts of albite, spessartine, rhodocrosite, phlogopite etc, showing different synthetic condition of temperature and Mn composition. Synthetic Mn-tourmalines are of site deficiency in X-site ($0.53{\sim}0.68$) more than that of natural ones (approx. $0.2{\sim}0.3$) and show Mn cations occupying Y-site less than expected with initial experiments, leading to failure in synthesis of end-member tsilaisite. Rietveld structural refinements reveal that $R_{wp}$ ($R_{p}/R_{exp}$) is in the range of 13.35 and 18.62%, $R_{B}$ and S (CofF) are $4.85{\sim}6.25%$ (S-18: 8.57%), $1.31{\sim}1.59$ (S-18: 1.81), respectively. Unit cell parameters (space group R3m, z=3) are ${\alpha}=15.8994\;{\AA}$ and $c=7.1846\;{\AA}$ in average (S-18: ${\alpha}=15.9491\;{\AA},\;c=7.1773\;{\AA}$). Average bond lengths of and are $2.67{\sim}2.69\;{\AA}$ (S-18: $2.65\;{\AA}$) and $2.00{\sim}2.02\;{\AA}$ (S-18: $1.96\;{\AA}$), respectively. Ditrigonality (${\delta}$) are in the range of 0.022 and 0.031 (S-18: 0.061), indicating degrading symmetry with increase of Mn content.

Optimization of Hyaluronidase Inhibition Activity from Prunus davidiana (Carriere) Franch Fruit Extract Fermented by its Isolated Bacillus subtilis Strain SPF4211

  • Strain SPF4211, having hyaluronidase (HAase) inhibition activity, was isolated from P. davidiana (Carriere) Franch fruit (PrDF) sugar extract. The phenotypic and biochemical properties based on 16S rDNA sequencing and an API 50 CHB kit suggested that the organism was B. subtilis. To optimize the HAase inhibition activity of PrDF extract by fermentation of strain SPF4211, a central composite design (CCD) was introduced based on three variables: concentration of PrDF extract (X1: 1-5%), amount of starter culture (X2: 1-5%), and fermentation time (X3: 0-7 days). The experimental data were fitted with quadratic regression equations, and the accuracy of the equations was analyzed by ANOVA. The statistical model predicted the highest HAase inhibition activity of 37.936% under the optimal conditions of X1 = 1%, X2 = 2.53%, and X3 = 7 days. The optimized conditions were validated by observation of an actual HAase inhibition activity of 38.367% from extract of PrDF fermented by SPF4211. These results agree well with the predicted model value.
