Effects of Essential Oils Supplementation on Growth Performance, IgG Concentration and Fecal Noxious Gas Concentration of Weaned Pigs

  • Ninety six crossbred pigs (Landrace${\times}$Yorkshire${\times}$Duroc) were used to determine the effects of essential oils (Fresta F $Conc^{(R)}$) supplementation on growth performance, immune response and fecal noxious gas of weaned pigs. Treatments were 1) NC (negative control; basal diet without antibiotics), 2) PC [positive control; basal diet+CSP (CTC+Sulfathiazole+Penicillin) 0.1%], 3) NCF (basal diet+Fresta F $Conc^{(R)}$ 0.03%) and 4) PCF [basal diet+CSP (CTC+Sulfathiazole+Penicillin) 0.1%+Fresta F $Conc^{(R)}$ 0.02%]. From d 0 to 14, ADFI was increased in pigs fed PCF diet (p<0.05). From d 14 to 28, pigs fed PCF diet had greater ADG and ADFI than pigs fed NC diet (p<0.05). From d 28 to 49, ADG and ADFI in pigs fed PCF diet were higher than in pigs fed NC diet (p<0.05). Through the entire experimental period, ADG and ADFI in pigs fed PCF diet were the highest compared to pigs fed NC and PC diets (p<0.05). There was no significant difference in fecal consistency score among the treatments (p>0.05). No statistical differences (p>0.05) were found in red blood cells (RBC) counts, white blood cells (WBC) counts, lymphocyte counts, total protein and albumin. Serum IgG concentration of PCF treatment was greater than that of other treatments (p<0.05). From d 0 to 14, there was no significant difference in digestibility of dry matter and nitrogen among the treatments (p>0.05). From d 14 to 28, digestibility of dry matter in pigs fed PC, NCF and PCF diets was higher than that of pigs fed NC diet (p<0.05) and treatments with added essential oils were higher than other diets on digestibility of nitrogen (p<0.05). Also, from d 28 to 49, digestibility of nitrogen in pigs fed PCF diet was the highest among others (p<0.05). On d 14 and 28, no statistical differences (p>0.05) were found in volatile fatty acid (VFA), ammonia nitrogen ($NH_3$-N) and hydrogen sulfide ($H_2S$) concentrations among treatments. On d 49, there was no significant difference in VFA concentration among the treatments (p>0.05). $NH_3$-N concentration in pigs fed PCF diet was lower than in pigs fed other diets (p<0.05). $H_2S$ concentration in pigs fed diets with added essential oils was lower than others. In conclusion, the results suggest that the dietary addition of essential oils and antibiotics into diets for weanling pigs improved growth performance, IgG concentration and nitrogen digestibility and decreased noxious gas concentration. Essential oils can be used to partly replace antibiotics in diets for weaned pigs without negative affects on growth performance.

Anti-Inflammatory and PPAR Transactivational Effects of Oleanane-Type Triterpenoid Saponins from the Roots of Pulsatilla koreana

  • In this study, 23 oleanane-type triterpenoid saponins were isolated from a methanol extract of the roots of Pulsatilla koreana. The NF-${\kappa}B$ inhibitory activity of the isolated compounds was measured in $TNF{\alpha}$-treated HepG2 cells using a luciferase reporter system. Compounds 19-23 inhibited $TNF{\alpha}$-stimulated NF-${\kappa}B$ activation in a dose-dependent manner, with $IC_{50}$ values ranging from $0.75-8.30{\mu}M$. Compounds 19 and 20 also inhibited the $TNF{\alpha}$-induced expression of iNOS and ICAM-1 mRNA. Moreover, effect of the isolated compounds on PPARs transcriptional activity was assessed. Compounds 7-11 and 19-23 activated PPARs the transcriptional activity significantly in a dose-dependent manner, with $EC_{50}$ values ranging from $0.9-10.8{\mu}M$. These results suggest the presence of potent anti-inflammatory components in P. koreana, and will facilitate the development of novel anti-inflammatory agents.

Continuous Ethanol Production Using immobilized Baker's Yeast (고정화 효모를 이용한 연속적 에탄올 생산)

  • - Ethanol production by calcium alginate-immobilized baker's yeast was studied in the continuous shaked-flask reactor (CSFR) using glucose medium as a feed. Immobilized cells were stable at 30~$37^{\circ}C$ and pH 4~8. Fermentation characteristics of immobilized baker's yeast were examined changing the initial glucose concentration employed were 50, 100 and 150 g/l, respectively. It was investigated that the influent glucose concentration and the dilution rate have an influence on the ethanol fermentation characteristics at steady state in continuous culture of immobilized baker's yeast. The optimum conditions for high ethanol productivity and low residual glucose output in ethanol prodution were shown to be 0.2 h ' for the dilution rate and 150 g/l for the influent glucose concentration. The maximum ethanol productivity, ethanol yield, specific growth rate and glucose conversion rate were around 7.12 g/$l\cdot h$, 0.23, 0.366 g/$l\cdot h$ and 78.43, respectively.

  • $CdS_{0.69}Se_{0.31}$ single crystal grown by sublimation method. Hall effect measurement were carried out by the Van der Pauw method. The measurement values under the temperature were found to be carrier density $n=1.95{\times}10^{23}m^{-3}$, Hall coeffcient $RH=3.21{\times}10^{-5}m^3/c$, conductivity ${\sigma}=362.41{\Omega}^{-1}m^{-1}$, and Hall mobility ${\mu}=1.16{\times}10^{-2}m^2/v.s.$ Heterojunction solar cells of $n-CdS_{0.69}Se_{0.31}/p-Cu_{2-x}S_{0.69}Se_{0.31}$ were fabricated by the substitution reaction. The open-circuit voltage, short-circuit currint density, fill factor and power conversion efficiency of $n-CdS_{0.69}Se_{0.31}/p-Cu_{2-x}S_{0.69}Se_{0.31}$ heterojunction solar cell under $80mW/cm^2$ illumination were found to be 0.41V, $19.5mA/cm^2$, 0.75 and 9.99%, respectivity.

Effect of indole acetic acid on the contents of various phosphate compounds in the growth phase of saccharomyces uvarum (Saccharomyces uvarum의 배양시기에 따른 여러가지 인산화합물의 함량에 미치는 IAA의 효과)

  • In order to interpret the effect of IAA on the phosphate metabolism and biosynthesis of organic compounds, Saccharomyces uvarum were cultured in the media treated with various concnetration of IAA $(10^{-3}M,\;10^{-5}M,\;10^{-7}M)$. Sampling at the beginning and intervals of culture, yeast cells fractionated were traced the contents of inorganic phosphate and organic compounds of various fractions. 1. Growth of Saccharomyces uvarum were enhanced by IAA $(10^{-3}M,\;10^{-5}M)$ and phosphate contents in DNA and RNA fractions treated with IAA were accelerated 2.3 times and 2 times in comparison with those of control. 2. Amounts of poly-P"A" and poly-p"B" were increased but poly-P"C" decreased during the culture. Therefore, it is considered that poly-P"C" play on most important role as a phosphate pool. 3. It is suggested that because phosphate contents in DNA, protein and lipid fractions increased, inorganic phosphates required phosphates required RNA were transferred from phosphates in cytoplasm, because these increased slowly during the culture. 4. Alkali-labile protein were accelerated by IAA and alkali stable protein only were inhibiction were enhanced by IAA while, ethanol : ether soluble fraction was induced by $10^{-7}M$ IAA in comparison with those control.X> IAA in comparison with those control.

  • Objectives : We investigated the effects of Bojungikgi-tang (BJIGT) on P450 cytochrome enzyme and oxidative stress in the cells. Methods : We enrolled the HepG2 hepatocyte cell line to assess MTT assay, flow cytometer, and immunoblotting analysis. Expression of CYP450 was confirmed by immunoblotting analysis in the Huh7 cell line. Results : We determined that BJIKT markdely changed the expression of the CYP2C19, CYP2D6, and CYP2E1. Moreover, BJIKT inhibited the cell toxicity induced by arachidonic acid + iron treatment, as assessed by FACS analysis. BJIKT induced AMPK activation, which increased the phophorylation of ACC. Conclusions : This study verified the effects of BJIKT, on P450, ROS production, mitochondrial damage and AMPK signaling pathway, which might give us the scientific information about the traditional herbal prescription.

  • We optimized dextranase culture conditions by batch fermentation using Lipomyces starkeyi KCTC 17343. Furthermore, dextranase was purified by an ultra-membrane, and then dextran hydrolyzates were characterized. Cell growth and dextranase production varied depending on the initial culture pH and temperature. The conditions of optimal dextranase production were met in a pH range of 4-5 and temperature between $25-30^{\circ}C$. At optimal fermentation conditions, total enzyme activity and specific enzyme activity were about 4.85 IU/ml and 0.79 IU/g cells, respectively. The specific growth rate was examined to be $0.076\;hr^{-1}$. The production of dextranase in culture broth was very stably maintained after mid-log phase of growth. The enzyme hydrolyzed dextran into DP (degree of polymerization) 2 to 8 oligodextran series. Analysis of the composition of hydrolysates suggested that the enzyme produced is an endo-dextranase.

Inductoin of Radioresistance by Overexpression of Glutathione S-Transferase K1 (hGSTK1) in MCF-7 Cells (MCF-7 세포주에서 Glutathione S-Transferase K1 (hGSTK1) 과발현에 의한 방사선 내성의 유도)

  • Purpose : This study was conducted to assess the effects of x-irradiation on the expression of the novel glutathione S-transferase K1 gene. Materials and methods : Human glutathione S-transferase K1 (hGSTK1) DNA was purified and ligated to a pcDNA3.1/Myc-His(+) vector for the overexpression of hGSTK1 gene. MCF-7 cells were transfected with or without the recombinant hGSTK1 gene, and irradiated with 6 MV x-ray. After incubation of 14 days, cell survival was measured and compared. The expression of hGSTK1 and the effect of x-irradiation on hGSTK1 expression were also estimated in MCF-7 cells transfected with or without the hGSTK1 gene by RT-PCR. Results : Following 2 to 12 Gy of x-irradiation, the cell survivals were higher in the MCF-7 cells transfected with the hGSTK1 gene than in those without transfection. Despite the higher cell survival in the hGSTK1-transfected cells, RT-PCR for hGSTK1 mRNA revealed no significant differences according to radiation dose, fractionation, and time after irradiation. Conclusion : The MCF-7 cells transfected with the hGSTK1 gene showed higher cell survival than those without transfection of the gene. The hGSTK1 gene might be associated with the radiosensitivity of MCF-7 cell line and further analysis should be needed.

Epigenetic modification of long interspersed elements-1 in cumulus cells of mature and immature oocytes from patients with polycystic ovary syndrome

  • Objective: The long interspersed elements (LINE-1, L1s) are a group of genetic elements found in large numbers in the human genome that can translate into phenotype by controlling genes. Growing evidence supports the role of epigenetic in polycystic ovary syndrome (PCOS). The purpose of this study is to evaluate the DNA methylation levels in LINE-1 in a tissue-specific manner using cumulus cells from patients with PCOS compared with normal controls. Methods: The study included 19 patients with PCOS and 22 control patients who were undergoing controlled ovarian hyperstimulation. After oocyte retrieval, cumulus cells were extracted. LINE-1 DNA methylation levels were analysed by bisulfite treatment, polymerase chain reaction, and restriction enzyme digestion. The Connection Up- and Down-Regulation Expression Analysis of Microarrays software package was used to compare the gene regulatory functions of intragenic LINE-1. Results: The results showed higher LINE-1 DNA methylation levels in the cumulus cells of mature oocytes in PCOS patients, 79.14 (${\pm}2.66$) vs. 75.40 (${\pm}4.92$); p=0.004, but no difference in the methylation of cumulus cells in immature oocytes between PCOS and control patients, 70.33 (${\pm}4.79$) vs. 67.79 (${\pm}5.17$); p=0.155. However, LINE-1 DNA methylation levels were found to be higher in the cumulus cells of mature oocytes than in those of immature oocytes in both PCOS and control patients. Conclusion: These findings suggest that the epigenetic modification of LINE-1 DNA may play a role in regulating multiple gene expression that affects the pathophysiology and development of mature oocytes in PCOS.

Activation of Intestinal Immune System by an Orally Administered Methanol Extract from Pine Needles (솔잎 메탄올추출물의 마우스 경구투여에 의한 장관면역 활성)

  • When pine (Pinus densiflora) needles were fractionated into cold water (PD-CW), hot water (PD-HW) and methanol extract (PD-M), PD-M showed potent simulating activity (1.19-fold of the saline control) for proliferation of bone marrow cells mediated by Peyer's patch cells in vitro. MeOH extracts were prepared by homogenization, stirring or reflux to identify the method of methanol extraction, and MeOH extract by reflux method showed significantly highest intestinal immune system modulating activity (1.30-fold) in vitro. The intestinal immune system modulating effect of orally administered PD-M fractionated from pine needles also were studied in mice. Analyzing intestinal immune system modulating activity mediated Peyer's patch cells from C3H/He mice which had been fed with PD-M at different doses for 7 days, 1.0 g/kg of BW/day indicated that the bone marrow cells had proliferated (3.65-fold of 3% EtOH administered group). In addition, the amounts of IL-6 in the culture supernatant of Peyer's patch cells at 1.0 g/kg of BW/day were increased (1.13-fold) whereas the production of GM-CSF was not dose dependent. These results indicate that oral administration of PD-M enhances the secretion of hematopoietic growth factors such as GM-CSF and IL-6 from Peyer's patch cells, and these cytokines also act on modulator of bone marrow cell proliferation.
