Stimulator of interferon gene (STING) is induced by various inflammatory agents, such as lipopolysaccharide and microbial pathogens, including virus and bacteria. In this study, we obtained a full-length cDNA of a STING homolog from olive flounder using rapid amplification of cDNA ends PCR technique. The full-length cDNA of Paralichthys olivaceus STING (PoSTING) was 1442 bp in length and contained a 1209-bp open reading frame that translated into 402 amino acids. The theoretical molecular mass of the predicted protein sequence was 45.09 kDa. In the PoSTING protein, three transmembrane domains and the STING superfamily domain were identified as characteristic features. Quantitative real-time PCR revealed that PoSTING expressed in all the tissues analyzed, but showed the highest level in the spleen. Temporal expression analysis examined the significantly upregulated expression of PoSTING mRNA after viral hemorrhagic septicemia virus (VHSV) stimulation. In contrast, no significant changes in the PoSTING expression were detected in Edwardsiella tarda-challenged group compared to the un-injected control. The expression of P. olivaceus type I interferon (PoIFN-I) was also highly upregulated upon VHSV challenge. These results suggest that STING might be involved in the essential immune defense against viral infection together with the activation of IFN-I in olive flounder.
The aim of this study was to evaluate the genetic parameters of growth traits for improvement breeding in olive flounder Paralichthys olivaceus at the 8th generation of selective breeding in April 2021. Growth traits such as total length, body weight and condition factor at 11 months of age were measured for 7,508 individuals with confirmed paternity. Data were analyzed using the restricted maximum likelihood method applied to a multiple traits animal model. The effects of sex and family were significantly different across traits (P<0.05). The heritability values of total length, body weight and condition factor were estimated to be high as 0.479, 0.457, and 0.466, respectively. Correlation analysis between phenotypic and breeding values, indicated that the selection accuracy was 75.9-85.2% for all traits. To increase the selection accuracy for parent fish selection, the sex and pedigree characteristics that affect each trait should be considered. Moreover, further improvement of multiple traits can be achieved if the correlation between each trait is appropriately considered.
The aim of this study was to evaluate genetic parameters of growth traits for improving selective breeding in olive flounder Paralichthys olivaceus produced in April 2019. Since 2004, we have been conducting a selective breeding program to improve growth traits in the olive flounder. The effect of sex and production period showed significant differences in all traits (P<0.05). The heritability of all traits was 0.428-0.520, which is relatively high by measurement month. Therefore, it is considered that individual selection will be more advantageous than family selection. However, to maintain an appropriate degree of inbreeding and genetic diversity for future generations, it is necessary to consider family selection adequately. Results of the correlation analysis between the same traits according to the measurement period indicated that considering production costs such as feed cost, selection at 18-months-old will be advantageous. Olive flounder is a major aquaculture species in Korea, and continuous selective breeding research is essential to improve productivity.

Concerns about foodborne illnesses caused by Kudoa septempunctata are steadily growing, but reports of K. septempunctata in clinical and food specimens related to food poisoning in Korea are limited. This study aimed to genetically identify K. septempunctata in patients with acute diarrhea and in clinical and food samples related to food poisoning caused by sashimi consumption. Both real-time and nested polymerase chain reaction assays were performed to detect K. septempunctata 18S and 28S rDNA genes in the stools of 348 patients with acute diarrhea, 11 samples (6 stool and 5 rectal swab samples) from patients with food poisoning, and 2 raw Paralichthys olivaceus samples collected from a restaurant where a food poisoning incident occurred. K. septempunctata was identified in 5 clinical specimens (4 stools and 1 rectal swab) and 1 P. olivaceus sashimi sample. All detected K. septempunctata were of genotype ST3. This is the first study to identify K. septempunctata in both patients and food samples with epidemiological relevance in Korea, providing evidence that it is a pathogen that causes food poisoning. Also, this is the first study to confirm the presence of K. septempunctata genes in rectal swabs. Despite continuing suspected occurrences of Kudoa foodborne outbreaks, the rate of identification of K. septempunctata is very low. One reason for this is the limitation in obtaining stool and vomit samples for the diagnosis of Kudoa infection. We strongly suggest the inclusion of rectal swabs among the diagnostic specimens for Kudoa food poisoning.
The surge in online seafood consumption has increased parcel delivery, leading to a need to implement effective preservation methods. As the cold chain system is not fully established in Korea, styrofoam boxes and cold packs are commonly used for low-temperature seafood distribution. The impact of cold packs on product preservation depends on the number utilized. Herein, the freshness of vacuum-packed frozen fish fillets (Paralichthys olivaceus and Scomber japonicus) stored at 25±0.5℃ for up to 84 h was measured. Chemical (pH and volatile base nitrogen), microbiological (viable cell count), and physical (odor intensity) properties were assessed using 2 or 4 cold packs in a styrofoam box. Four cold packs yielded lower values, indicating superior freshness, and extended fish freshness by approximately 12 h compared with two cold packs. Therefore, it is recommended to use a minimum of 4 cold packs (-350 g/cold pack) in a styrofoam box for distributing approximately 300 g of frozen fish fillets at room temperature during the summer, considering an average delivery period of 2 days in Korea.
활성산소종(ROS)은 환경 스트레스 및 병원체의 침입에 의한 산화 대사의 자연적인 산물로써 생물에서 생산된다. 산화적 스트레스에 의해 생성되는 superoxide 음이온 및 과산화수소와 같은 ROS는 세포와 조직에 독성을 나타낼 수 있고, 이 과정에 관여하는 superoxide dismutase (SOD)는 중요한 metalloenzyme이다. 최근 연구는 올리브 넙치(Paralichthys olivaceus)에서 SOD의 부분 유전자가 benzo[a]pyrene에 의해 강하게 발현이 유도되고 산화 스트레스 반응의 지표라고 확인하였지만, 바이러스성 감염에 대한 전사적 반응에 대해서는 조사되지 않았다. 본 연구에서는 항바이러스 반응에서 넙치 SOD의 기능을 알아보기 위해 공간 및 시간적 발현 프로파일을 분석하였다. 넙치 SOD 전사체는 정도의 차이는 있지만 다양한 기관에서 보편적으로 발현되었으며, 근육, 간, 뇌에서는 높게 발현되었고, 위와 비장에서는 상대적으로 낮게 발현되었다. VHSV 감염 후 넙치 콩팥에서 SOD 발현은 3시간 이내에 증가하였으며 점차적으로 감소하여 감염 2일째 원래 수준으로 돌아갔다. 검사 조직에 따라 발현이 유도되는 시간의 차이는 있지만 근육, 간, 뇌에서도 콩팥과 유사한 발현양상을 보였으며, 공통적으로 급성적 면역반응에서는 발현이 증가하지만 만성적 면역반응에서는 감소하였다. 이상의 결과들을 종합해 볼 때, 넙치 SOD는 넙치(P. olivaceus)의 면역 방어 시스템에 중요한 역할을 하고 넙치의 산화 스트레스에 대한 보호 효과에 기여할 것으로 기대할 수 있다.
실험에는 Bacillus subtilis BGA, Bacillus cereus var. mycoides ATCC 11778, Micrococcus luteus ATCC 9341을 상법에 따라 제작하여 muller hinton agar(Difco)에 각각 접종한 뒤, 3가지 조건의 pH가 조정된 검사용 평판배지(plate)인 B. subtilis(pH 6.0), B. cereus(pH 6.0), B. subtilis(pH7.2), B. subtilis(pH 8.0), M. luteus(pH 8.0)의 EEC 4-plate변법을 이용하였다. 넙치에 옥시테트라싸이클린(OTC)을 농도 100 mg/kg/day으로 단 1회 강제 경구 투여한 후 1일, 3일, 5일, 10일, 15일, 20일, 25일 30일마다 3마리씩 채취하여 시료의 처리를 각각 달리한 근육직접법, 추출디스크법, 직접디스크법의 3종류 간이스크리닝법으로써 OTC의 잔존유무를 분석하였다 B. subtilis(pH 6.0)배지는 투여 후 1일째 근육직접법에서만 양성반응으로 의심되는 의양성 반응(${\pm}$)을 나타내었을 뿐 30일까지 간이스크리닝법 3종류가 모두 음성 반응을 나타내었다. B. subtilis(7.2), B. subtilis(8.0), M. luteus(8.0)배지는 투여 후 1일째부터 30일까지 간이스크리닝법 3종류가 모두 음성반응을 나타내었다. 그러나 B. cereus(pH 6.0)배지는 1일째부터 15일까지 간이스크리닝법 3종류가 모두 명확한 양성반응, 투여 후 20일째 근육직접법과 직접디스크법은 의양성반응(${\pm}$)을 보였으나 추출디스크법은 분명한 음성반응, 그리고 투여 후 25일째 및 30일째는 시험조작법 모두가 음성반응을 나타내었다. 따라서 근육직접법, 추출디스크법, 직접디스크법의 간이스크리닝법 3종류는 OTC의 양성반응 판정에 커다란 차이가 없었으며, B. cereus는 OTC의 모니터링에 유효한 균주라는 사실을 확인하였다.

To identify and quantify the effects of a combination of dietary $1{\times}10^8CFU/g$ Lactococcus lactis subsp. lactis I2 ($LI_2$) and 0.1% ${\beta}$-glucooligosaccharides (BGO) on the growth and immunity of olive flounder (Paralichthys olivaceus), a feeding experiment was conducted. Flounder ($14{\pm}0.5g$) were divided into two groups and fed control and synbiotic feeds for 8 weeks. Investigations were carried out on growth and feed utilization, innate immunity, serum biochemical parameters, intestinal lactic acid bacterial (LAB) viability, microvillus length, and changes in the expression levels of genes encoding pro-inflammatory cytokines (tumor necrosis factor $[TNF]-{\alpha}$, interleukin $[IL]-1{\beta}$, and IL-6). Results demonstrated the synbiotic diet had significantly better (p < 0.05) responses in terms of weight gain and specific growth rate, three innate immune parameters (respiratory burst, serum lysozyme, and superoxide dismutase), intestinal LAB viability, and the relative $TNF-{\alpha}$ expression level (p < 0.05). Moreover, after challenge with Streptococcus iniae ($1{\times}10^8CFU/ml$), the synbiotically fed group exhibited significantly higher (p < 0.05) protection against streptococcosis, validating the observed changes in immune parameters and induction of the cytokine-encoding gene. Therefore, according to the results of the present study, synbiotic feed ($LI_2+BGO$) increased growth, modulated innate immune parameters and protected olive flounder against streptococcosis.
유기주석화합물인 tributyltin chloride (TBTC), tributyltin oxide (TBTO)와 triphenyltin chloride (TPTC)를 넙치 간장으로 만든 미크로좀에 in vitro적으로 노출시켜서 이들 화합물의 대사에 관여하는 mixed function oxidase (MFO) 중 cytochrome P450 (CYP) 농도와 7-ethoxyresorufin deethylase (EROD) 활성의 변화를 조사하였으며, 또한 in vivo 실험에서는 TPTC를 넙치에게 복강주사(7.5 mg $kg^{-1}$ BW)하여 간장의 MFO (CYP농도, NADPH cytochrome c 환원효소 활성, NADH chtochrome b5 환원 효소 활성, EROD 활성) 반응을 경시적으로 조사하였다. 그 결과, in vitro에서는 TBTC, TBTO 및 TPTC가 모두 CYP 농도와 EROD 활성을 저해하였으며, 저해력은 TPTC가 가장 컸고 이어서 TBTO, TBTC의 순이었다. 유기주석화합물의 노출농도와 노출시간과 비례하면서 저해정도가 커졌으며, 특히 EROD활성의 저해는 노출농도에 크게 의존적이었다. 그리고 in vivo실험에서도 유기주석 화합물은 CYP농도, NADPH cytochrome c 환원효소 활성, NADH cytochrome b5 환원효소 활성, EROD 활성을 억제하였다. EROD 활성은 오염물질에 의한 반응이 민감하고 재현성도 있어 바람직한 측정지표로 이용될 수가 있을 것이다.
This study aimed to examine the effect of external pop-up satellite archival tags (PSATs) attachment and water temperature on the oxygen consumption rate (OCR) of the olive flounder (mean body weight 2281.7 g). The OCRs of fish were measured under conditions of three different water temperature conditions (15, 20, and 25℃) and two different tagging methods [non-tagging, control; bio-logger external attachment with a miniature PSAT (dummy mrPAT), BEA] using a closed flow-through respirometer. The OCRs of fish linearly increased with the increase in water temperature in both the control and BEA (P<0.001); however, the OCRs of BEA were approximately 1.8-1.9 times lower than those of the control at each water temperature (P<0.001). The Q10 values of the control and BEA were the highest in the water temperature range of 15 to 20℃, but sensitivity to water temperature changes was higher in BEA than in the control. The metabolic energy loss rate (MEL) of fish increased with increasing water temperature regardless of external tagging, but the MEL of the control was higher than that of BEA (P<0.001). These results demonstrate that OCR, thermal sensitivity, and energy expenditure are all affected in adult olive flounder with external PSAT attachment.
