16S rDNA sequence에 대한 종특이성 primer를 이용한 중합효소연쇄반응증폭에 의한 Porphyromonas endodontalis의 동정에 관한 연구 (A STUDY ON THE IDENTIFICATION OF Porphyromonas endodontalis BY PCR USING SPECIES SPECIFIC PRIMERS FOR THE 16S rDNA)

  • P. endodontalis which was known to be associated with the infected root canals and periapical lesions is very difficult to detect by culture methods or traditional methods. Detection of bacteria using polymerase chain reaction(PCR) for 16S ribosomal DNA(rDNA) is fast, simple, and accurate with relatively small amount of target cells. 16S rDNA consist of conserved regions those are same to all species, and variable regions which represent species specificity. The 16S rDNA sequences of P. endodontalis and P. gingivalis were aligned and two highly variable regions were selected as a pair of species specific oligonucleotide primers for P. endodontalis. And then the pair of primers for PCR amplification was synthesized to identify P. endodontalis. The sequences of the species specific primers for the 16S rDNA of P. endodontalis were as follows ; sense primer[endo1]: 5'-CTATATTCTTCTTTCTCCGCATGGAGGAGG-3' antisense primer[endo2]: 5'-GCATACCTTCGGTCTCCTCTAGCATAT-3' In this study, for the identification of P. endodontalis without culture from the mixed clinical samples, PCR was done with species specific primers for the 16S rDNA sequences of P. endodontalis. The results were as follows : 1. The species specificity of the primers for the 16S rDNA of P. endodntalis was determined by the PCR methods. About 490bp amplicon which was specific only for P. endodntalis was produced with P. endodontalis. No amplicon was produced by PCR with other strains similar to P. endodontalis. 2. The synthesized species specific primers reacted with conventionally identified P. endodontalis which we have in conservative dentistry laboratory. 3. The identification of P. endodontalis using PCR technique with samples collected from infected root canals or periapical lesions was more sensitive than that of culture methods. 4. Seven samples revealed including P. endodontalis by PCR technique. Five of them were related with pains, two of them with sinus tract, three of them with foul odor, and three of them with purulent drainage. P. endodontalis was shown to have great relation with pains.

연구개발(R&D)집중도의 결정요인 및 영향에 관한 연구 : S-C-P 패러다임 관점에서의 접근 (Antecedents and Effects of R&D Concentration : An Analysis from the Perspective of the Structure-Conduct-Performance paradigm)

  • This paper examines, from the perspective of the structure-conduct-performance (S-C-P) paradigm, the structural factors that determine R&D concentration in industries. The results are as follows. First, an industry's R&D concentration is directly related to its market concentration, R&D intensity, capital intensity, and technological opportunities. In contrast, the higher an industry's performance, the more likely the diffusion of R&D investment is for firms belonging to that industry. Second, an industry's R&D concentration has a positive effect on its market concentration but a negative effect on its performance, suggesting that governments should adopt R&D policies that would induce more firms to invest in R&D instead of focusing only on a few firms to enhance industry performance.

요양보호사의 응급처치 수행자신감 예측요인 (Predictors of Caregivers' First Aid Confidence)

  • 본 연구는 요양보호사를 대상으로 의사소통 자신감, 자기효능감, 응급처치 수행자신감 정도와 상관관계를 확인하고 응급처치 수행자신감 예측요인을 알아보고자 실시하였다. 요양보호사 202명을 대상으로 2022년 3월 1일부터 3월 31일까지 자료수집 하였으며, 수집된 자료는 SPSS 29.0 프로그램을 이용하여 t-test, ANOVA, Scheffe's test, Person's correlation coefficients, Multiple linear regression으로 분석하였다. 자기효능감은 의사소통 자신감과 정적 상관관계(r=.54, p<.001), 응급처치 수행자신감은 의사소통 자신감(r=-.18, p<.05), 자기효능감과 부적 상관관계를 보였다(r=-.31, p<.001). 응급처치 수행자신감 예측요인으로 간호조무사가 없을 때(β=-.18, p=.009)와 자기효능감(β=-.30, p<.001)으로 나타났고, 설명력은 11.0%였다(Adj R2=.110, p<.001). 본 연구결과를 근거로 요양보호사의 응급처치 수행자신감 향상을 위한 응급교육 프로그램을 개발하여 적용할 것을 제언한다.

COVID-19 팬데믹 상황에서 사무직 남성근로자의 건강증진행위에 영향을 미치는 요인: Pender의 건강증진모형을 적용하여 (Factors affecting the health promoting behaviors of office male workers during the COVID-19 pandemic: Using Pender's health promotion model)

  • Purpose: The purpose of this study was to analyze the factors influencing the health promoting behavior(s) of office worker males in the COVID-19 pandemic by applying Pender's health promotion model. Methods: The participants in this study were 149 male office workers at companies located in S, G and S cities. The collected data were analyzed using descriptive statistics, t-test, ANOVA, Pearson's correlation coefficient and a stepwise multiple regression using the SPSS Window 25.0 program. Results: The subject's health promoting behaviors and prior related behaviors (r=.58, p<.001), perceived benefits of action (r=.41, p<.001), self-efficacy (r=.53, p<.001), social support (r=.39, p<.001), self-esteem (r=.47, p<.001) and commitment to a plan of action (r=.67, p<.001) showed a high positive correlation. The factors affecting the subjects' health promoting behaviors were the commitment to a plan of action (𝛽=.35, p<.001), self-esteem (𝛽=.27, p=.005), prior related behavior (𝛽=.26, p<.001), health status (good) (𝛽=.20, p=.001) and self-efficacy (𝛽=.14, p=.047). These variables explained 63.0% of the subjects' health promoting behaviors. Conclusion: During the COVID-19 pandemic, subjects are more likely to be exposed to disease due to reduced outdoor activity time and irregular eating habits due to the strengthening of social distancing. Health promoting behaviors are an important concept that can maintain health and prevent diseases. To improve the health promoting behaviors of men engaged in office work, it is necessary to develop and operate a health promotion behaviors program considering those variables.

Bacillus subtilis의 Pho Regulon을 통한 인산 결핍 스트레스 반응 (Phosphate Deficiency Stress Response Mediated by Pho Regulon in Bacillus subtilis)

  • 인산 결핍기에 직면한 Bacillus subtilis는 PhoP-PhoR twocomponent system (TCS)를 통해 이러한 상황을 인식하고 생존을 유지하기 위해 Pho regulon으로 불리는 일련의 유전자들의 발현을 조절한다. 이때 histidine kinase인 PhoR은 자동 인산화되어, 인산을 response regulator인 PhoP에 전달한다. 인산화된 PhoP (PhoP~P)는 Pho regulon 유전자의 프로모터(promoter) 부위에 존재하는 반복되는 6 bp의 잘 보존된 PhoP 결합서열에 결합하여 해당 유전자의 발현을 활성화시키거나 억제한다. 이러한 Pho regulon 신호전달 시스템은 최소한 세 개의 TCS (PhoP-PhoR, ResD-ResE TCS, SpoOA phosphorelay), 광범위한 탄소대사 조절자(CcpA), 전위기 조절자(AbrB, ScoC) 등을 포함하는 신호전달 시스템과 밀접하게 상호 연결되어 있을 뿐만 아니라, 생육에 필수적인 YycF-YycG TCS와 상호조절을 통한 밀접한 관련을 가지고 있다. Pho regulon에 의한 인산결핍 스트레스 반응을 이해하는데 많은 진척이 있었으나, 많은 의문들은 여전히 남아있다. 이러한 의문들을 푸는 일은 B.subtilis의 응용연구에 중요한 정보를 제공할 것이다.

Detection of Pectobacterium chrysanthemi Using Specific PCR Primers Designed from the 16S-23S rRNA Intergenic Spacer Region

  • The 16S-23S rRNA intergenic spacer regions (ISRs) were sequenced and analyzed to design specific primer for identification of Pectobacterium chrysanthemi. Two types ISRs, large and small ISRs, were identified from three strains (ATCC 11663, KACC 10163 and KACC 10165) of P. chrysanthemi and Pectobacterium carotovorum subsp. carotovorum ATCC 15713.Large ISRs contained transfer RNA-Ile(tRNA$^{Ile}$)and tRNA$^{Ala}$, and small ISRs contained tRNA$^{Glu}$. Size of the small ISRs of P. chrysanthemi ranged on 354-356 bp, while it was 451 bp in small ISR of P. carotovorum subsp. carotovorum ATCC 15713. From hypervariable region of small ISRs, species-specific primer for P. chrysanthemi with 20 bp length (CHPG) was designed from hypervariable region of small ISRs, which was used as forward promer to detect P. chrysanthemi strains with R23-1R produced PCR product of about 260bp size (CHSF) only from P. chrysanthemi strains, not from other Pectobacterium spp. and Erwinia spp. Direct PCR from bacterial cell without extracting DNA successfully amplified a specific fragment, CHSF, from P. chrysanthemi ATCC 11663. The limit of PCR detection was 1${\pm}10^2$ cfu/ml.

  • Let $R_e\,=\,\frac{{\mathbb{F}}_{p^m}[u]}{{\langle}u^e{\rangle}}$, where p is a prime number, e is a positive integer and ue = 0. In this paper, we first characterize the structure of cyclic codes of length ps over Re. These codes will be classified into 2e distinct types. Among other results, in the case that e = 4, the torsion codes of cyclic codes of length ps over R4 are obtained. Also, we present some examples of cyclic codes of length ps over Re.

Prion Protein Genotypes in Pakistani Goats

  • The PCR-amplified prion protein (PrP) gene was sequenced to determine the frequency of scrapie-associated as well as novel PrP genotypes in 72 healthy goats representing five breeds. A total of six genotypes were detected, resulting from the three reported 143 (H/R), 154 (R/H) and 240 (S/P) and the two novel 39 (S/R) and 185 (I/F) amino acid polymorphisms. Of the four silent mutations 42 (a$\rightarrow$g), 138 (c$\rightarrow$t), 231 (c$\rightarrow$a) and 237 (g$\rightarrow$c) detected in this study, 237 (g$\rightarrow$c) is novel. A genotype (SIP/RFP) harboring three amino acid polymorphisms 39 (S/R), 185 (I/F) and 240 (S/P) was found in few goats. Although both scrapie-associated genotypes with 143 (H/R) and 154 (R/H) polymorphisms and others with 39 (S/R), 185 (I/F) and 240 (S/P) polymorphisms were present in the studied Pakistani goats, their frequency was lower than that of the wild-type genotype SHRIS/SHRIS (34.7%). These results emphasize the need for further sequencing of the PrP gene in a large number of goats representing the five studied breeds, so that overall PrP variability can be assessed in these breeds in research addressing future concerns about scrapie.

복부비만 고령여성의 대사증후군 위험요인과 고감도 C-반응성 단백의 관련성 (The Relationship between Metabolic Syndrome Risk Factors and High Sensitive C-reactive Protein in Abdominal Obesity Elderly Women)

  • 이 연구에서는 복부비만을 가진 고령여성을 대상으로 대사증후군 동반 유무에 따른 대사증후군 위험요인과 hs-CRP와의 관련성에 대해 알아보고자 하였다. 대사증후군 진단은 AHA/NHLBI (American Heart Association/National Heart, Lung and Blood Institute) 2005년 기준에 따라 5가지 기준 중 3개 이상 해당되는 경우 대사증후군 진단군(MetS, N=77), 2개이하의 위험요인에 해당하는 경우 대조군(Absent, N=97)으로 분류하였다. hs-CRP 농도는 대사증후군 위험요인과 밀접한 관련이 있으며, 특히 복부비만(r=0.190, p=0.014), 공복혈당(r=0.240, p=0.002), HDL-콜레스테롤(r=-0.164, p=0.035)과 연관이 있음을 확인할 수 있었다. 또한 대조군보다 대사증후군 진단군에서 hs-CRP가 높게 나타났으며(p=0.007), 복부비만 상태일지라도 높은 혈당(p=0.006)과 낮은 HDL-콜레스테롤혈증(p=0.010)의 위험요인이 있는군에서 hs-CRP가 높았다. 결론적으로 복부비만이 있더라도 대사증후군 위험요인의 동반 유무에 따라 염증관련 위험도가 달라짐을 알 수 있었다.
