목적 : 방사선조사에 의한 apoptosis에서 나타나는 각종 세포주기관련 유전자들의 발현 양상을 RNA와 단백 수준에서 분석하여 방사선조사에 의한 apoptosis에서의 세포주기 조절의 변화를 규명함으로서 방사선치료의 기전에 대한 분자생물학적 이해를 도모하고자 본 연구를 시행하였다. 대상 및 방법 : promyelocytic leukemia 세포주인 HL60 세포주를 배양하여 선형가속기(6MV X-선)로 세포에 8Gy의 방사선을 조사하였다. 조사후 다양한 시간 간격으로 Apoptotic DNA Fragmentation Assay법을 이용하여 apoptosis를 확인하고 동시에 세포주기관련 유전자들(cyclinA, cyclin B, cyclin C, cyclin Dl, cyclin E, cdc2, CDK2, CDK4, $p16^{INK4a}$, $p21^{WAF1}$, $p27^{KIP1}$, E2F, PCNA와 Rb)을 단백질과 RNA 수준에서 분석하기위해 western blot analysis와 반정량적 RT-PCR을 시행하였다. 결과: 8 Gy의 방사선 조사에 의해 HL60세포에서 apoptosis가 관찰 되었다. 방사선 조사군에서 cyclin A단백은 조사후 48시간까지 시간이 갈수록 증가하였으며, cyclin E, E2F, CDK2 및 Rb 단백은 증가되었다가 다시 감소를 보였다. Rb단백의 증가는 대부분 비활성형인 ppRb (phosphorylated Rb protein)의 양적변화에 의한 것이었다. cyclin Dl, PCNA, COC2, CDK4, $p16^{INK4a}$단백은 발현의 차이를 보이지 않았으며 $p21^{WAF1}$과 $p27^{KIP1}$ 단백은 검출되지 않았다. cyclin A, B, C mRNA는 방사선 조사 직후 감소하였다가 12시간부터 발현이 증가되었으며 cyclin Dl mRNA는 조사후 바로 증가하여 48시간에 다시 감소하였다. cyclin E mRNA는 조사 후 시간이 경과함에 따라 감소하였다. CDK2 mRNA는 3시간째는 감소하다가 6시간부터 많은 증가를 보였으며 CDK4 mRNA는 조사후 6-12시간에 급격한 발현증가를 보였다. $p16^{INK4a}$ RNA는 발현의 변화가 없었으며, $p21^{WAF1}$ 및 $p27^{KIP1}$ RNA의 발현은 관찰되지 않았다. 결론 : 이상의 결과로 미루어볼 때, 방사선 조사에 의한 HL60세포의 apoptosis와 세포의 Gl/S transition는 밀접한 관계가 있는 것으로 생각되며 Rb단백의 증가와 활성형 Rb단백의 감소 현상도 관련이 있는 것으로 사료된다. 이는 E2F의 비정상적인 과발현 및 cyclin E/CDK2의 발현 증가와 관련이 있는 것으로 추측된다. 또한 $p21^{WAF1}$ 및 $p27^{KIP1}$는 방사선에 의한 apoptosis에는 관여되지 않는 것으로 사료된다.
본 연구는 참치의 석유 에텔 추출물로 부터 항암 효과가 있는 활성 성분을 분리 정제하고, 흰생쥐의 임파모 세포인 LI210 과 P388, 인체 장암세포인 HCT-48, HT-29 및 HRT-18를 대상으로 단계별로 얻어지는 성분의 항암 활성올 측정하고, 그 물질의 생체내에서의 작용 기전을 밝히기 위해 여러가지 면역학적 실험을 수행하였다. In vitro배양액에서의 인체 결장암세포인 H HCT-48에 대해, crude extract는 배양액 1ml강 $60\mu\textrm{g}$ 첨가시 증식이 2배로 억제되었으며(1 unit), 이를 column chromatography 수행하여 얻은 fraction D의 1 unit은 $2 22\mu\textrm{g}$이며, 다시 TLC를 하여 얻은 Spot I 의 1 unit은 $9\mu\textrm{g}$으로서 정제 했을때 277배 증가하였다. Spot I 은 GC/ M5에 의해 7가지 성분이 섞여있는 혼합물로서 이 중 4가지 생분은 $C_{14:0},\;C_{16:0},\;C_{17:1},\;및\;C_{18:0}$인 지방산으로 확인이 되었으며, 나머지 3가지는 지방 성분으로 분자식은 확인되지 않았다. 다른 인체 장암 세포인 HT-29과 HRT-18에 대한 crude extract의 1 unit은 $64\mu\textrm{g}\;및\;54\mu\textrm{g}$이었으며 frattion D의 1 unit에 해당하는 양은 두 세포 모두 약 $34\mu\textrm{g}$이었다. 또한, 생쥐의 백혈병 성 임파모 세포인 L1210과 P388의 경우 crude extract의 1 unit에 해당하는 양은 $2-2.5\mu\textrm{g}$이며, fraction D의 경우는 $1.2-1.4\mu\textrm{g}$으로서 참치 추출물의 세포 종식 억제력은 백혈병성 임파모세포에서 인체 장암세포보다 높았으며, 암세포의 종류에 따라 효과의 차이가 있었다. 참치 추출물(fraction D)을 첨가한 배양액에서 배양한 HCT-48 세포획 크기 분포도는 배양시간이 경과함에 따라 세포의 크기가 작아지며, 현미경 사진에서 보면 본래의 형태가 위축되거나, 세포의 주위가 흐트러지면서 사면 현상을 보였다. 참치 석유 에텔 추출물에는 DHA가 18.8%를 차지 하며 EPA는, 3.25% 포함되어 있었다. 대조군 생쥐에 비해 S-180을 접종한 후 참지 추출물을 투여한 생쥐에서 용혈반 형성 세포수와 혈청 단백질 중 immunoglobulin의 relative %가 현저히 증가되었다. 그러나 비장의 자연 살해 세포의 활성에는 변화를주지 않았다. 이상의 결과로 보아, 참치의 석유 에델 추출물중에 in vitro에서 인체 장암세포 및 백혈병성 임파모세포의 증식 억제 및 생체내에서 면역 증강 효과를 나타내는 성분이 존재함을 알 수 있다.
오미자의 면역조절작용에 미치는 효과를 살펴본 결과, 오미자(SZX)는 T 및 B세포의 증식을 유의성있게 촉진시켰으며, in vivo 및 in vitro 실험에서 비장내의 T 및 B림프구를 증가시켰고, T세포 중 특히 $T_H$세포를 증가시켰다. 또한 흉선 T림프구 중의 $T_H$세포의 수를 증가시키는 작용을 나타냈다. 아울러 SZX는 in vitro계에서 L1210세포의 apoptosis를 유도하였으며, 복강대식세포에서의 탐식능 및 nitric oxide(NO)생성을 촉진시켰다. 이 결과는 복강대식세포로부터 생성된 NO가 L1210세포의 apoptosis를 유도하는데 중개역할을 하고 있을 가능성을 시사한다. 이상의 결과 오미자는 T, B림프구 및 대식세포 등의 면역세포의 활성을 증강시키는 작용을 보유하고 있으며, 암세포의 apoptosis를 유도하는 기능을 가지고 있음으로써 면역조절제로서의 역할을 가지는 것으로 추정된다.
8-Azaxanthine(1), 3-${\beta}$-D-ribofuranosyl-8-azaxanthine(2), 3-${\beta}$-D-ribofuranosyl-8-azaxanthine-5'-monophosphate(3) 그리고 3-${\beta}$-D-ribofuranosyl-8-azaxanthine-5'-(3-pyridinylcarbonyl)monophosphate(4)를 합성한 후 Mouse leukemia P388, Murine mammary carcinoma FM3A, Human histiocytic lymphoma U937 암세포들에 대해 시험관내 항암활성을 MTT를 이용한 방법으로 예측하고 $IC_{50}$(${\mu}mol/mL$)으로 나타내었다. 그 결과, 비정상적인 염기의 N-3와 당의 C-1' glycoside 결합을 가진 2는 세가지의 암세포에 대해 감수성이 좋아 $IC_{50}$이 각각 0.05, 0.06, 0.06 ${\mu}mol/mL$이었으나 항세균 활성 예측에서는 감수성을 보이지 않았다. Human histiocytic lymphoma U937 세포에 대한 $IC_{50}$은 1이 0.33, 2는 0.66, 3은 0.25 그리고 4는 0.33 ${\mu}mol/mL$로 구조 변화에 따라 서로 다른 값을 보였다
몇가지 nucleoside 5'-monophosphate 유도체들과 지질 친화성을 증가시킨 nucleoside 5'-(3-pyridinyl carbonyl)monophosphate 유도체들을 합성한 후 Mouse leukemia P388, Murine mammary carcinoma FM3A, Human histiocytic lymphoma U937 세포들에 대해 시험관내에서 항암활성을 MTT를 이용한 방법으로 나타내었다. 그 결과 uridine 5'-(3-pyridinylcarbonyl) monophosphate(7)와 2',3'-didehydro-3'-deoxythymidine-5' -(3-pyridinylcarbonyl) monophoshate(8)의 inhibition이 uridine 5'-monophosphate(1)와 2',3'-didehydro-3'-deoxythymidine-5'-monophosphate(4) 보다 각각 증가하였다. 이는 nucleoside 5'-(3-pyridinylcarbonyl) monophosphate 유도체들이 임상적 한계를 극복할 수 있는 가능성을 보인 것이다.
The effects of potassium, sodium, calcium, magnesium, and iron on cyanobacterial bloom potentials were investigated in Daechung Reservoir, Korea. Potassium showed the highest correlation with the cyanobacterial cell number (r=0.487, P<0.05) and phycocyanin concentration (r=0.499, P<0.05). However, it was not likely that the potassium had directly affected the bloom formation, because the variations of its concentration were not significantly large. In contrast, the Fe concentration fluctuated drastically and exhibited a negative correlation with the cyanobacterial cell number (r=- 0.388, P<0.1) and phycocyanin concentration (r=-0.446, P<0.05). Accordingly, the K:Fe atomic ratio would appear to reflect the extent of cyanobacterial bloom more precisely than K or Fe alone. The K:Fe ratio specifically correlated with cyanobacterial percentage, the cyanobacterial cell number and phycocyanin concentration (r=0.840, P<0.001; r=0.416, P<0.05; r=0.522, P<0.01, respectively). With the K:Fe atomic ratio of over 200, the chlorophyll-a concentration, cyanobacterial cell number, and phycocyanin concentration exceeded $10\mu$g $1^{-1}$20,000 cells $ml^{-1}$, and 20 pM, respectively, the general criteria of eutrophic water.
목적 : 사람의 유방암 세포인 MCF-7 세포주를 대상으로 gultathione S-transferase K1 (hGSTK1) 유전자의 발현 정도 및 방사선 조사에 의한 hGSTK1 유전자의 발현 변화를 관찰하고 hGSTK1 유전자를 과발현시킴으로써 hGSTK1이 방사선 감수성에 어떤 영향을 미치는 지를 관찰하였다. 재료 및 방법 : 사람의 모유두 세포 pBluescript phagemid cDNA library로부터 선별한 hGSTK1 cDNA를 pcDNA3.1/Myc-His (+) vector에 결합시킨 후, 사람의 유방암 세포인 MCF극 세포에 이입시켰다. hGSTK1 유전자를 이입시키지 않은 MCF극 세포와 hGSTK1을 이입시킨 MCF-7 세포에 $2\~12\;Gy$의 엑스선을 조사하여 생존 분획을 비교하였다. hGSTK1 유전자를 이입시키지 않은 MCF-7 세포와 hGSTK1을 이입시킨 MCF-7 세포에서 방사선량, 분할조사 여부, 방사선 조사 후 경과 시간에 따른 hGSTK1 mRNA 발현의 차이를 보기 위하여 RT-PCR 분석을 시행하였다. 결과 : hGSTK1 유전자를 이입시키기 전의 MCF-7 세포보다 hGSTK1 유전자를 이입시킨 MCF-7 세포에서 생존 분획이 유의하게 높은 것으로 나타났다. hGSTK1 유전자를 이입시키지 않은 MCF-7 세포에서 2 Gy 생존 분획은 $0.3250{\pm}0.0319$였고, hGSTK1 유전자를 이입시킨 MCF-7 세포에서 2 Gy 생존 분획은 $0.4125{\pm}0.0325$였다 (p<0.05). 그러나 RT-PCR에 의한 hGSTK1 mRNA 분석에서는 방사선량, 분할조사 여부, 방사선 조사 후 경과 시간에 따른 발현의 차이를 볼 수 없었다. 결론 : MCF-7 세포주에서 hGSTK1의 과발현은 방사선 감수성에 영향을 미쳐서 MCF-7 세포의 생존 분획을 증가 시켰다고 볼 수 있으나 이에 대한 정확한 기전을 알기 위해서는 더 많은 연구가 필요할 것이다.
$K^+$ outward currents in the outer hair cells (OHCs) of circling mice (homozygous (cir/cir) mice), an animal model for human deafness (DFNB6 type), were investigated using a whole cell patch clamp technique. Littermate heterozygous (+/cir) mice of the same age (postnatal day (P) 0-P6) were used as controls. Similar slow rising $K^+$ currents were observed in both genotypes, but their biophysical and pharmacological properties were quite different. The values of Vhalf for activation were significantly different in the heterozygous (+/cir) and homozygous (cir/cir) mice ($-8.1{\pm}2.2mV$, heterozygous (+/cir) mice (n=7) and $-17.2{\pm}4.2mV$, homozygous (cir/cir) mice (n=5)). The inactivation curve was expressed by a single first order Boltzmann equation in the homozygous (cir/cir) mice, while it was expressed by a sum of two first order Boltzmann equations in the heterozygous (+/cir) mice. The $K^+$ current of homozygous (cir/cir) mice was more sensitive to TEA in the 1 to 10 mM range, while the 4-AP sensitivities were not different between the two genotypes. Removal of external $Ca^{2+}$ did not affect the $K^+$ currents in either genotype, indicating that the higher sensitivity of $K^+$ current to TEA in the homozygous (cir/cir) mice was not due to an early expression of $Ca^{2+}$ activated $K^+$ channels. Our results suggest that the $K^+$ outward current of developing homozygous (cir/cir) mice OHCs is different in both biophysical and pharmacological aspects than that of heterozygous (+/cir) mice.
Panax ginseng has been extensively used in the traditional oriental medicine as a restorative, tonic and Prophylactic agent. Recently, several reports regarding to anticancer effects of Panax ginseng has accumulated. These studies emphasized the fact that the anticancer activities might be due to a glycoside group called ginsenoside or pan.u saponin which has a water soluble characteristic. However, the authors and collaborates demonstrated that a highly lipid soluble component in extract of Panax ginseng roots contains a considerable cytotoxic activities against marine leukemic cells (L1210, P388) and human censer cells (HRT-18, HT-29, HCT48). This study was devised to observe the cytotoxic activities of Petroleum-ether extract of Panax giuseng roots (crude GBD and its Partially Purified fraction from silicic acid column chromatography (7 : 3 GX) against sarcoma-180 (5-180) and Walker carcinosar- coma 256 (Walker 256) in vivo, and murine leukemic Lymphocytes (L1210) and human rectal cancer cells (HRT-18) and human colon cancer cells (HT-29 and HCT48) in vitro. Each cell-line was cultured in medium containing serial concentration of the crude GX or 7 : 3 GX in vitro. A highly lipid soluble compound in the extract of Panax ginseng root was cytocidal to murine leukemic cells and human colon and rectal cancer cells in vitro. In the meantime, ginseng saponin derivatives did not have cytotoxic effects at its corresponding concentration. The growth rates of the cancer cells in medium containing ginseng extracts were inhibited gradually to a significant degree roughly in proportion to the increase of the extract concentration. The cytotoxic activity of 7 : 3 GX was about 3 times more potent than that of crude GX, one unit of cytotoxic activity against L1210 cells being equivalent to 2.54 Ug and 058 Ug for the crude GX and 7 : 3 GX, respectively. The Ri value of the active compound on silica- gel thin layer chromatography with petroleum-ether/ethyl ether/acetic acid mixture (90 : 10 : 1, v/v/v) as a developing so lvent was 053. While, the Panaxydol and Panaxynol as active compounds were purified from Petroleum-ether extract of Panax ginseng root by Drs. Ahn and Kim, and author found out that the one unit of cytotoxic activity of the Panaxydol and Panaxynol against L1210 cells being equivalent to 056 Ug and 0.3918 respectively. The survival times of mice inoculated with S-180 cells were extended about 1.5 to 2 times by the 7 : 3 GX treatment compared with their control group. The significantly decreased hemoglobin values of rats after inoculation with Walker 256 were recovered to normal range by oral administration of the crude Gt The synthetic levels of protein, DNA and RNA in human colon and rectal cancer cells were significantly diminished by treatment with the crude GX, which can explain a part of the origin of its anticancer activity.
Background: Studies of the prognostic role of circulating tumor cells (CTCs) in early-stage non-small cell lung cancer (NSCLC) are still limited. This study investigated the prognostic power of CTCs from the pulmonary vein (PV), peripheral blood (PB), and bone marrow (BM) for postoperative recurrence in patients who underwent curative resection for NSCLC. Methods: Forty patients who underwent curative resection for NSCLC were enrolled. Before resection, 10-mL samples were obtained of PB from the radial artery, blood from the PV of the lobe containing the tumor, and BM aspirates from the rib. A microfabricated filter was used for CTC enrichment, and immunofluorescence staining was used to identify CTCs. Results: The pathologic stage was stage I in 8 patients (20%), II in 15 (38%), III in 14 (35%), and IV in 3 (8%). The median number of PB-, PV-, and BM-CTCs was 4, 4, and 5, respectively. A time-dependent receiver operating characteristic curve analysis showed that PB-CTCs had excellent predictive value for recurrence-free survival (RFS), with the highest area under the curve at each time point (first, second, and third quartiles of RFS). In a multivariate Cox proportional hazard regression model, PB-CTCs were an independent risk factor for recurrence (hazard ratio, 10.580; 95% confidence interval, 1.637-68.388; p<0.013). Conclusion: The presence of ≥4 PB-CTCs was an independent poor prognostic factor for RFS, and PV-CTCs and PB-CTCs had a positive linear correlation in patients with recurrence.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
