Source Tracking of PCDD/Fs in Ambient Air Using Pine Needles (소나무 잎을 이용한 대기 중 다이옥신/퓨란 발생원 추정)

  • Objectives: Pine needles were used as a passive air sampler (PAS) of atmospheric persistent organic pollutants (POPs). This study was performed to investigate concentrations of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) deposited on pine needles near a waste incinerator and PCDD/Fs source contributions using principal component analysis (PCA). Methods: Two-year-old pine needles were sampled at 11 points with respect to distance and wind direction from the incinerator. PCDD/Fs deposited on pine needles were analyzed with HRGC/HRMS. The source contribution of PCA was calculated with SPSS. Results: The average concentration of PCDD/Fs deposited on pine needle was 0.79 (0.27-1.76) pg TEQ/g dry, PCDDs with 0.24 (0.01-0.95) pg TEQ/g dry and PCDFs with 0.56 (0.27-0.82) pg TEQ/g dry, respectively. The average concentration fraction of PCDDs was 29.7%, that of PCDFs was 70.3%, and PCDFs were more prevalent than PCDDs. The contributions of PCDD/Fs sources were estimated as incineration at 58.3% and automobiles at 28.4%. However, a relation and regulation between PCDD/Fs concentrations deposited on pine needles and distance from incinerator or wind direction was not shown. Conclusion: It was concluded that atmospheric PCDD/Fs concentrations near an industrial complex with a waste incinerator were affected by multiple sources. However, PCDD/Fs concentrations were lower than in other inland cities with the exception of background area.

Enhanced Effect of Gluten Hydorlysate on Solubility and Bioavailability of Calcium in Rats (글루템 가수분해물에 의한 칼슘의 가용화 및 체내이용성 증진 효과)

  • Dietary peptides have recently received attention regarding their beneficial effects on nutrient metabolism since the caseinphosphoptides obtained from casein hydrolysate are generally believed to enhance the intestinal absorption of Ca. The two experiments were conducted to investigate the effects of various hydrolyzed fractions of gluten on Ca bioavailability. The gluten hydrolysate of dietary components was produced by enzymatic hydrolysis of gluten whereas gluten hydrolysate supernationt and its precipiate resulted from centrifugation. In experiment I, the rats were for 4 weeks fed the 4 kinds of diets containing same amount of nitrogen and calories and diffeing only in the forms of nitrogen sources. The diets were gluten (G), gluten hydrolysat(GH), gluten hydrolysate supernatant(GHS) and gluten hydrolysate precipitatie(GHP). Determination was made for the body weight gain, serum Ca concentration, Ca solubility in small intestinal contents, bone weight, length and stength, bone ash and Ca content, and Ca balance, respectively. No significant difference was noticed as regards growth, serum Ca, and bone dimension and Ca content among rat groups. More significant increase was observed with regard to Ca absorption and intestinal solubility in the rats receiving the GH or GHS diet which containe crude gluten peptides, than in those subjected to G or GHP diet. In experiment II, in vitro determination for Ca solubility was made to ascertain the mechanism responsible for the effects of gluten peptides on Ca absorption. The 10mM Ca in potassium phosphate buffer solution(pH 7.0) incubated for 3 hours at 37$^{\circ}C$ by the GHS fraction, was observed to be capable of increasing the Ca solubility at 5-25mg/ml concentration of gluten peptides. These observations suggest that the gluten peptides from gluten hydrolysate may enhance the Ca absorption efficiency by increasing the solubility of Ca in small intestine.

Results of Extracorporeal Membrane Oxygenation (ECMO) Support before Coronary Reperfusion in Cardiogenic Shock with Acute Myocardial Infarction

  • Despite aggressive treatment, the mortality rate of cardiogenic shock with acute myocardial infarction (AMI) is high. We performed extracorporeal membrane oxygenation (ECMO) prior to coronary reperfusion, and evaluated the early clinical results and risk factors. Materials and Methods: From May 2006 to November 2009, we reviewed the medical records of 20 patients in cardiogenic shock with AMI (mean age $67.7{\pm}11.7$ yrs, M : F 14 : 6). After initially performing ECMO using the CAPIOX emergency bypass system ($EBS^{(R)}$Terumo, Tokyo, Japan), patients underwent coronary reperfusion (coronary artery bypass grafting, 13; percutaneous coronary intervention, 7). Results: All patients were in a cardiogenic shock state, cardiopulmonary resuscitations (CPR) were performed for fourteen patients (mean CPR time $20.8{\pm}26.0$ min). The mean time from vascular access to the initiation of ECMO was $17.2{\pm}9.4$ min and mean support time was $3.8{\pm}4.0$ days. Fourteen patients were able to be weaned from ECMO and ten patients were discharged (mean admission duration $50.1{\pm}31.6$ days). Patients survived on average $476.6{\pm}374.6$ days of follow-up. Longer CPR and support time, increased cardiac enzyme, lower ejection fraction, lower albumin, and major complications were the risk factors of mortality (p<0.05). Conclusion: The early application of ECMO prior to coronary reperfusion and control of risk factors allowed for good clinical results in cardiogenic shock with AMI.

Fiber Type Effects on the Flexural Behavior of Steel Fiber Reinforced Concrete Beams (강섬유의 형태에 따른 SFRC보의 휨거동에 관한 연구)

  • This paper presents a comparative evaluation of five different types of steel fibers used as reinforcing material in concrete beams. Two types of plain and RC beams were prepared to compare the relative flexural behavior. The fibers used were dog bone (paddled), both ends hooked. コ-type straight. crimped and wavy type with aspect ratio of 43 to 75. Fiber volume fraction of 1 to 2% were used while shear span to depth ratio (aid) and steel ratio p were fixed. Fiber reinforcement effect index Ef and effective toughness index Te were adopted to evaluate fiber reinforcing effects. The effect of fiber reinforcement on flexural strength is higher in plain beams than in RC beams. Hooked and dog bone type fibers were found to be more effective than the other type ones in enhancing the flexural strength and post-peak energy absorption capacity of concrete beams.

The Study on the Thyroglobulin in Thyroid Diseases (갑상선질환(甲狀腺疾患)의 Thyroglobulin에 관(關)한 연구(硏究)(예보(豫報)))

  • In recent years with development of immuno-electrophoresis, more acurate analysis of the serum protein became possible. However, there is few reports in the literature which investigated the changes of the immunoglobulin compared with electrophoretically fractioned serum thyroglobulin in the patients with various thyroid diseases. The purpose of this report is to investigate the changes of thyroglobulin in various thyroid diseases by the method of immuno-electrophoresis and to compare the results with.serum protein fractionated by the method of agar-gel micro-electrophoresis. Materials and Methods: Sera from 9 patients with diffuse toxic goiter, 2 nodular nontoxic goiter, 2 thyroiditis, 3 hypothy, roidism, 1 thyroid cancer, 7 cystic degeneration of the thyroid gland, and 10 normal subject were taken. All cases were confirmed by various laboratory thyroid function tests and thyroid needle biopsy. Immuno-electrophoretic analysis of the serum were performed by Scheidegger's modified micro-immuno-electrophoretic method. The antiserum was obtained from the Travenol Laboratories International, Hyland Products Division and was rabbit anti-human thyroglobulin. Microscope slide agar-gel electrophoresis for serum protein fractionation was performed at $4^{\circ}C$ using veronal buffer, pH 8.6 and ionic strength 0.05, with 54 volts and 2.8 mA for 60 minutes. The fractionated slide was stained with 0.1% thiazine red. The results were as follows: 1) Increase of immune-globulin macroglobulin (IgM), alphaglobulin, and immune-globulin A (IgA) by 95.8%, 100%, 29.2% respectively was found in the serum from various thyroid diseases. 2) Thyroglobulin fraction was found to be increased in 50%, no change in 41.7%, and no line in 8.3% with all of the various goiter patients. On the other hand, 10 normal control group showed only 2 cases of increase, 5 cases of no change and 3 cases of no line.

Correlation Estimation between Geochemical Metal-fraction and Soil Properties in Agricultural and Industrial Soils (농경지 및 공장지역 토양 내 중금속 존재형태와 토양 특성과의 상관성 평가)

  • The Standards, Measurement and Testing Programme (SM&T-formerly BCR) extraction procedure was applied to fractionate Cr, Cu, Ni, Pb and Zn in 23 top soil samples into: (i) exchangeable phase; (ii) reducible phase; (iii) oxidisable(sulfides and organics bound) phase; and (iv) residual phase. Fractions of Cr and Ni were in the order of residual > oxidisable > reducible > exchangeable phase. The oxidisable phase was identified as dominant for Cu and Pb. Zn had the highest ratio of exchangeable phase in comparision to the other metals. The bioavailability and mobility were assessed to be the greatest for Zn, followed by a decreasing order of Pb, Cu, Ni and Cr. All metal average concentrations in topsoil samples was higher in industrial sites than in agricultural sites. Our results revealed higher concentrations in topsoil samples (0~15 cm) than in sub soils (15~30 cm, 30~60 cm) for most metals at six sites (No. 5, 6, 17, 19, 20, 23). The fractions of exchangeable, reducible ad oxidisable phases showed relatively high correlation with soil pH, Fe/Mn oxide concentrations and organic matter contents, respectively.

Antimicrobial Activity of Caesalpinia sappan against Animal husbandry disease. (가축질병 균주에 대한 소목의 항균활성)

  • Antimicrobial activity of Caesalpina sappan L. extract (CS extract) against animal husbandry disease-related bacteria was studied. The CS extracts showed a significant antimicrobial activity against Gram(+) bacteria and this antimicrobial activity was most significant against Staphylococcus epidermidis. Minimum inhibitory concentrations (MIC) of CS was in the range of 0.8∼16 mg/ml and 0.8∼10 mg/ml, in the case of MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with EtOAc layer. The antimicrobial activities of CS extracts against most microbial strains were stable by either heat treatment or acid treatment, whereas those against S. aureus and Sal. typhimurium were reduced by acid treatment. The inhibitory effect of CS extracts on microbial cell growth was further examined by the addition of 0, 100, 300, and 500 ppm of CS extracts into growth medium. The growth of gram(+) bacteria, S. aureus and S. epidermidis, was inhibited for 72 hours in all ranges of CS extracts added, but the growth of gram(-) bacteria was only inhibited when at least 100 ppm of CS extracts were added. Taken together, the antimicrobial activities of CS extracts were more effective against gram(+) bacteria compared to those against gram(-) bacteria.

Evidence of an Alternative Route of Cellobiase Secretion in the Presence of Brefeldin A in the Filamentous Fungus Termitomyces clypeatus

  • Secretion of cellobiase occurred in a brefeldin A (BFA) uninhibited manner in the filamentous fungus Termitomyces clypeatus. Fluorescence confocal microscopy revealed that application of the drug at a concentration of 50 ${\mu}g$/ml caused arrest of Spitzenkorper assembly at the hyphal tip. This resulted in greater than 30% inhibition of total protein secretion in the culture medium. However, the cellobiase titer increased by 17%, and an additional 13% was localized in the vacuolar fraction en route secretion. The secretory vacuoles formed in the presence of the drug were also found to be bigger (68 nm) than those in the control cultures (40 nm). The enzyme secreted in the presence and absence of BFA revealed a single activity band in both cases in native PAGE and had similar molecular masses (approx. 120 kDa) in SDS-PAGE. The BFA enzyme retained 72% of native glycosylation. It also exhibited a higher stability and retained 98% activity at $50^{\circ}C$, 93.3% activity at pH 9, 63.64% activity in the presence of 1M guanidium hydrochloride, and 50% activity at a glucose concentration of 10 mg/ml in comparison to 68% activity, 75% activity, 36% activity, and 19% activity for the control enzyme, respectively. The observations collectively aimed at the operation of an alternative secretory pathway, distinct from the target of brefeldin A, which bypassed the Golgi apparatus, but still was able to deliver the cargo to the vacuoles for secretion. This can be utilized in selectively enhancing the yield and stability of glycosidases for a successful industrial recipe.

Enhanced Controlled Transdermal Delivery of Hydrochlorothiazide from an Ethylene-vinyl Acetate Matrix

  • Repeated oral administration of hydrochlorothiazide, a loop diuretic, due to transient high blood levels, may cause adverse effects such as gastric disturbance, nausea, high blood sugar, and hyper lipidemia. Transdermal administration could avoid some of these systemic side effects and gastric disorders. We have developed a matrix using ethylene-vinyl acetate (EVA), a heat-processible and flexible material, for transdermal delivery of hydrochlorothiazide. Drug solubility was highest at 40% PEG-400 volume fraction. Drug release increased as concentration increased with a linear relationship between the release rate and the square root of loading dose. Increasing temperature increased drug release from the EVA matrix. The activation energy, measured from the slope of log P versus 1000/T, was 11.9 kcal/mol for a 2.5% loading dose from EVA matrix. Diethyl phthalate had the highest plasticizing effects on the release of hydrochlorothiazide. To increase the skin permeation of hydrochlorothiazide from the EVA matrix, enhancers such as the saturated fatty acids, the unsaturated fatty acids, and the non-ionic surfactants were added to the EVA matrix, and skin permeation was evaluated using a modified Keshary-Chien diffusion cell fitted with intact excised rat skin. Polyoxyethylene 23-lauryl ether showed the highest enhancing effects. In conclusion, transdermal delivery of hydrochlorothiazide could be improved from an EVA matrix containing plasticizer and permeation enhancer.

Cardiac function associated with home ventilator care in Duchenne muscular dystrophy

  • Purpose: Cardiomyopathy is becoming the leading cause of death in patients with Duchenne muscular dystrophy because mechanically assisted lung ventilation and assisted coughing have helped resolve respiratory complications. To clarify cardiopulmonary function, we compared cardiac function between the home ventilator-assisted and non-ventilator-assisted groups. Methods: We retrospectively reviewed patients with Duchenne muscular dystrophy from January 2010 to March 2016 at Gangnam Severance Hospital. Demographic characteristics, pulmonary function, and echocardiography data were investigated. Results: Fifty-four patients with Duchenne muscular dystrophy were divided into 2 groups: home ventilator-assisted and non-ventilator-assisted. The patients in the home ventilator group were older ($16.25{\pm}1.85years$) than those in the nonventilator group ($14.73{\pm}1.36years$) (P=0.001). Height, weight, and body surface area did not differ significantly between groups. The home ventilator group had a lower seated functional vital capacity ($1,038{\pm}620.41mL$) than the nonventilator group ($1,455{\pm}603.2mL$). Mean left ventricular ejection fraction and fractional shortening were greater in the home ventilator group, but the data did not show any statistical difference. The early ventricular filling velocity/late ventricular filling velocity ratio ($1.7{\pm}0.44$) was lower in the home ventilator group than in the nonventilator group ($2.02{\pm}0.62$. The mitral valve annular systolic velocity was higher in the home ventilator group (estimated ${\beta}$, 1.06; standard error, 0.48). Patients with Duchenne muscular dystrophy on a ventilator may have better systolic and diastolic cardiac functions. Conclusion: Noninvasive ventilator assistance can help preserve cardiac function. Therefore, early utilization of noninvasive ventilation or oxygen may positively influence cardiac function in patients with Duchenne muscular dystrophy.
