Effects of purinergic nerve on relaxation of pig coronary artery (돼지 심관상동맥의 이완작용에 대한 purinergic 신경의 효과)

  • To elucidate the effects of purinergic nerve on relaxation of pig coronary artery, the effects of ATP, 2-methylthio ATP and electrical perivascular nerve stimulation were investigated from physiograph on the isolated coronary artery of pig. The results btained were as follows; 1. The relaxative responses induced by perivascular nerve stimulation(20V, 0.5msec, 10sec) were the frequency(1~8Hz) dependent manner with phentolamine($10^{-5}M$) and atropine($10^{-6}M$) on isolated coronary artery of pig. 2. The relaxative responses induced. by adenosine($10^{-7}{\sim}5{\times}10^{-3}M$) or ATP($10^{-7}{\sim}5{\times}10^{-5}M$) on precontraction with histamine($10^{-5}M$) were the dose-dependent manner, but the contractile responses were often induced by ATP($10^{-4}M$ and $10^{-3}M$). 3. The relaxative responses induced by 2-methylthio ATP($2.5{\times}10^{-8}{\sim}2.5{\times}10^{-6}M$) on precontraction with histamine($10^{-5}M$) were the dose-dependent manner. 4. The relaxative response induced by 2-methylthio ATP($10^{-7}M$) on precontraction with histamine($10^{-5}M$) was completely blocked by the pretreatment with $P_{2Y}$-purinoceptor blocker, reactive blue 2($10^{-4}M$). 5. The neurogenic relaxative response induced by perivascular nerve stimulation(20V, 8Hz, 0.5msec, 10sec) was weakly inhibited by the pretreatment with ${\beta}$-adrenoceptor blocker, propranolol($10^{-5}M$) and blocked by the addition with $P_{2Y}$-purinoceptor blocker, reactive blue 2($10^{-4}M$). The results suggest that the purinergic nerve is innervated, and its relaxative response was mediated by $P_{2Y}$-purinoceptor on isolated coronary artery in pig.

  • In the treatment of the wastewater containing metals($Cu^{2+}$, $Zn^{2+}$, $Ni^{2+}$, $Cr^{3+}$) by using batch precipitation and flocculation followed by membrane filtration, permeate flux and removal efficiency were investigated according to by the effect of pH and coagulants, and the type of membranes used and pore size. It was found that it is most effective to use $0.45{\mu}m$-polysulfone membrane and coagulant(PAC) at the conditions of the pH of 10.0~10.5 for the case of copper containing wastewater, $0.1{\mu}m$-PVDF membrane and coagulant(PAC) at the conditions of the pH of 10.0~10.5 for the case of zinc containing wastewater, $0.1{\mu}m$-PVDF membrane and coagulant at the conditions of the pH of 11.0~11.5 for the case of nickel containing wastewater, $0.2{\mu}m$ membrane and coagulant at the conditions of the pH of 8.0~8.5 for the case of chromic containing wastewater, and $0.2{\mu}m{\sim}0.45{\mu}m$ membrane and coagulant at the conditions of the pH of 11.0~11.5 for the case mixture wastewater. The permeate flux could higher as to be used coagulants except for the case of copper containing wastewater.

  • InGaAsP/InP PBH-DFB-LD emitting at 1.55${\mu}$m wavelength has been fabricated for 2.5Gbps optical fiber communcations. For fabrication of PBH-DFB-LD, Interference expose for grating formation 3-step LPE epitaxial growth were used. Fabricated PBH-DFB-LD operates in single longitudinal mode with larger than 35dB SMSR and wider than 3dB bandwidth of 3GHz. A 8${\mu}$m mesa structure was introduced by channel etching to reuce parasitic capacitance. To reduce pad capacitance, we designed a small electrode. 0.27mW/mA in the case of spectrum shows single logitudinal mode operation with larger thatn 30dB SMSR measured at 5mW.

Inhibition and Chemical Mechanism of Protocatechuate 3,4-dioxygenase from Pseudomonas pseudoalcaligenes KF707 (Pseudomonas pseudoalcaligenes KF707에서 유래한 protocatechuate 3,4-dioxygenase 의 저해 및 화학적 메커니즘)

  • We carried out pH stability, chemical inhibition, chemical modification, and pH-dependent kinetic parameter assessments to further characterize protocatechuate 3,4-dioxygenase from Pseudomonas pseudoalcaligenes KF707. Protocatechuate 3,4-dioxygenase was stable in the pH range of 4.5~10.5. L-ascorbate and glutathione were competitive inhibitors with $K_{is}$ values of 0.17 mM and 0.86 mM, respectively. DL-dithiothreitol was a noncompetitive inhibitor with a $K_{is}$ value of 1.57 mM and a $K_{ii}$ value of 8.08 mM. Potassium cyanide, p-hydroxybenzoate, and sodium azide showed a noncompetitive inhibition pattern with $K_{is}$ values of 55.7 mM, 0.22 mM, and 15.64 mM, and $K_{ii}$ values of 94.1 mM, 8.08 mM, and 662.64 mM, respectively. $FeCl_{2}$ was the best competitive inhibitor with a $K_{is}$ value of $29{\mu}M$. $FeCl_{3}$, $MnCl_{2}$, $CoCl_{2}$, and $AlCl_{3}$ were also competitive inhibitors with $K_{is}$ values of 1.21 mM, 0.85 mM, 3.98 mM, and 0.21 mM, respectively. Other metal ions showed noncompetitive inhibition patterns. The pH-dependent kinetic parameter data showed that there may be at least two catalytic groups with pK values of 6.2 and 9.4 and two binding groups with pK values of 5.5 and 9.0. Lysine, cysteine, tyrosine, carboxyl, and histidine were modified by their own specific chemical modifiers, indicating that they are involved in substrate binding and catalysis.

  • The addition products of glutathione to ${\beta}$ -nitrostyrene derivatives were synthesized. ${\beta}$ -Nitrostyrene (1a), p-methyl-${\beta}$-nitrostyrene (1b), 3,4,5-trimethoxy-${\beta}$-nitrostyrene (1c), o-, m- and p-chloro-${\beta}$-nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) undergo addition reactions with glutathione to form S-(2-nitro-1-phenylethyl)-L-glutathione (5a), S-[2-nitro-1-(p-methyl)phenylethyl]-L-glutatione (5b), S-[2-nitro-1-(3', 4', 5'-trimethoxy)phenylethyl]-L-glutathione (5c), S-[2-nitro-1-(o-chloro)phenylethyl]-L-glutathione (5e), S-[2-nitro-1-(m-choro)phenylethyl]-L-glutathione (5f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-glutathione (5g), S-[2-nitro-x-(o-methoxy)-phenylethyl]-L-glutathion e(5h), S-[2-nitro-x-(m-methoxy)phenylethyl]-L-glutathion e (5i), and S-[2-nitro-1-(p-methoxy)phenylethy])-L-glutathione (5j), respectively. The structure of adducts were identified by UV and IR-spectra, molecular weight measurement, and elemental analysis.

Dual Regulation of R-Type CaV2.3 Channels by M1 Muscarinic Receptors

  • Voltage-gated $Ca^{2+}$ ($Ca_V$) channels are dynamically modulated by Gprotein-coupled receptors (GPCR). The $M_1$ muscarinic receptor stimulation is known to enhance $Ca_V2.3$ channel gating through the activation of protein kinase C (PKC). Here, we found that $M_1$ receptors also inhibit $Ca_V2.3$ currents when the channels are fully activated by PKC. In whole-cell configuration, the application of phorbol 12-myristate 13-acetate (PMA), a PKC activator, potentiated $Ca_V2.3$ currents by ~two-fold. After the PMA-induced potentiation, stimulation of $M_1$ receptors decreased the $Ca_V2.3$ currents by $52{\pm}8%$. We examined whether the depletion of phosphatidylinositol 4,5-bisphosphate ($PI(4,5)P_2$) is responsible for the muscarinic suppression of $Ca_V2.3$ currents by using two methods: the Danio rerio voltage-sensing phosphatase (Dr-VSP) system and the rapamycin-induced translocatable pseudojanin (PJ) system. First, dephosphorylation of $PI(4,5)P_2$ to phosphatidylinositol 4-phosphate (PI(4)P) by Dr-VSP significantly suppressed $Ca_V2.3$ currents, by $53{\pm}3%$. Next, dephosphorylation of both PI(4)P and $PI(4,5)P_2$ to PI by PJ translocation further decreased the current by up to $66{\pm}3%$. The results suggest that $Ca_V2.3$ currents are modulated by the $M_1$ receptor in a dual mode-that is, potentiation through the activation of PKC and suppression by the depletion of membrane $PI(4,5)P_2$. Our results also suggest that there is rapid turnover between PI(4)P and $PI(4,5)P_2$ in the plasma membrane.

A Seismic Study for Anisotropy and Engineering Property of Rocks at Boeun Area. (탄성파를 이용한 보은지역 암석의 공학적 성질과 이방성에 관한 연구)

  • The Okchon Formation and the Mesozoic granite of the Boeun, Chungbuk are compared in terms of seismic wave velocities estimated from the field experiment, and seismic wave velocities in 3-D measured from the rock specimen. P-wave velocity for the field data ranges from 861 m/s (Guryongsan-2 Formation) to 2697m/s (Bulguksa Granite). P-wave anisotropy also ranges from 46% (Changri Formation) to 81% (Bulguksa Granite), with an average value of 68.5%. P-wave velocities for the rock specimens from Guryongsan-1, Guryongsan-2, Changri, and Munjuri Formations are greater than 5000m/s. S-wave velocities for those specimens are approximately 3500m/s, which is 3-5 times grater than the ones estimated from the field experimental data. P-wave anisotropy for the specimens from Bulguksa Granite and Guryongsan-1 Formation exceeds 60%, which is compared to 30% for the other specimens. This value is much smaller than average P-wave anisotropy (69.5%) for the field data. It is suggested that velocity difference, associated with the propagation direction, is much greater for the field data than for the specimens.

  • Pyrrolidone-anilide derivatives from L-glutamic acid and anilines were synthesized as follows: The products were identified by elementary analysis, IR, NMR and Mass spectra with (R)-2-pyrrolidone-5-carbox-anilide, (R)-2-pyrrolidone-5-cabox-p-chloroanilide, (R)-2-pyrrolidone-5-carbox-o-toluidide, (R)-2-pyrrolidone-5-carbox-m-toluidide and (R)-2-pyrrolidone-5-carbox-p-toluidide. The products were testes for their phytotoxicity on the germination and the seedling growth of radish and rice plants. Among them, (R)-2-pyrrolidone-5-carbox-anilide and (R)-2-pyrrolidone-5-carbox-p-chloroanilide derivatives were strongly inhibitory especially on the germination and the seedling growth of radish seeds. All the compounds also showed an inhibitory activity upon the germination of rice seeds. Additionally, the inhibiting rate of radish growth differs according to the isomeric position(ortho, meta and para) of the methyl group; (R)-2-pyrrolidone-5-carbox-m-toluidide derivative was more effective than both (R)-2-pyrrolidone-5-carbox-o-toluidide and (R)-2-pyrrolidone-5-carbox-p-toluidide derivatives.

Control of Bermudagrass (Cynodon dactylon) Causing Weedy in Zoysiagrass matrella Merr (금잔디에 잡초성 버뮤다그래스 방제)

  • Bermudagrass (Cynodon dactylon.) is one of the most difficult weedy species to control in turfgrass because it's high tolerant to various environmental and management stresses. This experiment was performed to find the integrated weed management including cultural practices to suppress bermudagrass in Zoysiagrass matrella (L) Merr. As results, two sequential applications of Fluazifop-P-butyl 0.05 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-2}$ and Fenoxaprop-P-ethyl 0.1 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-2}$ applied on 20 days intervals were evaluated the primary option for bermudagrass suppression and turfgrass injury was acceptable in zoysiagrass. In both treatments, turf injury was observed during 30days after the first application and almost recovered at 40days. While Fenoxaprop-Pethyl 0.1 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-22}$ were lightly phytotoxic to zoysiagrass in chlorophyll content test, there was no growth inhibition of zoysiagrass. Verticut practice (4 mm depth) just before herbicides application where zoyisagrass is contaminated with bermudagrass was not helpful to reduce turf injury in this experiment. However, alone verticut management was utilized to decrease about 12-14% bermudagrass population. Thus the application of Fenoxaprop-P-ethyl 0.1 ml $m^{-2}$ + Triclopyr-TEA 0.5 ml $m^{-2}$ which are permitted for turfgrass after zoysiagrass is perfectly recovered from turf injury by verticut practice should be utilized for bermudagrass reduction in zoysiagrass.

Effect of salicylic acid and its analogues on stomatal closing in Commelina communis L. (닭의장풀의 기공닫힘에 미치는 살리실릭산과 그 유사물의 효과)

  • ABA and SA showed different effect on stomatal closing on same condition. The addition of 1 M salicylic acid to fully opened stomata resulted in a significant reductionn of 22 % in stomatal aperture. However, 1 mM ABA reduced 73 % of stomatal aperture. The light absorption spectra of the salicylic acid solution showed that SA was degraded within 1 hour. Therefore, SA solution was resupplied % the detached epidermis every 30 min. during incubation and it was found that even at 10 $mu$M SA induced stomatal closing significantly. Its effect was also greatly pH dependent. The reduction of stomatal aperture caused by 1 mM SA was most effective at lower pH (pH 7.2, 5 %: pH 6.2, 40 %; pH 5.2, 78 %). Therefore, if SA was properly treated to the epidermal strips in the medium, the effects of SA on stomatal closing were similar with those of ABA.
