Purification, Characterization and Application of a Cold Active Lipase from Marine Bacillus cereus HSS

  • Lipases (triacylglycerol acylhydrolases [EC 3.1.1.3]) are water-soluble enzymes. They catalyze the hydrolysis of fats and oils. A cold-active lipase from marine Bacillus cereus HSS, isolated from the Mediterranean Sea, Alexandria, Egypt, was purified and characterized. The total purification depending on lipase activity was 438.9 fold purification recording 632 U/mg protein. The molecular weight of the purified lipase was estimated to be 65 kDa using sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. The optimum substrate concentration, enzyme concentration, pH, and temperature were 1.5 mM, 100 µl, pH 6 and 10℃, respectively. The lipase was tolerant to NaCl concentrations ranging from 1.5 to 4.5%. The lipase was affected by the tested metal ions, and its activity was inhibited by 16% in the presence of 0.05 M SDS. The application of the cold-active lipase for the removal of an oil stain from a white cotton cloth showed that it is a promising biological agent for the treatment of oily wastes and other related applications. To the best of our knowledge, this is the first report of the purification and characterization of a lipase from marine B. cereus HSS isolated from the Mediterranean Sea.

Machinability of the cutting tools of powder metallurgy and conventional high speed steel (분말 및 용해고속도 공구의 절삭성)

  • Under the optimal cutting conditions, determined the machinability difference of cutting tools are by two major factors. One is the geometric shape of the cutting tool, and the other is the tool materials or heat treatment or coating of the cutting tool. In this research, we evaluated the machinability of cutting tools with conventional HSS and P/M(powder metallurgy) which was made from the different of materials and manufacture processes. Tool wear, surface roughness, cutting force and squareness of machined workpiece were evaluated.

  • High$\cdot$speed steels (HSS) with a combination of good wear resistance and toughness are finding new, non-cutting applications such as rolls and rollers. In this paper, the research interests are focused on the microstructural evolution of a SMo-6W series high speed steel during HIPping and the effect of HIPping process parameters on its microstructure and properties. HIPping process variables includes; temperature, pressure and hold time. The microstructures of the HIPped HSS were examined by SEM, OM and X-ray diffraction whereas the properties measured were the relative density, hardness, and bend strength at room temperature. In HIPped materials, MC and M6C were the major carbides formed in a matrix of martensite. The effect of powder size on the microstructure and mechanical properties of HIPped materials was insignificant. However, HIPping temperature and hold time strongly affected the carbide size and distribution. The results show that at proper HIPping temperature and pressure conditions, the final products approach the full density ( > 99% RD). The particle boundaries were completely eliminated without an eminent microstructural coarsening. The bend strength was about 2.3 Gpa, which is superior to cast HSS. At excessive HIPping temperatures, rapid carbide coarsening occurred, thus deteriorating the mechanical properties of the P/M steels.

Characteristics of Heat Stable Salts Treatment Using Anion Exchange Resins in CO2 Absorption Process (음이온교환수지를 이용한 CO2 흡수 공정시 발생하는 열안정성염 처리 특성)

  • In this study, we studied the characteristics of ion exchange for treatment of HSS (heat stable salts) which cause performance reduction in CO2 gas capture amine solution using anion exchange resins. The optimum HSS removal efficiency, 96.1% was obtained when using strong base anion exchange resin SAR10 at dosage 0.05 g/mL, 316 K, pH 12 and the best resin regeneration efficiency, 78.8% was obtained using NaOH solution of 3 M at 316 K. The adsorption data were described well by the Freundlich model and the sorption intensity(n) was 2.0951 lying within the range of favorable adsorption. The adsorption selectivity coefficients were increased by increasing valences and size of ion and desorption selectivity coefficients showed a contradictory tendency to adsorption selectivity coefficients. By continuous HSS removal experiments, 13.3 BV of HSS contaminated solution was effectively treated and the optimum NaOH solution consumption was 5.2 BV to regenerate resins.

Hard TiN Coating by Magnetron-ICP P $I^3$D

  • A 30-kV plasma immersion ion implantation setup (P $I^3$) has been equipped with a self-developed 6'-magnetron to perform hard coatings with enhanced adhesion by P $I^3$D(P $I^3$ assisted deposition) process. Using ICP source with immersed Ti antenna and reactive magnetron sputtering of Ti target in $N_2$/Ar ambient gas mixture, the TiN films were prepared on Si substrates at different pulse bias and ion-to-atom arrival ratio ( $J_{i}$ $J_{Me}$ ). Prior to TiN film formation the nitrogen implantation was performed followed by deposition of Ti buffer layer under A $r^{+}$ irradiation. Films grown at $J_{i}$ $J_{Me}$ =0.003 and $V_{pulse}$=-20kV showed columnar grain morphology and (200) preferred orientation while those prepared at $J_{i}$ $J_{Me}$ =0.08 and $V_{pulse}$=-5 kV had dense and eqiaxed structure with (111) and (220) main peaks. X-ray diffraction patterns revealed some amount of $Ti_{x}$ $N_{y}$ in the films. The maximum microhardness of $H_{v}$ =35 GN/ $M^2$ was at the pulse bias of -5 kV. The P $I^3$D technique was applied to enhance wear properties of commercial tools of HSS (SKH51) and WC-Co alloy (P30). The specimens were 25-kV PII nitrogen implanted to the dose 4.10$^{17}$ c $m^{-2}$ and then coated with 4-$\mu\textrm{m}$ TiN film on $Ti_{x}$ $N_{y}$ buffer layer. Wear resistance was compared by measuring weight loss under sliding test (6-mm $Al_2$ $O_3$ counter ball, 500-gf applied load). After 30000 cycles at 500 rpm the untreated P30 specimen lost 3.10$^{-4}$ g, and HSS specimens lost 9.10$^{-4}$ g after 40000 cycles while quite zero losses were demonstrated by TiN coated specimens.s.

Quality Characteristics of Factory-Style and Handmade-Style Ssamjang (공장 및 수공업 생산 쌈장의 품질 특성)

  • This study investigated the quality characteristics of factory-style ssamjang (FSS) and commercial handmade-style ssamjang (HSS) products. Moisture, crude protein, and crude fat contents were significantly higher in the HSS groups (49.37~62.12%, 9.39~13.46%, and 4.40~8.35%) than the FSS groups (41.94~45.83%, 7.50~9.09%, and 1.81~3.36%). Salt content was higher in the HSS groups (6.33~11.18%) than the FSS groups (6.10~7.57%). Moreover, the average salt content (7.51%) of the HSS groups and the FSS groups was lower than that of commercial ssamjang (8.73%). Hunter's color value was also significantly higher in the FSS groups. However, free sugar, organic acid, and free amino acids contents varied greatly between the FSS groups and the HSS groups, which was likely due to the different manufacturing method, ripening degree of doenjang and the main material used for ssamjang.

Gene Transcription in the Leaves of Rice Undergoing Salt-induced Morphological Changes (Oryza sativa L.)

  • We describe the gene expression profile of third leaves of rice (cv. Nipponbare) seedlings subjected to salt stress (130 mM NaCl). Transcripts of Mn-SOD, Cu/Zn-SOD, cytosolic and stromal APX, GR and CatB were up-regulated, whereas expression of thylakoid-bound APX and CatA were down-regulated. The levels of the compatible solute proline and of transcripts of its biosynthetic gene, ${\Delta}^1$-pyrroline-5-carboxylate synthetase (P5CS), were strongly increased by salt stress. Interestingly, a potential compatible solute, ${\gamma}$-aminobutyric acid (GABA), was also found to be strongly induced by salt stress along with marked up-regulation of transcripts of GABA-transaminase. A dye-swap rice DNA microarray analysis identified a large number of genes whose expression in third leaves was altered by salt stress. Among 149 genes whose expression was altered at all the times assayed (3, 4 and 6 days) during salt stress, there were 47 annotated novel genes and 76 unknown genes. These results provide new insight into the effect of salt stress on the expression of genes related to antioxidant enzymes, proline and GABA as well as of genes in several functional categories.

Search for Novel Stress-responsive Protein Components Using a Yeast Mutant Lacking Two Cytosolic Hsp70 Genes, SSA1 and SSA2

  • Heat shock proteins (Hsp) 70 are a ubiquitous family of molecular chaperones involved in many cellular processes. A yeast strain, ssa1/2, with two functionally redundant cytosolic Hsp70s (SSA1 and SSA2) deleted shows thermotolerance comparable to mildly heatshocked wild type yeast, as well as increased protein synthesis and ubiquitin-proteasome protein degradation. Since mRNA abundance does not always correlate well with protein expression levels it is essential to study proteins directly. We used a gel-based approach to identify stress-responsive proteins in the ssa1/2 mutant and identified 43 differentially expressed spots. These were trypsin-digested and analyzed by nano electrospray ionization liquid chromatography tandem mass spectrometry (nESI-LC-MS/MS). A total of 22 non-redundant proteins were identified, 11 of which were confirmed by N-terminal sequencing. Nine proteins, most of which were up-regulated (2-fold or more) in the ssa1/2 mutant, proved to be stress-inducible proteins such as molecular chaperones and anti-oxidant proteins, or proteins related to carbohydrate metabolism. Interestingly, a translational factor Hyp2p up-regulated in the mutant was also found to be highly phosphorylated. These results indicate that the cytosolic Hsp70s, Ssa1p and Ssa2p, regulate an abundance of proteins mainly involved in stress responses and protein synthesis.

Effect of CeO2 on piezoelectric properties of PSN-PZT ceramics for a hypersonic sound speaker application (지향성 스피커용 PSN-PZT 세라믹스의 압전 특성에 미치는 CeO2 첨가 효과)

  • The effect of $CeO_2$ as a sintering additive on the microstructure and the piezoelectric property of yPb$(Sb_{0.5}Nb_{0.5})O_3$-(1-y)Pb$(Zr_{0.52}Ti_{0.48})O_3$ ($0{\leq}y{\leq}0.1$, PSN-PZT) for a hypersonic sound speaker (HSS) application was investigated. The samples were sintered at $1250^{\circ}C$ for 2 h. The crystal structure and surface morphology of the samples were examined using XRD and FE-SEM, respectively. Study on the influence of $CeO_2$ additives on the dielectric and piezoelectric properties indicated that the $CeO_2$-added PSN-PZT system had a high piezoelectric properties. The optimized results of ${\varepsilon}_r=1209$, $K_p$=52% $d_{33}$=351(pC/N) and $Q_m$=1230.16 were obtained at 0.4 wt.% $CeO_2$-added PSN-PZT.
