포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과 (Effects of the Grapevine Shoot Extract on Free Radical Scavenging Activity and Inhibition of Pro-inflammatory mediator Production in RAW264.7 Macrophages)

  • Free radical scavengers or quenching agents for reactive oxygen species (ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract (GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory mediator production. Utilizing 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and xanthine oxidase (XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the $IC_{50}$/ values of 34.5 and 155 $\mu\textrm{g}$/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E$_2$ (PGE$_2$) and nitric oxide (NO) production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells with the $IC_{50}$/ value of 6.4 and 14.5 $\mu\textrm{g}$/ml, respectively. This result suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

Effect of Ethanol on $Na^+-P_i$ Uptake in Opossum Kidney Cells: Role of Membrane Fluidization and Reactive Oxygen Species

  • This study was undertaken to examine the effect of ethanol on $Na^+ -dependent$ phosphate $(Na^+-P_i)$ uptake in opossum kidney (OK) cells, an established renal proximal tubular cell line. Ethanol inhibited ^Na^+-dependent$ component of phosphate uptake in a dose-dependent manner with $I_{50}$ of 8.4%, but it did not affect $Na^+-independent$ component. Similarly, ethanol inhibited $Na^+-dependent$ uptakes of glucose and amino acids (AIB, glycine, alanine, and leucine). Microsomal $Na^+-K^+-ATPase$ activity was not significantly altered when cells were treated with 8% ethanol. Kinetic analysis showed that ethanol increased $K_m$ without a change in $V_{max}$ of $Na^+-P_i$ uptake. Inhibitory effect of n-alcohols on $Na^+-P_i$ uptake was dependent on the length of the hydrocarbon chain, and it resulted from the binding of one molecule of alcohol, as indicated by the Hill coefficient (n) of 0.8-1.04. Catalase significantly prevented the inhibition, but superoxide dismutase and hydroxyl radical scavengers did not alter the ethanol effect. A potent antioxidant DPPD and iron chelators did not prevent the inhibition. Pyrazole, an inhibitor of alcohol dehydrogenase, did not attenuate ethanol-induced inhibition of $Na^+-P_i$ uptake, but it prevented ethanol-induced cell death. These results suggest that ethanol may inhibit $Na^+-P_i$ uptake through a direct action on the carrier protein, although the transport system is affected by alterations in the lipid environment of the membrane.

애기땅빈대의 항산화 활성 성분 (Antioxidative Constituents from the Whole Plants of Euphorbia supina)

  • Eight compounds were isolated from the EtOAc soluble fraction of Euphorbia supina MeOH extract as the radical scavengers for antioxidant activity. Their structures were identified as kaempferol (1), quercetin (2), juglanin (3), avicularin (4), astragalin (5), isoquercitrin (6), hyperin (7), and nicotiflorin (8) by spectroscopic analysis. The antioxidant activity was evaluated by the ORAC (oxygen radical absorbance capacity) assay, which measures scavenging activity against peroxy radicals induced by 2,2'-azobis (2-methylpropionamidine) dihydrochloride, and the ORAC value is expressed as relative trolox equivalent. Compounds 4, 6, and 7 exhibited potent antioxidant activity, whereas the other compounds showed weaker activity than trolox.

백서 태자의 배양 피부세포에서 Adriamycin의 세포독성에 관한 연구 (Cytotoxic Effect of Adriamycin in Cultured Skin Cells of Fetal Rat)

  • Adriamycin (Doxorubicin HCl)의 혈관밖 유출에 따른 조직의 손상, 특히 피부괴양 및 괴사 기전을 규명하기 위한 연구의 일환으로 흰쥐 피부세포를 이용한 in vitro 실험에서 adriamycin에 의한 산소라디칼 생성 및 그와 관련된 세포독성 기전으로 지질과산화를 검토하였다. Adriamycin은 흰쥐 태자 피부의 배양세포에서 lactic dehydrogenase(LDH) 유리를 용량 및 시간 의존적으로 증가 시켰으며, NADPH 및 NADH 첨가 조건에서 $superoxide\;anion(O^-\;_2{\cdot})$ 생성을 현저히 증가시켰다. Adriamycin은 지질과산화 반응의 척도인 malondialdehyde(MDA) 생성을 역시 NADPH, NADH 존재하에서 용량의존적으로 증가시켰고, 산소라디칼 제거물질들인 superoxide dismutase (SOD), catalase 및 thiourea와 항산화물질인 butylated hydroxytoluene(BHT), ${\alpha}-tocopherol$은 MDA 생성증가를 현저히 억제하였다. 1, 3,-bis(2-chloroethyl)-1-nitrosourea(BCNU)를 처리하여 산화성 공격에 대한 방어기전의 하나인 glutahione 체계를 억제할 경우 adriamycin에 의한 MDA 생성은 더욱 현저히 증가하였고, 이는 역시 항산화 물질들에 의하여 억제되었다. 이상의 연구성적에서 adriamycin은 산소라디칼 생성의 증가와 그에 따른 지질과산화를 촉진하므로서 피부세포에 손상을 줄 것으로 사료되었다.

Evaluation of Genotoxicity of Three Antimalarial Drugs Amodiaquine, Mefloquine and Halofantrine in Rat Liver Cells

  • The genotoxic effect of antimalarial drugs amodiaquine (AQ), mefloquine (MQ) and halofantrine (HF) was investigated in.at liver cells using the alkaline comet assay. AQ, MQ and HF at concentrations between $0-1000{\mu}mol/L$ significantly increased DNA strand breaks of rat liver cells dose-dependently. The order of induction of strand breaks was AQ>MQ>HF. The rat liver cells exposed to AQ and HF (200 and 400 ${\mu}mol/L$) and treated with (Fpg) the bacterial DNA repair enzyme that recognizes oxidized purine showed greater DNA damage than those not treated with the enzyme, providing evidence that AQ and HF induced oxidation of purines. Such an effect was not observed when MQ was treated with the enzyme. Treatment of cells with catalase, an enzyme inactivating hydrogen peroxide, decreased significantly the extent of DNA damage induced by AQ, and HF but not the one induced by MQ. Similarly quercetin, an antioxidant flavonoid at $50{\mu}mol/L$ attenuated the extent of the formation of DNA strand breaks by both AQ and HE. Quercetin, however, did not modify the effects of MQ. These results indicate the genotoxicity of AQ, MQ and HF in rat liver cells. In addition, the results suggest that reactive oxygen species may be involved in the formation of DNA lesions induced by AQ and HF and that, free radical scavengers may elicit protective effects against genotoxicity of these antimalarial drugs.

Cell-free Myeloperoxidase/$H_2O_3$/Chloride System에서 Singlet Oxygen이 관여한다는 실험적 증거 (Evidence for Singlet Oxygen Involvement in Cell-free Myeloperoxidase/$H_2O_2$/ Chloride Sytem: Exclusion of Hydroxyl Radical Involvement)

  • 인체 백혈구에서 추출한 myeloperoxidase(MPO)에 의한 NADH산화 및 methional에서의 ethylene 생성을 관찰하여 cell-free MPO/$H_2O_2/Cl^-$ system에서 관여하는 oxygen metabolites가 무엇인지를 규명하기 위하여 본 실험을 수행하였다. 1) NADH는 MPO/$H_2O_2/Cl^-$에 의하여 산화됨을 확인하였다. 즉 MPO, $H_2O_2$$Cl^-$가 존재하는 medium에서 NADH의 산화는 azide와 catalase에 의하여, 그리고 medium에서 $Cl^-$를 제거하였을 때 완전히 제거되었다. 2) 이와같은 MPO/$H_2O_2/Cl^-$에 의한 NADH산화는 $^1O_2$ 제거물질인 1,4-diazabicyclo(2,2,2) octane(DABCO)에 의하여 완전히 억제 되었으나 $OH{\cdot}$의 제거물질인 mannitol, benzoate, formate 및 methanol에 의해서는 영향을 받지 아니하였다. 3) 또한 methinal을 MPO/$H_2O_2/Cl^-$으로 처리하였을 때는 ethylene이 전혀 검출되지 아니하였으나 기타 $OH{\cdot}$을 생성할 것으로 알려진 산화계인 xanthine/xanthine oxidase 및 $Ca^{++}-H_2O_2$에 의해서는 현저한 ethylene생성을 관찰하였다. 이상의 결과는 Cell-free MPO/$H_2O_2/Cl^-$ 산화계에서는 $^1O_2$이 산화반응에 관여하는 주된 산소 대사물이며 $OH{\cdot}$은 생성되지 아니함을 알 수 있었다.

저준위 방사선에 의해 유도된 방사선저항의 기전 (Mechanism of Radioresistance Induced by Low-Dose Irradiation)

  • 림프구와 림프종에 고준위 방사선(8Gy)을 조사했을 때. 저준위 방사선을 먼저 조사한 경우 세포 생존률이 3.7배 증가하는 것이 관찰되었다 세포 생존률은 고준위 방사선 조사로 발생되는 산소 라디칼의 제거. 변성된 유전자와 단백질의 제거, 변화된 세포의 제거 등으로 증가할 수 있다. 본 연구에서는 저준위 방사선 조사로 유도되는 방사선 저항기전을 알아보기 위하여, 고준위 방사선 조사시 발생되는 산소 라디칼을 제거하는 효소들의 활성과 방사선 보호제로 알려진 glutathione의 양을 측정하였다 림프종의 경우 저준위 방사선 조사시 peroxidase의 활성이 133.3%로 증가하였다. 림프구의 경우는 superoxide dismutase, glucose-6-phoshpate dehydrogenase와 glutathione의 활성이 각각 138.5%. 122.4%. 120.8%로 증가하였으며. 이들 효소들의 활성은 저 준위방사선 조사 간격이 7 시간일 때 가장 증가되었다.

Mutation, DNA Strand Cleavage and Nitric Oxide Formation Caused by N-nitrosoproline with UVA & UVB

  • N-Nitrosoproline(NPRO) is endogenously formed from proline and nitrite. NPRO has been reported to be nonmutagenic and noncarcinogenic. In this study, we have detected the direct mutagenicity of NPRO with UVA and UVB towards S. typhimurium. Formation of 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG), a mutagenic lesion, was observed in calf thymus DNA treated with NPRO plus simulated sunlight. Furthermore, the treatment with NPRO and sunlight induced single strand breaks in the superhelical replicative form of phage M13mp2 DNA. An analysis using scavengers suggested that both reactive oxygen species and NO radical mediate the strand breaks. The formation of nitric oxide was observed in NPRO solution irradiated with UVA. The co-mutagenic and co-toxic actions of NPRO and sunlight merit attention as possible mechanisms increasing the carcinogenic risk from UVA irradiation.

Effect of Antioxidant Flavonoids (Quercetin and Taxifolin) on In vitro Maturation of Porcine Oocytes

  • Quercetin (QT) and taxifolin (TF) are structurally similar plant-derived flavonoids that have antioxidant properties and act as free radical scavengers. The objective of this study was to investigate effects of QT and TF on nuclear maturation of porcine oocytes. Effects of TF at 0, 1, 10, and $50{\mu}g/mL$ on oocyte nuclear maturation (polar body extrusion) were investigated. After incubation for 44 h, there were no significant differences between the treatment and control groups except in the $50{\mu}g/mL$ group which was significantly lower (59.2%, p<0.05) than the other groups (control: >80%). After parthenogenetic activation, further in vitro development of QT- or TF-treated vs control oocytes was investigated. A significantly higher proportion of QT-treated ($1{\mu}g/mL$) oocytes developed into blastocysts compared to controls (24.3% vs 16.8%, respectively); however, cleavage rate and blastocyst cell number were not affected. The TF-treated group was not significantly different from controls. Levels of reactive oxygen species (ROS) and intracellular glutathione (GSH) in oocytes and embryos in a culture medium supplemented with QT or TF were measured. Both treatment groups had significantly lower (p<0.05) levels of ROS than controls, however GSH levels were different only in QT-treated oocytes. We conclude that exogenous flavonoids such as QT and TF reduce ROS levels in oocytes. Although at high concentration ($50{\mu}g/mL$) both QT and TF appear to be toxic to oocytes.

Magnesium 결핍이 항산화효소의 활성유도에 미치는 영향 (Effects of Magnesium Deficiency on Induction of Activity of Antioxidative Enzymes)

  • 유해(有害)한 활성산소종(活性酸素種)들을 생성하는 환경 stress에 대한 식물의 적응과정(適應過程)을 파악하기 위한 일환으로서, 호박을 식물재료로 공시하고 Mg이 결핍(缺乏)된 영양액(營養液)에서 재배하여 산화적(酸化的) stress 조건을 부여하였을 때, 나타나는 과산화(過酸化)의 피해정도와 항산화효소(抗酸化酵素)들의 활성변동(活性變動)을 조사하였다. 충분한 양의 Mg이 공급된 영양액에서 재배된 호박의 잎에서는 Mg, 엽록소(葉綠素), 단백질(蛋白質) 함량(含量) 및 항산화효소(抗酸化酵素)들의 활성(活性)이 다소 증가 되었거나 거의 일정하게 유지되었다. 그러나 Mg이 결핍(缺乏)된 영양액(營養液)에서 재배된 호박의 잎에서는 Mg, 엽록소(葉綠素), 단백질(蛋白質) 함량(含量)은 현저히 감소된 반면, 항산화효소(抗酸化酵素)인 AP, GR 및 SOD의 활성(活性)이 크게 증가되었다. 본 연구에서 얻어진 결과들은 식물에서 Mg의 결핍(缺乏)으로 이하여 해로운 활성산소종(活性酸素種)이 생성되며 동시에 이들 활성산소종(活性酸素種)의 유해(有害)한 작용을 소거(消去)하기 위한 항산화효소(抗酸化酵素)의 활성(活性)도 유도(誘導)되는 것을 시사한다.
